Does someone have experience with eluting the CRISPR RNPs from gold nanoparticles?
I found a few articles that suggest using the Vivaspin 300 kDa concentrator: https://www.nature.com/articles/s41551-018-0252-8#Sec40
Chatgpt suggested: Citrate Buffer, High Salt Solution (e.g., 300 mM NaCl or Higher), thermal elution (e.g., 37-50°C) for 10-20 minutes.

I'm not sure I fully understand the desired outcome, but I can offer some insight. The high salt concentration and heat will probably lead to gold nanoparticles (AuNPs) aggregation. If the RNPs are bound to the AuNPs, this will lead to RNP aggregation, which I assume is a bad outcome. Low buffer concentration of 20mM without excess salts would be preferable for the AuNPs.
Are there free AuNPs in the solution and RNP-AuNP complexes that require separation? If both complexes are large, they would both be blocked by the Vivaspin filter. You could try purification via CL6B Sepharose matrix - it is suitable for separation of large complexes. Separation via agarose gel electrophoresis may also be viable.
If you wish to remove the AuNPs completely, a chelator could specifically break down the AuNPs. Examples include EDTA, potassium cyanide and thiosulfate, but they have disadvantages. The literature may provide useful insight and protocols. After chelating the AuNPs, use Vivaspin or a G-25 column to remove excess Au salts and chelator.

I recently encountered an issue on ResearchGate: one of my scientific publications disappeared due to a presumed bug, along with its statistics and access link. Naturally, I contacted ResearchGate’s support via the “Contact Us” form, providing all necessary details. However, weeks have passed, and I have yet to receive any response.
This raises a broader question: How responsive and effective is ResearchGate’s support in handling user queries? Given that researchers may face issues ranging from minor inconveniences to critical concerns (such as lost data, account problems or even plagiarism), one would expect timely assistance. Yet, my experience so far suggests otherwise (I hope, it is different in general).

I have contacted them many times, A few cases, I did not receive any answers! but for the rest of the times that they answered, there was not a fix time table, a day to several days and more. Also all the time that I received an answer there was just a "Sam" in signature.

Please I have been struggling for days on estimating some spatial diagnostic test on my model. The stata command I am using is spatdiag command.
I am working on a panel data with 759 observations. However, I am using a 33x33 matrix. Each time I run the spatdiag command after the regression model, I get this error "Matrix W is 33x33, regression has been carried out on 759 obs. To run -spatdiag- weights matrix dimension must equal N. of obs"

Hi,
I am a beginner in bioinformatics. I have more than 500 genomes to analyze and I already got the results from Roary. I want to visualize these results by showing the tree compared to a matrix with the presence and absence of core and accessory genes. Roary provides us with roary_plots.py, which can achieve this. However, If I have hundreds of genomes, the plot will be a mess and nor very clear.
If there are any tips to make the tree look more clear in the cases of having hundreds of genomes, I'd be grateful to hear them.

I'm not sure if there is an optimal way to do it with roary_plots.py, since hundreds of genomes will still be many rows. Looking at the code of roary_plots.py, you should be able to increase the width of the tree to improve the clarity by changing the “colspan” parameter in the tree display. You can also improve the size and resolution of the image with the “figsize” and “dpi” parameters.

In research, I am simply looking to gain more information about a given topic. Through research I can learn a magnitude of different things and it can have a very broad range. I see evaluation as being more fine tuned. Evaluation involves testing out a question and seeing how it works, and then utilizing that information to inform decisions.
I feel that my definition of each is heavily influenced by my career as a teacher and how I utilize each one daily. So I've got high purity 5-MeO-DMT Freebase oil, tested at 99+ by GCMS.
Regarding 5-MeO-DMT, I've spoken with my colleague (our head chemist). We've got the actual synthesis optimized quite nicely now, however the problem is that the end product is an oil (freebase). We've tried so many different techniques to induce crystallization but nothing works. We've found literature that details different polymorphs of DMT and 5-MeO-DMT and indeed some of these polymorphs are oils. The purity by GCMS of the material is extremely high and no residue is left on vaporizing. If you have any suggestions to induce crystallisation I'd be happy to hear. We've tried vacuum distillation as per Shulgin but that doesn't work. The HCl salt is easy to form by I can't get the Freebase to crystallize.

