Hi all,
I am having trouble deciding if this is cell contamination or something else. I keep seeing this fiber like bodies in my media (not on the same level as my adherent cells). There is no smell or media color change that indicates contamination. I do also see these fibers when i attempt to do a cell count with trypan blue. Has anyone seen something similar? I dont see any movement at all and they dont move when I jiggle the flask gently. I am concerned since I keep seeing these even after I rinse with PBS and place in new media. Is something off with my sterile filter when making my media or is this some type of yeast or bacterial contamination?

Dear Ashley,
It looks to me like fibers from, for example, pulp or a similar material. Check the procedure of your experiment and the handling to see if such a fiber entry is possible at that point.

Hi, everyone. I have some issues with THP-1 cell culture. I have little experience in cell culture and I hope some of you could share some experiences in culturing this type of cell.
I'm currently trying to grow non-differentiated THP-1 cells for MTT experiments. The chosen media is composed of RPMI-1640, glutamine, antibiotics, sodium bicarbonate and FBS (10%). These cells either don't grow enough or die over the weeks. So here are my questions:
1 - Should I replace the old media with a new one, or just add the new media to the old media? I've had success in the past by just adding new media to the flasks, but for some reason, it stopped working over time.
2 - How many times should I submit these cells to centrifugation? In order to dispose of the old media and add a new one, I need to submit the suspended cells to centrifugation. But in my experience, this process ends damaging cell structure of some of the cells and leaves cell fragments, which could be detrimental to the culture. I'm currently submitting them to 1200RPM, during 7 minutes at 4ºC, once a week.
3 - Is it normal for these cells to form clusters/clumps?
4 - Should I leave the culture flasks standing on a vertical position? I've seen some people recommending to leave the initial, post thaw, T-25 flasks on a vertical position instead of in a horizontal position. The reasoning behind this is to increase direct contact between cells in a smaller space, as these cells are dependant on each other to grow.
5 - At which confluence should I replace the T-25 flask with a T-75 one?
6 - Should I use 20% FBS for initial growth instead of 10%?
7 - Feel free to write any other tips you consider crucial while dealing with these cells, I really appreciate the help.

The following are the answers to your questions.
1 - Should I replace the old media with a new one, or just add the new media to the old media?
When THP-1 cells start to proliferate, they excrete growth factors that aid their growth. So, THP-1 cells grow better in conditioned media. Therefore, whenever you passage the cells, leave 1-2ml of old media and add fresh media to the culture.
2 - How many times should I submit these cells to centrifugation?
Spinning THP-1 cells out to split them can cause stress as the cells will be subjected to centrifugal force. Submitting the cells to 1200rpm for 5-7 minutes once a week is acceptable.
3 - Is it normal for these cells to form clusters/clumps?
Sometimes cells may clump. But if they are growing fine and have a normal morphology, it should not be a problem. Small clumps can be taken care of by just pipetting up and down a couple of times.
However, if they get denser than the required cell density, then they will be very unhappy, may slow down or stop dividing and may clump or have an irregular or blebbing appearance. THP-1 cells are sensitive to cell density. The recommended seeding density is between 3-7 x 10^5 cells/ml. Generally, 5 x10^5 cells /ml is used. Do not allow the cell concentration to exceed 1 X 10 ^6 cells/ml. Change media every 2 to 3 days. Dead cells if they are present should be removed as sometimes these dead cells could be the cause of clumping.
4 -Should I leave the culture flasks standing on a vertical position?
Yes, keep the culture flask in a vertical position until the cells reach the exponential phase of growth. This may take up to 6-7 days.
5 - At which confluence should I replace the T-25 flask with a T-75 one?
You may continue growing the cells in T-25 flask. If additional cells are required, larger flasks/culture volumes may be used.
6 - Should I use 20% FBS for initial growth instead of 10%?
Yes. You need to increase the FBS content to 20% for the first two to three passages, then you can switch over to 10% FBS for successive passaging.
7 - Feel free to write any other tips you consider crucial while dealing with these cells, I really appreciate the help.
a. The first propagation of cells should be for generating stocks for future use. This will ensure stability and performance of cells for subsequent experiments.
b. Growth medium for THP-1: RPMI 1640, 10% FBS, 2 mM L-glutamine.
c. As far as possible, avoid using antibiotics in the growth media.
d. Use THP1 cells with less than 20 passages for cell-based assays.

