Background and Purpose Staphylococcus aureus is a common pathogen responsible for causing various human and animal infections and is well known for its ability to develop resistance to multiple antibiotics. This study aimed to evaluate the occurrence of methicillin-resistant Staphylococcus aureus (MRSA) in bulk milk and dairy farms in northwestern Ethiopia and to determine their phenotypic and genotypic antimicrobial susceptibility patterns. Methods We collected 50 bulk milk samples from 50 dairy farms and 50 hand swabs from dairy milkers. The cefoxitin disk diffusion test and PCR-based assays were used to identify MRSA isolates. In addition, cefoxitin-resistant isolates were tested for susceptibility to other antibiotics using the Kirby-Bauer disk diffusion method. Results The results showed that MRSA was detected in 8 samples: 6 from bulk milk samples (12%) and 2 from hand swabs (4%). All MRSA isolates exhibited a high resistance rate to penicillin (100%), followed by tetracycline (75%), ciprofloxacin (25%), chloramphenicol (25%), erythromycin (25%), gentamycin (12.5%), and trimethoprim-sulfamethoxazole (12.5%). Moreover, 72% of the isolates showed resistance to three or more antibiotic classes and were classified as multidrug-resistant. Conclusion This study identified methicillin-resistant Staphylococcus aureus and multidrug-resistant MRSA in bulk milk and dairy farms in northwestern Ethiopia. These findings highlight the potential risk of transmission of these antibiotic-resistant bacteria to humans and the need for improved antibiotic stewardship in the dairy sector using the One Health approach.

Macau, recognized as a global tourism hub and the world’s most densely populated region, provides a unique environment conducive to methicillin-resistant Staphylococcus aureus (MRSA) transmission in healthcare and community settings, posing a significant public health concern both locally and globally. The epidemiology and molecular characteristics of MRSA in the distinct city of Macau remain largely unelucidated. This five-year longitudinal study (2017–2022) examined the local prevalence and molecular typing of MRSA in Macau, with future MRSA type distribution predicted through ARIMA modeling. We subsequently analyzed the epidemiological characteristics of MRSA, including specimen source, clinical department, collection year, season, patient age, sex, and the annual number of tourists. Comprehensive antibiotic resistance profiles of the strains were also assessed. Of 504 clinically isolated S. aureus strains, 183 (36.3%) were identified as MRSA by the cefoxitin disk diffusion method and validated through multi-locus sequence typing (MLST). The MRSA detection rate showed an upward trend, increasing from 30.1% in 2017 to 45.7% in 2022. SCCmec type IV was predominant (28.9%), followed by types II (25.4%), III (22.1%), and V (22.1%). The primary sources of MRSA isolates were sputum (39.2%) and secretions (25.6%). Older age emerged as a risk factor for MRSA infection, whereas no significant associations were found with seasonal variations, gender, or the annual number of tourists. Despite displaying universal resistance to cefoxitin, oxacillin, and benzylpenicillin, MRSA isolates in Macau remained fully sensitive to vancomycin, tigecycline, quinupristin, nitrofurantoin, and linezolid. Continuous surveillance and analysis of MRSA distribution in Macau could provide invaluable insights for the effective management of MRSA prevention and control measures within healthcare settings.

The cefoxitin disk diffusion test and PCR-based assays were used to identify methicillin-resistant S. aureus isolates. In addition, cefoxitin-resistant isolates were tested for susceptibility to other antibiotics using the Kirby-Bauer disk diffusion method. MRSA was detected in 8 out of the 100 individual samples tested. Out of 8 MRSA isolates, 6 were obtained from pooled milk samples, and two were from personal hand swabs. On the other hand, MSSA isolates were detected in six of the specimens. All eight MRSA isolates exhibited high percentages of resistance to cefoxitin (100%), penicillin (100%), and tetracycline (75%). Moreover, 72% of isolates showed resistance to three or more antibiotic agents. This study was the first to document the presence of methicillin-resistant Staphylococcus aureu s and multidrug-resistant MRSA in raw milk and farm workers in northwestern Ethiopia.

Cow milk is a nutrient-dense food containing various nutrients, i.e., carbohydrates, fat, and protein. However, it can potentially spread several pathogenic bacteria, such as staphylococci. Staphylococci cause public health problems. Staphylococcal strains were divided into two groups based on coagulase characteristics, namely coagulase-positive and coagulase-negative staphylococci (CoNS). Coagulase-positive was represented by Staphylococcus aureus. S. aureus and CoNS can become resistant to several antibiotics, known as multidrug resistance (MDR) bacteria. S. aureus and CoNS bacteria are known as causal agents in human infections. This study aims to identify the presence of multidrug resistance S. aureus and CoNS bacteria in several dairy farms in East Java, Indonesia. We collected 332 milk and 125 swab samples from the hand of farmers from several dairy farms in Probolinggo, Tulungagung, and Blitar of East Java, Indonesia. The collected S. aureus and CoNS were tested for antibiotic susceptibility using the disc diffusion method. The antibiotic used were cefoxitin, erythromycin, gentamicin, tetracycline, and oxacillin. The results of isolation and identification based on morphological and biochemical characteristics showed that out of 457 samples, 291 samples (63.68%) were positive for S. aureus, and 166 samples (36.32%) were positive for CoNs. There were 15 S. aureus isolates (5.15%) and 7 CoNS isolates (4.22%) confirmed as MDR; therefore, it is confirmed that S. aureus and CoNS resistance to various drugs was very high in the province of East Java. Intensive livestock systems, high livestock densities, and excessive use of antibiotics in modern dairy systems may predispose the developing growth of MDR S. aureus and CoNS infections. In conclusion, the results of this study indicate the presence of very high multidrug resistant S. aureus and CoNS in the East Java provinces. MDR bacteria can be prevented from spreading by improving biosecurity, hygiene in the milking process, and regular health care for animals and dairy workers.

