Because when I count the %viability I get more than 100% and upon consulting my supervisor, she told me to normalize the data first.

I would recommend you to use alamarBlue dye to measure cell viability as it is completely non-toxic and does not interfere with mitochondria as MTT.

2. Write an outline of your proposal

3. Write a proposal introduction

5. Sign off with a summary and call to action
Before you finish your consulting proposal, it's important to include a summary and any relevant facts and figures, such as estimates of costs, budget, schedule and other significant figures. End your proposal on a call to action that specifies what next steps a potential client can take if they're interested in your proposal. In this section, you could encourage clients to reach out to you if they have any questions and include clear contact details. You can also include a brief note of thanks to the client for reading your proposal.
6. Send your proposal to the client

Hi,
I'm using the Malvern Zetasizer to measure size and Zeta potential of calcium carbonate particles. I expect them to be in the order of microns, and so I followed the manual, saying I should make a solution of around 0.1 mg/mL for effective measurement.
I am checking for the effect of pH and Ca-ion concentration on size and Zeta potential. Size distribution measurements seem to work correctly. But when I use a clean folded capillary cell to measure the Zeta potential, I keep getting peaks at 0 +/- 4 mV. Another student experienced the same with this device. Even a standard supplied by Malvern (which is supposed to be at -68 mV) produces a zero value.
I am about to consult with the apparatus supervisor here, but at this point I am just wondering if anyone else here has experienced the same phenomenon, and if so, if you managed to resolve the issue?

Dear Alan F Rawle , I thought since my question sounds exactly the same, my solution would also belong here.
I did contact the support - and now the issue seems to be resolved! In fact, the reason was pretty simple: loose contact between the cell electrodes and the device (more precisely, the electrodes on the cell were not pushing the electrodes inside the measurement chamber fully against the walls - even though it didn't look loose). According to the support agent's advise, I bent the electrodes on the cell a little and came up with some tricks to insert the cell inside.
I can't say my measurements are now 100% clean and reliable, but at least I have some signal.
John Francis Miller , I see your point. This must be one of the reasons you avoid the disposable DTS1060 cells...

I saw that there are PhD about it, but I'm not sure to be qualified for them.
How could I increase the chances of being admitted to one without pursuing another master?
I'm finishing a double master in plant sciences and plant biotechnology at Wageningen University & Research. My specialisations are functional plant genomics and phytopathology and entomology, with a minor in nematology and one still to define (around biophysics and omics).
I did a thesis in nematology at the ETH Zurich, one in plant molecular biology at INRAE (French national institute for Agro-Environmental research) and one still to define at SLU (Swedish Agricultural University).
I'd like to pivot toward policy making, but I only have a few courses about transversal skills such as "Negotiation skills", "Communication & Persuasion", "Scientific Writing" etc. and none about policy itself.

Great suggestion by Sally Elkady . I would suggest that you get into a PhD program focusing on Transdisciplinary research which straddles both policy-making and Biology. You have studied Phytobiology ( plant ecology), entomology, and genomics. I think it is a great combo to explore the impact of GM seeds and variants on friendly insects and plant ecology. This is exactly what is needed to influence policy. Your research comes would be suggested policy direction or changes that would ensure that ecology and friendly insects are protected for sustainable farming. Hope this helps.

About an year ago, I intended to submit my article in International Conference on Advanced Learning Technologies (ICALT 2017). I Submitted the paper and got acceptance letter within a week. Despite being a young researcher I became curious about the originality of the conference. I consulted with my supervisor who told me about the fake Organization (WASET) which was replicating the original conferences with the same name as of famous conferences. Then I stopped any further conversation with the ICALT 2017, the fake one without submitting the conference fee and submitted the paper in Original conference. Today, I encountered that my article was published and was available on the web under WASET organization. I intend to submit the full version of the same paper to well reputed journal but curious to know the following points.
1. Can I withdraw the article published under WASET
2. Can I submit the full version of the same paper to an indexed journal.
3. If I can not submit this article to an original conference or journal what could I do with my hard earned research work which has been lost ?

Tlhere is good research in WASET periods, as well as bad work. In reputable magazines, sometimes there are bad works. The person meeting someone from the magazine may overlook bad work. That way, a reputable magazine can contain bad work. On the other hand, predatory magazines can contain good work.

