    Phase vs frequency in Bode plot?

    Hello everybody,

    I only has Nyquist plot ( -Im Z vs Re Z) when I measured electrochemical behavior of supercapacitor while I want to change to Bode plot (phase vs frequency). Can I change from Nyquist plot data ( frequency , -Im Z and Re Z) to Bode plot ( phase vs frequency)?

    I am evaluating Graft V Leukemia in chimeric mice - how long should I wait to observe significant rejection of leukemia in the mice?

    Hello everyone, I am evaluating the graft vs leukemia effect in chimeric mice. The chimeric mice were created by transplantation of hematopoeitic stem cells (H2kb) of wild type mice into BALB/c mice (H2d) the mice are now mixed chimeric and i am investigating if the T cells of H2b origin capable of rejecting the H2d leukemia.

    My plan is to inject the CFSE labelled WEHI3 (H2d leukemic cells) into the chimeras and observe if the leukemia has proliferated or diminished by FACS analysis.

    I want to know what would be the appropriate time to do CFSE FACS analysis on blood of the chimeric mice injected with WEHI3 cells? Is 3 days too early to see any rejection of the leukemia? How long should i wait to observe a significant rejection of leukemia in the allogeneic/chimeric mice?

    Thank you Davide. The chimeras that we developed are through Hematopoeitic stem cell transplantation and we used a semi-lethal model so they became stable and exhibit mixed chimerism.

    In invitro the T cells from the chimeras exhibited good amount of CD8-cytotoxicity against the WEHI3 leukemic cells compared to just BALB/c T cells, which was our objective from the beginning, and i just want to see if the anti-luekemic activity is replicated in vivo conditions.

    I will read more into the WEHI3, but i was hoping that the minor antigen differences with leukemic cells should be enough to activate the CD8 chimeric cells against them.

    As you suggested i will check the blood on day 3 and 5. but do you think CFSE will be stable if i go beyond that day?

    Can anyone suggest possible solutions for cell culture contamination problem?

    We seem to have a very serious problem in our cell culture lab. Lately, we haven't been able to culture cells due to contamination by bacteria. The cells are thawed in one day and the next day, although cells are apparently ok, there are already some bacteria in solution. In two days we have a  cloudy medium. We always throw away all contaminated flasks and also medium and plastics that were used at the same time the contaminated cells were in culture. Everything else, including incubator and flow chambers were sterilized before thawing new cells. All the protocols were revised to assure aseptic technique and only experienced workers are handling the cells. Initially, we thought there was a contamination problem with our stock, but it seems that no matter which cell line we use, we always get contamination. This has never happened before in our lab.

    Can you give me some advice of how to handle this situation? Is there anything else we should be doing?

    Dear Angela,
    I agree with the previous advices. In addition to the open LB petri dishes (without antibiotic) into the hood, you should add one in the cell incubator.
    Check also the water you use in the cell incubator, and the air coming in to the cell culture room.
    Finally, you should check the medium +/- serum without cells, 24-36hs in the incubator.

    Gravitomagnetism, created by O. Heaviside (1893) also predicted gravity waves by analogy with electromagnetism.

    Hence, telling that "Einstein was right" is of course presumptuous and a proof of bias.

    First they will need to prove that the "ripple" exists, then prove that the "ripple" is the "universe itself", and then that the "ripples" comply with Einstein's equations.

    What would Edmund Burke say about Human Rights?

    There has been much attention to the political thought of Edmund Burke of late, and this arises, in part, out of the long felt tension between Burke on the American crisis of the late 18th-century, Burke on Irish emancipation and Burke on India, vs. Burke as the most famous opponent of  the French Revolution. One key to this is to understand Burke on rights. He makes many appeals to rights, and clearly gives them a high moral status, yet he opposes doctrines of "abstract" rights, and this comes out in his criticisms of the French Revolution.

