I would suggest to check the whole mechanical alignment of your diffractometer and compare with measurements of the exact same samples (to exclude effects of contamination) at the same temperature (presumably room temperature?) on a well calibrated diffractometer.

I use ferrous sulfate as a chromium reducer. As we know from the chemical reaction equation 3 mol Ferrous reacts with 1 mol of cr(vi) which means 3.22 mg ferrous reacts with 1 mg of cr(vi), am I right ?. the sample contains 5.28 ppm cr(vi), how much reducer(by mass ) should be added to 1 kg of sample to reduce the amount of cr(vi) by 1 ppm ??

So I am doing an analysis of the Four Factors created by Dean Oliver which are important metrics for basketball teams to control if they want to win basketball games. Essentially I wanted to test if the weights that are often mentioned alongside the Factors are the same as when he estimated them in the early 2000's. The issue is he doesn't really explain how he got them and states that he used what I assume is a proprietary program he created called "Roboscout" to calculate them without giving any explanation for how he got them.
Given this I just decided to do what seemed to make the most sense to me which is using the four factors (both offensively and defensively) as the predictors for a glm with a binomial regression of wins and losses using statistics for each team since the introduction of the three point line. This worked well and I got great results where each predictor had a vif below 2, the model I predicted to be best in each of my datasets was the lowest aic and bic by between 20 and 250, there was no multicollinearity or overfitting and my assumption proved right in that the shooting factor became more important in recent years.
My issue is I think it is important in my paper to explain how much of the variance is explained by my independent variables. Obviously I can't just use R-squared because it is a logistic regression but I have only used pseudo R-squared in very minimal capacities and don't really know how to pick the right pseudo R-squared for my analysis. I read this article (https://stats.oarc.ucla.edu/other/mult-pkg/faq/general/faq-what-are-pseudo-r-squareds/) by UCLA and that provided me with 4 options, Efron’s, McFadden’s, McFadden’s (adjusted), and McKelvey & Zavoina pseudo R-squareds, but I don't know what is best or what I should use. I have found some places saying that McFadden's is really popular and is commonly used in logistic regression. Is it popular because it is really good or just popular? Should I use the adjusted Mcfadden's or not bother with it because I only have 8 ind. variables? Also I am not doing model comparison, I already did that in my paper using AIC and BIC and got really clear results to that point that it made me worry about overfitting and multicollinearity though I did tests for both and got good results. Am I overthinking this and it doesn't even matter that much?

Broadly speaking you are overthinking it and no pseudo R^2 measure is really that useful. Indeed R^2 in normal linear models is not that useful or important and tends to be over-interpreted.

I compared protocols from SigmaAldrich and ATCC and there are still some points to clarify.
1. What is the full medium for growing BEAS-2B?

I have total 12 lakh records dataset. From that i have taken 100000 records. From these 1 lakh records I have taken 10k records as test set and from remaining 90k i have created 9 batches each consists of 10k records. out of these 10k 9k i m using for training and 1k i m using for validation records, which eventually i will use for parameter tuning. after performing parameter tuning on each validation set from each of the 9 batches , I m testing on test dataset of 10k records. after that i m using best parameters to train full dataset. then original 12lakh records i m dividing into 80% training and 20% test ratio. after training on full dataset , i m testing my model on 20% of the test data. so what i m doing here, is it right approach?

Dear all,
I'm learning and taking the same genotyping PCR experiments and want to get / share some advice or experience on obtaining the right bands for the data.
I ran 3 samples of Stra8-iCre and 2 out 3 samples were not wild type, which was different genotype compared to the actual data from our laboratory and all of them were supposed to be wild type.
There was no contamination nor mistakes on performance of the protocol and I also regulated annealing temperature into 58 or 60 degrees Celcius for this whole times.
I wonder how can I get the right bands for Stra8-iCre and what was I missing except for the accuracy of the protocol and regulation of the temperature.

choose an annealing emperature that does amplify and run a pcr set with 0. 1%, 2% up to 8%dmso and at some dmso concentration the pcr will probably be a clean band. If your gene has pseudogenes or is one of a family of similar genes then touchdown pcr or new primers might be needed.. You can perform primer gradient PCR to optimize the concentration of primers. Also, You can use pico molar concentration of primers. or use different gradient pcr such as with annealing and denaturation etc.

