I have an preliminary study with 5 subject and I intend to find out the effect of a massage therapy on blood pressure. In total, I have 3 session, each massage session last 20 mins. For every session, pre and post data blood pressure data will be recorded. The 3 massage session will run consecutively for 3 days.
As for now, I will be using non-parametric test since I have small sample of data. I am thinking to conduct the Wilcoxon Signed rank test for each of the session separately. What statistical test should I use?

Possible issues: 1. not all model degrees-of-freedom are actuated,
2. there are tracking tasks for locked coordinates, and/or
3. there are unnecessary control constraints on reserve/residual actuators.
to solve this problem ,i had double checked Possible issues 1-3 above, however it did't work.

I've just finished a brilliant book called 'Art of Thinking Clearly ' by Rolf Dobelli and it introduces the concept of the 'Authority Bias' where when making decisions we carry out irrational tasks out of sheer obedience to authority. Authority also apparently reinforce their authority with symbols (e.g. White coats). Rolf's advice of course is to naturally not bow to authority if you feel threatened or ordered to carry out irrational tasks. Challenge him/her instead, and of course there are huge cultural issues that prevent people from 'standing up' for themselves. My other great writer Malcolm Gladwell has described this phenomenon very well in his also brilliant book 'Outliers' where plane crashes in the past have been attributed in part to the fact that the 2nd in command were unable to speak up against the captain when a disaster was looking using mitigating language instead.
It makes me wonder does the same 'Authority Bias' exist in research teams and if so how can we prevent it. I know that at all times I encourage my Postgraduates to drive their projects forward independently and of course this is the essence of a PhD that the person becomes a Level 10 independent thinker with an ability to apply knowledge gained etc into practical life and society etc etc but of course students often lack confidence and can sometime behave submissively. Being aware of this 'Authority Bias' and alerting students to its concept is probably wise in any research group but what I also wonder is when should an academic with a PhD feel confident in calling themselves Dr.... outside of the research world???
Is the use of calling yourself a 'Dr' even within your own research group a form of 'Authority Bias' being exerted?????

I'm trying construct a phylogenetic tree using the analysis method maximum parsimony (Max-mini-branch and bound) for different protein families (each family contain set of protein sequences with the same domain) using MEGA-X software. I've just finished a brilliant book called 'Art of Thinking Clearly ' by Rolf Dobelli and it introduces the concept of the 'Conformation Bias' where we have a tendency to interpret new information so that it becomes compatible with our existing theories, beliefs and convictions. In fact subconsciously we filter out new information that contradicts our existing views. Rolf's advice is to always look for dis-confirming evidence and never use the word 'exception'. Apparently, what a human being is best at doing is interpreting all new information so that their prior conclusions remain intact.
This gets me thinking about my experiences with supervising Postgraduates and how difficult it can be to get them to refute their original ideas/hypotheses/predictions, and the awkwardness when it comes to deciding to include/exclude obvious outliers when analyzing research data.
This makes sense, because after making the effort to write a research proposal and then looking up the literature (especially if cherry picking for a focus article/mini-review), it can be extremely difficult to see the signs that the original predictions are incorrect or correctly interpret 'dis-confirming evidence'.
I'm interested to hear opinion about this, because I have a feeling it also contributes towards our huge problem with 'publication bias' etc.

It is often an unconscious tendency. I tend to guard against where possible. Researchers do have some fixed notions that become accepted wisdom then guide expectations, perpetuating bias.

Hi,
I have question regarding cross sectional-dependence in panel data please.
In case there is evidence for cross sectional dependence in my data, how can I correct it please so it does not affect me results.
I know we can apply xtscc/FGLS in STATA but when I applied it and apply xtcsd pesaran to check the results it generate error massage as I should use xtreg.
Additionally, is there any way to correct the cross sectional dependence and generate the data without cross sectional dependence so I can use the data in another software.

During meshing the COMSOL model you have to remember one think is that the minimum mesh size should be smaller than your smallest domains in your model.
Example, suppose in your model consists of 9 domains. Lets assume domain 1 is having small size among the domains i.e. 2cmx1cm, them your meshing size should be ( maximum mesh element is 0.5 and in minimum mesh element is 0.2).
Or
You can mesh all the domains separately according to their size.

