Dear peers,
Recently we purchased some NSG mice from JAX lab (#005557) to establish an orthotopic lung tumor model by intratracheal injection. However, we came across some enigmatic issues in tumor development.
Our study has 2 cohorts (7 mice in each cohort), and those mice received luciferase+ A549 cells through intratracheal cell administration. After the cell administration, the mice were monitored under the IVIS system, and the bioluminescence signal indicated successful cell delivery. Subsequently, the mice underwent regular weekly IVIS imaging to monitor tumor development. As expected, some cells will die after the injection, and we confirmed the cell loss by reduced luciferase activity. Then, during the following 2 months, 5 mice kept showing negative signals all the time, but the rest mice developed tumors. So, we paused the IVIS monitoring and kept the mice for 1 more month, followed by an autopsy.
To our surprise, all 5 mice showing negative luciferase activities developed tumors, and some were quite large. We are confused why the tumors were not developed in the first 2 months but rapidly grew in the last month.
We are aware that if we take batch imaging, the mice with larger tumors will display strong signals, which can make the mice with sporadic tumors show negative signals. So we also took the images individually but still didn't see any luciferase activity.
Are there any possible reasons? NSG mice have functional neutrophils and partially functional monocytes. Could these cells suppress the tumor cell proliferation at the monitoring window?

Hello.
We are currently observing the in vivo environment of mice at different glucose levels and
For this reason, we are currently measuring blood glucose levels in mice that have been administered anesthesia beforehand and treated with an open abdomen.
When we perform the laparotomy, we inject 30% glucose through a needle into the stomach of the mice.
However, the blood glucose level did not rise at all even after waiting 30 minutes after administration, and the mice died during the procedure after being warmed up to the 38°C range with a heater.
We are looking for a way to raise the blood glucose level in order to finally observe the glucose uptake in the pancreas, but do you have any good ideas on how to administer it?
Incidentally, we are currently considering direct glucose administration via tail vein, but no one has tried it and we are still in the consideration stage.

Applying 30% Glucose into the abdominal cavity should result in serious osmotic stress, but not in glucose uptake.
For simplicity, you could try to infuse a glucose solution by the tail vein, using a syringe pump (hospital style or homemade, e.g. taking advantage of sth like a Cavro XP-3000 with a small volume syringe, which you might find on the used market. They are quite easy to control from a laptop through a USB-to-serial converter and sending commands from a terminal program).
An alternative injection point might be the jugular vein, but this approach requires some surgery. For an example with rats, please refer to this paper:

Hey friends, I am trying to conduct research on the effects of intraperitoneal administration of cannabinoids (THS: CBD) on an animal model of cerebral ischemia as part of my graduate thesis. During my studies, I have come across some questions, particularly about which substance to use to do the most effective research. I hope that these questions can be answered here.
As a matter of fact, I noticed that the drug Sativex (Nabiximols) contains THC and CBD in a 1:1 ratio, so will it be a good choice for a project as I mentioned above?

STZ is generally used to induce type 1 diabetes mellitus by intraperitoneal or intravenous injection at a dose of 45 to 65 mg/kg depending on the route of administration.

In case of T2DM, the main factor is to develop insulin resistance rather than hypoinsulenemia. If you have to develop moderate prediabetic stage then first try using HIGH FAT DIETS. As because STZ only destroy the beta cells creating a scarcity of insulin in the system thereby elevating the plasma glucose levels. But in case of T2D you have to create a state of systemic insulin resistance. So you should try HFD. You can follow my research article where we used this HFD regime to create hyperglycemic state. However, there are constraints that are/should be imposed via public policy. All sectors of higher education cannot be supported equally as long-term projections regarding the particular country's needs should be incorporated, both in the total number of students accepted and in numbers accepted in particular disciplines which should be commensurate with the country's projected needs in the production/research process and in relation with international production/research market.
A classical failure in this way of thinking is Higher Education in Greece. 89% of highschool graduates gain entry into higher education without any preplanned selection of entry in disciplines that are either important to the national economy or are of market value abroad. As a result, there was massive immigration in the past 8 years of over 450,000 graduates, those with higher qualifications in grades/marketability. The simple idea that there is no economy that can support cohorts of 80% university graduates did not play any role as the main motive behind this policy was a political gain for the parties in power, which are seen to dispense an economic good for free.