    As an opening to discussion, I recommend the following short (45 Min.) video from Trinity College, Dublin, given by the British philosopher Onora O'Neill:

    This talk is an excellent probing of Burke on this issue, and some considerable sympathy with Burke is expressed at the end of the talk.

    show how much the views of Burke and Tocqueville on the French Revolution resemble each other and enlighten each other.  

    Accordig to Tocquevville : ''The condition of these writers prepared men to like general and abstract theories in government, and to trust to them blindly. In the almost infinite distance at which they lived from practical life, no experience came to temper their natural ardour; nothing warned them of the obstacles which existing facts might oppose to the most desirable reforms; they had no idea of the dangers which always accompany the most necessary revolutions.’

                Their animosity to Christianity was a feature which would, of course, attract the attention of every observer. M. de Tocqueville ascribes it entirely to want of political experience, and agrees with Burke in the conclusion that the least degree of practical knowledge would have prevented it.

                Two passages from the Reflections and the Ancient Régime on this subject are strikingly similar. Burke, whilst insisting on the fact that experience had brought Englishmen to reverence religion as the basis of society, says that his experience has lived down scepticism: ‘Who, born within the last forty years, has read one word of Collins and Toland, and Tindan and Chubb and Morgan, and that whole race who called themselves free-thinkers? Who now reads Bolingbroke? Who ever read him through?’

                After referring, not to this passage of Burke, but in general terms to the Deistical controversy in England, M. de Tocqueville says: ‘Why look for examples out of France? What Frenchman in the present day would think of writing such books as those of Diderot and Helvetius? Who would read them? I should almost say, who knows their titles? The incomplete experience which we have acquired in sixty years of public life has sufficed to disgust us with this dangerous literature.’

    Hello there! Does anybody have experience in opening up Oxford Instrument cryostat for X-band EPR?

    I am having trouble with one: it seems helium is passing jolly well through the whole system but it appears it is missing the sample (and the thermocouple). My theory is there is a blockage somewhere after the He inlet (to the cryostat body) which does not allow helium to go to the sample but diverges it back to the dedicated out path. It would be nice to look inside and blow several paths with air.

    1 Check if the lever is not clogged (I assume you did it)
    2. Check if you can create vacuum in the system. You will be shure that the glas part that is inside of the resonator is not broken and properly sealed with apiezon in the metal socket.
    3. In this glas part there are holes that need to fit to the same holes in the metal part to have vacuum in the glass part.
    4. Maybe the glass part is not put inside correctly. You can take it out and put new apiezon and put it back. The glass part is expensive if you brake it.
    5. If that won't work than the round black box can be open. It is vacuum sealed with imbus screws cup and a gasket. I think that a hole somwhere there would force problems with vacuum. But who knows...

    General theory of relativity (GRT) threatens gravity as a phenomenon resulting from the curvature of spacetime.  In that context, gravitational waves are ripples in the geometry of spacetime which propagate as waves travelling outwards from an accelerating object.

    Gravitoelectromagnetism (GEM) threatens gravity as a phenomenon analogous to electromagnetism (EM).  In that context  gravitational waves are fluctuations in the gravitational field which propagate as waves travelling outward from an accelerated mass (in analogy with EM waves that manifest themselves as generated by an accelerating electric charge).

    Now gravitational waves have been observed, again one can ask the question which of this two ways (GRT or GEM) is the most plausible to describe and to understand gravity.

    Is there some contradiction between Feynman's path integral and the relativity?

    A well known formula in Feynman's path integral formalism is

    (1) K(rA, rB) = K(rA, rC) K(rC, rB) drC,

    where K(rA, rB) is the amplitude of probability to reach the position rB at the time tB, given that at the time tA the system was at the position rA.  rC is the position of the system at a given intermediary time tC between tA and tB.

    A strange fact is, however, that the integral in (1) is from -∞ to ∞. That means, the point C sweeps all the space, even out of the region [rA, rB]. A contradiction with the relativity is hidden here, because for jumping from a finite position in space, rA, to infinity, in a finite interval of time tC - tA, implies an infinite velocity. But the theory of relativity sets an upper bound to velocities, the light velocity.