You assigned some surface with a kinematic constraint and a contact constraint. So they may conflict each others.
A similar message appear when I have 2 surfaces having a common line. And I assigned each of them with a boundary condition, so the common line have two different assignment from both surfaces and the program provides warnings.
The warning may not effect the model results and is acceptable but you could use General contact option instead of specifying a particular surface with contact constrain to get rid of this message.

Right away: This is a question I am struggling with, and it is asked for a PhD program that I want to get into. So maybe I am too stupid and shouldn’t participate since I can’t answer this question. However, I will ask the community anyway since I am always eager to learn more and I want to solve this puzzle. If this is considered cheating in any way, I would like to ask the authorities to delete this post, and I’m sorry. If not, I would like to communicate, especially if I can learn and talk with people that are more experienced than me and I hope that someone can help me and would start a discussion with me.
So here is the question:
I have a protein of interest that has acetyltransferase activity and binds to promoters of actively transcribed genes. The knock-out shows no phenotype in mammalian cells. However “an inhibitor of its catalytic activity caused impaired DNA replication and transcription.”
My task is it to interpret the results and suggest further experiments to validate my hypothesis. Well, I am struggling with the theory already. The main problem that is puzzling me is the fact that I have no phenotype in the knock-out but with a present but catalytically inactive protein. Suggesting, that the protein of interest is not essential (or the right condition for the phenotype to emerge hasn’t been tested yet). However, the inactive form causes some problems.
During the reading, I got stuck with the idea that this protein might be somehow involved in the p53 pathway or alternatively causes problems on the chromatin itself when expressed but inhibited. However, I do not find a lot of information about acetylation besides Histones and N-Terminal regions of newly translated proteins (which are a lot I’m assuming, but I don’t have the time to study all the possible proteins modified by acetyltransferases and their effect).
1) Since one of the prerequisites of p53 function is the acetylation of p53, I thought it might bind to p53 but is unable to modify it and therefore is kind of titrating the p53 away. Therefore, p53 can’t support DNA replication, can’t promote Mdm2 transcription (that also seems to support DNA replication according to a paper I read) and influences also the stress response upon, e.g. DNA damage.
2) Alternatively, the protein with inhibited catalytic activity might bind to Histones but remains stuck which leads to problems, since these proteins will accumulate and block the Histones for “reader” proteins? This, in turn, leads to aberrant transcription since TFs don’t receive the massage to get activated.
BUT, if the hypothesis above were true why does, the knock-out doesn’t show a phenotype. Might the protein just be sufficient by enhancing some processes like transcription by further opening chromatin through acetylation of histones or p53, but is not necessary for these processes since the knock-out obviously doesn’t show a phenotype? Or does this protein might have some moonlighting effect and might bind/regulate some entirely different proteins/pathways? If so, how can I know?
I would really appreciate tips, help or constructive criticism that would help me understand the problem here.

Hi all,
i am trying to run fusedTriplets.py . i have run the following commands in order to get my input
makeblastdb -in all.fas -dbtype prot -title all -out allseqs -parse_seqids
And for the input file I used
blastp -query all.fas -db allseqs -out all-vs-all_dbsize_defult.tsv -outfmt 6
blastp -query all.fas -db allseqs -out all-vs-all_dbsize_defult.tsv -outfmt “6 qseqid sseqid evalue pident bitscore qstart qend qlen sstart send slen”
and I get the error Assertion error: network header should start with 'qseqid sseqid'
I have tried using blast outfmt 7 (edited it too) and 10. but still get the same error massage can anyone help with the appropriate thing to do to overcome this

blastp -query all.fas -db allseqs -out all-vs-all_dbsize_defult.tsv -outfmt “6"
use only this and remove other flags, your sequences in the blast database seems to miss the id in header.

