Effectivity of Carrot (Daucus carota) Seed Oil as Hair Growth Promoter on Rats (Rattus novergicus)

Abstract

Hair quality and health are important not only for the esthetical reasons but also to maintained the hair’s function in protecting the head. Carrot seed oil (CSO) is rich in vitamin A and beta-carotene, which influence hair quality. CSO also contains carotol, which has anti-fungal properties that treat dandruff. This research aimed to observe the potential of CSO as a hair growth promoter. Twenty male rats were used in the study, with their backs shaved and divided into two areas, each with different frequencies of CSO application. The backs of the rats were given CSO every day for 14 consecutive days with different concentrations, i.e., 25%, 50%, and 75%. Hair growth was assessed based on its length, root, shaft, and tip thickness on days 9,12, and 14 and the results were recorded. On day 15, the rats were euthanized and their skin was collected to observe the number of follicles. Results showed that the application of CSO with different concentrations had no effect on hair length growth but it did affect the thickness of the root, shaft, and tip of the hair, with the best concentration being 75%. The frequency of application did not seem to affect the growth in terms of the length or thickness of the root, shaft, and tip of the hair; however, the application of CSO with 75% concentration and an application frequency of once a day produced the best amount of follicle regeneration.

The hair cycle is composed of four primary phases: anagen, catagen, telogen, and exogen. Anagen is a highly mitotic phase characterized by the production of a hair shaft from the hair follicle, whereas catagen and telogen describe regression and the resting phase of the follicle, respectively, ultimately resulting in hair shedding. While 9% of hair follicles reside in telogen at any time, a variety of factors promote anagen to telogen transition, including inflammation, hormones, stress, nutritional deficiency, poor sleep quality, and cellular division inhibiting medication. Conversely, increased blood flow, direct stimulation of the hair follicle, and growth factors promote telogen to anagen transition and subsequent hair growth. This review seeks to comprehensively describe the hair cycle, anagen and telogen balance, factors that promote anagen to telogen transition and vice versa, and the clinical utility of a variety of lab testing and evaluations. Ultimately, a variety of factors impact the hair cycle, necessitating a holistic approach to hair loss.

Carrot (Daucus carota L.) is a vegetable plant that has many uses for public health services in the world. Fresh carrots contain water, protein, carbohydrates, fat, fiber, ash, anti-cancer nutrients, natural sugars (fructose, sucrose, dextrose, lactose, and maltose), pectin, glutathione, minerals (calcium, phosphorus, iron, potassium, sodium, magnesium, chromium), vitamins (beta carotene, B1, and C) and asparagine. The purpose of this study was to characterize simplicia and carrot seed oil extract, and to determine the antioxidant activity of carrot seed oil extract with formulations of concentrations of 3%, 6% and 9% seeds on the effectiveness of anti-aging Moisture, Evenness, Pore, Spot and Wrinkle. Carrot seed oil can be formulated in a homogeneous cream dosage form with an oil-in-water emulsion type, pH 6.7-7.7; does not cause skin irritation and is stable on storage for 90 days at room temperature. Carrot seed oil cream with a concentration of 9% showed the best anti-aging effectiveness by increasing water content, reducing skin hardness, shrinking pores, reducing blemishes and reducing wrinkles compared to other cream formulas.

As one of the main appendages of skin, hair follicles play an important role in the process of skin regeneration. Hair follicle is a tiny organ formed by the interaction between epidermis and dermis, which has complex and fine structure and periodic growth characteristics. The hair growth cycle is divided into three continuous stages, growth (anagen), apoptosis-driven regression (catagen) and relative quiescence (telogen). And The Morphogenesis and cycle of hair follicles are regulated by a variety of signal pathways. When the signal molecules in the pathways are abnormal, it will affect the development and cycle of hair follicles, which will lead to hair follicle-related diseases.This article will review the structure, development, cycle and molecular regulation of hair follicles, in order to provide new ideas for solving diseases and forming functional hair follicle.

