I have used this repeater in the past and it's been my favorite: https://www.pipettes.com/brandtech-handystepr-705002-electronic-repeating-dispenser-110v-us-plug
It's easy to use and has the features that we need and none of the ones that we don't. I'd love to purchase this one again, but it's sadly been discontinued.
The manufacturer's replacement is this one: https://brandtech.com/product/new-handystep-touch/ I've used it in the past and frankly do not care for it. It has a lot of features that my lab does not use and I found the touchscreen's interface to be confusing.
I was also looking into this one: https://online-shop.eppendorf.us/US-en/Manual-Liquid-Handling-44563/Manual-Pipetting--Dispensing-44564/RepeaterE3-RepeaterE3x-PF-135444.html While this one seems easier to use than the current Brandtech repeater, it uses proprietary tips. Our lab actually has the repeater in the first link I posted, which uses these tips: https://brandtech.com/product/new-pd-tip-ii-dispenser-tips/ and I'd hate to have to order two different kinds of tips. I suppose it is possible that the Eppendorf repeater can use the Brand repeater tips, but we already have several boxes of the Brand tips.
Any help would be much appreciated!

Hi Zakary,
Which pipette did you end up buying? We are just considering of buying a repeating pipette for dispensing mastermix for a qpcr reaction (13 uL per well in 384 well plate). Would you have any advice on that?

Scientists know that the second law of thermodynamics is not consistent with experiments and intentionally conceal it.
  • Is the second law of thermodynamics correct? Just use experiments to explain it,
  • The formula for the second law of thermodynamics: dE/dV=(dP/dT)*T-P. , Measure E, P, By substituting this formula, we can determine whether the second law of thermodynamics is correct.
  • Under stricter conditions such as low temperature, high pressure, and critical point, theoretical predictions are relatively large, and experimental values will also be larger. Whether a theory is consistent with an experiment depends on the data.
  • In these cases, if the theory does not match the experiment, you can ask scientists who specialize in thermal properties. These phenomena that do not match the experiment can be concealed by scientists,
  • Scientists like to make technical arguments: there are fluctuations in the critical point, hydrogen bonds, strong interactions, and low temperatures.,, I hate this kind of technical justification the most.
  • Scientists like to talk about entropy without considering thermal properties. DE/dV=(dP/dT) * T-P does not match the experiment (fact), so entropy is meaningless.

Answer
First of all it has to be stated that the formula above is not the second law of thermodynamics, it is just the consequence of it. To add, it is obtained at some rude approximations, namely, assuming that everything goes from equilibrium to equilibrium state. This is entirely wrong for any practical system. On the contrary, the essence of 2nd law is pretty simple - when two bodies with diffrent temperatures are put in contact the heat flow will start from the one that is hotter to the one that is colder. If you observe in any correct experiment the opposite, immediately publish in Nature, Nobel prize will be guaranteed. Of course, you may face some problems with the authorities (as many scientists in the past), but some times afterwards scientific community will recognize your astonishing achievment, because nobody can argue against correctly made experiment.
Being serious I suggest you reading modern papers on thermodynamics that are much more rigorous and postulate up to 16 independent laws (see doi.org/10.1016/S0370-1573(98)00082-9), so that to correctly derive every mathematical expression without the omissions and flaws of the classical picture.

I just came across some answers to the question on whether students enjoy learning. Most answers are NO. And one answer is along the line that when learners feel engaged, then they would love to learn. That idea really resonates with how I feel about learning. When I was in school, I hated sitting in class and listening to things that didn't matter to me. Having to reach highscore due to parents' and peers' pressure still left such a trauma for me as being abusively forced to do things I found no meaning. Doing assignments and writing essays to be marked always sounded like a kind of torture to me. I didn't enjoy any of that. However, whenever I read something I am interested in, it feels so effortless and marvellous. Whenever I write about something that powerfully drives me to investigate, I feel like have more energy and my spirit is lifting.
So I reckon that being able to like or really want to do anything, including learning, that person must voluntarily find that activity meaningful, so much so that when they are involved in it, it naturally feels just like eating or breathing, without the need to "try to do it". "Like to learn or not" seems to be the question on whether you feel "in the flow" isn't it?
How do you suggest teachers and learners create that flow?