There I'm simulating the gas adsorption of CH4 and CO2 on a MOF.
The simulation works well from 1 to 5,5bar but above it's not working. I tried to change Flowrate and initialize specification. For isotherm I'm using the I.A.S Langmuir Isotherm.
The error massage is "integrator corrector failed. Check your model equations and variable bounds."
Do you have an idea how to overcome the problem? It would be nice to be able to simulate up to 10bar.

Indurative mastitis is associated with fibrosis of udder parenchyma. Edema is most likely emanating from obstruction of blood flow by fibrous tissue, in which antibiotics are no longer the solution . I would suggest surgical intervention. Even with that, this cow is unlikely to have adequate alveolar parenchymal regeneration for the next lactation.

Our lab is doing zebrafish research and my job is to maintain the health of zebrafish. I found a female fish with a very big belly so I let her mate with another male. However, there was no egg came out when I checked it the other morning and the belly remained big. Does anyone know what cause this problem? Should I do a belly massage to help her strip the egg? If yes, how to do it?

I agree that it sounds like your female is egg-bound. Basically, the ovaries are producing eggs but the female has stopped releasing them, perhaps because the ovarian ducts are blocked and the oocytes are not reabsorbed efficiently, hence the increase in the size of her abdomen. It can be painful for the fish and in extreme cases can cause the skin to start splitting.
When something is off with our fish, the first thing I do is check all of the water quality parameters. Next, look at other parameters like food and light. You could be seeing this response if you have changed the type of food you are using, if the food is very expired, or the female is obese or has not been bred very much (especially if you house according to gender). Also, check the timer on your lights to make sure that it is working properly.
Most sources recommend gently stroking the abdomen from head to tail, massaging or squeezing the abdomen. This is very similar for procedures used for egg stripping for in vitro fertilization (https://youtu.be/ziNAXS1E0vE). You can do this with or without anesthesia (I usually choose without since it is fairly quick). If you choose to go without, make sure you use a soft net and keep her in it during the massage. You will know if this works in a relatively short amount of time after she has been returned to her tank and/or recovered from anesthesia.

hello every body, I was trying to simulate fluid flow and heat transfer in axisymmetric vertical channel but when I set all condition and solve the case I got this massage (Divergence detected in AMG solver: ads-0). and when I changed 2D space to planar case is solved. what is the problem? could anyone tell me what is the problem? My proposed answer is that they know what it is that they want to know. They do not know that the optimal statistic exists, let alone that it is linear! Irving Fisher told them that estimation is a choice of functions and so it is a problem in stochastic optimal control or just simply calculus of variations. Instead of doing the worl, they massage the data set until it tells them exactly what the pay masters wish for it to tel. Thus they pride themselves by being his/her master's voice. I ran this by two econometrician who are economics Nobel acolytes.  They told me essentially that the only objections then had to econometrics (thy advocate that no none should teach econometrics!). Their reason is that the "results" do not confirm the economic prejudices of either one of them.  Can you help?

I don't mind sharing at all.  The primary rationale for our dosage and frequency were to make the treatments clinically feasible within our primary field of athletic training were providers sometimes have limited time with individual patients.  However, we also wanted to build in multiple treatment sessions so that we could potentially train/improve the sensory system on top of treating the symptoms.  Much of the literature suggests that while a single session, of shorter duration, could be effective, multiple treatments result in a more meaningful response in select outcomes.  This was our first effort to determine if the larger responses were train to altering motor control programs within the patient by improving the degrees of freedom available to them.  Based on these results, we believe that is possible and are now working on the next steps to confirm these results.  I hope this helps

IRR for 45 mins will be too long as it can cause burns. Usually it is given for 10 mins, to the max 15 mins and it is very good combination after massage espicially in palliative care

I use an ANFIS toolbox to predict a parameter with anfis and this error has appeared.
Warning: genfis1 has created a large rulebase in the FIS. MATLAB may run out of memory if this FIS is tuned using ANFIS.

This error happens when number of FIS rules is greater than 250. You should use a smaller FIS (smaller Number of MFs) and use genfis2 instead of genfis1
GENFIS1 uses the grid partitioning and it generates rules by enumerating all possible combinations of membership functions of all inputs; this leads to an exponential explosion even when the number of inputs is moderately large.