In Research Methods, the focus is on the traditional research process and the methods used. In the consulting process, one of the major concerns is the need of the stakeholders- the clients. Usually, the method used is that of action research as differentiated from traditional research. You need to differentiate too the role of the management consultant from the organization development consultant. The process is different since there is entry and contracting, diagnosis and feedback, implementation of interventions, evaluation and institutionalizing the change effort. Consulting can be a one time thing focused or limited to a specific intervention, but the OD work is concerned with the whole process.

There is no doubt that determining criteria is important, and also their relative importance
Importance for what?
To select alternatives? No.
They are used to establish balances between criteria, determining gains and losses, and nothing else, therefore, they are not useful to evaluate alternatives
The only weights that are useful are those that measure the intrinsic discrimination in each criterion, such as Entropy and Standard Deviation

Dear Alex,
My point was that such a question could not really be answered precisely. Ultimately, it is up to a negotiation between the consultant and the contractor. I have done very little consulting in the health or business world, so I don't really have an answer for you. However, I suggest this for people starting out. That is to ask the contractor is they have a standard range they pay. After one gains some experience, they learn a reasonable range to ask.

I want to thank those that request copies of my publications and many people have written me asking for them. It is an honor and a satisfaction that people think that they can help
However, those publications are books or chapters of books or articles in a journal, and are protected by copyright, and if I make copies I would be violating my contracts with the publishers.
For this reason, please don't ask me for copies since I am forbidden by contract to do that.
I suggest consulting your university library, there are chances that some of them are there.
Until now, and for years, I have answering each requester, but I will start discontinuing this procedure.

Hope you all doing great...
I was doing some experiments in lab and found a descrepancy of an estimated 15-20% which to my surprise shouldnt have been there and finally after consulting my friends and colleagues i got the equipment cleaned and placed at a location where it was more stable causing less vibrations (Did do the calibrations myself) and what i had presumed was absolutely right. We sometime go on with the flow and dont give improtance to the error inclusion that might creep in due ti variius reasons. the point of discussiing this is to know if some of you have encountered the same problem and how did you rectify it. this surely is one area where we need to b vigilant enough esp in large labs where maintenance ia done once a year for bulky machines etc..

Of course you need to calibrate your equipment and perform good maintenance. Like the pipetting error in the old days, there will always be an analyser error nowadays, mainly bias for wrong settings but also imprecision due to wear.
What kind of equipment / kind of lab are you dealing with? Best option is to perform regular (time interval dependent on the purpose) both internal and external quality control and keep your equipment within the specs your manufacturer and your professional organisation have laid down

If you have data on the location of the trees which you inventoried (and I mean precise geo-coordinates), then simply download VHR imagery (or consult with your project partners about acquiring UAV imagery for your area of interest, photogrammetrically pre-process it to extract an Orthomosaic) of your area of interest. This could be used for your OBIA to extract tree CPA which you can then match to your tree height dataset using their locational information. Once you have these matched, you can then proceed to using a species-specific allometric model ( you can extract this from the GlobAllomeTree website). You can even model the DBH of your current trees out, extract their wood density while at it and lots more. It should be very interesting but if you encounter further problems or need more technical advice you can contact me personally on this platform. Best of luck.

Hi, I have a quick question. My variable (for which i have collected some data through surveys) is called importance of x factor in consulting success and is ranked between 1-5.
Suppose, I want to prove a hypothesis that the importance of factor x in consulting success is greater than 3 (or the mean is greater than 3). Therefore, I want to reject the hypothesis that the importance of factor x in consulting success is less than or equal to 3. For this kind of analysis, should I just use the mean value to prove or reject the hypothesis, like if the mean is 4, i say it is proven that the importance of x factor is indeed greater than 3? or is there some other method I can use like for instance a one-tailed one sample t-test? would this test be relevant in my case? Hi, so I'm doing some final analysis for my thesis and the basis of my thesis is that there are certain crucial factors that need to be present for success in consulting. The only data that I have is the importance of these crucial factors in consulting success on a scale of 1-5, so only independent variables and no proper dependent variable.
My question is, can I make a hypothesis statement such as "The importance of factor x in consulting success, on average, is at least a 4 out of a 1-5 scale" or ""The importance of factor x in consulting success, on average, is at least very important." I say "very important" because on my scale, very important means a 4. Can I use this kind of a hypothesis? and this accept or reject it based on the mean value?
So if the mean is at least a 4, I accept it. Based on my work and the data that I have, I feel this to be the only solution. Thanks.

