Phytochemical profiling and acute oral toxicity of Suregada zanzibariensis (Baill) root extract

Abstract

Background: Traditional healers utilize the roots of Suregada zanzibariensis for managing diabetes mellitus. Therefore, evaluation of toxic properties of this plant is important. Objective: To evaluate acute oral toxicity of S. zanzibariensis root extract on Wistar rats and to screen phytochemical compounds of the EAESZ. Methods: GCMS analysis of the plant extracts were performed by using GCMS-2010 Shimadzu and mass spectra of the compounds found in the extract was matched with the data in the library of National Institute of Standards and Technology (NIST). Acute oral toxicity testing was carried by administering a single Distilled water extract (DWESZ) and EAESZ to four different groups of rats at dosage of 300mg/kg and 2000mg/kg in each extract to the separately group of rats Results: The GC-MS analysis of S. zanzibariensis roots extract revealed the presence of 10 major compounds. A higher single dose (2000mg/Kg) of EAESZ and DWESZ extract did not produce any sign of toxicity throughout 14 days of study, in terms of changes in behaviour or mortality in tested rats. No significant (p > 0.05) hematological, liver histological, biochemical changes were noticed between rats treated and control rats Conclusion: The results obtained suggest that the plant extract can be classified as non-toxic. Background: Duranta erecta is used in traditional medicine for the treatment of myriad of diseases in most developing countries. The potential safety concerns associated with the administration of hydroethanolic fruit extracts of the plants were investigated in rats. Materials and Methods: Extracts were screened for their possible antioxidant activities by the DPPH scavenging activity. Bioactive compounds present in methanolic extracts of ripe and unripe fruits of D. erecta were identified using Gas chromatography-mass spectrometry (GC-MS). Male and female rats were grouped taking their body weights into consideration to achieve approximately equal conditions among the groups. A freshly prepared solution of DRR or DRU extract was administered orally at 100, 250 and 500 mg/kg b.wt. to different groups while normal group received distilled water daily for 28 days. Toxicity assessment was done using relative organ weight, haematological, and biochemical parameters and histological assessment. Results: The administration of extracts resulted in overall body weight increase, significant change in relative organ weight of the liver, changes in haematological index such as platelet and biochemical parameters namely ALT, AST, ALP, TBil, DBil, IBil, creatinine and urea of the tested group relative to the normal. Histological observations showed normal hepatocytes. Conclusion: The findings suggest that hydroethanolic Duranta erecta fruit extracts is safe but its prolonged use may have some level of adverse effect on the liver.

Seaweed has tremendous potentials as an alternative source of high-quality food products that have attracted research in recent times, due to their abundance and diversity. In the present study, three selected seaweed species commonly found in the Malaysian Peninsular, Ulva intestinalis, Halimeda macroloba, and Sargassum ilicifolium, were subjected to preliminary chemical screening and evaluated for α-glucosidase inhibitory and cytotoxic activities against five cancer cell lines. Chemical composition of U. intestinalis, H. macroloba, and S. ilicifolium methanolic extracts was evaluated by Gas Chromatography-Mass Spectrometry (GC-MS) analysis. Our results revealed the highest total carotenoids (162.00 μg g-1 DW), chlorophyll a (313.09 ± 2.53 μg g-1 DW), and chlorophyll b (292.52 ± 8.84 μg g-1 DW) concentrations in U. intestinalis. In the α-glucosidase inhibitory activity, S. ilicifolium demonstrated the lowest efficacy with an IC50 value of 38.491 ppm compared to other species of seaweed. H. macroloba extract, on the other hand, was found to be the most cytotoxic toward MCF-7 and HT 29 cells with IC50 of 37.25 ± 0.58 and 21.32 ± 0.25 μg/mL, respectively, compared to other cell lines evaluated. Furthermore, H. macroloba extract was also found to be less toxic to normal cell (3T3) with IC50 of 48.80 ± 0.11 μg/mL. U. intestinalis extract exhibited the highest cytotoxicity toward Hep G2 cells with IC50 of 23.21 ± 0.11 μg/mL, whereas S. ilicifolium was less toxic to MDA- MB231 cell with IC50 of 25.23 ± 0.11 μg/mL. Subsequently, the GC-MS analysis of the methanolic extracts of these seaweed samples led to the identification of 27 metabolites in U. intestinalis, 22 metabolites in H. macroloba, and 24 metabolites in S. ilicifolium. Taken together, the results of this present study indicated that all the seaweed species evaluated are good seaweed candidates that exhibit potential for cultivation as functional food sources for human consumption and need to be promoted.