Methicillin-Resistant Staphylococcus aureus (MRSA) is the name given to Staphylococcus aureus that has multidrug-resistance (MDR) characteristics and is resistant to β-lactam drugs. Compared to other livestock, companion animals have been mentioned as potential MRSA reservoirs more frequently. This study aimed to identify MDR and detects MRSA strains from cats in Surabaya. A total of 150 nasal swab of cats were taken from several clinics and veterinary hospital. Samples were swabbed using Amies Medium Transport then identified using microbiological standard method. Kirby-Bauer diffusion method was performed for S. aureus antibiotic resistance profile on five different antibiotic discs. As a confirmatory test for MRSA, S. aureus isolates that were resistant to cefoxitin continued to grow on Oxacillin Resistance Screening Agar Base (ORSAB). The findings of the isolation and identification and process revealed 18 (12%) S. aureus isolates. The antibiotic resistance test revealed 3 (2%) MDR S. aureus isolates and 4 (2.6%) MDR S. aureus isolates that were ORSAB positive. It has been discovered that there are MDR and MRSA S. aureus isolates. It could be used as an indicator of irrational usage of antibiotics and it explained that cats could be source of infection to public health.

Asthma is a chronic respiratory disease that affects children worldwide. Increasing evidence suggests that Staphylococcus aureus contributes to the pathology of asthma. The aim of this study was to evaluate the nasal carriage, antimicrobial susceptibility profile, and presence of enterotoxin genes from S. aureus isolated from children with asthma. Nasal swab samples were collected from 158 children, including 98 children with asthma and 60 healthy controls. S. aureus isolates were identified using phenotypic methods and the presence of the nuc gene. Antimicrobial susceptibility testing was performed using the Kirby-Bauer disc diffusion method. Polymerase chain reaction (PCR) confirmed the presence of the mecA gene and enterotoxin genes. The nuc gene was confirmed in 83 isolates, resulting in a nasal carriage of 52.5% (83/158). The nasal carriage of S. aureus was higher among asthma cases (72.4%), with a significant association of S. aureus nasal carriage observed among asthma cases (OR 0.201, 95% CI 0.063–0.645, p = 0.007). Methicillin-resistant S. aureus (MRSA) nasal carriage was 11.4%. The S. aureus isolates showed high resistance to cefoxitin (99%) and penicillin (92%) but were sensitive to gentamicin (25%). Furthermore, 67.5% of the isolates were multi-drug resistant. The staphylococcal enterotoxin c gene (sec) was the most prevalent enterotoxin (19.7%) among cases and controls. These findings highlight the need for improved antibiotic stewardship in paediatric medicine and implementation of infection control policies.

Background Methicillin-resistant Staphylococcus aureus (MRSA), is considered a potential and aggressive nosocomial pathogen. It accounts for 50% of S. aureus isolates in tertiary hospitals in Iran, however, there is no sufficient evolutionary and epidemiological investigation about this medically important bacterium. We aimed to study the lineage and evolution of MRSA in Northwest Iran during 2021–2022 based on the obtained phenotypic and genotypic characteristics. Materials and methods Seventy-two non-duplicate MRSA isolates were collected from 3 referral hospitals in Tabriz, Ardebil, and Urmia cities. The antimicrobial susceptibility patterns were determined by disk diffusion test and micro broth dilution methods. Thereafter 4 virulence genes (eta, etb, pvl, tst) and 5 types of staphylococcal cassette chromosome mec (SCCmec) were detected by PCR. In the final step, representative isolates were selected to be studied by Multilocus sequence typing (MLST). Results The highest resistance was observed to erythromycin and clindamycin at a rate of 76.4%, followed by ciprofloxacin (61.1%), gentamicin (54.2%), rifampin (38.9%), and co-trimoxazole (27.8%). All isolates were susceptible to vancomycin. The virulence genes of etb, pvl, tst, and eta were detected in 50%, 29.2%, 21.8%, and 13.9% of isolates, respectively. SCCmec types III and I were the most prevalent types, followed by types IV, II, and V. MLST analysis revealed 6 sequence types: ST6854, ST5282, ST127, ST7804, ST1607, and ST7784. Two MLST-based clonal complexes (CC8, and CC97) were identified as well. Conclusion The ST numbers were non-repetitive. CC8 as a pandemic clone and an individual lineage and clinically significant clade was reported as the most prevalent clonal complex. It is essential periodic evaluations of antibiotic susceptibility patterns and study the evolutionary characteristics of medical-challenging microorganisms in particular MRSA to effectively treat and restrict the outbreaks.
