Self-Assembly and Adhesion of DOPA-Modified Methacrylic Triblock Hydrogels

Marine mussels anchor to a variety of surfaces by secreting liquid proteins that harden and form water-resistant bonds to a variety of surfaces. Studies have revealed that these mussel adhesive proteins contain an unusual amino acid, 3,4-dihydroxy-L-phenylalanine (DOPA), which is believed to be responsible for the cohesive and adhesive properties of these proteins. To separate the cohesive and adhesive roles of DOPA, we incorporated DOPA into the midblock of poly(methyl methacrylate)-poly(methacrylic acid)-poly(methyl methacrylate) (PMMA-PMAA-PMMA) triblock copolymers. Self-assembled hydrogels were obtained by exposing triblock copolymer solutions in dimethyl sulfoxide to water vapor. As water diffused into the solution, the hydrophobic end blocks formed aggregates that were bridged by the water-soluble midblocks. Strong hydrogels were formed with polymer weight fractions between 0.01 and 0.4 and with shear moduli between 1 and 5 kPa. The adhesive properties of the hydrogels on TiO2 surfaces were investigated by indentation with a flat-ended cylindrical punch. At pH values of 6 and 7.4, the fully protonated DOPA groups were highly adhesive to the TiO2 surfaces, giving values of approximately equal to 2 J/m2 for the interfacial fracture energy, which we believe corresponds to the cohesive fracture energy of the hydrogel. At these pH values, the DOPA groups are hydrophobic and have a tendency to aggregate, so contact times of 10 or 20 min are required for these high values of the interfacial strength to be observed. At a pH of 10, the DOPA groups were hydrophilic and highly swellable, but less adhesive gels were formed. Oxidation of DOPA groups, a process that is greatly accelerated at a pH of 10, decreased the adhesive performance of the hydrogels even further.

    ABSTRACT: The idea of being able to glue bone fragments with a suitable biocompatible adhesive remains highly attractive to orthopaedic surgeons. Yet despite decades of research, no suitable system that fully meets all the many requirements for such an adhesive has yet been identified. This article reviews the requirements and challenges of developing a bone adhesive for fracture repair and also the substantial progress that has been made. Developments in bone adhesives can roughly be classified into two groups: synthetic and biologically-derived/inspired. Early examples of synthetic adhesives include poly(methyl methacrylate) and related polymers, cyanoacrylates and polyurethanes. These materials are characterised by relatively good mechanical properties but often lack the required biocompatibility and biodegradability. More recent adhesive systems based on lactide–methacrylate chemistry therefore attempt to address these issues. Similarly, there is renewed interest in glass ionomer cements and calcium/magnesium phosphate cements. Biological adhesives include fibrin and gelatin-based systems. These have good biocompatibility and biodegradability but lack the cohesive strength to have good adhesion to bone and are used chiefly in soft tissue applications. Newer examples that are looking more promising include adhesives inspired by mussel adhesive proteins and the “sandcastle glue” of the marine worm Phragmatopoma californica. As well as the challenge to develop the adhesive systems themselves a further need is for greater consistency in the testing of adhesion to bone both in vitro/ex vivo and in vivo. Test methods in the literature are reviewed together with considerations for the design of such tests.

    ABSTRACT: o-Benzoquinonemethacrylate QMet is prepared by the reaction between 3,6-di-tert-butyl-o-benzoquinone and monomethacrylic ethers of ethylene glycol. Free radical polymerization of this monomer allows to synthesize poly-o-benzoquinones Poly-QMet with molecular weight depending on the polymerization method and concentration of initiator. Polymerization in solution leads to polymer with 22–30 o-benzoquinone fragments in polymeric chain whereas bulk polymerization results in polymer with 110–190 o-benzoquinone fragments in polymeric chain. These polymers can be used in the preparation of metal-containing polymeric catecholate complexes. Triphenylantimony(V) polycatecholate Poly-SbCatMet is prepared by two ways – oxidative addition of SbPh3 to poly-o-quinone and polymerization of monomeric triphenylantimony catecholate SbCatMet. This polymeric catecholate Poly-SbCatMet is able to the reversible absorption of molecular oxygen on air both in solution and in thin film with conversion about 50–65 % (by NMR-spectroscopy and UV–vis data).

    ABSTRACT: Microgels/nanogels are crosslinked polymeric particles, which can be considered as hydrogels if they are composed of water soluble/swellable polymer chains. They possess high water content, biocompatibility, and desirable mechanical properties. They offer unique advantages for polymer-based drug delivery systems (DDS): a tunable size from nanometers to micrometers, a large surface area for multivalent bioconjugation, and an interior network for the incorporation of biomolecules. Present and future microgel applications require a high degree of control over properties. They include stability for prolonged circulation in the blood stream, novel functionality for further bioconjugation, controlled particle size with uniform diameter, and biodegradability for sustained release of drugs for a desired period of time and facile removal of empty devices. This review describes the recent developments of microgel/nanogel particles as drug delivery carriers for biological and biomedical applications. Various synthetic strategies for the preparation of microgels/nanogels are detailed, including photolithographic and micromolding methods, continuous microfluidics, modification of biopolymers, and heterogeneous free radical and controlled/living radical polymerizations.
