the importance of those approaches for teaching grammar in the EFL classroom
I am personally against having a separate class for teaching grammar for ESL or EFL students. I believe that grammar should be taught through contexts. That is, we may teach grammar as part of a listening, speaking, reading or writing class. This is my opinion regarding grammar and vocabulary teaching.
As we all know, grammar and vocabulary are language systems not skills. So we should teach these systems through the skills. Teaching the systems separately will make it hard for student to connect what they learned to Language use in real contexts. .Following
- 6Any advice on a MTT assay false positive result?
I am using MTT assay to test the toxicity effect of a substance on A549 cells (adherent cell).
I seed 5x10^4 cells/well of a 96 well-plate, incubate overnight for cell attachment. Then I treated cells with the substance for 24hrs.
After treatment I observed that cells in the treated wells detached from the culture plate, changed the morphology to be round shape, and the cell confluency in treated well was significantly lower than control wells (observed under a microscope).
However, when used MTT to measured the cell viability, the treated wells gave higher signals than the control wells. I tried to observe the well after incubating with MTT for 2hrs, I saw the cells in treated well had more MTT crystals in the cytoplasm than those in the control wells.
Is it false positive data? How could I solve this problem?

Further to Vladimir's comment, I need to clarify as clearly I wasn't precise enough and want to avoid misunderstandings:
1. diazonium solution from p-nitroaniline must not be frozen, hence refrigeration only at usual temp of approx 4-5C, and not placing in freezer, where temp <0C;
2. the glass container (best is simply the beaker in which it was made) is sealed with a readily removed lid (petri dish or watch glass). This way over-pressurisation is not possible whilst stopping evaporation of the solution.Following

Try to exported with another format first (ex. GRID) and then convert it from raster ti ASCII.

- 10cell viability interpretation issues in MTT Assay. Is compound cytotoxic or not?
In MTT Assay, Cell viability is approx 50% when concentration of Toxicant is increased from 0 micro-molar to 50 micro-molar. But when the concentration is further increased from 50 micromolar to 500 micromolar the Viability is almost same, it hardly reached 40%. Can anybody suggest me, how to interpret the toxic behavior of compound used. Whether it will be considered cytotoxic or not?HeLa cells were used for MTT Assay.
If you want to know whehter your compound is cytostatic versus cytotoxic having used a colorimetric assay (MTT, SRB, etc...), we have to "play" with three parameters, i.e. the GI50, the LC50 and the TGI which have been developed and validated by the US NCI on their 60-cell-line panel. You can go on their public website and look at thousands of compounds for which these parameters have been calculated.

hi, I am working on some what identical research in Vietnam's Mekong Delta.
I dont think CVM or other stated preference methods would be helpful, especially when it comes to the realization of ecosystem services for farmers. My suggestion would be using the changes in crop yields as a proxies. However, you should be careful since this is an over simplification of terms and further in-depth analysis is needed.Following

The situation is as follows:
I have a dependent variable (Y), measured on a scale from 1 to 11
I have an independent variable (X) with possible values from 0 to 6
I have a dichotomous moderator variable (Z) with the possible outcomes 0 and 1 (Group 0 and Group 1)
I hypothesize that the relationship between X and Y can be characterized by non-linear curves. I assume that the nature of this curve differs between the two groups of the moderator.
My hypothesis is that in group 0 the curve grows quickly in the beginning and then reaches a plateau but in group 1 the curve grows slower but keeps on growing and cuts the other curve at some point and reaches the plateau later and at a higher point.
Put another way: For low values of X I expect to find higher values of Y in group 0 compared to group 1. For high values of X I expect to find higher values of Y in group 1.
It should look like the function in the attached picture.
How can I test this hypothesis with a regression? My approach would be:
I assume that there is a regression of the type:
Y = ß0 + ß1X + ß2Z + ß3X1/2 + ß4X*Z + ß5X1/2*Z
Then I would look at the tests for ß4 and ß5. I would assume ß4 to be bigger in Group 1 resulting in higher Y values for high X values and bigger ß5 for Group 0 resulting in higher Y values for low X values.
Does this make sense? In my field of research there is not much research using higher order regression terms (except X²) let alone interactions with those terms. I´m not sure if it is correct to include the square root term with a linear term in the same equation.
This problem is quite specific and it has been hard finding proper advice so I am thankful for any thoughts or advice on this topic.