Andrographis paniculata is widely used in traditional herbal medicines for the treatment of common cold, fever and diarrhea, in many regions of Scandinavia and Asia, including Thailand. The pharmacological activities of A. paniculata are mainly attributed to active diterpenoids including 14-deoxyandrographolide, which is uniquely high in first true leaf ethanolic extract (FTLEE)of A. paniculata. In this study, the acute toxicity of the standardized FTLEE of A. paniculata was examined according to the OECD test guideline No. 420. Mice were divided into four groups of each sex and orally received the standardized FTLEE of A. paniculata (0, 300, 2000, or 5000 mg/kg BW). Post-treatment, body weight, signs of toxicity, and/or mortality were observed for 14 days. At Day 15, animals were euthanized, internal organs were observed grossly, and blood samples collected were subjected to hematology and clinical biochemistry analyses. The results showed that all treated animals survived and no apparent adverse effects were observed during the duration of the study. Gross necropsy observation revealed no lesion in any organ of all the standardized FTLEE-treated mice. Although significant alterations in BUN, lymphocytes, neutrophils, hematocrit and hemoglobin were observed, these alterations were not treatment-related toxic effects. Therefore, we concluded that a single oral administration of the standardized FTLEE of A. paniculata with an upper fixed dose of 5000 mg/kg BW has no significant acute toxicological effects.

Background: The plant Rotula aquatica Lour. was traditionally well known due to its large number of pharmacological action and medicinal uses. The plant is a necessary component of many Ayurvedic drug preparations since historical times. It is widely used as a crucial ancient drug for kidney and bladder stones. Objectives: The main objective of the study was to evaluate the acute toxicity and anti inflammatory efficacy of methanolic extract of R.aquatica Lour. in in vivo models. Materials and methods: The qualitative phytochemical analysis and invitro antioxidant activity of the roots of methanolic extract of R.aquatica Lour. (MERA) was evaluated. The acute toxicity effect of MERA was evaluated with two different doses (550, 2000 mg/kg body weight), were administrated orally to Wistar rats. The rats were observed for sign and symptoms of toxicity and mortality for 14 days. The parameters measured including relative organ weight, blood, biochemical and histopathological parameters of hepatic and renal toxicity. The anti-inflammatory effect of MERA was also evaluated in carrageenan and dextran-induced paw edema models. Results: The phytochemical evaluation of MERA shows the presence of secondary metabolites like alkaloids, flavonoids, phenolics and tannins, phytosterols, reducing sugars, proteins and terpenoids. The results of in-vitro antioxidant evaluation of MERA reveal its capability to scavenging free radical at a lower concentration. The MERA did not show any visible signs of toxicity up to the dose of 2000 mg/kg body weight. The results obtained from our carrageenan and dextran-induced paw edema model study also proved the anti-inflammatory effect of MERA in rat model. Conclusion: The result shows the potential of MERA as an anti-inflammatory drug to reduce the signs of inflammation devoid of any toxic effect.