Do companies running social media portals consciously shape the general social awareness of citizens, Internet users through the specific information policies applied?
In recent years, there have been an increasing number of examples of situations of deliberate practices in which companies operating social media portals consciously shape the general social awareness of citizens, Internet users through specific information policies applied. The Senate Committees of Inquiry at the U.S. Capitol, which have been taking place for several years, address, among other things, the issue of verifying the use of, for example, algorithms on Facebook platforms that promote certain content, including not only socially positive content, but also socially negative content. The aforementioned algorithms are then changed so that the scale of social negativity is reduced. However, recently there have been an increasing number of similar socially negative cases of algorithms promoting specific political content, e.g. promoting content typical of right-wing political options and limiting the spread of certain social media sites typical of left-wing political content. Thus, these are situations of intentional discrimination against a part of the community of citizens holding certain political views, which the owners of certain companies operating social media portals have deemed to be contrary to the information policy applied in their social media and/or the specific ideology promoted in these media. This type of activity does not correlate with the issue of freedom of speech, unrestricted development of the information society, democracy.
Recently, companies running social media sites have been improving the aforementioned media through the implementation of new Industry 4.0/5.0 technologies, including Big Data Analytics and generative artificial intelligence. The aforementioned technologies can also be used to technically improve the algorithms that control and promote selected content typed and passed on by Internet users, users of the aforementioned online media, which is an important part of shaping information policy in these media.

I would be interested in your opinions and own experiences on Jaron Lanier and Tristan Harris' case on how social platforms and the modern age of (monetized) big data manipulate behavior and deteriorate the ability to think critically in an individual.
I don't mean how sophisticated algorithms suggest the best-placed advertisings, that's a commonly known fact and everybody is aware of that nowadays.
What I mean is the perceived "stupification" of younger generations, fueled by overuse of technology in general, and social platforms in particular. The sophisticated tailoring of those platforms to the short attention spans, the mainly senseless content, the biased bubbles and so on, to me, appear to lessen the critical thinking abilities (and -willingness), paired with the Dunning-Kruger-Effect, that those who are the least informed have overwhelming confidence in their ill-informed opinions.
I have a very strong opinion on this, but it may also be biased by experiences I have made throughout the past years as an educator, and deterred by the sort of students I have worked with. I would like to see a bigger picture, not research trends.
So, what are your subjective impressions on the phone- and social platform addicted young generation and their cognitive abilities, their willingness to achieve excellence, and the influence new media has on them?

Initially, ebooks are offered free of charge as part of the promotion, which are usually developed with the help of ChatGPT, are of poor content quality, usually containing only general, well-known, popular science knowledge, which can be found independently on the Internet on various websites. Besides, the ebooks created with the help of ChatGPT or other similar intelligent chatbots do not contain all the sources correctly listed. Besides, the resulting ebooks contain many passages, whole sentences, paragraphs taken from other publications that are not shown in the bibliography, and plagiarism thus occurs. In addition, in the field of expertise, there are factual errors and irrational, random, random, combined content from different sources, and thus inconsistent with the facts, ridiculous content and/or descriptions of "fictitious facts" occur. This is because much of the database that constitutes the sources of data and information for ChatGPT is factually outdated, as it dates from late 2021 or January 2022. It may happen that in the ChatGPT-generated text there may be some sensitive data of specific companies or public institutions, which found themselves there accidentally by being mistakenly entered into ChatGPT by an employee of a specific company or institution. In view of the above, there are still gaps in paragraphs in the legal norms defining the rules for the correct use of tools such as ChatGPT, and still the adaptation of legal norms to the rapidly developing technology is often done with too much delay. Besides, popular online social media even feature partly free and partly paid training courses and webinars, where Internet users learn how to create texts for articles, columns, essays, etc., as well as chapters for books, which can be published as ebooks, in a relatively easy way with the help of applications available on the Internet based on generative artificial intelligence technology. Besides, in addition to text, graphics, photos, drawings, etc., which are included in the texts of chapters in ebooks can also be generated semi-automatically in applications based on generative artificial intelligence technology. It also happens that during the aforementioned online trainings and webinars, access is sold to specially created websites that act as intermediary platforms, overlays that contain links to various AI-based web applications, which have been classified in a certain way on a specially created platform and, to make identification more difficult, the links are called by different names relative to the web applications to which they direct. In addition, many of these web-based applications based on generative AI technology are made available on the Internet on the original source sites free of charge. On the other hand, on intermediary platforms created by companies or sole proprietors that contain links referencing these applications, access is paid for and is often sold as part of so-called promotional offers during the conducted, aforementioned online training courses and webinars. Internet users usually learn about such online trainings and webinars from banners and advertising posts posted on popular online social media sites.

