My Idea is making Procurement in smarter way with systematic knowledge directly interconnected with computer softwares Globally like a chain to reduce human interaction like physical counts and monitoring the stocks to reorder the Items with the help of IOT interconnecting all departments in serial and dynamic model to provide information to the managers to make just a decision with received info like order qty xxx -> Existing stock xxx -> Automatic lean Material plan comparing with other items -> Exact needed Qty xxxx to order.
( Incoming Job Order ---> Lean Production Plan with,
--->Incoming RM Stocks Counts & Qty in Store or warehouse and also in unused Vendor & Supplier consignment stock,

Hi all,
I received a vial of HEK293T cells from a vendor. Based on some published research, the recommended passage number for those cells should be, ideally, lower than 20 to get good downstream experiment results, as I plan to do some genetic engineering with those HEK293T cells. However, based on the information from the vendor, the passage number for the cells I received is about 28 ~ 32 since the company started the screening. Thus, I am communicating with the vendor regarding whether they could send me some cells with a lower passage number. However, I am also wondering, for those who have worked with the HEK293T cell line before, have you worked with HEK293T cells with a high passage number? if so, is there any negative effect you have observed / experienced with those high passage number cells?

I just installed OpenChrom software (Version 1.5). However, I am having problems installing Plug-ins. I received the following message: You curently no proprietary converters installed. These are required to open instrument vendor files. Do you want to install converter plug-ins now?
However, I followed the software's instructions but could not fix this problem. Configuring probably didn't complete because it finished too quickly and ended before completion.
Can someone guide me to fix this error?

I often find Streptococcus sp. that show alpha hemolysis and grow on Trypticase Soy Agar with 5% sheep's blood (standard blood plates used for clinical microbiology). The colonies are small, clear, round, smooth with an area of partial hemolysis. Our incubation temperatures depend on the source animal, and fish culture plates are held at 27C. I ask because different countries have different solutions to this issue. In the country where I have been operating for many years, the main, dominant, meanstream, public news media are under government control, are state-owned (e.g., companies in which the treasury dominates as the main or sole shareholder), are referred to as public, and are used by the government as a mouthpiece for pro-government propaganda, including generating disinformation to citizens so that citizens will once again vote for the same ruling political party in parliamentary elections. On the one hand, this has been operating for many years and nothing has changed in this regard. And on the other hand, it is criticized by media experts, researchers and scholars, by media scholars, but also by many citizens who are not professionally or scientifically involved in the media. The criticism points to the lack of information objectivity, to the politicization of information, to the creation of pro-government propaganda, consisting of constant and incessant praise of what the government is doing, criticism of what the opposition parties are proposing, etc. on the unreliability of information, on the violation of the rules of journalistic ethics, on the public practice of hegemony against people working in opposition political circles, on the practice of disinformation even against people representing the so-called "hardcore electorate" of voters who usually vote in parliamentary and other elections for the ruling PIS political option.
In view of the above, I address the following question to the esteemed community of scientists and researchers:

Dear All,
I did the extension experiment with 20mer RNA primer annealing with 40mer RNA template in the presence of recombinant human coronavirus SARS-CoV-2 RNA-dependent RNA polymerase (RDRP) which was purchased from the vendor. No extension product was observed but some products which were lower than primer shown were found (please see the attached screenshot). Does anyone know the possible reason to cause this? The primer and template were purchased directly from IDT and they are still good (I checked the purity via HPLC). And the polymerase was purchased in July and are stored under -80C until using. Thanks in advance!

RNA Polymerase should have 3’-5’ exonuclease proof reading activity and it is simultaneously degrading the primer and extending it with ATP. If the exonuclease site probably glutamates and aspartates that bind divalent magnesium are mutated to alanine this should knock that out.

I have an assessment vendor who is claiming that his AI scoring works almost as good as 'trained assessor' scoring on generated responses on language assessment.
We have some open ended questions and recording of test taker's answers on those questions. These are then scored by trained 'Human Evaluators' (2-3 evaluators independently evaluating each recording) and by machine.
The vendor is saying that 40% mismatch between Human and machine evaluation is acceptable (mismatch = difference of more than 1 point of a 6 point scale) and 30% is good, 20% is very good and 10% is acceptable. For me 10% is the maximum we can accept, beyond that it seems problematic.
I am looking for research references in this line. Any help/reference is most welcome and will be highly appreciated.