Devin Lee, you may have already read this by now, but apparently the freebase can be induced to crystallize by recrystallization from boiling hexane, as per Shulgin, mentioned in the book TiHKAL, under the entry for 5-methoxy-DMT: "There was thus obtained 12.8 g (78%) 5-methoxy-N,N-dimethyltryptamine (5-MeO-DMT) which on recrystallization from hexane had a mp 69–70 °C." Recrystallization in this manner has been suggested to yield "waxy colorless crystals", according to at least one post on an online discussion board.

I analyzed some samples by LC-MS/MS, and I got a pretty good peak for quantifier ion, but the peak intensity for qualifier ion is not good enough so I got "ion ratio" as outlier reason.

We have examined Fusarium toxins in cereal products in our laboratory as part of a project.
I can confirm Zvonimir Mlinarić 's statement that these toxins have structural isomers that do not or hardly differ in their retention times, masses and fragments. However, we did not focus on metabolites but only on the toxins. As MS we used the Thermo ID-X Orbitrap, so that we could calculate a sum formula for the LC-peaks with the help of the accurate mass, which then coincided with the sum formula of the toxins. The MS/MS spectra could also be found in Thermo Fisher's MZCloud database and thus the toxins could be clearly identified. This was the aim of the students' work. If you have the possibility to analyze your samples with HRMS, I would recommend it. I agree with Zvonimir Mlinarić that a shift in ionic ratios can be explained either by matrix effects or by the presence of various structural isomers. I consider the latter to be the probable cause.

I think this machine tries to do too much, and that eats into cost and practicality...
Personally, the flexibility of having a PC operate the machine is a much better option rather than having an integrated GUI. Sure, you may need a bit more bench real estate for a small PC workstation, but I'd like to highlight a few points on how its better and how Thermo may be losing sight of what costumers need and benefit from. First, configuring an imager to a network or transferring large files via USB is an unnecessary, and possibly tedious step. Relying on a USB stick is likely to be what most occasional users might do and this also increases the chances of cross-contamination with potentially harmful reagents. Secondly, from my personal experience of using the highest-end model (iBright FL1500 Imaging System), I immediately noticed how slow and clunky the interface is. Why charge extra premiums by adding all the extra peripherals to run the imaging and analysis software when it only eats into the cost of the device and the hardware is sluggish? It ultimately makes for a very, VERY unpleasant operating experience. Removing the onboard interface would reduce the price by a large fraction and just a small portion of those savings can afford the user a PC that's going to perform better in every way.
Emptying the stored data to make room for new data? Tedious extra step. Processing units and digital peripherals need a firmware or software update or repair costing you thousands in tech support from Thermo? A tedious (and costly) extra step. Having to wait for a colleague to not only image their gel but also analyze results on the touchscreen? Tedious and time-consuming extra steps; I don't know why anyone would want to stand in front of a machine and play with parameters (that take literal tens of seconds to load) after imaging when it makes a lot more sense to do it at a desk with a faster, more powerful computer... Why Thermo encourages its equipment users to do this and thinks it's convenient is beyond me, now you've got buffer spots all over the touchscreen! Alternatively, a computer hooked up to the machine which only provides the essential hardware to operate is much easier. Run out of space? No problem, swap out a drive, or the whole computer, I dont care and nobody else does, but that's highly unlikely because affordable PCs routinely come equipped with terabytes of storage which would likely last a busy lab years (in comparison to the 256 GB of storage onboard the highest-end model).
Now to the worst part of the entire machine: the automated drawer... When I have a busy day at the lab, the last thing I want to do is wait for a sluggish interface to decide to initiate the drawer opening function when my brain and hand can execute the same task efficiently (I don't actually know if my brain and hand are more efficient, you'd have to run some serious metabolic calculations and compare that to the wattage of the thermo system), oh and it also takes time to initiate the drawer closing function. I just wish we could have a company that realizes that all we want is the same hardware functionality without all the silly bells and whistles that 90% of users won't touch or appreciate and that ruin science for everyone and steal tax money from everyone. Dont buy it

I just wonder if I can determine the mechanical properties of my gel by using G' and G" taken from the rheological test. For instance, I want to determine how stiff or soft my material is at 20C or how viscous my material goes after experiencing amplitude at a certain %strain by just looking/translating G' and G".

