Hi all
I was wondering if anyone ever came across a scenario where S aureus ΔRM Systems were working for electroporation and all of a sudden stopped. I had a batch stop working, so I threw it out streaked a new plate from the glycerol stock and redid the competent cells multiple times including the 1 I primarily used for the cells I used for a full year. All conditions were met the cells were stored in a -80, removed as needed and thawed on ice, DNA ranging 0.1 - 1.5 ug was used but no colonies, the Abx was tested on non plasmid carrying bacteria and its working when the cells themselves are grown with no selection pressure they grow, can anyone give any opinions as to what could be wrong?.
Ps the electroporator was tested on E coli and it works,

Hello,
I am trying to implement an attitude filter that can help to estimate roll/pitch angles from gyro/accelerometer/magnetometer sensor for a helicopter with several manoevres. The problem is that when external acceleration appears, the attitude estimation is disturbed and gives large error. I tried to find the solution from several articles (reject correction from accelero when high acceleration or adapt the covariance matrix in function of accelero output) but they didn't work. Another idea is to use magnetometer to correct gyro drift but it didn't work either (or I got a bad method).

btw, Additionally, a cost function can be incorporated in the revenue function, which is then maximized again for a more optimal (and conservative) discount.

Yeah you absolutely can for GPCMs, selecting the most "informative items" means that you prioritizing those that provides most precision in estimating θ (for the relevant range of interest).
However, this also means you're potentially discarding items that actually capture important aspects of a construct that theoretically is needed in order to fully interpret the construct, i.e., content validity is potentially reduced. But more importantly, the item information function depends on the θ distribution, so items you find to be non-relevant in your population could turn out to be pretty important in other populations.
So yes, it is a valid approach, but these are some "drawbacks" you absolutely should address as weaknesses or limitations

Once the plasma samples have been collected, they must be frozen within 8 hours. To ensure the quality and efficacy of plasma samples, fresh plasma samples must be frozen to very low temperatures. Plasma samples stored below -30 degree C will work for a period of one year. However, if you wish your plasma samples to last for more than 5 years, then you should store your plasma samples well below -60 degree C.
When you have to use frozen plasma samples, after thawing, you may refrigerate them at 4 degree C and use the samples within 24 hours.
I have a little doubt whether your plasma samples will work when stored at -20 degree C for such a long period (one and a half years).

I am currently doing a study where I try to model the theory of planned behavior via structural equation modeling. I have longitudinal data (4 waves) with items assessing behavior so I want to include a cross-lagged effect from intention in one wave on behavior in the following wave. My problem is that I don't know if I should model the effects of attitude, subjective norm, and perceived behavioral control (predictors) on intention within the same wave or if these predictors should have an effect on intention in the following wave. I'm leaning towards the first option since it makes sense to me that the effects of the predictors on intention would be immediate (e.g. if I already have a positive attitude towards a behavior it wouldn't take until the next wave to have the intention to do it). Unfortunately, I couldn't find any information on whether the effects of the predictors on intention are immediate or not. The original papers by Ajzen didn't give me a concrete answers and almost all of the longitudinal studies I looked at model the predictors and intention within the same wave without explicitly justifying an immediate effect.
Does anyone have an idea how an immediate effect could be justified or can cite useful papers on this topic?

you don't have to decide about the model but simply can analyze all sorts of synchronous and lagged effects (1yr, 2yr etc.). A four wave study is an excellent possibility to address that empirically.

The experiment normally reports a viscosity value, but behind the measurement, what is really this viscosity value for such case - is it the viscosity after mixing or happens to be one of the oxide liquid?

Thanks for the replies. Want to update that I found two more ways 1) send isolates in Dna-Rna shield (but needs to be cleaned up before sequencing with extra cost) 2) dry isolates in gen-tegra rna tubes (to recover, add the final volume of elution solution such nuclease free water and directly use for sequencing). The second option was more cheaper so I sent samples to Macrogen in those tubes.

Background: The expansion of artificial intelligence (AI) across diverse sectors worldwide demands an understanding of its impact on future generations. The studies of its influence on university students’ behavior and application in Lebanon are still limited. The present study aimed to explore the knowledge, attitudes, and practices (KAPs) of university students regarding AI and to identify factors affecting these dimensions. Methods: An online questionnaire (n = 457) was distributed to university students who were at least 18 years of age across Lebanon. Results: The results revealed that a significant majority (97.2%) of the participants were familiar with AI, from which 43% demonstrated a high level of knowledge. Furthermore, attitude toward AI role and integration in academic and professional paths was moderately satisfactory (43%), although it was reportedly used by 75% of students throughout their university years. There was a significant association between knowledge levels and sociodemographic factors such as age, sex, source of AI-related information, and knowledge rating (p < 0.05), whereas the academic major and knowledge rating affected attitudes toward AI (p < 0.05). Conclusion: These findings support the incorporation of AI education within the curriculum to increase acceptance of AI as a modern tool enhancing various sectors and serving as a facilitator for teaching and learning processes.

Hello! I am running a Multigroup analysis with Amos 29 using the Theory of plan behaviour. My latent constructs are Attitude, subjective norms, perceived behavioural control, and intention (intention would be my dependent variable). My CFA and Structural models have a reasonable Fit. However, during the construct validity analysis, the AVE of some latent variables is below 0.5, and the construct reliability is between 0.6 and 0.88.
How can this impact the interpretation of the results? The structural model has a reasonable fit. CFI 0.93 IFI 0.93 TLI 0.89 RMSEA 0.06 NFI 0.92.

Hi,
as far as I know AMOS still cannot print the "standardized residuals"--that is, the cell-wise differences between the empirical covariance matrix and the model implied matris (stanardized). These are essential for any diagnostic where (with regard to which covariances) the model fails. I would really spend one week (you don't need more) to learn R (especially "tidyverse" framework) and then use the lavaan package. It is worth it!
The lavaan.org website has many tutorials which make it easy to run a model.

Usually 8min at room temperature will work with 0.1% pFA concentration. Then you may have to sonicate the samples to have the lysate before IP. After IP you have to reserve the cross-linking before SDS-PAGE by boiling at 95C for 20min...Hope this helps you...

About time dilation. We remember the descriptive diagram which includes, for example, the train which moves at a speed "v". The two mirrors installed in the train between which a beam of light circulates constantly. There is also an external observer who, according to the descriptive diagram, observes what is happening. According to Einstein this observer who is outside the train, this observer being stationary, observes that the time it takes the light to travel the distance between the two mirrors dilates! In my opinion we cannot observe a dilation of time, moreover we do not yet know what is the definition or the concept of time is!!! In my opinion the outside observer can notice that the speed of light is a constant which is, c,!! But according to me the light circulating between the two mirrors "sees" its time dilates because of the speed of the train!! thus the speed of light circulating between the two mirrors is no longer, c, but less than, c!!! And in my opinion, light has an intrinsic property which is the dilation and each dilation corresponds to a reduction in its speed so that it becomes less than, c! and in my opinion this must be the principle of atomic clocks which delay when they are moving at one speed, v!! In other words it is not gravity that slows down an atomic clock in mouvement at a speed, v !!

Time dilation, as described by Einstein's theory of relativity, occurs due to the relative motion between observers or the presence of gravitational fields.
Light itself travels at a constant speed (approximately 299,792,458 meters per second in vacuum), and from its own perspective, it experiences no passage of time because, according to relativity, time does not pass for massless particles like photons.
Until a superior theory emerges, we can agree that light itself does not cause time dilation. Instead, time dilation effects manifest when comparing the experiences of observers in various frames of reference (due to relative motion) or at different gravitational potentials.