Pistia stratiotes L. commonly known as water cabbage, water lettuce, Nile cabbage, or shellflower. Its leaves are traditionally used against ringworm infection of scalp, boils and syphilitic eruptions. Traditionally, oil extracts is used for treatment of tuberculosis, asthma and dysentery. Eichhornia crassipes (water hyacinth) is an invasive weed that causes serious issues for rivers, lakes, and other reservoirs around the world, although it can be an excellent source for bioactive compounds such as phytosterols and some steroids found in many plants. The aim to study the phytochemical screening from Pistia stratiotes L. and Eichhornia crassipes (Mart.) Solms and analysis of components present in it by gas chromatography-mass spectrometry (GC-MS). The plant were sequentially extracted in different solvents viz., ethanol, methanol, ethyl acetate, petroleum ether, chloroform, acetone, hexane, aqueous, and 1%HCl. The ethanolic crude extract of weed, Pistia stratiotes L. and Eichhornia crassipes (Mart.) Solms showed different types of high and low molecular weight compounds by GC-MS analysis. n-Hexadecanoic acid (7.18%), E-11-Hexadecenoic acid, ethyl ester (1.04%), Hexadecanoic acid, ethyl ester (13.29%), L- Glutamine (0.38%), Linolelaidic acid, methyl ester (2.41%), 9,12,15-Octadecatrienoic acid, methyl ester,(Z,Z,Z) (2.7%), Palmitic acid (12.09%), Phytol (2.12%), 9,12-Octadecadienoic acid, ethyl ester(3.79%), Linolenic acid, ethyl ester (26.26%), Stearic acid, ethyl ester (0.98%), α-Glyceryl linolenate (1.35%), Diisooctyl phthalate (53.84%), Stigmasterol (11.39%), 1-Monolinoleoylglycerol trimethylsilyl ether(1.52%). Most of the isolated and identified compounds by GC-MS in the crude extracts exhibit following bioactivities. Anticancer, Anti-inflammatory, Antimicrobial, Diuretic, Hepatoprotective, Anti-arthritic, Antiasthma, Antioxidant, Hypocholesterolemic, Nematicide, Pesticide, Lubricant, Anti-androgenic, Flavor, Hemolytic, 5-Alpha reductase inhibitor, Insectifuge, Antihistaminic, Anti-eczemic, Anti-acne, Anti-coronary, Antifouling effects so that they can be recommended as a plant of phytopharmaceutical importance. Therefore ethanol extract of Pistia stratiotes and E. crassipes proves as a potential source of bioactive compounds of pharmacological importance.

The normal hematological values in various phases of the rat life provide a valuable guide to researchers and could be useful for experimental works. However, database information available on the literature are incomplete. Aim: This study aimed to present normal hematological parameters of young and aged rats. Methods: Male and female rats were distributed into seven experimental groups with 1, 2, 3, 6, 12, 18, and 24 months of age. Blood samples taken from the tails were analyzed. Normal hematological values were determined for each age group. Results: Rats showed a progressive weight gain with advancing age, predominantly after 3 months of life. With advancing age, differences were found on hematological parameters: some of them showed a progressive rise with age and others did not. Hemoglobin levels and hematocrit did not change while the number of circulating red blood cells suffered slight increase. Conclusion: The present study determined the normal values for absolute and relative hematological parameters in Wistar rats from 2 to 24 months for male and female rats. The results can be used in studies of effects of aging, feeding, and medications on growing and aging rats.

Background Ajuga remota Benth is traditionally used in Ethiopia for the management of diabetes mellitus. Since this claim has not been investigated scientifically, the aim of this study was to evaluate the antidiabetic effect and phytochemical screening of the aqueous and 70% ethanol extracts on alloxan-induced diabetic mice. Methods After acute toxicity test, the Swiss albino mice were induced with alloxan to get experimental diabetes animals. The fasting mean blood glucose level before and after treatment for two weeks in normal, diabetic untreated and diabetic mice treated with aqueous and 70% ethanol extracts were performed. Data were statistically evaluated by using Statistical Package for the Social Sciences software version 20. P-value <0.05 was considered statistically significant. ResultsThe medium lethal doses (LD50) of both extracts were higher than 5000 mg/kg, indicating the extracts are not toxic under the observable condition. Aqueous extracts of A.remota (300 mg/kg and 500 mg/kg body weight) reduced elevated blood glucose levels by 27.83 ± 2.96% and 38.98 ± 0.67% (P < 0.0001), respectively while the 70% ethanol extract caused a reduction of 27.94 ± 1.92% (300 mg/kg) & 28.26 ± 1.82% (500 mg/kg). Treatment with the antidiabetic drug, Glibenclamide (10 mg/kg body weight) lowered blood glucose level by 51.06% (p < 0.05). Phytochemical screening of both extracts indicated the presence of phenolic compounds, flavonoids, saponins, tannins, and steroids, which might contribute to the antidiabetic activity. The extracts, however, did not contain alkaloids and anthraquinones. Conclusion The aqueous extract (500 mg/kg) showed the highest percentage reduction in blood glucose levels and the ability of A. remota extracts in reducing blood glucose levels presumably due to the presence of antioxidant constituents such as flavonoids. The effect of the extract supported the traditional claim of the plant.