Thank you for the compliment. I am glad to have met a like-minded person in the field of pedagogical approaches. Speaking about existential pedagogy, I meant elements of pedagogical approach based on philosophical and psychological direction of existentialism. In this approach, the subconscious mind is the main target of the pedagogue's efforts; the appeal to the intellect and logic is relegated to the background. The foundations of existential pedagogy are quite diverse and have been developed in different countries by different researchers (Carol C. Gould, Breisach Ernst, William Barrett, Shoma Morita). Although I am not close to the excessively individualistic approach of existentialists to education, at the same time, the general sense and orientation of their actions in the field of pedagogy seem to me to be correct. And if we return to your initial question, then, yes, in my opinion, this approach can lead (not always, but in important key moments of learning) to the state of ‘flow’, at least in its classical definition by M.Csikszentmihalyi.

The theory of the Genesis of earthquakes has been created.
Based on this theory, the main fields, the main forces involved in the development of earthquakes, are determined.
The participation of the same fields in the creation of anomalies and in the creation of the earthquake itself ensures 100% reliability. The low speed of the wave that creates an earthquake ensures high accuracy in determining the location, start and end time, and the strength of a future earthquake.
This makes it possible to save people's lives.
This makes it possible to receive high income by reducing the losses of countries in earthquakes.
This gives sustainable growth through the development of the Global Earthquake Prediction Network.

Should we prohibit the false statements from the papers published in academia journals?
The words that “Einstein's famous equation E=mc2” is a sentence directly copied from internet. It is a general sentence about E=mc2. But, it is clear, this is a false statement. Now, it is well known that it is De Pretto who originally presented E=mc2 from the energy of atomic nuclei. His explanation about E=mc2 is the current explanation in today’s theory of relativity. And, Ohanian showed that in 7 papers, Einstein’s mathematics derivations about this equation are wrong.
And, these sentences are also false statement: 1) Einstein predicted the invariance of the speed of light or Einstein’s postulation of the invariance of the speed of light, 2) Einstein predicted the gravitational light bending or Einstein’s gravitational light bending.
(PDF) Anti-ethics and pseudoscience: On Albert Einstein's theory of relativity (V2) (researchgate.net)
these problems are introduced in detailed.
I was astonished at that, in the books or articles about the theory of relativity, this kinds of statement: Einstein predicted xxx, Einstein’s principle/postulation about xxx, Einstein obtained xxx, and so on, are all false statement. Could you find out one of such kinds of statement is true?
In the past 100 years, we obtained the maximum scientific innovations. These innovations are much more than the sum of that before 100 years. But, it is very unfortunate, the development of theoretical physics is stopped or slowed. Some people think that “Physics is dead”. Here we emphasize, the main reason for the death of physics is that the false statements are prevailing in the papers and books published in the famous academia journals.
We think, we are all the honest people. We don’t like or hate lie and false statement. Therefore, could we reject the false statement in scientific papers with the journals, magazines, books and so on that publish the false statements?
How can we do?

Fascinating question. In the current middle eastern political climate, do Palestinins need Israelis and do Israelis need Palestinians since they deny each other the most. My response will be yes, they need each other the most. I think denial comes from lack of knowledge, fear of rejection, and lack of exposure to each others' way of life. Edward Said, when describing othering groups of people as a means of conquest, and Fanon, describing the oppressed becoming the oppressor are describing current middle eastern societies. Perhaps we deny the entity the most, because of how desirable it appears to us.

