The present rice market requires a range of rice products with increased eating and nutritional values. The presented work was carried out within the framework of the breeding program for developing new rice cultivars with desirable grain quality traits based on an integrated approach, using modern methods of trait phenotyping and the genomic approach, post genomic, and cellular technologies. The latest study aimed to phenotype rice cultivars from the Unique Scientific Installation (USI) - Collection of Federal Scientific Rice Centre, Krasnodar, Russia, as sources of valuable grain quality traits. Studied traits included vitreosity, fracturing, and protein and grain amylose content. The coefficient of variation determined the variability of traits and helped identify the best cultivars as sources of valuable traits. The grain quality of harvested sample cultivars showed significant differences. The rice cultivar Thaibonnet gave the best results in terms of grain quality traits and variability. By combining low variability of technologically-tested quality traits and protein content, the rice cultivars Elbrus and Thaibonnet led all the samples. Cultivar Svetlana was characterized by high traits of vitreosity and fracturing and having less variability. The cultivars Leader and Patriot both had high protein content and low trait variability. These cultivars are recommended for use in parental crosses in breeding programs to develop rice genotypes with desirable grain quality traits and nutritional values.

Gametoclonal changes are an additional source of genetic variation suitable for use in crop improvement programs for a range of agronomic traits. The aim of the study was to accelerate development of genetically stable DH rice lines with high morphological and genetic uniformity by the method of experimental haploidy in vitro based on breeding valuable samples with specified characteristics and carrying genes of a wide spectrum of blast resistance, as well as samples with racespecific genes for resistance to the pathogen, and subsequent assessment of agronomic traits of androgenic lines. The data on the callusogenic and regenerative capacity of 30 genotypes of domestic rice and samples of Chinese introduction with genes for blast resistance in anther culture in vitro are presented. Genetically stable (homozygous) androgenic lines were developed on the basis of these breeding valuable samples. The characteristics of economic and biological traits and elements of panicle productivity of 30 DH-line developed from three samples with blast resistance genes are given. The variability of traits arising as a result of gametoclonal variability in androgenic lines was noted for such traits as plant height, panicle length, blast resistance, and weight of 1000 grains.

The aim of the study was to study the development of rice microspores in anther culture in vitro , study the structure of androclinic callus to identify cyto-embryological features of the formation of morphogenic structures in anther culture, obtain doubled rice haploids ( Oryza sativa L .) and accelerate the development of valuable breeding material with desired properties. Within the framework of this study, the results of a cyto-histological study of rice haploid androgenesis in vitro were obtained, which indicate that it is induced under the influence of phytohormones from rice anther microspores at the mononuclear or early binuclear stage of development. The abnormal development of microspores on nutrient media with phytohormone 2,4-D was traced, in which nuclei, having lost their characteristic functions, acquired the ability to unlimited division and growth with the formation of microcallus. The morphological structure of calli was assessed. The main morphotypes of callus tissues and the pathways of morphogenesis, leading to the formation of androgenic structures, up to full-fledged regenerant plants, were identified. Homozygous androgenic lines based on F 1 and BC 1 - rice generations obtained in the course of hybridization and backcrossing between Chinese samples carrying blast resistance genes and Russian accessiond were rapidly developed.


Comparative characteristics of the introduced Chinese rice germplasm and adapted local varieties of Kuban breeding under the environmental conditions of Southern Russia

Under the environmental conditions of the Kuban region, morphological, vegetative and economic characteristics of the Russian and Chinese genotypes of Oryza sativa L. were studied; adapted productive forms were identified. In the course of research, productive introduced forms were selected for further breeding work. Sixteen lines were tolerant to Pyricularia and demonstrated the strong biological potential and adaptability to the conditions of the region.

Within the framework of this study, the first Russian-Chinese joint program for development of rice varieties with long-term blast resistance was launched on the basis of rice germplasm exchange and the subsequent comprehensive study of the obtained breeding samples, hybridization of Russian and Chinese varieties with specified traits. The genetic diversity of the Chinese rice samples obtained by exchange was studied by biological and morphological traits of plants, taxonomic belonging to a botanical variety, elements of the yield structure, and resistance to lodging. 31 hybrid combinations (F1) from the crossing of Russian and Chinese varieties were used to obtain the BC1 generation. The technologies of cultivation of isolated rice anthers in vitro have been optimized in relation to Chinese genotypes in order to accelerate the genetic stabilization of breeding material obtained from crossing. Phenotyping of Russian and Chinese breeding samples was carried out on the basis of resistance to the local population of the blast pathogen ( Magnaporthe grisea (T.T. Hebert) M.E. Barr)), on natural and artificial infectious backgrounds. DNA identification of genes for blast resistance was carried out for the same breeding samples. Based on the experimental data obtained, promising samples were selected - prototypes of new rice varieties.