Tonik rambut merupakan produk perawatan terhadap kerontokan rambut yang praktis, mudah terserap kulit kepala, serta tidak menimbulkan iritasi. Berbagai ekstrak tumbuhan diformulasikan untuk mengatasi kerontokan rambut sebagai alternatif dari tonik rambut berbahan sintetis yang dapat menimbulkan efek samping. Bidara (Ziziphus nummularia) berpotensi mengatasi kerontokan rambut karena mengandung senyawa antiinflamasi dan antioksidan yang mengurangi efek negatif dari stres oksidatif pada sel papila kulit kepala. Namun, sampai saat ini belum ada pengujian ekstrak daun bidara terhadap pertumbuhan rambut mamalia. Penelitian ini bertujuan menentukan formula terbaik dari tonik rambut ekstrak daun bidara dan menguji aktivitasnya terhadap pertumbuhan rambut kelinci jantan ras New Zealand. Penelitian dilakukan di Laboratorium Fakultas MIPA Universitas Garut dari Maret 2020 hingga Maret 2021. Perlakuan yang diuji adalah formulasi tonik rambut yang menggunakan ekstrak etanol daun bidara dengan konsentrasi 3%, 6% dan 12%; akuades (kontrol negatif) dan minoxidil (kontrol positif). Semua perlakuan diulang tiga kali. Parameter yang diamati adalah karakteristik tonik rambut yang meliputi bau, warna, viskositas, homogenitas, pH, iritasi, pertumbuhan rambut pada kelinci. Formula tonik ekstrak etanol daun bidara 3%, 6%, dan 12% memiliki kualitas bau, warna, viskositas, homogenitas, dan pH yang seragam. Ketiga formula tidak menimbulkan iritasi terhadap kulit kelinci. Konsentrasi ekstrak terbaik untuk formula tonik rambut adalah 3% yang menghasilkan pertumbuhan rambut sebesar 10,67 mm selama 28 hari. Ekstrak etanol daun bidara terbukti meningkatkan pertumbuhan rambut kelinci jantan putih New Zealand lebih baik daripada kontrol dan minoxidil 2%.

Alnus sibirica Fisch. ex Turcz (ASFT), belonging to the family of Betulaceae, grows naturally in Asia, Europe, and America. The aims of this study are determining the efficacy of various biomarkers related to hair loss, evaluated by extracting the branch with 60% alcohol, and purely separating diarylheptanoid oregonin, an indicator and active substance, from 60% alcohol extract of the tree. To determine the preventive effects on hair loss, we investigated the anti-oxidative and anti-apoptotic effects on hydrogen peroxide-induced cytotoxicity on human hair dermal papilla cells using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and Western blotting analysis for proving of apoptosis-related marker alteration, respectively. Moreover, we examined the ameliorative effects of 60% alcohol extract of the tree and oregonin against changes of oxidative stress-induced cytokine and testosterone-induced dihydrotestosterone production as crucial pathways of the hair loss mechanism. These results suggest that 60% alcohol extract of the tree and oregonin were available as novel natural materials for maintaining hair health in mammals.

Hair loss by excessive stress from work and lifestyle changes has become a growing concern, particularly among young individuals. However, most drugs for alopecia impose a plethora of side effects. We have found the powerful impact of Malva verticillata seed extracts on alleviating hair loss. This study further isolated effective chemicals in M. verticillata seed extracts by liquid silica gel column chromatography. Under the screening for the growth rate (%) of human follicles dermal papilla cells (HFDPCs), we identified linoleic acid (LA) and oleic acid in n-hexane of M. verticillate (MH)2 fraction. LA treatment activated Wnt/β-catenin signaling and induced HFDPCs growth by increasing the expression of cell cycle proteins such as cyclin D1 and cyclin-dependent kinase 2. LA treatment also increased several growth factors, such as vascular endothelial growth factor, insulin-like growth factor-1, hepatocyte growth factor, and keratinocyte growth factor, in a dose-dependent manner. Besides, LA significantly inhibited Dickkopf-related protein expression (DKK-1), a primary alopecia signaling by dihydrotestosterone. Our findings suggest that LA treatment may alleviate a testosterone-induced signaling molecule and induces HFDPCs growth by activating Wnt/β-catenin signaling.