I have created a mental health measurement that is designed to be more accurate in assessing an international group of Asian people when compared to an assessment that has been created and standardized on a western population.
I have run a large group of said group through an administration of the test that we have created and an assessment that was created in the west. We have good realiability btween the two tests on the overall score. However, the sub tests that are quite different, because they are measuring divergent sub constructs definitely do not correlate with the sub scales of the established western test.
A confirmatory factor analysis has conformed a very good fit for this new test CFI=.925 and
a RMSEA .0990 (.0890 - 0.109)90%CI, ...AIC 6146 and BIC 6362. with a population from this country.
However, the The Western based test goodness of fit tests for the same subject are: CFI=0.856, RMSEA .0679 (.0522-.0825) 905CI, ....AIC 6146 BIC 6362.
My question is: Can this relative poor fit among these subjects be a demonstration of divergent validity.....at the same time that I used the pearson's correlation coefficient for the overall score as a demonstration of convergent validity.
Essentially, I am saying that there is a common universal construct that both tests identify. However, I am suggesting that this new model fits better than the old. can the CFI, RMSEA, AIC, BIC be used in this type of argument?

There you are more interestedin how many patients and their associated costs - but there is a large overlap (intesection) and the structured data and codification between the two. This is why you see some places use, for example, ICD-10 coding (the World Health Organisation's statistical codification scheme) being used for clinical coding in some part sof the world for some clincal sub-groups.
I think, by the way, that when the accountants' view is the smae as the clinician's view then there is a good corrependence between "health" and "value". But since accountants like to generalise and average, and since politics plays a role in the value of health in every country, I don't see that the two views will ever overlap 100%. But this is just my very subjective point of view - no science behind it :)

I've read that there are several methods for the administration of exosomes to mouse models (e.g. intravenous/tail-vein, retro-orbital, intraperitoneal, intranasal, and subcutaneous) and I was just wondering what the advantages/disadvantages of each are – or in other words, how to select the right method for a particular study? Moreover, if you are checking Exo targeting/biodistribution towards a specific organ, then routes of administration depending on its outcomes could be different, for example, if you're studying Exosomes exposure in the brain, then intra-cerebral and intranasal administration routes must be checked for systemic biodistribution studies.

I had submitted my paper to the IEEE Access journal, after 19 days (very fast), my paper was rejected to address the reviewer’s concerns. The journal has a binary peer-review. I received two reviews that were different. The first reviewer accepted the paper and the second one rejected it (and suggested the resubmission). The editor rejected the paper and gave the opportunity of a resubmission. The reviews were very professional and helped improve the paper’s quality, and a reviewer asked me to add his/her citations. After 17 days, I have resubmitted, and after 5 days, the status has changed to ‘Under Review.’ However, after a month, the status hasn’t changed. I had emailed the article administrator and got a reply saying,
“Your manuscript is still undergoing the peer review process. At this time, the editor is still waiting to receive reviews. The editor is doing everything possible to expedite the process. As soon as a decision can be made, you will receive a decision letter via email.”
I have been waiting for two months. I am a bit worried if I am giving impressions like urging them. But this paper is a base of my doctoral thesis; thus, the paper must be accepted within this school year (i.e., by the end of March). I have decided to email the article administrator again and inform him that I need the decision for my graduation and request him to expedite the process if possible, but I still got the same reply. (same as the previous email)
What should I do? I need the decision for graduation. Should I withdraw and submit to another journal (lower impact factor)? What if the reviewer rejects the paper after two months of waiting? Please advise. Thank you in advance.

I am investigating how (dis-)similar bachelor students interested in pursuing a Master's degree perceive according offers (i.e. Master programmes from different universities). Altogether, I am comparing 15 Master degree programmes. Hence overall there are (15*14)/2 = 105 pairwise comparisons to be made. For obvious reasons, I cannot ask each respondent to make similarity ratings for each of the 105 pairings. I wonder if it is legitimate to have each repondent rate one (and only one) pair to which they are randomly assigned. As I do have access to a sample of several thousand students, I expect to have at least 10 similarity ratings for each pair of objects compared. I wonder if this procedure is statistically sound or if, in multidimensional scaling analysis, each participant must rate all possible pairings of objects (Master degree programmes in this case).