Do you still believe that forgetting someone's Signature is a crime in the USA?
Fifth Press Release – Third Crime Report
December 11, 2023
Documentary Books series: Collective Injustice (Wells Fargo)
Fourth Book -- Wells Fargo: a symbol of denial and irresponsibility
Second Crime Report Title: Wells Bank Involvement in Illegal Affairs: Forged Signature at Wells Fargo
Please read the story at the end of this Crime Report.
Who Involved: Wells Fargo Bank Illegality, Ex-Wife, Family Court, Lawyers
Dates: During 2012
Discovered in 2018 by pure chance.
Damage and Impacts: A bad reputation with the bank, loss of time and money, and effort with adverse effects are catastrophic for me, my business, and my health. The damage and impacts are still continuous.
Legal and Principal Parties
1) My ex-wife, Madam Raefa Mostafa Al-Mehelmy, whom I divorced in Egypt in January 2018 and has been separated from in an unfair California family court for almost six years now.
2) Wells Fargo, in which all my personal and commercial accounts are in which all bank transactions
3) The family court is very unfair and harsh in dealing with American citizens of Arab or Islamic origin
4) Notorious credit bureaus Sub-legal parties
Sub Legal Parties:
2 Lawyers in California and 3 Lawyers in Egypt
In one of my visits to the bank at the beginning of 2012، a deputy Waverly Branch OF WELLS FARGO, PALO ALTO, I opened a Visa Card account because I am one of the excellent customers in the bank. I told her I agree.
She said - We will know in just one minute. The response came with a refusal. We were surprised. I asked her to look for the reasons. Of course, the bank refused to know what the real reasons were against the bank's refusal and the intervention of the head of the branch, and with my strong insistence on knowing the reason for this very harsh refusal. Although I accepted that refusal, I learned from my life in America that refusal occurs in a semi-normal way with American citizens of Islamic and African origins as forms of legalized racism in most cases, unfortunately.
The vice president was in a challenging situation, and she referred me to another person in the bank to look behind the bank's denial. And in a very short time after, he drew himself and his arrogance on him. I saw a document with signatures on it. And he asked me with rudeness and false arrogance - are these your signatures? I told him: The signatures are not my Signature. I almost exploded from rage and held firm. He let go of the false arrogance and said to me in a moderate voice - Can you sign your signatures on a piece of paper so that I may recognize your Signature? I told him: The bank has my Signature. I said it to God alone and drew my Signature. He adjusted and said I am very sorry because you have an old problem with the bank. They opened two accounts for you and your wife a long time ago (my ex-wife was in Egypt in January 2018), and they have signatures other than your signatures in your house, which do not match your signatures now as well, and I am not able to give you the documents. I erupted, became mad, and asked the police to visit the bank. The bank's vice president realized the situation was not in the bank's favor.
I mean, I have been living with a filthy demon since 2012. No morals. There is no creed, no origin, and all my thoughts are on my children and grandchildren after me, God willing. And I screamed at the top of my voice, and my heart cried and told them that this is a significant crime as we know it in America.
I tried to get the documents from the vice president of the bank, who spoke urgently with the central bank and told them that I insisted on calling the police to prove the incident. I took a copy of it with the accounts with a forged signature on it and asked her what had happened with the bank employee. She said the bank fired her - I took the documents and left, sadness and sorrow fill my heart, and the rest will come, God willing, soon.
God suffices me, and He is the best agent.

Chapter 11: A Forged Signature
"The husband is the last one to know" is a French proverb. Although I am of Egyptian origin and live in America, this proverb applies. I say this because I only learned what my wife (now my ex-wife) did in 2012 by pure coincidence in 2018. Raefa Mustafa Al-Mehelmy was divorced in Egypt and is still separated from me before the "unfair" California Family Court. On one of my visits to Wells Fargo Bank, which contains all my personal and commercial accounts, at the Palo Alto branch, the Vice President of the Bank spoke to me about opening a Visa Card account, especially since I am one of the privileged customers. I agreed, and we began the usual procedures. Then, the response was confronted by a refusal. I asked her to let me know the reasons for the denial, but the bank management refused to disclose the cause. The head of the branch intervened, but it was in vain. I insist on finding out the reasons and opinions for this unfair refusal, despite my prior knowledge of the rejection caused by racism that was taking place against Americans of African and Islamic origins. Because of hateful and legalized racism, the bank's vice president is in an unenviable position. She referred me to another employee to discuss the reasons behind this unjustified refusal. In his office, I saw a document in front of him with many signatures. He tried to show off himself with arrogance.
Showing a grudge and malice, he asked," Do these signatures belong to you?" I categorically denied it and said that my signatures were forged. He asked me after he had given up his arrogance and spoke moderately:" If you, please put your Signature approved by the bank on this paper?" I told him that they had my authorized Signature already. However, in the end, I submitted to his request and put my Signature on a separate paper. After that, he said, "You have an old problem with the bank because of opening two accounts for you and your wife (my ex-wife now). We have papers with signatures that do not match your Signature." He pointed with his hand to the paper on which I drew my Signature. I tried to get a copy of these papers, but he refused vehemently, which forced me to break my calm, raged, and went crazy. I demanded that the police come to the bank. The bank's vice president sensed the situation's seriousness, and it was not in the bank's interest. Therefore, he acquiesced and gave me a copy of the documents with the forged signatures. I took the papers and left with a worn-out heart squeezed by grief and despair. What lousy luck put in my path this devil whom I honored by marrying and raising her family's status and hers, my ex-wife, Raefa Mustafa Al-Mehelmy. That woman has no morals, belief, origin, or loyalty to a man who does not fault that he trusted her and entrusted her with his money, house, and children.