This study investigated the hair regeneration promotion and hair loss prevention properties of Nelumbinis Semen (NS) extract in vitro and in vivo. The effect of NS on the proliferation and migration of human dermal papilla cells (hDPCs) was measured in vitro via CCK-8 and scratch migration assays, after which the antioxidant activity of NS was also quantified. NS extracts were then applied to the back of 7-week-old C57BL/6 mice for 3 weeks to monitor hair growth patterns and hair follicle (HF) histology. The mice were divided into three groups: negative control group (NC; DMSO), positive control group (PC; 3% minoxidil), and experimental group (NS extract 1,000 ppm). Moreover, to study the molecular mechanisms by which NS extract regenerates hair growth, real-time PCR was used to analyze factors related to the hair growth cycle. The NS extracts were found to possess high antioxidant properties due to their high flavonoid contents and electron-donating ability. Moreover, NS extracts enhanced hDPC proliferation and migration in a concentration-dependent manner (15.63–125 ppm). Watercress oil (WCO) is the seed oil of Nasturtium officinale, Brassicaceae family. WCO is a cosmetic for hair growth approved by the Saudi Food and Drug Authority (SFDA). The study aimed to analyze by GC-MS, the phytochemical profile and the safety of WCO as a cosmetic according to SFDA, validates the traditional method of applying hot oil mixtures to the scalp, and also reports the pH, UV absorption, and in-silico binding mechanism of fatty acids for hair growth. The absorption of UV-B light (284.50 nm and 290.60 nm) by WCO suggests that it acts as a sunscreen for the scalp. The pH of the marketed WCO, 5.53 ± 0.02, shall not damage the scalp and hair. The GC-MS analysis confirmed fatty acids as the principal constituents (65.65%) of WCO composed of monounsaturated fatty acids (MUFAs): cis-11-Octadecenoic acid (17.14%), cis-9-Octadecenoic acid (17.11%), cis-13-Eicosenoic acid (11.13%), cis-11-Eicosenoic acid (3.48%), and cis-13-Docosenoic acid (1.64%); saturated fatty acid: n-Hexadecanoic acid (12.89%); saturated fatty alcohol: Docosanol (2.26%). The GC-MS results also confirmed the absence of colorants, toxic solvents, and therapeutic substances in WCO, a cosmetic requirement, according to SFDA. The densities of mixtures of WCO+Coconut oil was 0.801 ± 0.03 mg/mL, and WCO+Olive oil was 0.885 ± 0.01 mg/mL; the kinematic viscosities of WCO+Coconut oil was 21.5 ± 0.0 mm²/s, and WCO+Olive oil was 28.6 ± 0.1 mm²/s at 40℃, which was lesser than the mixtures at 27℃. The decrease in density and viscosity of hot oil mixtures can improve the penetration of oil into the hair filament, which substantiated the traditional application method. In-silico, molecular docking results revealed that all the fatty acid constituents except Docosanol contributed to the hair growth promotion effect by activating MAPK1, MAPK3, and MAPKp38 receptors. This work is the first report on WCO to establish the phytochemical profile to confirm SFDA standard by GC-MS and its in-silico efficiency of activation of MAPK receptors for hair growth.

Essential oils have been used in multiple ways, i.e., inhaling, topically applying on the skin, and drinking. Thus, there are three major routes of intake or application involved: the olfactory system, the skin, and the gastro-intestinal system. Understanding these routes is important for clarifying the mechanisms of action of essential oils. Here we summarize the three systems involved, and the effects of essential oils and their constituents at the cellular and systems level. Many factors affect the rate of uptake of each chemical constituent included in essential oils. It is important to determine how much of each constituent is included in an essential oil and to use single chemical compounds to precisely test their effects. Studies have shown synergistic influences of the constituents, which affect the mechanisms of action of the essential oil constituents. For the skin and digestive system, the chemical components of essential oils can directly activate gamma aminobutyric acid (GABA) receptors and transient receptor potential channels (TRP) channels, whereas in the olfactory system, chemical components activate olfactory receptors. Here, GABA receptors and TRP channels could play a role, mostly when the signals are transferred to the olfactory bulb and the brain.

Hair loss (alopecia) is a universal problem for numerous people in the world. The present study was conducted to investigate the effects of red ginseng oil (RGO) and its major components on hair re-growth using testosterone (TES)-induced delay of anagen entry in C57BL/6 mice and their mechanisms of action. Seven-week-old C57BL/6 mice were daily treated with TES for 1 h prior to topical application of 10% RGO, 1% linoleic acid (LA), 1% β-sitosterol (SITOS), or 1% bicyclo(10.1.0)tridec-1-ene (BICYCLO) once a day for 28 days. Hair regenerative capacity was significantly restored by treatment of RGO and its major compounds in the TES-treated mice. Histological analysis showed that RGO along with LA and SITOS but not BICYCLO promoted hair growth through early inducing anagen phase that was delayed by TES in mice. Treatment of mice with RGO, LA, or SITOS up-regulated Wnt/β-catenin and Shh/Gli pathways-mediated expression of genes such as β-catenin, Lef-1, Sonic hedgehog, Smoothened, Gli-1, Cyclin D1, and Cyclin E in the TES-treated mice. In addition, RGO and its major components reduced the protein level of TGF-β but enhanced the expression of anti-apoptotic protein Bcl-2. These results suggest that RGO is a potent novel therapeutic natural product for treatment of androgenic alopecia possibly through hair re-growth activity of its major components such as LA and SITOS.