Background: In sub-Saharan Africa, chronic kidney disease (CKD) is being recognized as a non-communicable disease (NCD) with high morbidity and mortality. In countries like Tanzania, people access many sources, including traditional medicines, to meet their healthcare needs for NCDs, but little is known about traditional medicine practices among people with CKD. Therefore, we sought to characterize these practices among community members with CKD in northern Tanzania. Methods: Between December 2013 and June 2014, we administered a previously-developed survey to a random sample of adult community-members from the Kilimanjaro Region; the survey was designed to measure traditional medicine practices such as types, frequencies, reasons, and modes. Participants were also tested for CKD, diabetes, hypertension, and HIV as part of the CKD-AFRiKA study. To identify traditional medicines used in the local treatment of kidney disease, we reviewed the qualitative sessions which had previously been conducted with key informants. Results: We enrolled 481 adults of whom 57 (11.9 %) had CKD. The prevalence of traditional medicine use among adults with CKD was 70.3 % (95 % CI 50.0-84.9 %), and among those at risk for CKD (n = 147; 30.6 %), it was 49.0 % (95 % CI 33.1-65.0 %). Among adults with CKD, the prevalence of concurrent use of traditional medicine and biomedicine was 33.2 % (11.4-65.6 %). Symptomatic ailments (66.7 %; 95 % CI 17.3-54.3), malaria/febrile illnesses (64.0 %; 95 % CI 44.1-79.9), and chronic diseases (49.6 %; 95 % CI 28.6-70.6) were the most prevalent uses for traditional medicines. We identified five plant-based traditional medicines used for the treatment of kidney disease: Aloe vera, Commifora africana, Cymbopogon citrullus, Persea americana, and Zanthoxylum chalybeum. Conclusions: The prevalence of traditional medicine use is high among adults with and at risk for CKD in northern Tanzania where they use them for a variety of conditions including other NCDs. Additionally, many of these same people access biomedicine and traditional medicines concurrently. The traditional medicines used for the local treatment of kidney disease have a variety of activities, and people with CKD may be particularly vulnerable to adverse effects. Recognizing these traditional medicine practices will be important in shaping CKD treatment programs and public health policies aimed at addressing CKD.

Objective: Structure and mechanical properties of red blood cells are markedly influenced by pathophysiology of many diseases which in turn potentially impair microcirculatory blood flow. The physiological association between blood rheological parameters and red blood cell indices was investigated in otherwise healthy unselected mid-age women prior to elective gynaecological surgery. Methods: Red Blood Cell-deformability (RBC 1.2, 3.0; 6.0, 12.0; 30.0, 60.0) during exposure to low (RBC 1.2, 3.0), moderate (RBC 16.0, 12.0) and high shear forces (RBC 30.0, 60.0; Rheodyn; Myrenne), -aggregation (MA1; Myrenne) during low shear (E1; 4 -1 S) and in stasis (E0) and plasma viscosity (Pv; KSV 1; Fresenius) were correlated with red blood cell indices (RBC-I: MCV, MCH and MCHC) and subjects' characteristics in 286 healthy women the day before undergoing gynaecologic standard surgery. Women with known pregnancy, malign-, infective-, chronic-disease or extreme BMI (<16; >40 Kg/m2) were excluded from this trial. Results: From June 2014 to December 2014 a total of 286 healthy women (age: 46.5±17.6 y; BMI: 25.5±5.2 kg/m2) were eligible for inclusion into this prospective evaluation. Pv (mean±SD: 1.17±0.12 mPa s) and RBC aggregation (E0:12.6±6.3; E1:17.9±7.3) were not significantly correlated with RBC-I but with age and BMI. In contrast, RBC-deformability correlated significantly with MCV and MCH but significantly inversely correlated with MCHC. Deformability significantly increased with age but was unaffected by BMI of women. The correlation between RBC-I and RBC deformability was most remarkable during moderate shear force exposure. Neither haemoglobin nor haematocrit were correlated with RBC deformability or RBC-I. Conclusions: Cell volume and haemoglobin content had a strong impact on deformability in apparently healthy mid age women, whereas low MCHC and large MCV were associated with an increase in deformability while high MCHC and small MCV correlated with increased rigidity of RBC. BMI had no impact on deformability while age was associated with an increase in all determinants of blood viscosity. RBC aggregability was not affected by MCV, MCHC or MCH in mid-age women.