In my opinion, academic conferences (ACs) are (very) necessary in the tertiary institutions of higher learning, as it creates a platform for knowledge sharing and exchange. Scholars from all walks of life dare to attend because, they believe they will gain useful "something." Sadly, these conferences run for only two or three (maximum) days: a duration some scholars and institutions may find unnecessary to spend a lot of money for.
If the impact of AC is to network for further collaboration, look for external examiners for your faculty/department/students, peer review of pedagogical practices for the sake of standardisation, then 2/3 days are just not good enough.
Therefore, I think/suggest that:
  • AC should have minimum of 5/7 days, whereby, experts in their various fields can be invited to come and share their experiences and how they made it, new trends and current research areas that can advance our lives.
  • Experts in analytical software that are more appropriate for the chosen field to come and empower scholars in that discipline (I hate giving my work to someone to analyse for him to tell me the outcome). When you analyse yourself, you gain more insights that can lead to multiply knowledge creation.
  • The suggested duration can help to achieve the listed impacts in the second paragraph.
Who can afford not to attend a conference of such benefits? and who will dare to say that such a conference is of little benefit?
I invite comments!

Relevant answer
Answer

The average American has always had a love-hate relationship with the IRS-with hate predominating. The so-called Exit tax kicks in the moment you think of expatriating from the U.S. The current maximum capital gains rate is 23.8%, which includes the 20% capital gains tax and the 3.8% net investment income tax. Why its unfair? Currently, federal income tax brackets range from 10% to 37%. Nearly half of the American population pays taxes and in fact, the top 50% account for nearly the entire federal income tax. So the question of levying an exit tax on Americans relinquishing their citizenship really means I am going to get what I can from you before you make a break for it. In other words, the IRS ensures that the relinquishing citizen would never feel grateful for giving and doing his bit for the great country. The current threshold is so less ($12,550 and $28,500.) that almost everyone who holds a reasonable job pays his federal taxes by filing an income tax return each year. Then consider that inflation in the millennium year was around 3.3% whereas today it's over 8%, so where has real income disappeared? Then when someone decides to head west (after paying all his legal dues of a lifetime), IRS slaps him or her with the Exit Tax. Hence the question.
Relevant answer
Answer
Not everyone who wants to leave the USA or gives up their citizenship is subject to the exit tax; there is a relatively high wealth allowance. Incidentally, this tax is well founded in terms of content and can therefore be understood as quite appropriate. See the following link for details:

A question about models of recall and recognition based upon personal experience:
I have had the experience of not being able to recall a specific words (I hate growing older ;) ), but still can pull up enough context to do a search for the word. But here has been my experience--before I begin to scan the results of the search I suddenly am able to recall the word; it is almost as if the mere act of beginning the scan there is some level of 'pre-activation' that enables me to recall, but it feels as a gray area between recall and recognition. Now, I have not been active in cognitive psychology for a while, but my quick research review has not provided insight. So for those cognitive psychologists out there, including developmental psychologists, as it seems to be there are developmental processes that must be at play here, is there a body of specific to these processes? I have a single-cell clones from a CRISPR cell line that have become contaminated. I would like to not have to re-generate the line and instead recover what we have. They are cultured in DMEM with 10% FBS and 1% Pen/Strep. I have also added Normocin as well, but still have some contamination issues leading to poor cell health. I think I may have identified the source of the contamination, butI hate to loose the 6 months of time it took to get these cells. I also recognize that I might need to start over, but was hoping that there might be some tricks of the trade out there that we could try first.

Answer
Hi Kanwal,
I subcultured my cells into two 35mm dishes. One dish I changed the media every day for a week and the other dish I changed every other day. The cells I was using did not like the daily media changes. When I made the change I alternated DMEM with 10% FBS and psg (complete media) without normocin and complete media with Normocin. I had some recovery but I never quite got rid of the contamination. When I pelleted the cells and washed the pellet with 1xPBS 2-3times, centrifuging in-between to completely remove the trypsin , I had better results. What I thought was contamination may have been a result of incomplete removal of trypsin. I also realized thatch cell health issues could be due to mycoplasma. Normycin will help prevent mycoplasma and maybe even treat low level mycoplasma, but it does not completely eradicate it. Unfortunately I did not have a way to test for mycoplasm in my lab to confirm it, as I was working remotely in another lab. I did not treat for it since a lot of those treatments can be harmful to the cells over time if you are not careful. The lab where they came from has had issues with mycoplasm. I hope my experience helps you to navigate yours.

