Anti-inflammatory activities of aqueous/ethanol and methanol extracts of Perna viridis Linn. in mice

Anti-inflammatory effect of the edible mytilid bivalve Perna viridis was evaluated using formalin, carrageenan and dextran-induced paw oedema in mice. The whole mussel tissue without shell was first extracted with ethyl acetate and then successively with methanol and water:ethanol, 7:3 (aqueous/ethanol). Three doses (100, 500 and 1,000 mg/kg) of the extracts (methanol and aqueous/ethanol) were administered orally prior to the injection of inflammatory agents. The inflammation induced by formalin was inhibited by the administration of both extracts for 7 days and the activity was found to be higher for methanol than aqueous/ethanol extract. A dose-dependent reduction in paw thickness was observed for carrageenan-induced paw oedema on treatment with methanol and aqueous/ethanol extracts which was comparable to the standard drug diclofenac. The effect of extracts on dextran-induced oedema was less than that produced by the other two inflammatory stimulants.

    ABSTRACT: Objective: Invertebrates are the most explored groups in the field of cytotoxic/ anti-cancer marine natural products research, considering their ecological and evolutionary significance. Hence an attempt has been done to evaluate the possible antioxidant and cytotoxic activities of the Indian green mussel Perna viridis. Methods: Qualitative analysis of the secondary metabolites present in the ethyl acetate extract of P. viridis was done using general detection reagents and spray reagents on TLC. Antioxidant activity of the same was assessed by DPPH radical scavenging assay. Brine shrimp, Dalton's Lymphoma Ascites (DLA) and Ehrlich Ascites Carcinoma (EAC) were employed for evaluating cytotoxicity of the extract. MTT assay of the extract was performed with two human cancer cell lines namely, HCT 116 and MCF 7. Results: General detection reagents and spray reagents showed the presence of different secondary metabolites in the ethyl acetate extract of P. viridis. The extract showed good scavenging activity against DPPH radical (IC50 0.616 mg/ml). It also proved significantly cytotoxic towards brine shrimp, DLA and EAC cells. MTT assay on human cell lines proved to be more cytotoxic towards MCF 7 than for HCF 116 for the extract. Conclusion: The results shows that ethyl acetate extract of P. viridis possess both antioxidant and cytotoxic activities, which can be attributed to the presence of secondary metabolites like phenolics and alkaloids.

    ABSTRACT: Stachys pilifera Benth has long been used to treat infectious diseases as well as respiratory and rheumatoid disorders in Iranian folk medicine. Antioxidants, antitumor, and antimicrobial properties of the plant have been reported. This experimental study was designed to evaluate systemic and topical anti-inflammatory effects of the hydro-alcoholic extract from aerial parts of Stachys pilifera (HESP). Anti-inflammatory effects of HESP was studied in four well-known animal models of inflammation, including carrageenan- or formalin-induced paw edema in rat (thirteen groups, 6 rats per each group), and 12-O-tetradecanoylphorbol-13-acetate (TPA)- or xylene-induced ear edema in mouse (ten groups, 6 mice per each group). The rats received HESP (50-400 mg/ kg) orally 45 minutes before the subplantar injection of carrageenan or formalin. In TPA or xylene tests, HESP (1, 2.5, and 5 mg/ear) was applied topically simultaneous with these phlogistic agents on the ear mice. Finally, pathological examination of the inflamed tissues (paw and ear) was carried out. Acute toxicity study of the extract showed that no rats were killed at 5000 mg/kg (LD50 > 5000 mg/kg). The extract (100 and 200 mg/ kg) significantly suppressed carrageenan-induced paw edema 1, 2, 3, and 4 hours after carrageenan challenge in comparison with the control group (P < 0.001). The HESP (100 and 200 mg/kg) also produced a considerable antiedematogenic effect in the formalin test over a period of 24 hours (P < 0.01). Furthermore, topical administration of the HESP (1, 2.5, and 5 mg/ear) inhibited TPA- and xylene-induced ear edema in comparison with the control group (P < 0.001). The pathological analysis of the paws and ears revealed that HESP was capable of reducing tissue destruction, cellular infiltration, and subcutaneous edema induced by the indicated phlogistic agents. The present data confirmed systemic and topical anti-inflammatory effects of Stachys pilifera which is comparable to indomethacin.
