I have in vitro transcribed my gene (downstream a T7 promoter). I do get a product of 3ug/ul but when the rna is injected I see no expression... any ideas on how to solve this problem?

I want to assess luciferase activity in my cells. I would like to knockdown a protein of interest using siRNA (usually I do double transfection) and then transfect with the vector containing the luciferase gene. I am wondering if anyone knows whether it is better to first treat the cells with the siRNA (double transfection) and then transfect with luciferase and renilla or if you know of other ways to do it? I am just thinking that cells might be quite stressed and I do not know if they will be able to handle 3 transfections.