Good day,
I am a student trying to work on Autodock for a project regarding Ligand-DNA interaction so i am quite new to molecular docking. i have followed tutorials and did all the steps recommended for the docking to work with PDBQT files for both the ligand and DNA, but every time i run AutoGrid the window for the process opens for less than 1 second and then shuts down immediately with no output file. i made sure the previous steps were working correctly and to open the ligand and DNA files in their respective input category and the paths for each part in the run window right before the launch have no spaces and are correct. Same problem happens with Autodock.
Does anyone know if there is a fix for this problem?

I routinely see requests under the questions category for particular PDF patterns. What is behind that? Do these researchers not have access to the database for themselves?
Many if not most compounds have more than one card for them. Different sources, different preparations lead to slightly different patterns. Maybe it is in the peak positions or relative intensities. Some samples may have included a second phase. Some records have more information than others.
Therefore, it seems that they should be matching your sample to the available patterns and citing the one that best fits. I don't think they should be citing a particular card just because that it what someone else referenced in their paper. They may not have it right. (I have seen that error before.)

FWIW, I just had to correct a researcher who cited the wrong PDF number in their published paper. (They have had many problems with their details.) If you had asked for that PDF record based on their say-so, you would have gotten a record that was of no use to you. It is better to be able to search for yourself.

Keep in mind, in a normal liberal democratic environment based on majority rule and the independent rule of law, power moves from one normal democratic outcome to another normal democratic outcome through a peaceful transfer of power, where the normal democratic outcome is the one that reflect the majority view rule.....If the current normal democratic outcome is not working in the best interest of the majority as it should, then voters can vote with their HEADS AND FEETS and keep electing normal democratic outcomes that actually reflect the best interest of the majority....If somebody lies to you and you have the power to get rid of him/her with your vote, they you vote him/her out at the first opportunity and bring someone else committed to the best interest of the majority. The voter can use happiness to re-elect RESPONSIBLE normal democratic outcomes or to vote out irresponsable ones to bring in a responsible or more responsible one....
If the majority allows unhappiness to lead to natural full complacency or be the victim of effective targeted chaos induced full complacency, then it allows an extreme democratic outcome to win instead of another normal democratic outcome and then the majority rule process will be held hostage by the minority view as the minority view wins the democratic contest....
The minority view is the exism view, with values contrary to the normal democratic thinking and rule of law as it places the best interest of the exism movement over the best interest of the country and places the best interest of the minority over the best interest of the majority.
So Ludo, to understand the direction where informed and independent rational voter discontent should go to ensure the preservation of the best interest of the majority is always the outcome or where they go to lead to the lost of the best interest of the majority you need to think in terms of NORMAL DEMOCRATIC OUTCOMES/NORMAL LIBERAL DEMOCRACIES and EXTREME DEMOCRATIC OUTCOMES/EXTREME LIBERAL DEMOCRACIES, then you can understand how they can compete for power and persist in power as a competition between normal democratic outcome vrs extreme democratic outcome instead of the USUAL normal democratic outcome agains normal democratic outcome.

I find it odd, for example, when I hear in the news that a parent forgives someone who intentionally caused severe injuries to their child. It seems to me that only the child has the right to forgive in such a situation.
Addendum: Let me also add for consideration the more extreme case of the parent or spouse of a murder victim forgiving the murderer. Although the state may have the right to pardon the perpetrator at some point, that is different from forgiveness, and it seems to me that no one has the right to forgive the perpetrator for the murder itself. To suggest otherwise would be tantamount to regarding the victim as property of which one has been deprived. It would be tantamount to forgiving someone for stealing and totalling your car.

Hey everyone,
In my sample preparation of protein sample I am using simultaneous TCEP and CAA for a reduction and alkylation in one step.
Using DTT and CAA a simultaneous reduction and alkylation is not possible - the DTT and the CAA will react with each other (right?) and the disulfide bridges are still there.
Now my question - Why DTT and CAA react with each other, but TCEP and CAA not? Why is this simultaneous reduction and alkylation possible? What are the differences?
I know..
- TCEP reacts with the free electron pair of the phosphorus
- thiol of DTT gets rid of a proton and the sulfur anion reacts
- both reactions are nucleophillic attacks
