ABSTRACT: Dendritic cells (DCs) detect and respond to microbes or their components by producing cytokines and other molecules that can activate the proliferation and differentiation pathways of T cells. Investigation of DC responses to pathogens would thus provide important insights into how T-cell responses most appropriate for the pathogen are induced. Here, we describe methods for the use of mixed leukocyte reactions, to determine the proliferative and cytokine responses of murine splenic T cells in response to co-culture with bone marrow-derived DCs stimulated with Candida albicans.