Hello, I am currently working on finding antecedents of brand hate. I have put forth a construct that leads to brand hate. So, the antecedent which leads to brand hate has three different dimensions (Political, Economic, and Military). And brand hate is made up of three different emotions (Anger, Disgust, and Contempt).
Independent Variables (Antecedent)
- Treatment 1: Political
- Treatment 2: Economic
- Treatment 3: Military
- Treatment 4: Control
Dependent variable (Brand Hate)
- Anger
- Disgust
- Contempt
We are going to develop three different essays (scenario) that will represent each of the independent variables. And there will be a survey that will be filled by the participants after reading the scenario. Three different treatments will be given to different groups.
I would like to know the following:
Type of Experimental design?
Population?
Sample size?
Hypotheses type: Directional or Nondirectional?
Data collection: Random, Nonrandom? (the study will distribute the survey online)
Analysis?
And that whether if there is a need for a control group or not.
Thank you in advance, if there is any information which you believe is missing please message me and I would provide it. It would be very helpful if someone with experience in experimental research could message and guide me through this.
Thank you

This is the best explanation. Between-group is going to work the best in my option. I have already started my work with Between-group design where i have 3 conditions (treatment groups) from animosity dimensions and the dependent variable is brand hate. And for the outcomes brand hate becomes a mediating variable for animosity.

The question is chaotic yet very relevant. Why would a country hate its own government?. A good example to note from the elections of May 2020 where countries in Europe send their representatives to EU. A number of far right parties and others who didn't believe in the EU principles got a large share of votes. Countries like Greece, Moldova and Bulgaria have among the least government popularity percent's. There are others too, outside of EU-Saudi Arabia for one, Pakistan , Israel, Jamaica, and even PRC (though they haver no elections). Its important that the governments woke up to the fact that people ultimately determine the direction of the governments-and its people who will ultimately decide how can government stay in power-with or without the use of force.
Relevant answer
Answer
Its true; in true democracy, people are powerful and can topple the government through election. In dictatorship, it is the other way around. The controlling junta prefers the pursuit of their own interest over the needs of the ruled society.

The reason for starting this discussion is a very arrogant, abusive, hateful (and of course anonymous) review that we received for our manuscript, submitted in Arachnology https://bioone.org/journals/arachnology lately. Almost all of the reviewer’s comments started with phrases like: very boring; useless; non-essential; a fairy tale as for example “Alice in Wonderland”; limited experience in spider taxonomy; very limited knowledge about spider taxonomy by the authors, or even “preoccupied with nationalism” ??? Apart from us, the reviewer abuses also some other authors that we cited in our manuscript.
The point here is not about positive or negative review, but for the ethics in peer reviewing and the language of the reviewer. And my question is should the editors accept such nonsense as a valid review and send it to the authors at all? In many high impact Journals like https://plos.org/resource/ethics-for-peer-reviewers/ , there are reviewer’s guidelines and respectful and professional language is explicitly required. Although such guidelines are missing in Arachnology, or at least I couldn’t find them, I still think that such hateful remarks should not be accepted by the editor. It’s interesting to hear the colleagues’ opinions about this.

Dear Friends,
It is not acceptable to repeat mistakes committed in the dark ages even in 21st century. Isn’t it a failure of the system, if such mistakes were committed with impunity? Many great researchers such as Giordano Bruno and Galileo were hated, humiliated, prosecuted, imprisoned or even killed, when they tried to expose flawed 2300-year-old basic presumption (the earth is at the center) in the very foundation of then dominant paradigm (known as geocentric paradigm).
The pre-paradigmatic foundation for the dominant paradigm existed in 16th century was created more than 1800 years ago, which comprised of unproven basic 2300-year-old presumption such as “the Earth is static at the center”. It is a basic presumption, since large chunks of know lodge, observations or evidence in the mankind’s BoK (Body of Knowledge) until 16th century had been acquired and accumulated by relying on the unproven flawed presumption.
Is it acceptable to hate, denounce or persecute anyone for tiring to expose such unproven flawed presumptions in the pre-paradigmatic foundation of any exiting dominant paradigm for any scientific or technological discipline that is playing vital role and widely used in 21st century? Is it acceptable, the research community acts or behaves no differently that the researchers, who tried every thing in their power to suppress and sabotage the efforts of anyone trying to expose such flawed unproven basic presumptions in the pre-paradigmatic foundation of the dominant paradigm?
Any scientific or technological discipline end up being a fake or pseudo science, if its pre-paradigmatic foundation comprises of flawed presumptions, since it is impossible to create real hard science by relying on flawed presumptions. Any scientific discipline end up being a paradox, if the BoK for the discipline has been acquired and accumulated by relying on flawed presumptions.
The pre-paradigmatic foundation for software and computer science created more than 50 years ago comprising of unproven flawed presumptions and large chunks of knowledge, observations and evidence for existing dominant paradigm for software engineering has been acquired and accumulated by relying on the flawed presumptions in the pre-paradigmatic foundation.
No technological problem can ever be solved by relying on flawed or invalid knowledge relevant to the problem. Hence, certain large problems in software stood unsolved for decades. Isn’t pathetic, if the research community can’t understand this simple logic that it is impossible to acquire relevant valid knowledge by relying of flawed basic presumptions?
Many software researchers argue computer science is not a science. I agree, since it is impossible to acquire and accumulate valid scientific knowledge by relying on flawed presumptions. Even the basic science end up as a paradox (i.e. geocentric paradox) in 16th century, since it is rooted in flawed presumption. The basic science was transformed by exposing the flawed presumption in its pre-paradigmatic foundation.