Hello everyone! Throughout my master's thesis and Ph.D. studies, I have encountered difficulties in achieving optimal clean-up of the UHPLC-ESI-MS/MS system.
My research focus lies in the analysis of peptides derived from plasma samples, and despite implementing an extensive pretreatment procedure aimed at eliminating proteins, particularly albumin, the chromatographic system consistently exhibits issues such as increased noise in the chromatogram and elevated column backpressure. Even the use of a pre-column has not provided a complete resolution to these challenges.
Notably, these problems seem to intensify when my colleagues, particularly those working in the metabolomic field, use the instrument following my experiments. Despite employing a thorough cleaning protocol involving a prolonged gradient elution (lasting approximately 1.5 hours) to ensure the removal of all potential samples residues from the system, the results remain the same.
I would like to know if you have come up with the same problems and if you ahve any insights on potential strategies to overcome these persistent issues. Whether it involves refining the pretreatment procedure, exploring alternative stationary phases, optimizing the cleaning protocol of the system, or considering additional precautions for shared instrument use.

It is pretty common to have carry-over analyzing plasma/serum samples. Someone recommended me the use of Trifluoroethanol to wash out all retained peptides. Basically you just use it as a sample and inject 1 or 2ul direct onto the column with a relative short gradient. It does not damage the column in my experience, so maybe you can give it a try in the future.

But can it be caused by conflicting sensory information within one sense? I am curious if there is a way in Mendeley to have the 'description' of the media referenced in the bibliography in brackets, following the title, without using italics, while still before the period ending the 'title element.' For example, in the reference examples area of the 7th edition of APA, 88. TED Talk (American Psychological Association, 2020, p. 343) shows (please excuse the missing hanging indents):

How the Current Mendeley Citation is Entered
In the current listing example, I entered the description - [Video] - at the end of the title since it needs to be before the period "in the title element of the reference" (American Psychological Association, 2020, p. 342). However, in Mendeley, this puts the description improperly in italics. I have yet to find an area where we can add the media description for a web page.
Solutions I have Tested
I have begun looking at changing the 'type' I used to classify the work to see if this could solve the issue. However, none seemed to format it correctly after using generic, television broadcast, and film. Is there something I have yet to try that you believe may work? Does Mendeley have a little-known way to format something differently in the 'title element' (i.e., similar to how on some sites, if you enter * before and after your entry, it makes it bold)?
Do you have any other ideas or suggestions? I would love to hear them and give them a try. If unable to find a solution, I may have to manually adjust the bibliography before submissions and/or whenever the document has been refreshed.

My colleague and I are looking to run an online study that requires assessment of general reading level of the participants. We're looking to assess reading level in hearing and deaf samples. I'm finding it hard to find a measure to use. The test just needs to give us a proxy estimate of reading level and be straight forward to deliver in an online format. So far all of the research articles I've been able to find that use a reading test are dated and the tests are no longer available. Does anyone have a suggestion for a test we can use and can point us in the right direction to obtain the test?

"Conclusion A library is a storehouse of information and a key to the knowledge, but the era of information technology revolution has relegated the librarians especially in schools to the background. The audiovisual media specially the Internet has lured the budding readers away from the printed works like the Pied Piper. There is a great need for the upliftment of school libraries in India. If the reading habits of the students have to be changed, the conditions of the school libraries have to be improved. For this purpose, the central as well as the state government, the school authorities, the school librarian and the teachers will all have to work together. Firstly, the government of India must ensure that the school library has a well-stocked active collection managed by a qualified librarian. For this purpose, a school library legislation should be passed as soon as possible Secondly, the positive attitude of the school principal is very important. He should clearly lay down policies regarding the school library services including its goals, priorities and services as well as its relation to the school curriculum. In fact, CBSE has recently brought out a book entitled „Organizing school libraries – Guidelines'. It provides useful information for the school principals to upgrade their school libraries and make them more functional. They should organise their school libraries according to the guidelines provided. The school librarians in India must play a positive role of being the information providers. Librarians must assist the teachers and students to search out their information needs, critically evaluate the materials and use technological means to synthesize their findings into new knowledge . Such an agency should highlight the basic responsibilities of the school librarians as well as the responsibilities of the teachers and the school authorities towards the library by drafting standards keeping in mind the requirements of the present day school students. Since replacing CO2 tanks today, we've been hearing a squeaking noise every time the incubator replenishes its CO2 (like a squeaky grocery cart wheel). I see the pressure dip briefly when the noise starts (indicating that the gas is being used) & once the incubator reaches 5% CO2, it stops. It seems like the noise is coming from the regulator, but it's hard to pin point the source. I'm usually the one who changes out the tanks, but somebody else did it for the first time today; it seems like she did everything right, but maybe she overlooked something. Our system doesn't usually make noise...how do I make it stop?