Taking into account the available technological solutions and applications offered by ICT service providers, as well as the growing scale of psychological problems of children using smartphones and online social media, the following question arises that is relevant today: How to effectively organize parental control of what a child does in the aforementioned social media?
Children and adolescents represent the youngest generations using online social media. The youth currently attending schools and studying in universities are mainly the so-called Generation Z, who grew up with smartphones equipped with Internet access, including online social media. Today, the children and young people of Generation Z are thus at the greatest risk of being negatively influenced by online social media, including various types of misinformation, fake news, misleading unreliable offers of advertised products and services, influencers and youtubers promoting themselves, etc., which are increasingly appearing in them. In addition, there are many untrustworthy offers of products and services in social media, offers presented by influencers and youtubers, offers presented as part of advertising campaigns conducted in these media, offers promoted through spots, animations and advertising videos, in which generative artificial intelligence technology is increasingly used. It is increasingly common for advertising companies to create AI-generated influencer avatars based on intelligent chatbots for their online advertising campaigns. AI-generated fictional influencer personas look, speak and behave like real people in videos posted on social media. Internet users watching these digital influencers often don't realize that they are watching not real people but digitally generated non-sisterly real characters. In addition, in recent years there has been a growing scale of hate speech most often generated by peers from class, school. The developing hejt was the reason for the increasing scale of child and adolescent suicides in some countries in recent years. In addition, many people, especially young girls, have psychological problems due to a sense of low self-worth and self-esteem which is related to spending a lot of time on online social media and watching influencers promoting certain sublime standards, what is in a certain subculture of youth recognized and promoted as informal canons of beauty, attitudes, possession of certain material goods, etc. In addition, such psychological problems were exacerbated during the Covid-19 pandemic, which was associated with locdowns imposed in some countries on selected sectors of the economy, bans on being in certain types of public places, periodically introduced national quarantines and education conducted remotely in the form of e-learning. In some countries, the scale of such bans and restrictions, which were intended to slow down the transmission of the SARS-CoV-2 (Covid-19) coronavirus, was exceptionally large. This was the case, for example, in the country where I operate. Unfortunately, the mortality rate of citizens during the Covid-19 pandemic in Poland, despite the large-scale introduction of the aforementioned anti-pandemic bans and restrictions, was exceptionally high. Besides, the mentioned anti-pandemic bans and restrictions were introduced without adequate public consultation, on the basis of special legal regulations in the form of laws and ordinances, with the bases of prior research and analysis of the potential negative effects of such controversial measures. At present, in 2024, it is known that in the country in which I operate the negative effects of the aforementioned introduced on a record large scale so-called anti-pandemic measures, including bans, lockdown-type restrictions, periods of national quarantine were much more in comparison with the expected but not realized positive effects. In view of the above, taking into account the available technological solutions and applications offered by ICT service providers, as well as the growing scale of psychological problems of children using smartphones and online social media, it is necessary to improve the computerized systems and applications running on smartphones that enable effectively conducted parental control of what a child does on the aforementioned social media. On the other hand, citizens should influence politicians, and politicians should influence the technology companies that run online social media, so that these companies also take much greater care of the safety of children and young people using these media. The aforementioned technology companies should not treat children and young people merely as potential customers for product and service offers presented during advertising campaigns conducted on social media. The technology companies running these media should not create algorithms that promote posts, posts, comments, banners, animations, videos, etc. that contain negative and socially harmful content. That this is how this kind of media works was proven during the Senate committee hearings of former managers who previously worked at Meta, for example, and developed certain solutions within Facebook and Instagram. TokTok has also grown rapidly in recent years, which also features many examples of disinformation, factoids, posts, memes, entries, banners, videos, etc., containing unreliable and factually incorrect content, as well as many advertisements presenting various product and service offers aimed mainly at children and young people.

