Clarity of solution by turbinometer
Question
  • Feb 2022
Did someone faced problem with clarity measurements with turbinometer?
We encountered a problem with the analysis of clarity of solutions. As per Ph.Eur. reference suspension I should have 3 NTU, reference II 6 NTU, however we measured around 4.7 and 9.6 respectively.
We prepared several solution, all the same.
We bought Primary opalescent suspension with Assay on CoA of 4094 NTU, obtained standard of opalescence is 79.4 NTU.
We use a 4 points calibration.
I cannot understand what more should be done?
Any suggestions? Thank you in advance!

Is anyone in the UK using the CLARITY method?
Question
  • Oct 2013
I'd like to try the CLARITY method on some of my knock-in mice with fluorescent reporter molecules attached to their post synaptic proteins. It is impractical for me to set up the entire method when I don't know if it will kill the fluorescence, so it would be excellent if I could come and borrow someone's set up for a short period to test the method on my samples.

Need clarity on analysis of ordinal phenotypic data?
Question
  • Mar 2017
Any leads to how this data can be transformed to distances? Transformation techniques? software? Currently reading on Mahalanobis distances and  Gowers (dis)similarity coefficient but there is still too much ground to cover. All productive detours are welcome. Thank you

Is CLARITY compatible with tracing dyes (dextrans, etc)?
Question
  • May 2015
Can't seem to find whether or not CLARITY is able to be performed on intact brains without quenching or washing away injected tracers (non-viral or genetic expression of fluorescence) such as dextrans or beads.
Does anyone know if any clearing method is compatible with tracer injections?

Staining Neurons and counting them using CLARITY?
Question
  • Apr 2017
we are using the CLARITY technique to clear Rat brains and we would like to label astrocytes and neurons in the cerebral cortex and to be able to count them. We tried to stain it using NeuN and GFAP, but it didn’t stain. Do you have any recommendations/thoughts on why it did not work, or any specific steps we should follow? we followed steps on

Has anyone seen CLARITY tissue clearing with no electrophoresis?
Question
  • Jul 2013
I just posted this in the CLARITY resource center forum, but posting it here too. We found this hydrogel perfused/embedded right hemisphere of a rat cortex to be nearly completely clear after sitting in clearing solution in 37 degree incubator for ~5 weeks completely unattended. Next to zero tissue damage and it looks by far better than any brain that we have run through our ETC chamber. Has anyone else seen this?

Has anybody done optical clearing and/or clarity on skin?
Question
  • Sep 2014
I want to image skin tissue in depth by confocal microscopy and my target is dermal-epidermal junction. So, thinking that optical clearing/clarity might help in this regard.
Thanks.

Can I have a protocol for paste clarity of starch?
Question
  • Dec 2014
I am conduction a research on physico chemical characteristics of starch.  I have got this protocol online.
Paste clarity was determined by the method of Singhal and
Kulkarni (1990) by measuring the percentage light transmitted
by different concentrations of starch (0.5–4.0%) at 660 nm on a
UV/visible spectrophotometer. Distilled water was used in the reference
cell.
Do we have to let the paste cool down ?
