Two important things about this method confuse me.

1. some of the researcher say ascorbic acid and citric acid are heat sensitive, so cannot be add to MS medium before autoclave.

2. 2nd thing is pH, of medium, if i add these antioxidant after autoclave in laminar flow unit, how can i manage the pH of medium, because it will change the pH of the medium.

How much fertilizer can be placed safely with seed?

I want to irradiate the seeds of okra(Abelmoschus esculentus)seeds for inducing mutant plants. Kindly let me know how to prepare the seeds for mutation treatment. And I would like to know which physical mutagens(gamma rays/ ion beam/ neutron beam)  or chemical (EMS) are better for inducing mutations in cytoplasmic genes (mitochondrial or chloroplast DNA) of plant cells I mean if I take seeds as mutation material? When I am going for selection in kanamycin the selected seedlings are not having well formed roots because of the effect of antibiotic so I am facing problem while transferring to soil. Can anybody help me with their suggestions in this regard?