In your opinion, in a country with a democratic system of government, should the main, dominant, meanstream, public news media be independent, private or state-owned, under government control?
I ask because different countries have different solutions to this issue. In the country where I have been operating for many years, the main, dominant, meanstream, public news media are under government control, are state-owned (e.g., companies in which the treasury dominates as the main or sole shareholder), are referred to as public, and are used by the government as a mouthpiece for pro-government propaganda, including generating disinformation to citizens so that citizens will once again vote for the same ruling political party in parliamentary elections. On the one hand, this has been operating for many years and nothing has changed in this regard. And on the other hand, it is criticized by media experts, researchers and scholars, by media scholars, but also by many citizens who are not professionally or scientifically involved in the media. The criticism points to the lack of information objectivity, to the politicization of information, to the creation of pro-government propaganda, consisting of constant and incessant praise of what the government is doing, criticism of what the opposition parties are proposing, etc. on the unreliability of information, on the violation of the rules of journalistic ethics, on the public practice of hegemony against people working in opposition political circles, on the practice of disinformation even against people representing the so-called "hardcore electorate" of voters who usually vote in parliamentary and other elections for the ruling PIS political option.

In your opinion, does the development of new online media, including online social media and the new technologies Industry 4.0 implemented into these media, including the use of artificial intelligence in these media, increase the issue of objectivity and transparency of information or rather generate more disinformation?
On the one hand, online social media, which has been developing for 2 decades now, is making a significant contribution to the development of remote online communication, social remote communication, open communication of sending content created on the fly, sending information to friends, promoting oneself and/or specific product or service offers, informal data transfer, expressing one's emotions in the information sent, including positive as well as negative emotions, and so on. In this way, online social media on a local, regional or global scale have also contributed to the objectification of information in the context of news reported in official, meanstream media. In this regard, online social media are fulfilling their role of social and objectification of media information both in countries with democratic power systems and in non-democratic, dictatorial power systems as long as they are not blocked and restricted by the power system.
On the other hand, there has long been a lot of fake news and disinformation in online social media, the transmission and forwarding by more Internet users of memes, posts, videos, banners, comments containing unverified, unconfirmed content, data and information. In addition, new ICT and Industry 4.0 information technologies, new versions of graphic and other applications are being used to generate disinformation. Also, cyber criminals and hackers breaking into social media user profiles, stealing sensitive data of social media users are also using new technologies. Recently, machine learning, deep learning and artificial intelligence technologies have also been used by cybercriminals and untrustworthy social media users to generate disinformation. As part of this, cybercriminals, hackers and unethical Internet users generating disinformation take advantage of security gaps in information systems, exploit diagnosed inadequacies in cyber security tools operating in the aforementioned social media sites. At the very beginning, cell growth was good after taking out from liquid nitrogen. But after two days, the cell morphology turned terrible and a lot of black dots floated as in the attached pics. May I ask if anyone faced the same problem before? Please kindly suggest some solution to this issue.
In my personal opinion, I would like to re-culture the cell will brand new media with a double dose of antibiotic. Would it be a good solution for this situation? Since I am not quite sure if it is contamination or not.

Hello, I isolate PMBCs from blood using Ficoll and plate the cells in a 96-well plate for different treatments for a few hours before T cell activation. At the end of the treatments I centrifuge the plate, discard media and use new media to pipette the cells several times and transfer to an anti CD3 coated 96 well plate for 3 days, new media is also supplemented with anti CD28 ab and IL2.
I can't seem to get the cells into the new plate and rarely get any cells in the plate after the 3 day incubation. I was under the impression that these cells detach easily with pipetting. Any suggestions?

