Good day,
I am working on the molecular detection of Rubella virus. For the positive control I tried extracting RNA from the MMR vaccine(Tresivac). I have not seen any RNA after extraction. Its a attenuated form, Since RNA is sensitive could it be that the vaccine don't have RNA anymore?

Its live vaccine so you can extract RNA easily. Just see your extraction kit is working or not.

Hello,
I'm developing an assay to determine TCID50 titre of rubella virus by combining the competitive ELISA and cell culture. Initial part of my protocol involves infecting vero cells with rubella virus. If anyone share their TCID50 protocol, it would be great help to me.
Also, I was wondering if it would be possible to amplify the rubella virus in the cell culture plates- infecting Vero cells in the plates and after 4 days of infection transfer the supernatant to another Vero cell seeded plate. Subsequently, after 4 days, collect supernatant and run ELISA for Rubella virus glycoprotein E1.
Do you this can work?

Vero cells quite ok for Rubella you can see a several publication based on it.