@Timira, thank you for your answer. So, if I develop the hypothesis as "x factor is very important (4 on a 1-5 scale) in consulting success at 90% confidence," that would make more sense? If that is what you suggested, then what method can I use to prove or reject this statement? I have values right now for the variable on a 1-5 scale so I can only calculate the mean. Is there any other method or calculation that I can use to bring in the "confidence" element? Thanks.

Hi guys, I am doing a research about the welding under nitrogen atmosphere. I made an alloy and welded it using arc under nitrogen atmosphere. In order to quantify how much nitrogen was dissolved into weld area, I first cross-sectioned the weld area, polished and used XPS depth profiling at first due to its high sensitivity. I used multipek to do analysis and found that result was not convincing since it showed a very weird metal elemental compositions. After reading some literature and consulting technicians, it seems that XPS is suitable for qualitative analysis (relative amount) but not quantification. But for a rough guess there would be around 1% of nitrogen dissolved from XRD measurement.
Then I turned to WDX after reading some nitrogen steel literature published in 60s-80s. After consulting technicians, they said WDX was not accurate to measure nitrogen content too. It makes me confused since most of the papers studying nitrogen in alloy used WDX and WDX is designed to measure low Z elements with better detection limit.
Is it impossible to use WDX to measure nitrogen (around 1at %)? If not then how does the structural steel manufacturer give you production cert with nitrogen content as low as 0.2%? ToF-SIMS is not suitable for us since our financial fund is not enough and it is not worth it.

As I know, WDX can quantitively measure nitrogen up to detection limit around 0.01wt % with detector cyrstals having very higher sensitivity for Nitrogen. You also need good standard materials for accuracy.

But I must disagree (in part) with Mohammed Yousif Abbas Mohammed - rather use an experienced researcher in your own field than a professional statistician, unless they are familiar with your field. Too often, pure statisticians will take a purist line that ties the beginner in knots. You need someone who knows what will be accepted in the field in question, and what will be publishable.

I have several problems in LS-DYNA and must consult with you.
I study high velocity impact with Al2O3/6061-T6 Al composite plate which ceramics use MAT_110 JH-2 model of ANSYS/LS-DYNA software.
The thickness of ceramics was 11 mm and Al was 1mm by using 0.30 AP bullet.
I had once changed the value of A B C M N T FS HEL PHEL D1 D2 K1 K2 K3…etc in LS-DYNA, but the residual velocity didn't always decrease, the initial velocity was 868 m/s, the residual velocity was approximately 700 m/s.
And how does the ceramic model simulate the crack phenomenon ?

The "6 P's" of the Peripheral Vascular Assessment are commonly used as a neurological and neurocirculatory assessment. The "6 P's" are: pulselessness, (ischemic) pain, pallor, paresthesia, paralysis or paresis, and poikilothermia or "polar" (cool extremity). Some sources use delete poikilothermia for other "P's."

Dear all,
I am using the linkage-disequilibrium method implemented in the NeEstimator software; to compute Ne values for several fish species all collected from the same geographical area.
Fst values across the focal species are sometimes significant but relatively low: the maximum value is a 5-6% differentiation explained by sampling sites. Therefore, I wanted to estimate a "global" (~ meta-) Ne, i.e. at the total study area scale (one value per species), just to compare these Ne value between species on the area. Molecular markers are of the same nature (110 to 150 biallelic SNPs for each species, developed based on a discovery panel from the study area) but as we might expect, gene diversity varies between species, with expected heterozygosity ranging from on average 0.25 to 0.40.
So I had two questions:
(1) eventhough sub-structure might (slightly?) bias Ne estimation at the whole geographical area scale, what would you say about the relevance of comparing Ne between different species?
(2) LD-based estimation of Ne could also be biased by highly unbalanced allele frequencies (e.g., MAF for a locus very close to 0). Have you faced such an issue; and would you have any piece of advice regarding this particular feature and the resulting interpretations?

I could estimate Ne per sampling site or per cluster when these exist. However, I would suffer of low sample size (typically 15-40) and when giving this a try, I usually got nothing but infinite values and/or CIs.