Well, at the end of March this year 2023, a number of new technology developers, artificial intelligence experts, besides businessmen, investors developing technology start-ups, including, among others, Apple co-founder Steve Wozniak and the founder or co-founder of such technology companies as PayPal, SpaceX, Tesla, Neuralink and the Boring Company, i.e. Elon Musk, Stability AI chief Emad Mostaque (maker of the Stable Diffusion image generator) and artificial intelligence researchers from Stanford University, Massachusetts Institute of Technology (MIT) and other AI universities and labs have called in a joint letter for at least a six-month pause in the development of artificial intelligence systems more capable than the GPT-4 published in March. The aforementioned letter acting as a kind of cautionary petition was published on the Future of Life Institute centre's website, advanced artificial intelligence could represent "a profound change in the history of life on Earth" and the development of this technology should be approached with caution. In this petition of sorts, there are warnings about the unpredictable consequences of the race to create ever more powerful models and complex algorithms that are key components of artificial intelligence technology. The aforementioned developers of leading technology companies suggest that the development of artificial intelligence should be slowed down temporarily, as the risk has now emerged that this development could slip out of human control. The aforementioned petition warns that an uncontrolled approach to AI development risks a deluge of disinformation, mass automation of work and even the replacement of humans by machines and a 'loss of control over civilisation'. In addition, the letter suggests that if the current rapid development of artificial intelligence algorithm systems gets out of hand, then the scale of disinformation on the Internet will increase significantly, the process of work automation already taking place will accelerate many times, which may lead to the loss of jobs for about 300 million people within the current decade and, as a consequence, may also lead to a kind of loss of human control over the development of civilisation. Developers of new technologies point out that advanced artificial intelligence algorithm systems should only be developed when the development of artificial intelligence is under full control, the effects of this development are positive and the potential risks are fully controllable. Developers of new technologies are calling for a temporary pause in the training of systems superior to OpenAI's recently released GPT-4 system, which, among other things, is capable of passing tests of various kinds at a level close to the best results passed by humans. The aforementioned letter also calls for the implementation of comprehensive government regulation and oversight of new models of advanced AI algorithms, so that the development of this technology does not overtake the creation of the necessary legal regulations.
In view of the above, I address the following question to the esteemed community of scientists and researchers:

The open letter[1] is wishful thinking at best. Let us sample some of the statements provided in the letter:
  • "Powerful AI systems should be developed only once we are confident that their effects will be positive and their risks will be manageable."
The concept of manageable risk differs from individual to individual much less gather consensus on a population on what is a " manageable" risk
  • "we call on all AI labs to immediately pause for at least 6 months the training of AI systems more powerful than GPT-4."
This is the most naive statement of the letter. What can be accomplished in 6 months? We have not even gathered consensus on the simple philosophical approach to ethics in the history of philosophy, much less on how to approach AI from an ethical standpoint. stopping AI research for 6 months will not have a significant impact (there are even journals dedicated to AI ethics in print tackling the issue for several years and there is pretense to solve this in 6 months!).
  • "implement a set of shared safety protocols ...These protocols should ensure that systems adhering to them are safe beyond a reasonable doubt."
The concept of beyond reasonable doubt will eventually fall on what is reasonable practice in the field and this leads to weak safety protocols.
  • "AI research and development should be refocused on making today's powerful, state-of-the-art systems more accurate, safe, interpretable, transparent, robust, aligned, trustworthy, and loyal."
This statement cannot be more ambiguous in term definition and the references of the letter do not help.
The concept of stopping AI research should not be confused with regulating the commercial use of the technology. The letter aims at the wrong issue. Research cannot be stopped for ML, since anyone with a reasonable amount of computing power (which you can buy anyway in amazon) can continue to work in the subject. But, regulating business is easier since the major players will have the greatest impact in slowing down the misuse of the technology. also by regulating business more targeted restrictions on its use can be applied rather than vague lists of research topics.

The problem is more acute in the case of those social media which are among the most popular and on which mainly young people function, whose world view can be more easily influenced by factual information and other disinformation techniques used on the Internet. Currently, among children and young people, the most popular social media include Tik Tok, Instagram and YouTube. Consequently, in recent months, the development of some social media sites such as Tik Tok is already being restricted by the governments of some countries by banning the use, installation of this application of this portal on smartphones, laptops and other devices used for official purposes by employees of public institutions. These actions are argued by the governments of these countries in order to maintain a certain level of cyber security and reduce the risk of surveillance, theft of data and sensitive, strategic and particularly security-sensitive information of individual institutions, companies and the state. In addition, there have already been more than a few cases of data leaks on other social media portals, telecoms, public institutions, local authorities and others based on hacking into the databases of specific institutions and companies. In Poland, however, the opposite is true. Not only does the organised political group PIS not restrict the use of Tik Tok by employees of public institutions, but it also motivates the use of this portal by politicians of the ruling PIS option to publish videos as part of the ongoing electoral campaign, which would increase the chances of winning parliamentary elections for the third time in autumn this year 2023. According to analysts researching the problem of growing disinformation on the Internet, in highly developed countries it is enough to create 100 000 avatars, i.e. non-existent fictitious persons, created as it were and seemingly functioning thanks to the Internet by creating profiles of these fictitious persons on social media portals referred to as fake profiles created and functioning on these portals, to seriously influence the world view, the general social awareness of Internet users, i.e. usually the majority of citizens in the country. On the other hand, in third world countries, in countries with undemocratic systems of power, all that is needed for this purpose is about 1,000 avatars of these fictitious people with stories modelled, for example, on famous people such as, in Poland, a well-known singer claiming that there is no pandemic and that vaccines are an instrument for increasing control of citizens by the state. The analysis of changes in the world view of Internet users, changes in trends concerning social opinion on specific issues, evaluations of specific product and service offers, brand recognition of companies and institutions can be conducted on the basis of sentiment analysis of changes in the opinion of Internet users using Big Data Analytics. Consequently, this type of analytics can be applied and of great help in detecting factual news disseminated as part of the deliberate spread of disinformation on social media.

