So im doing solid phase extraction of a herbal extract. Then, i'm doing total phenolic content using the folin-ciocalteu assay. I.ve done the gallic acid standard with r2=0.98 with straight line eqn= 0.0085x -0.123. but then ive been wondering how do i use the tpc eqn? tpc=c v/m. c i can get from the equation. but for v and m? do i use the loading concentration data where i use 6 mg/ml or i use the concentration finished product of dissolve dried fraction (10 mg/ml). then the question also comes to mind should i use the concentration before putting the folin reagent which is 10 mg/ml or the diluted version (60 ul folin, 60 ul sodium carbonate, 150 ul water and 30 ul of extract which makes it 1 mg/ml)? please im so confused

I am currently investigating the cytotoxicity of a series of herbal extracts, and like many studies, I have been using the MTT assay to evaluate cell viability. However, I am encountering a significant challenge: my extracts are highly colored, which interferes with the colorimetric readings inherent to the MTT assay. This interference could potentially lead to inaccurate results.

i am working on the determination of the synergism activity of the herbal and allopathic drug. firstly i tried to find out the zone of inhibition of individual drug having lower (25, 50, 100 ppm), middle (250, 500, 1000 ppm) and upper (2500, 5000, 10,000 ppm) concentration against s. aureus bacteria. but the expected result was not obtained in case of herbal drug. kindly provide the valuable suggestions for the same with references. I tried the same experiment against E. Coli and Pseudomonas Aeruginosa & the expected result was obtained in both cases. I am facing a problem only in case of S. Aureus. I also like to add that i repeated the experiment 9-10 times by maintaining aseptic condition and other cares to be taken while performing the experiment. Kindly give the solution for S. Aureus to get desired result. Thank You
Colleagues, I am currently working on a project studying the toxicological effect of an n-Hexane fraction of a herbal extract using a clonogenic assay. However, I'm encountering difficulties as the herbal fraction does not dissolve in the cell broth, preventing me from measuring any cell toxicity effects. I have attempted different solvents, but each has presented its own challenges. n-Hexane placed upper the broth and evaporated quickly, Chloroform formed an insoluble hemisphere under the broth, and Acetone resulted in color dots at the bottom of the well and counting errors. Can anyone suggest alternative methods or solutions to overcome this issue? Your assistance would be greatly appreciated. Thank you."
We have been dealing with the bench-top spray drying of a hydroalcoholic herbal extract comprising around 30 GL ethanolic content and 5% solid content. The evaporatory capacity is 1.0 L/h and the equipment is operated in concurrent flow regime. The pneumatic (two-fluid) spray noozle available has 1.0 mm or 1.2 mm inner diameter. The nozzle air pressure is 0.4 MPa. The drying air is supplied by a blower (nominal flow rate of 1.0 m3/min) and electrically heated. The cylindrical drying chamber was made of borosilicate glass with 160 mm in diameter and 645 mm in height. Is it possible to determine which could be the safer experimental condiitons (i.e., range of drying air inlet temperature, extract feed rate, spraying air flow-rate, etc.)? Do you guys reccomend any literature regarding this issue?

I have obtained a herbal extract obtained by directly crushing the tissue (no solvent used). If I store said extract at 4’C; some constituents appear to settle down at the bottom of the storage tube which dissolve immediately upon lightly shaking the tube. Moreover, the scent of the extract is also comparatively milder upon storage. Literature reviews have indicated that the extract is composed of several volatile components which undergo spontaneous conversion.
It would be much appreciated if methods for comparing fresh extract and the stored extract for any change in composition (apart from HPLC) can be recommended?
(Ps.- I do not have extensive background in chemistry, in case that is something that you need to know to make recommendations).

I have gained something strange in the result of mtt assay of the PBMC.
I was testing an herbal extract that is declared to be safe for normal blood cells by researchers but I saw that ODs of the different doses on PBMc such as Ic50/4 , IC50/2 ,IC50 and 2 IC50 were the same!and all of them were half of the negative control!!!
Can any one interpret that for me?

Considering the results of an article which is similar to my current research, in a validated Soxhlet extraction method, the ratio of solvent to herbal powder should be 21.78 (v/w). But because of the small amount of the herbal powder and the large size of the apparatus that I have, it is not possible to follow this instruction.
I am wondering if using the same recommended solvent --but in a larger amount than the recommended volume-- while maintaining the same recommended duration of time of extraction can alter the efficiency of the extraction process.
I would appreciate it if you could help me with this question.

Interaction of various molecules in a herb and between various herbs in herbal compound formulation?
Traditional herbal medicines, which are often used as a fresh prepared herbal infusions as antypyretic agents, are used in a hot or a warm state. When the boiling water is used for making the infusion, the extraction temperature is at first a little below of boiling temperature. In this temperatures a spectrum of extractive substances is larger than in room temperature, when most phytochemical analysis are performed.
Reading the techniques used in phytochemistry I never found descriptions of analysis or chromatographies, made in temperatures of application of the infusions. On the other hand in many times when we look at the infusates they are clear in higher temperatures, when are hot and with descending temperatures is seen opalisation and precipitation.
To measure the entrapment efficiency we use UV not HPLC, is it enough to measure absorbance at any wavelength selected by me? one with max. absorbance for the extract, but not related to a certain compound coz we are not using a standard.

I am conducting a study to assess the quality of selected parts of some herbal materials and also develop acceptance criteria for their quality attributes. I am supposed to sample these materials from across the length and breadth of the country and I am hoping to stratify the country into strata and further divide each stratum into clusters, and then randomly sample the materials from each of the clusters picked up through systematic sampling.
My challenge is with the calculation of a 'realistic' sample size that can then be used to determine the number of clusters and the number of samples from each cluster. Very often what I see in literature tends to be convenient sampling, which may not be representative of the population. The focus of my study however requires that my sampling is representative of the population in the country (and also realistic), especially because of the part that has to do with setting acceptance criteria.