John Morreall says that when laughter is mentioned in the Bible, it is associated with one of three things. In descending order, they are: Hostility, Foolishness, and Joy. For laughter and hostility, consider Psalms 59:4-8 which implores God to “have no mercy on villains and traitors…. But you, O Lord, laugh at them, and deride all the nations.”
For laughter and foolishness, consider Genesis 17:17 when God tells Abraham at age 99 that he and his aged wife Sarah will have a son. Abraham “fell on his face and laughed.” On hearing the news, Sarah also laughed with disbelief, and “when God confronted her, she compounded her foolishness by denying that she had laughed.” When their child was born, they named him Isaac (meaning “He will laugh or rejoice”) (Genesis 18:12-15).
Laughter is again associated with foolishness in a Bible passage which reads: “Sorrow is better than laughter, for by sadness of countenance the heart is made glad.” “The heart of the wise is in the house of mourning; but the heart of fools is in the house of mirth” (Ecclesiastes 7:3-6). But laughter can also be associated with joy in the Bible as in: “When the Lord restored the fortunes of Zion, we were like those who dream. Then our mouth was filled with laughter, and our tongues with shouts of joy” (Psalms 126:2). In the New Testament, Jesus says, “Blessed are you who weep now, for you shall laugh” (Luke 6:21).

I disagree with the thesis of this article from the Physicians Committee for Responsible Medicine, and I am curious to hear thoughts from others.
The article claims that the APCs in MDPI journals results in lack of scientific quality, that MDPI journals self cite, and that MDPI is predatory. The article focuses on Nutrients, but implicates the entire publisher as well. I tend to disagree with these claims based on my own experience with this publisher.
Regarding the self citation issue, I find the idea ridiculous. The authors are independent actors, the publisher has no hand in what the authors choose to cite. Further, the journals within the publisher are not a single entity, each journal has it's own editors and research focus. I don't see how it's an issue if authors from an article from 1 MDPI journal cite another if the research is relevant.
Regarding the quality and predatory claims. we have published plenty in MDPI , including in Nutrients, and the peer reviews were thorough from my personal experience. Additionally, I have reviewed at least 23 papers from MDPI. I am very thorough, and find that I reject 40% of the time and request major revisions the other 60%, trying to maintain strict quality control with respect to the methods and legitimacy of the results. My rejections are almost always taken into account, in one recent case the academic editor of IJMS (international journal of molecular sciences) rejected based on my review report alone (due to serious methodological flaws) without any other review reports being submitted (there are usually at least 3 reviewers per article for this journal). The times it's not immediately rejected during the first review report (due to discrepancy in recommendations from other reviewers), it is often rejected in later reports if the issues are not addressed. I assume there are bad experiences from others, but that's likely true with any journal and publisher.

Dear ResearchGate community,
I am currently conducting research on the role of human alpha-klotho in blood and cerebrospinal fluid (CSF), and I am looking for recommendations on the most suitable ELISA kit to use for measuring alpha-klotho levels in these matrices. I am aware that there are several commercially available kits, but I am interested in hearing from other researchers who have used these kits and their experiences. Specifically, I would appreciate suggestions on which kit has been the most reliable, sensitive, and specific in detecting human alpha-klotho in blood and CSF samples. Additionally, I would like to know which factors should be considered when choosing an ELISA kit for this purpose, such as the type of sample matrix, cross-reactivity with other proteins or substances, and assay format. Any insights or experiences related to measuring alpha-klotho levels in CSF samples would be especially valuable.
