I recently performed PCR and observed peaks for HEX and FAM, but the ROX signal was flat. I am unsure how to interpret and present these results.
Could someone please arrange a Zoom meeting to provide detailed guidance on this analysis?

I might be wrong, but I think there is a miss understanding with the usage of the dyes. I assume that you have used TaqMan assay for your gen of interest (GOI) which might be with probes in FAM and house keeping gene (HK) which might have bound to HEX. In some older PCR-cyclers (i.e. bio-rad icycler) specially those with a camera detection system you would need to make a flat field correction (meaning to correct for the lower excitated border wells compare with the stronger wells in the middle of the plate) that can be archived with a calibration plate or with another third dye. Therefore some suppliers are selling q-PCR mixes premixed or with individual packed ROX dyes to acquire a reference prior to the q-PCR.
So if you have not ordered an assay with probes bound to ROX everything might be right. If you have used a ROX bound probe to multiplex with two GOI and one HK I'm wrong and the expression of your gene is absent.

Hello,
There's an article: Effects of Korean Red Ginseng on Semen Parameters in Male Infertility Patients, which I have uploaded. If someone can please look at it, I'm having some trouble with some statements being made in the article. It says that "All groups but not the non-V+P group, showed significant improvements in sperm concentrations, motility, morphology, and viability at the end of the study" But if you look at the table for the semen parameters, you see that sperm concentration, motility, morphology, and viability decreased significantly for those groups, except for sperm motility for V+KRG group, where it increased. Why are they claiming improvements when their results show none? Could someone please explain?

I understood that the authors compared their findings with the begining values presented in table 1, after that, the changes in theese values in post-treatment of the Non V+P group, table 2, were statistically not significant, that's whay.

how could an animal model be used to test wheather ginseng supplements could help with the problem of withdrawal from long term antidepressant use. how do we help people reducing and coming off antidepressants without the issues of withdrawal and a return to re-administration of antidepressants?

If love could be bought from the pharmacy, the problem would be solved. Also animals are sensitive to handling and "baby" talk. Even rats tend to survive lethal doses better if they like there care taker.