I've been having a lot of issues with my Raw 264.7 cultures--they haven't gotten higher than 60% viability and have slowly decreased in their replication rate and viability. I've used both RPMI and DMEM, both with 10% FBS. I've tried splitting every two days, as well as changing out media with new media every two days and splitting on the third day. I've heard this cell line is pretty sensitive, but I can't figure out how to optimize this culturing to get viability and confluency up to 90%.

Some facts that you should know about RAW264.7 cells while optimizing.
1. You could either use DMEM + 10% FBS or RPMI +10% FCS. Use of 2mM L-glutamine and Pen/strep is optional.
2. These cells are most suitable to culture in a T75 cm2 flask. Total volume in the flask should be around 10mL. The growth media should be refreshed every 2-3 days but no more than 4 days without causing significant cell death. The average doubling time of RAW264.7 cells is 15 hours.
3. When the cell confluency reaches around 60-70%, the cells need to be split. Aspirate all growth media by tilting flask to allow media to collect in a corner. Wash once with PBS (half of culture volume) by gently aspirating PBS into a corner, gently shaking the PBS around the flask and aspirate. Eject half of culture volume of trypsin directly onto the cells and incubate for 5 minutes at 37 degree C. Please note that RAW cells are quite adherent to tissue culture flask. So, if the trypsin does not immediately lift the cells, do not proceed to wait longer as this risks the viability of cells. Eject (half of culture volume) warm media directly onto the cells. Gently use the cell lifter and scrape cells off the flask. These cells do not lyse easily with a cell lifter. Collect the cell suspension from flask. Spin for 8-10 minutes at 1500 rpm.
4. Resuspend in fresh media. It is important to triturate repeatedly to allow for a single-cell suspension. Seed cells into a new flask with dilution and place back into the incubator. 1:3, 1:4 or 1:5 dilutions are all acceptable. Please note that greater the dilution, the longer the cells will take to reach confluence. Do not heavily dilute as cells seeded with extremely low confluence exhibit impaired growth and proliferation.

Hi everyone,
I have seen some very strange debris in my U87MG (glioblastoma cell line) culture. It seems that there are shards/fragments of glass that are not contaminating the cell culture and the cells are potentially trying to adhere to them. I attached some pictures of the debris, taken shortly after passaging, so they are not fully adhered.
When I first saw this, I bleached the cells, threw out the package of flasks I was using, made new media, etc. I figured that this originated from a glass pasteur pipette, so I thawed another vial of cells that was frozen down at a different time than the previous ones I used. However, I'm still seeing the same debris in the freshly woken up cells.
I'm confused because I didn't see the debris in the first set of cells until weeks into using them, and I didn't see them in the second set of cells until about two weeks since I thawed them. I may have just not seen it until two weeks in, but it just seems unlikely due to how much and how obvious it is.
This is the only cell line I use EMEM (w/ penstrep & 10% FBS) for, and I have never seen this in any of my other cell types that use other media (completed with the same batch of penstrep and FBS).
Please let me know if you have ever seen anything like this before! Any comments are appreciated - thank you all in advance.

I sometimes saw them after defrosting the cells. Such structures are formed by DMSO if it is unevenly mixed when cells are frozen.
When adding to the freezing medium, try to stir the DMSO as vigorously as possible. You can also add DMSO directly to the cryovials containing media and cells and mix thoroughly. A small volume of DMSO is easier to mix.

Hello all, I am culturing MCF7 cells that are in DMEM/High Glucose media. I split these cells once and the next day noticed these "gold foil"-like residues in the media plate. They seem to be attaching and growing with the cells because every time I wash the plate with DPBS, they go away. Everything in the culture room and culture hood have been cleaned with sterilizers. New bottles of media, DPBS, and trypsin are used. However, once new media is added and the plate is placed back in the incubator, they seem to grow back again. Are these contaminates? I appreciate all feedback!