Its been some time since i worked with enzymes. So as i now had a look at Enzymes by several vendors i wondered about the units the activity was given in and their meaning.
e.g browsing Novozymes products you find activity in units such as

We receive samples from different vendors from mouse studies. For the sample taken in life, some vendors can't tell us what volume is in the tube since it would take too much time to note every single volume. We know how much blood they take, but after clotting and centrifugation, they can not predict how much serum is going to be produced. For Sample Management and inventory purposes, I would like to put that information so that each time someone uses the sample we can subtract the volume taken and evaluate how much is left when we plan an experiment. I thought about following the scientist who thaw the sample for the first time and note the volume while it's thawed, but I was wondering if someone had thought of a better and more time efficient solution? I don't think there is much else you can do. If you could find out the manufacturer and model of the tube they use, you could try to get them, weight them and estimate volume by weight difference, but depending on the volume and variability in weight of the tubes this could be quite imprecise.

Dear all, would anyone help me to find a vendor, which can supply a human CD19 CAR vector?
We need a packaged lentiviral (or possibly retroviral) vector for use with Clinimacs Prodigy (Miltenyi).
We recently purchased Clinimacs Prodigy and some accompanying equipment. Currently need to organize training on using the equipment including Prodigy and cell analyser. It appears that Miltenyi will no longer supply customers of their equipment with the vector.
But we need the vector. If we have no one, we won't be able to prepare CAR-T cells and will not be able to test the process in our hands.
At this stage we do not plan to use the CAR-T cells for treatment of patients, only for training and in vitro testing. Thus, we can use non-GMP vector.
Do anyone know a commercial company which can sell us some sufficient amount of the human CD19 CAR vector? Kindly give contacts of any suitable vendor (in US, EU, China, any country).

Hello,
I am having trouble culturing and using SAEC in assays. They don't really seem to proliferate, but they don't die either...until I try to detach them from the flask and spin them down to use in an assay, at which point they all die.

There are cheaper and more efficient methods. You may precipitate the phosphates by adding a counter ion including buffer or anion exchange resins that can be used to get rid of anions. SPE anion exchange or gravity flow anion exchange may be preferred by using a SCX exchanger. Desalting and buffer exchange is a very good option too as Dr.Nick Gee adviced. If you are interested in a couple or a single protein of interest in the solution, it would be more appropriate to use immunoaffinity selective beads to immunocapture your targets. Simultaneously phosphate clean-up can be achieved by applying this approach.

Hello. I want to ask how can I translate weightages of "yes" or "no" of a criterion in terms of numbers in AHP calculations?
for examples, my goal is to select most suitable construction material vendor (Alternative) according to the set criteria including "ASTM Standards", with 4 other criteria.
This criterion means "Is the material vendor giving the material according to the Construction Standard ASTM, or not?" So the answer given to this by the alternative (vendor) would be a "Yes" or a "No", but not a value.
First, I've sent a questionnaire to the experts to give me pair wise comparison weightages for one criteria vs the other. And I've got great response with consistency of 0.9. Acceptable!
But when I ask the vendors to provide their responses, they put numerical values to the first four criteria, and I find the comparison matrix for these vendors (alternatives) for each criteria using simple maths division (value of one vendor divided by second vendor). It gives me the exact zero consistency, because I've found the alternatives weightages using exact math comparison. But I get numerical values of weightages to find ranks, so it works fine for me.
But when it comes to the 5th criterion (ASTM Standards), the responses are in "Yes" or "No" terms! My question is that what numerical value should I give to a Yes and a No so that I can get their comparison by dividing with each other?
for instance, if I use Yes =1 and No = 0, the system hangs because anything divided by zero gives an infinity answer, and it doesn't proceed further.
If I want to use a scale of 1 to 9, saying Yes = 9, and No = 1, it gives me good results. But I've no reason to use 1 or 9, for it could be any numerical value say 1000 for Yes, and 1 for No.
Please suggest me a reasonable value with a citation if possible.