If i want to calculate say global morans' I-index over the observation that is dispersed in administrative units to measure the level of clustering on a certain variable is it relevant to do it if i use the geographical data on the administrative units? So observations within the same administrative unit will have exactly the same position and I don't want to measure clustering between the observation across administrative units (i.e I set the observations to be neighbours within each administrative unit). However, for the second question, four percentages are mentioned. If it were just the GABA/L-theanine's effect on sleep duration and latency, one would expect only two percentages.
Also, Looking at Figure 4(C), why is the L-theanine bar showing the same height as the GABA/L-theanine but it's still considered insignificant compared to control while GABA/L-theanine is significant?

I have to conduct the analysis of data collected through phone survey by an external company.
The sample was pre-stratified based on administrative division, gender and area type (rural/urban). In addition, data weighting has been applied on the collected data, with the objective to compensate non-respondents bias; weighting variables have been gender and area type again, as well as age group and education level.
Is it correct to apply both pre-stratification and post data weighting? Do you undersee any concern with the methodology that has been applied?

These are two different things:
Stratification is used to address potential "confounding" issues in your study, such that the variation in the answers given in your survey could be (potentially) explained by the different strata.
What happens is that your stratified sample data may not be proportionally representative of the true stratification in the population. This may or may not be an issue, depending on the overall goal of the study.
Weighting is use to "make sure" that the proportion of each stratum in your data is the same as in the population. This can be done when the data collection is done when the participants were stratified before the beginning of the study (such as in an randomized design) or when stratification was done in the data after collection of the data (such as in an observational study).
Therefore, the goals of performing stratification and weighing are different. Hence, one does not exclude the other.
Finally, what I would like to re-emphasize is what I wrote at the end of the second paragraph: it depends on the overall goal of the study! Generally, in a randomized design we try to obtain strata that are similar in size (i.e., a somewhat balanced design), even if this is NOT representative of the population stratification. In a balanced design, the statistical analysis is "easier", allowing effects to be orthogonal to each other (great feature!).

I'm doing research on feline semen cryopreservation. Recently, my Local Ethical Committee disagreed to use privately owned cats for semen collection, arguing that a domestic cat is a laboratory animal and all the experiments must be performed on cats from experimental colonies.
There is no such colony of cats in Poland - does anybody know where it would be possible to perform an experiment in which I need to collect (by urethral catehetrization after medetomidyne administration) and cryopreserve semen three times from the same male?

We are seeking to know the preference of students-respondents in selecting their political candidate. When trying to identify language proficiency for cognitive test administration (Spanish vs English) in a clinical trial, what is the best way to ask a participant (via survey) which language they want to be tested in?
A colleague suggested we use the standard way asked by the Dpt of Education in the followig order:
(a) "What language did you first learn?" and then
(b) "What language is spoken most frequently in your home?"
If responses do not coincide then they ask
(c) "If you were to take a test, in what language would you want to take it in to give your best performance?"
Being a native Spanish speaker, I believe we should first ask option c to reach our goal (cognitive testing). It seems to me that asking a and b first is not a personal choice. For example, many of us were raised in Spanish and speak Spanish at home with our parents, but our education was in English and we live immersed in English so we may respond better in English on a cognitive test than if given in Spanish.
What are your thoughts? Is there a scientifically proven approach to this? Thank you for your responses.

We will soon run a dietary intervention trial and we need the participant to individually collect urine sample after 24 h from the administration of a dietary product. Unfortunately, the administration will probably take place on Saturday and thus they will collect the urine sample on Sundayand come to deliver it to us on Monday.
The question is: what is the best way to properly store the sample during Sunday?

I work in a clinical research area where we give study drugs, usually intravenously, then draw PK's and other drug levels at specific time points following the administration. We have some investigators that say all post-administration blood draws must be done peripherally. While others say that after flushing a CVAD well and discarding some "waste" blood, the CVAD can be used for blood draws. of our patients have very limited peripheral access, so a peripheral blood draw may be impossible. Does any know if using a CVAD can skew any results or not? And if so, how long or how much fluid must go through the CVAD before it is considered alright to use? Thanks for your input.