Raju

Aztecism holds when a society bets its existence on a commonly held belief which entails this society’s own annihilation – as the old Aztecs did.
This statement belongs into the field of the professional historians. However, it in addition also possesses an uncanny actuality to date. For all of us – the whole world – have ever since 1929 learned to live with the scientific conviction that there once existed a Big Bang and that the universe is still young and expanding to date. All high school kids and all newspaper readers across the world share in this belief which got reinforced by some ten Nobel medals over the decades. To place doubt in their conviction automatically raises pitiful eyebrows.
This self-righteous common belief is, of course, based on a majority consensus maintained for 90 years. It could in principle still turn out to be mistaken, but such a case has never occurred in modern history before.
“Do you really place doubt in a planet-wide scientific consensus acquired by the best minds and the most prestigious societies over 9 decades?” is the question. “If so, something is bound to be ticking wrongly with you. Presumably, the Moon landings are also put in question by you,” so every pupil is ready to laugh out loud.
The notion of Aztecism stands for an irrational conviction held by a whole society. So much more for the worse if I now confess to the reader that I do adhere to the outrageous conviction of Zwickyism.
May I elaborate? I begin by admitting that I am the first author on quite a few unchallenged scientific publications which claim to prove that maverick astronomer Fritz Zwicky was right in 1929 with his ridiculed so-called “tired light theory” of the cosmos. The whole world is chuckling because it appears highly unlikely that we all live in an Aztec society for 90 years – or do we?
The kids who follow me can stop reading here. What I will continue to say next is only of interest to adults who have experience with how to deal with ingrained ideologists. In other words: this is a case for the psychologist?
Unless it deserves to be shreddered, the present text presents a very disturbing message. We all have heard the story of the ghost driver who complains about the flood of ghost drivers encountered by him. On the other hand, this is also the secret dream of every scientist: to for once come into a situation in which you determine the direction of further scientific progress. Provided, of course, that you are not deluded. Therefore the above scenario is a very rare event to reckon with in reality – much too rare for it to occur in a lifetime, right?
But the unlikely does sometimes apply. The famous science of Thermodynamics from more than a century ago does not stand alone anymore for a few years, for example. It has acquired a sister science which had been overlooked for more than a century, called Cryodynamics. Cryós in Greek means cold, thermós means hot. A side effect of the existence of the new science is that there was no Big Bang, as this was first seen by Zwicky in 1929.
“But if you lean yourself as far out of the window as you just did,” so you will reply, “then further implications of a sensational character are bound to be announced by you in a minute?” Yes Sir, do I respond meekly, there is especially one point: “No CERN safety for earth anymore.” The for 11 ½ years non-renewed official planetary safety report LSAG of CERN’s comes to everyone’s mind here as a historical fact.
After my having just made this accusation, you will understand why I have for a decade become an outlaw in the scientific community, one who is unworthy even of the benefit of the doubt. Imagine: to claim that ten thousand living scientists (at CERN) are deluded in sync along with 9 decades of mainstream science – this is an absolutely ridiculous claim of mine, is it not? And to proclaim in addition that this new view puts the continued existence of earth at risk if it is not addressed, is bound to be a case of irresponsible fear mongering. Although this is exactly what a today unknown crackpot once tried to convince his fellow Aztecs of.
I sympathize with your unavoidable logical skepticism: Cryodynamics clearly is “too big a jump” to be easily absorbed by the community. I would nonetheless be very glad if there were a few high school students on earth ready to give me the benefit of the doubt, Greta Thunberg included. So far, the very existence of her movement points to the existence of a subconscious connection between my a decade old results and the heroic Friday movement of today.
You have now seen my worried soul, my dear adult reader. Please, do not hate me too much for my driving my lonely scientific car in an allegedly non-ghostly direction. And forgive me if I love your children more than I see you do in case I am not mistaken.

During electrophoresis, I observed a lagging read band on my gel. I have done many Westerns and have never encountered this before. I spoke with my PI and some others and they haven't observed it either. The only difference between this run and what I have done in the past, is the use of a 4x LDS Sample Buffer from a manufacture that we do not typically use. I contacted the manufacturer and was told that this is most likely from an issue with their product.
Has anyone encountered this before? If so, how did it alter your transfer and quantification?
This will not be used for publication. However, I would like to know if it would be possible to continue on so I can get some preliminary data. I hate the idea of wasting so much tissue as I had prepped lysates for several gels.