Very interesting your question. The problem lies in the fact that in AHP you determine artificial values, but if you use Yes or Nor, or 1 and 0, these are not only objective values but with only two possible answers
Coincidentally, less than a week ago, another colleague put the same question, something that I had not seen in years
This is one of the 'beauties' of AHP, so elemental a method, that does not allow for modelling frequently incidents like this.
In other systems, you simply put 1 and 0 respectively

I'm planning to use the MAB377 anti-NeuN (a very common antibody) for staining of primary hippocampal neurons in culture. The vendor website recommends diluting 1:10 - 1:100 for ICC and 1:100 - 1:1000 for IF, and all the references I checked only refer to using the antibody for IF purposes. I would have expected the concentrations used for ICC and IF to be reversed, so I don't know if I trust this! Based on your experience, is 1:10-1:100 a legitimate range for ICC? If not, what dilutions are you all using?

I concur with Jordan T White that using log-scale would give a better readout of the actual binding affinities. The differences in the layout of the two plots makes comparison of the values difficult.
I would determine the 'linear range' of the upper plot to be between 30 - 250 ng/ml, while the 'linear range' of the lower plot is <5 ng/ml -< 50 ng/ml; above 50 ng/ml, it is non-linear. But just looking at the two plots, the upper one, being sigmoidal, suggest co-operative binding, while the lower one looks like a saturation curve.
Perhaps there is a valency difference in the two proteins? I know that Fc chimeras will form dimerize to form bivalent chains. I do not know if Fc-Tag chimeras dimerize or not. I am unfamilar with that platform.

Generally when a vendor gives a spec for gradient strength, they are referring to the maximum gradient an individual gradient coil can produce. However, as Samuel mentioned there are 3 gradient coils(X, Y, Z), if you pulse them simultaneously you get a gradient along the diagonal that is stronger than what any of the individual coils can produce. if you pulse the three gradient coils at 45 mT/m, the gradient along the diagonal would be 78 mT/m. So the vendor advertising this vector amplitude, is trying to make it sound like their gradient coils are stronger than they actually are.
Maximum gradient strength really only matters for DTI. The higher your maximum gradient strength, the larger the maximum b-value you can achieve at a give TE. Or conversely you can achieve the same maximum b-value at a shorter TE, if you have stronger gradients. The shorter TE means higher SNR. For DTI experiments, you want to use the same b-value in many different directions, so the maximum gradient strength of the individual gradient coils is the important number. For the system you have mentioned, 78 mT/m can only be achieved for 3 direction.
For other demanding pulse sequences, such as EPI, the gradient coil slew-rate is much more important than the maximum gradient strength. The optimum gradient amplitude for typical read-out gradient pulses or slice selection gradient pulses, is usually well below the maximum gradient that available gradient coils can achieve. However the slew-rates are all software limited in order to avoid peripheral nerve stimulation, so the vendors don't talk much about slew-rate.

Lowering severity and recycling as much as possible is usually the first response but it is extremely difficult to do efficiently. Pinching tube is an option if the low rates are expected to continue for a prolonged period.

I'm currently searching for vendors to provide L-cystine dihydrochloride for use in plant and animal cell culture media. I'm attempting to obtain a version with no more than 0.5% loss on drying; however, the specifications listed by vendors vary widely. The vendor I'd like to order from has a maximum of 5.0%. I've had a hard time finding much information on whether the loss on drying value will have any significant impact on the performance of the various media we work with. For reference, the animal cells in my lab include BHK, CHO, and VERO attachment cells; and Sp2/0 and Sf9 suspension cells. there is no commercial vendor, since I have the sole international rights as developer of this cell line. If you still need the cell line you should write to me an application in order to receive the required agreement documents.

Hi there,
I am currently using Alfa Aesar fused quartz slides for my UV-vis and PL measurements of TiO2, but it seems that their slides emit in the UV around the same wavelength region where I would expect emission from TiO2.
Could someone recommend another type of slide? Maybe fused silica? A link to a vendor would be much appreciated.