Hi Chrystelle,
Interesting questions. Couple of things: what is the underlying population structure of these species without considering sample location? Have you performed analyses such as a PCA, clustering, or genetic distance? Estimated the number of migrants or migration rates? That way you can identify potential genetic groupings that would serve as a more appropriate units to estimate Ne. It would be more biologically-based than sample-based.
With LDNe it is often advisable to estimate Ne using different thresholds of MAF. I believe the default is usually 0.02. That will give an indication of how much MAF is affecting your estimates of Ne.
In theory Ne estimates for different species are comparable. Are the same exact loci being used across species? There are a number of studies that have pulled together Ne estimates from different species and compared them, so I would say they are comparable. The theory is the same.
Try estimating Ne at the site-level and across sites and report both with a fair discussion of what the limitations of combining site localities with slight substructuring might be. Check out some of Andrew Whiteley's paper's on brook trout: he has written extensively about this topic. Also, Wang's recent paper in Molecular Ecology suggests his sibship based estimate of Ne implemented in Colony might be more accurate and does not have the same assumptions of LD-Ne, but requires a larger sample size.

Hey Researchers!
I am a Professional Research Assistant with a BS in psychology who got his hands on a Tobii TX-300 eye-tracking system. I have been responsible for setup of the system, task design, and consultations with other researchers who want to use the system.
I am interested in learning standard approaches to analyzing eye-tracking data, specifically working with the output file from the Tobii and Eprime system where each row represents a single sample. I have experimented with the data in R using the EyeTrackingR package and in MATLAB.
Do you have any experience working with eye-tracking data? I find it all overwhelming, but would like to learn what I can for this position. I prefer to work in R or Python, and can likely write a script once I get my head around the processes. Namely, I want to extract the probability of first fixation for each stimulus type in my experiment. Which I think means I need to subset my data into trials and do some scripting.
As you can tell, I am quite confused. I appreciate any advice or guidance from you.

Hi Robert,
You probably might have realised that there’s no gold standard approach to analysing eye tracking data. Rather the approaches that the researchers used are primarily driven by their research questions, and the limitations of their eye tracking equipment (such as sampling rates, design flaws etc.).
Before you even go about extracting probability of first fixation for each of your stimuli etc. (though I'm not sure why you would need that variable), you'll definitely need to parse your eye tracking data first. Given that you’re interested in fixation data, perhaps this paper may be a great start in terms of understanding some of the broad approaches for parsing eye tracking data (saccades and fixations). This article even provides some pseudocodes for readers to get started -

This reader-friendly book has also been recommended to me previously when I just started eye tracking -
Holmqvist, K et al. Eye tracking: A comprehensive guide to methods and measures
Also if I’m not mistaken, the Tobii software uses a velocity-based threshold approach for parsing saccades from fixations. They have published their ‘white-papers’ with regards to their eye tracker under the authors Olsen & Matos. It is up to you to decide if the Tobii software and the filter used can provide you with the variables/ data you need to answer your research question.

I want the cost per GJ of process steam, cooling water and refrigeration used in a chemical plant. What reference should I consult? The attached image from Turton's book (Analysis, Synthesis, and Design of Chemical Processes) has shown the cost of steam to be greater than the cost of refrigeration which seems very unreasonable. It is written that the cost of refrigeration is maximum 13.11$/GJ whereas the cost of LP steam is shown as $14.05/GJ.
Please give some authentic reference to know the cost of steam and refrigeration.

I have seen many articles with bar graphs on different percentages of positive cells from immunofluorescent photos, but I need to know if to create these graphs it is only necessary to observe the image or there really is a specific intensity value to define which cell is positive or not. (I think this is the best method, but I can not find this methodology). Please, recommend me any document that you I consult. Thanks!!

Hi Daniela,
the intensity values you get depend on many factors (mainly laser intensity and image acquisition settings). This is why there is no general number for a minimal fluorescence intensity value that corresponds to positive cells.
A good protocol to count positive cells is (in my opinion) the following:
1. Make sure the samples you want to compare are stained together, to avoid differences in the quality of your immunofluorescence staining
2. Acquire all your images with the same laser, objective and imaging settings
3. Take one representative image and use ImageJ to set a threshold so that your positive cells are above the threshold and negatives are below. This is really very much like looking at your image and deciding what is the best value.
4. Analyse all your images with the same threshold settings.
In practice, this is not very different from looking at your images and counting cells; one does this to eliminate personal bias, that might bring you to include cells that are not so clearly positive in some of the samples.