Hi Chris,
Laemmli buffer usually Bromophenol blue as a dye. The dye is sensitive to pH, so you might have some pH-effect (e.g. if you re-used running buffer) and differently charged species of the dye.
An alternative explanation might be that the Bromophenol blue used by the manufacturer was not that pure and contained isomers or side products from the synthesis.
If the membrane looks good in Ponceau staining, I would not worry and proceed with the antibody incubation...
Good luck,

I am suspending a pre-labeled lyophilized Amyloid-beta[1-42] in DPBS -Ca/-Mg. I have this now in 37degree incubator to aggregate.
The protocol I am following has further pelleting and rinse steps following the 48hr aggregation/incubation. The protocol is designed for 5ml tubes and says to pellet at 14,000rpm.
I do not know what rotor they used to do this, so I do not know the actual force. I hate it when people put rpm in a protocol and don't say what rotor.
Does anybody have an idea at what force I should do these spins? The reagent is pretty expensive and I am already late on this. I do not want to spin way too hard or way to easily.
Relevant answer
Answer
Sounds like a clinical centrifuge (Hettich, Eppendorf etc.). Their maximum rate typically is 14000 rpm, equivalent to about 20000-25000 g, depending on rotor geometry. Apparently, this centrifugation step is not too critical ;-)

I was threaten by a French guy (not on this site) who kindly advise me not to play with fire, revelations from 16th C books could harm a lot of people. And Algerians were not ready to read their own history in Topography and General History of Algiers. So I'd rather spent my time educating them.
It's just too much for me, this kind of philosophy still prospering nowadays in 2018. As a matter of a fact, it's the French government's position regarding francophones: educate Africa. Vulgar and pretentious.
We all know how good French did the job. The book i'm translating, Topography and General History of Algiers, was translated in 1870. They forget the introduction, the publication license, the marginalias and took away a lot of details, transforming it in a "non-sense History". After that, they forced Algerian to remember how chaotic and illogical was their story at mandatory school, provoking hate, despair and discouragement among Algerians towards their own History. Now, 50 years after the Liberation, little by little, they are recovering and History is turning a passion in the country.
Do you think knowledge can be harmful?

Most of us speak fluently 3 or more languages. But do you realize in which language you're thinking? In which language do you talk to yourself? Personally, English is a great way to clarify and expose things; I also principally swear in English (because Spaniards don't understand it). Spanish is to me the only philosophical language, with two verbs for to be, but also the feelings language, the language that best fits to unpredictable things, the only language that evokes properly magic. French can be reduced to what makes it a "diplomatic language": the fabulous litote. When, in a French classical play, Chimene says don Joan: "Go, i don't hate you", all the French understand: "I'm crazy in love with you". How to signify the more saying the less. Apart from diplomacy, not very satisfying... So, in which language do you think?
Relevant answer
Answer
I think that each language represents a vision of the world and a specific way to express it. Nevertheless, experiments can be done. Spanish to express life can be adapted to English, which is bent to accept all influences. French is a jealous exclusive language and most French ain't that good at languages and claim French should be the international language. Catalan had been going through an horrible "normalization" that cut off the innate poetry of the language, to show how modern they are and, at Catalan school, they don't teach you Catalan authors (because none is respecting the stupid unlinguistic laws) but supermarket lists (supermarkets respect the stupid "Normalization"). I love Italian, understand almost everything and feel bent to it, like a natural thing that correspond to my way of being but I don't speak it in order to preserve my Spanish. I can also read Greek and Russian (I know cyrillic alphabet) and understand them basically by etymology (f.e. plants vocabulary). It's been a long time I've been interested in ideograms, but I never found time enough to seriously study Chinese. But maybe, because of my historical research, I'll have to study Arab, which is a language I feel bent to. I worked with some friend, that did the literate translation from Arab to French. My job was to find the poetic aspect and find out how it influence the meaning. I think nowadays Arab language isn't very well known, even if it's diversely spoken in many countries because poetry definitively escapes to capitalism.

I need to know about Shakespeare's opinion about women because I am interested in feminism and I want to know on which side Shakespeare was ?
Relevant answer
Answer
For me, Shakespeare's world did not inhabit any such bias, he dealt with specific social, human, interpersonal and personal problems and these loomed large; their outworking in his works was also complex. In most of his works, it was clear that men and women were villains, and he so depicted them, Macbeth, Anthony and Cleopatra etc. The biases in his works were the social biases of his day and his portrayals particularly because they were based mainly on the ruling elite's life conflicts depicted the social order and cultural environment of his time as they were. He queried a few hegemonies though (The Tempest). He was not a utopian artist and as such grounded his works on the existent social relations of his day.

