Description: Spirulina platensis is a blue-green alga with chlorophyll (green) and phycocyanin (blue) contained in phycobilisoms. Due to pharmaceutical and food applications, the best of the method of extraction is the vital. Objectives: Work focused on an evaluation of several phycocyanin extraction methods from two types of Spirulina, from two regions; Tamanrasset (Algeria) , Djerba (Tunisia) to deduce the optimal method. Methods: Six extraction methods were evaluated (water, freezing, sonication, solvent, biphasic separation, maceration with glycerol) on two spirulina strains from the Tamanrasset (Guelta at 1824m, (23° N. ,5° E) and Milita Djerba (33° 48 'N; 10° 51'E). Results: Two phase extraction gave higher yields versus other methods (water, solvent) (0.39 vs 0.19 mg / ml). By maceration with glycerol, algerian Spirulina gave phycocyanin concentration of 0.625 mg/ml, purity of 1.37. Tunisian Spirulina yielded 50% higher yields. Concentration of total polyphenol is between 4-22 mg/g/dm for algerian strain and 6-25 mg/g/dm for the tunisian strain. Conclusion: Maceration with glycerol is the optimal method of extracting. Characterization determine the yield of the phenolic compounds, flavonoids and antioxidant activity in the medical and cosmetic field.