Colored rice varieties, according to the results of studies, are characterized by various technological and biochemical grain quality traits (table 2). The red-grain variety Rubin (l / b 2.2) and the black-grain variety Mavr (l / b 2.5), based on the ratio of the grain length to its width, l / b, belong to the group of medium-grain rice varieties. Red-grain variety Mars and black-grain variety Gagat -to the group of long-grain (l / b 3.3-3.4), black-grain variety Yuzhnaya noch -to short-grain (l / b 2.0). The varieties have a medium-sized caryopses (21.5-27.0 g of 1000 grains); of them, the variety Yuzhnaya noch is characterized by the smallest grain. According to the content of the starch fraction -amylose, the varieties are different, which determines their different culinary advantages. Variety Mars -belongs to the group of low amylose, varieties Mavr, Rubin and Gagat -to the group of medium amylose, variety Yuzhnaya noch -to the waxy group, which becomes sticky when cooked. In terms of protein content in grain, all varieties are classified as medium protein. The grain size of all varieties was classified as medium. In the second variant, the weight of the caryopsis slightly increased in the variety Rubin, and remained at the same level in Mavr. Fracturing in Rubin and Mars in 2018 was slightly higher than in 2019 and 2017, and in the black-grain varieties Mavr, Gagat and Yuzhnaya noch it was the highest in 2019. The change in the grain size in Rubin, due to the different level nitrogen nutrition, unlike other varieties, was insignificant. In the variety Yuzhnaya noch, only in 2017, the difference in size was significant. The most important technological grain quality trait "fracturing" allows predicting the quality of produced rice products and determines the profitability of rice production. In the breeding process, the trait of fracturing is assessed at all stages in order to reduce its indicators in various agro-climatic growing conditions. The grain fracture is an integral indicator of the endosperm structure. The high grain quality is considered primarily due to the stability of varieties under changing conditions during the period of yield formation and the ability of the variety to maintain the integrity of the caryopsis without cracks. Colored rice varieties in the study showed different fracturing rates. The maximum grain fracturing was in the variety Mavr -within 19-25% and the minimum -in Mars and Gagat, respectively 2-5 and 2-6%. In 2017, all varieties had fracturing higher in comparison with the indicators of 2018 and 2019. In the varieties Rubin and Gagat, the grain fracture decreased by 2-8% with an increase in the nitrogen dose, in Mars it practically did not change, and in varieties Mavr and Gagat it increased.

In order to develop rice breeding material of a functional direction, technological and biochemical grain quality traits of varieties bred by the Federal Scientific Rice Centre (Rubin, Mars, Mavr, Gagat, Yuzhnaya noch) with a colored grain pericarp, high content of anthocyanins and intended for functional nutrition were studied. The varieties were grown in the valley agrolandscape zone of Krasnodar region (Russia) in 2017–2019. The studied rice varieties are included in the State Register of Protected Breeding Achievements. The grain size by weight of 1000 absolutely dry grains (weight of 1000 a.d.g.) was determined according to GOST 10842-89, fracturing on a DSZ-3 diaphanoscope, protein content was measured using an Infralum FT-10 device. The determination of amylose was carried out colorimetrically using the amylose-iodine reaction according to Juliano. The varieties have a medium-sized caryopsis (21.5-27.0 g of 1000 grains); of them, the variety Yuzhnaya noch is characterized by the smallest grain. The variety Mars belongs to the low amylose group, varieties Mavr, Rubin and Gagat to the medium amylose group, Yuzhnaya noch to the waxy group. In terms of protein content in grain, all varieties are classified as medium protein. The maximum grain fracture was in variety Mavr in the range of 19-25% and the minimum in Mars and Gagat, respectively 2-5 and 2-6%. Variations in grain size indicators, amylose and protein content of varieties are weak in all varieties of special purpose, which testified to their high stability under growing conditions.