Hello,
We are told that when it is hard to make a stable contact on a thin film while performing a IV-characteristic measurement, for example, sandwiching a small, flat sheet of indium between the thin film and the metal contact tip helps. However, I don't think I have a good physical explanation as to why it helps.
And, more importantly, I remember one of my PIs hated using indium, and instructed me to avoid it as much as possible. Is there any situation where it is not advisable to use indium? All I know is that the indium is put in series with the sample, and since the resistance is less than 1 ohm, the effect is negligible.

Though this can be perceived to be philosophical in retrospect, it is a valid behavioral question.
I contend that hate is more dominant. In observational research, not sanctioned, just in the world of social media, it seems that even the most benign of posts, say a yellow kitten, can become fodder for hateful attacks against others.
Is it that we live in the world of anonymous responses, or is it that hate is truly the more dominant emotion?
I am considering my dissertation topic, and believe Love versus Hate in social media would be a worthy topic for research.
Relevant answer
Answer
Consider: Are hate and love emotions? Are negative responses to posts on social media hate or are they merely temporary negative reactions? To what extend can you conceptualize a variety of different definitions for both love and hate? How can you measure someone's love or hate in a validated way? How can you account for the positive responses that may far out number the negative responses?
Just some questions I think you should consider. I think you should consider phrasing your dissertation around positive and negative responses instead of love and hate. Love and hate are accompanied by action where as these responses online are temporary and are typically not accompanied by action. Interesting topic! I look forward to hearing what else you come up with.

I guess that anyone faces at least unjustified hate from a hater in his/her life. IMHO, haters are weak, and psychologically ill people. Probably, the best way to deal with these personalities is to avoid them; however, in some cases avoiding them is impossible. Therefore, I am asking about the best scientific approach to deal with them provided that the violence option is excluded.

My tactic is to be extremely logical, and make it evident the 'hater' (I prefer using the term 'envious') is not logical. If the 'envious' are psychotic there is not much you can do to change them, since anything you do is 'wrong', just show yourself more intellectually logical so others disqualify their opinion and respect yours.

If I may,
the narrative is not relevant as a series of ideas, those ideas change,
especially given Trump's flip flopping. The narrative is based on groupthink around a personnality (trump) and leaders (pro trump figures, some of the republican leadership) though the identity itself is built, on opposition to mainstream elites, within the right wing. The most effective counter narrative would be to challenge the validity of the basis of the group or the narrative, to destabilize the current ideological bubble. Either showing that elites are just people that should not be opposed as a group, or to show that Trump is not deviating from the mainstream. That would be limited by the fact that the counter narrative cannot be deployed by people who would have interest in lying and can still be dismissed as a democrat plot. Ultimately, the only way to properly engage such a narrative is first on an individual basis, where group effects can be minimized, and then to put such objective and unquestionable points (to a Trump supporter not a liberal) that they would be listened to even if they are opposite to what the group's narrative is.

Created in 1972, when data digitization was non-existent, to help librarians choose the most relevant journals for users of scientific collections, impact factor became a competency factor in the 1990s. allowing to evaluate authors quality. Thus, in some countries, researchers are given premiums proportional to the impact factors of the journals in which they publish.
However, recently, some American journals have eliminated any reference to impact factor in their websites. In addition, The "Science" journal cited in its opening pages an article written by eight leading scholarly publishers with the title "Hate journal impact factors? New study gives you one more reason". The "Nature" journal has gone further: with this title: "Beat it, impact factor!".
Relevant answer
Answer
Does the impact factor realy provide good information on researchers quality work of ?
According to Lariviere et al. (2016), "This measure creates more confusion about the quality of scientific research than it illuminates, it should be abandoned completely". These authors wish that scientific community understand the limits of impact factor in research evaluation and they add that "the impact factor is highly influenced by a small number of widely cited articles". Thus, of the hundreds of articles published annually by the journals Nature (IF equal to 38.1) and Science (IF equal to 34.7), only about forty articles correspond to these averages. Larivière et al. (2016) calculated that citations from three-quarters of articles in 11 major journals in recent years are below the impact factor value. In other words, less than one in four items gets at least the average value. However, "The average is a relevant index when the data vary little and follow a normal distribution .... In the case of scientific articles, the majority are cited below the median and a minority of articles are highly cited, which makes the average very unrepresentative of articles individually.
Thus, "The unit of measurement must be the article itself and not the journal".