    Is there a remedy for this problem? Or, alternatively, the path integral theory admits indeed, faster-than-light velocities?

    Material costs in green buildings... Elements of this....?

    Actually, the construction cost of a LEED Platinum building is no higher than the LEED Bronze (eg, standard building code).  That is because in the truly 'green' buildings, you're not just swapping out windows and light bulbs, you're changing the design philosophy and the performance envelope.

    It's a pretty simple thing to design a building in the temperate zone that is entirely off-grid (well, except for cooking) by using STES, thermal mass, passive solar, solar thermal, and solar electric.  It would cost almost exactly the same as conventional construction - it wouldn't look the same, but it would cost the same.  We don't do that, in part because we want "to have our cake and eat it too" - we want our conventional looking home to be very high performing - we'll change the performance envelope, but not the design philosophy.

    If you assume an real increase in the cost of energy of 10% per year faster than inflation (not an unreasonable estimate for the near future, once this oil glut has passed), then any investment in energy efficiency that has a 44 year simple payback, or less, is a good investment.  Most of the 'low hanging fruit' has a simple payback in the order of 3 to 8 years (solar thermal, solar daylighting, thermal mass, STES, etc).  So even if you just swap out parts in a design for better energy efficiency, it will, in the end, save you substantially.

    Is the universe expansion rate constant in space at any given time, including at quantum scales?

    My understanding is that the cosmological constant, though poorly understood, is assumed to be uniform in space. Does this uniformity extend to small scales or is it only uniform at very large scales.

    Who would fund this falsification of GRT?
    Excellent suggestion, but it would never get funded, because it would falsify GRT. Light is a wave, not a particle and thus is unaffected by gravitation. A ref you might like to cite:

I am the first who understood and explained that the gravitons with the speed of light are too slow to keep the constellations and the galaxy together. I explained gravitation with the speed of the gravitons bigger than the speed of light. I discovered the Ferent gravitational force function. I calculated the volume of the universe at Planck wall and the volume of the universe at Ferent wall. My quantum gravity theory shows that the gravitons are too small to be detected by today’s technology.

    Hi, am trying to use approximated methods (Galerkin method). My understanding is, that I have to choose a comparison function that satisfies the Geometric boundary conditions. However, the beam configuration has only natural BC (one end has a torsional spring, the other is free).

    Why does Lanmuir always win?

    When looking for adsorption articles, majority of adsorption manuscripts are well fitting Langmuir isotherm equation rather than Freundlich. Even other known isotherm equations are used, finalists are always Langmuir and Freundlich.
    I wonder why? What is the reason of this result.

    While I was  developing a new method for measuring the adsorption of certain gases on zeolites I found that in the case of nitrogen the sum of two Langmuir equations could be very closely fitted to the measured data. The reason for this may be that the nitrogen atoms are electrical quadrupoles.  See my first article in 2005 !

    Depends on the type of disabilities, for instance eye tracking systems can provide inexpensive means for making multimedia systems accessible for people with disabilities that doesn't affect vision. Also EEG caps can be used for the sabe objective.

    I suggest measuring conductivity and TOC.  Then compare it against the ASTM Type I and Type II benchmarks.

    Ectopic Model vs Orthotopic Model, which is better to evaluate anticancer activity?

    Could anyone provide me some suggestions which xenograft model to use for evaluation of anticancer activity of a therapeutic agent - ectopic or orthotopic? I am involved in in vivo studies related to pancreatic cancer. For your information, I have read about these two models and have some idea on the pros and cons of both models. However, I could not make a decision on which xenograft model to use for my studies. I will use Ncr nude mice and the pancreatic cell line Capan-2.

    We faced many problems in trying to establish orthotopic transplantation of Capan-2 pancreas cancer cells in immunocompromized mice. In contrast, we had no problems with the BxPC-3 cell line (see the attached article by Van Quaquebeke et al., 2007).

    Pancreas cancer cell lines can behave very distinctly one from each other (see the attached article by Mijatovic et al., 2007).