I agree with Dr. Alexandr Chernov. Also, if you see it for yourself, the size of these cells is way larger than what a bacterial cell should be. It seems to be cell clumps that have remained together. I suggest that you prepare a single-cell suspension by gentle pipetting during sub-culturing and washing. Good luck!

While removing the used up media, by default we do tilt the flask so that media could collect in the corner and that does not cause any problem. But cells detachment happens because of the force of fresh media which is being poured directly over the cells surface. So you shall add media gently over the wall of the flask interior, while having the flask tilted.

Hello All, I work with INS1 cells and am able to grow properly and passage them. Howeve, I encounter difficulties when I attempt to culture them in 6 to 24 well plates. The primary issue is the cells tend to detach following administration of new media or other solutions, even when I add media to the wall slowly (15-30 sec/well). If cells don't all detach, the ones left over tend to be in the center.
Additionally, I will notice a morphology change in some of the cells that don't detach. They change from a full bodied, translucent cell with extensions to a small, rather opaque rounded cell with little to no extensions.
Has anyone encountered this issue? If so what would you suggest?
I have used poly-d-lysine coating to mitigate this, and while it has helped is hasn't completely erased the problem.
Any help would be greatly appreciated!

I am still trying to get a proper grasp on seeding cells into wells, and I have a formula that indicated I would need ~41 mL of concentrated cells (ST cells in my case). I am using nine 24-well plates to get a total volume of 216 mL.
Here is my formula, and if it seems incorrect or needs editing, please let me know as I am very much trying to learn the proper way of doing this:
  • Total volume of concentrated cells needed = ((desired total volume)*(desired final cell total per well)) / ((average cells counted in the hemocytometer)* (10^4 to get units of cells/mL)*(dilution factor)
So an example of what I would have is:
  • ((216)*(5x10^4)) / ((13)*(10^4)*(2)) = 41.53 mL of concentrated cells needed
Does this seem correct? Am I missing something?
For some more context that may be important:
  • When I centrifuge the cells and toss out the supernatant, I then resuspend the cells in 10 mL of new media.
  • I then take the 100 microL of the resuspended cells and put in 100 microL of trypan blue (so that's how I get my dilution factor of 2) to then count in the hemocytometer
Any help and guidance would be greatly appreciated!

You are right. You can also do the calculation as follows:
50,000 cells per well per ml is required.
You have 2,60,000 cells ( 13 x 10000 x 2 )
So by the formula
N1V1=N2V2
Where
N1 = Total number of cells = 2,60,000
V1= the volume to be calculated X
N2= Number of cells per well = 50,000
V2= Volume per well of the 24-well plate = 1ml
Therefore ,
2,60,000 x X = 50,000 x 1
X= 0.192 ml for one well per ml.
So for 216 wells the required number of cells will be 216 x 0.192 = 41.5 ml.
While taking the cell count using the hemocytometer you should use lesser volume of cell suspension (say 10ul and not 100 ul). So 10ul of cell suspension and 10ul of trypan blue.
You should always calculate 2 wells extra because of the volume effect. The last well will not receive the required volume and there is a possibility of an error.

Can cells be reused after I have seed them onto 96 well plates and test them with cell counting kit at OD450 (CCK8) at 12 hours, wash them with PBS x3 and add new media and let them incubate for another 12 hours to get a 24hour time point, same procedure for another 24 hours to get to 48-hour timepoint or is it necessary to do 3 parallel plates to get 3 timepoints?

I am trying to perform a modified MLR adding CD14+ monocytes to CD4+ T cells (both positive MACS isolation), under different conditions. However at 5 days, even with HEPES in culture media, the wells are yellow and I'm seeing significant CD4+ cell death.
I have been spiking the media with varying volumes of fresh media at intervals throughout 5 days however this doesn't change the outcome much.
Should I spike the wells with new media?
Should I completely exchange the media?
Could this be due to the ratio of APC to responder cells which appears to be crucial (currently 1:1 or 1:2 CD4:CD14)
Any other ideas?
Lastly I am also labelling the CD4 cells with CFSE to check for proliferation, is it ok to do this prior to the 5 days incubation or should this be performed at end of 5 days?