The effect of oral administration of the leaf extract of Chrysophyllum albidum G. on biochemical and haematological parameters were investigated in albino rats for 16 days. The extract did not show any significant effect (p > 0.05) on the plasma concentrations of total bilirubin, albumin and alkaline phosphatase (ALP) as well as the packed cell volume (PCV), haemoglobin (Hb), red blood cell (RBC), reticulocytes, neutrophils, lymphocytes, eosinophils, basophils, mean corpuscular haemoglobin concentration (MCHC) and mean corpuscular haemoglobin (MCH). The concentration of the platelets was significantly decreased (p < 0.05) at 1000 mg/kg body weight, while white blood cell (WBC) was significantly increased at 500 mg/kg body weight. The doses significantly reduced (p < 0.05) plasma levels of AST, ALT, total protein, glucose and creatinine while urea was significantly increased. While the extract significantly increased the lung, brain and liver-body weights, the kidney, heart, testis, spleen and epididymis-body weights were not significantly affected. The result suggests that the leaf extract of C. albidum contains antiplatelet and hypoglycemic properties and exhibited selective organ toxicity to the rats.

Obesity is characterized by an excess of adipose tissue mass due to an imbalance between caloric intake and expenditure leading to increased body weight and clinical complications such as insulin resistance, diabetes, hypertension and dyslipidemia. These complications not only increase the risk of cardiovascular diseases but are also responsible of certain types of cancer and osteoarthritis. A number of weight reducing drug are available in market, however they produce some serious adverse effects. Momordica charantia is one of the widely used traditional medicinal plants for loss of weight but scientific study has been carried out for that purpose. In this study the aqueous extract of Momordica charantia fruit was orally administered to overweight rats to assess the effect of the extract on body weight of rats. Five weeks treatment not only showed significant decrease in body weight of rats but there was also significant decrease in blood glucose, total cholesterol, triglyceride and LDL-cholesterol however there was increase in HDL-cholesterol levels in serum.

This study was designed to examine the effects of squalene on tissue antioxidant status in isoproterenol-induced myocardial infarction in male albino rats. Levels of diagnostic marker enzymes [alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), and creatine phosphokinase (CPK)] in plasma, lipid peroxides, reduced glutathione, and the activities of glutathione-dependent antioxidant enzymes [glutathione peroxidase (GPx) and glutathione-S-transferase (GST)] and antiperoxidative enzymes [catalase (CAT) and superoxide dismutase (SOD)] in the heart tissue of experimental groups of rats were determined. The prior administration of squalene at 2% level along with feed for 45 days significantly prevented the isoproterenol-induced elevation in the levels of diagnostic marker enzymes in plasma of experimental rats. Squalene also exerted an antioxidant effect against isoproterenol-induced myocardial infarction by blocking the induction of lipid peroxidation. A tendency to prevent the isoproterenol-induced alterations in the level of reduced glutathione and in the activities of glutathione-dependent antioxidant enzymes and antiperoxidative enzymes was also observed. The cardioprotective effect of squalene might be ascribable to its antioxidant property and membrane stabilizing action.

Diabetes mellitus is a growing problem in many developing countries and the financial burden associated with it is enormous. In traditional African communities, majority of people relies on traditional medicines and Traditional Health Practitioners as the primary source of health care. Hence, this study was undertaken in the Lower Eastern province of Kenya to document the medicinal plants used by the traditional practitioners to treat diabetes and to assess the existing knowledge in management of this condition. Data was collected using structured open- and close-ended questionnaires. Thirty-nine species belonging to 33 genera and 26 families were encountered and the most frequently cited species were from Caesalpiniaceae, Ebenaceae, Solanaceae and Labiatae families. Twenty-eight percent of the plant species are reported to have hypoglycaemic activity. Currently there is no data on medicinal plants used to treat diabetes in Kenya. Therefore, these findings are important in the management of diabetes and future research on traditional medicine in drug development.

Selective low (15 mg sodium fluoride (NaF)/L) and relatively high (150 mg NaF/L) doses of in vivo fluoride (F) treatment to Swiss albino mice through drinking water elicited organ-specific toxicological response. All the F-exposed groups showed severe alterations in both liver and kidney architectures, but there was no significant change in the rate of water consumption and body weight. Vacuolar degeneration, micronecrotic foci in the hepatocytes, and hepatocellular hypertrophy were evident in the mice exposed to low dose (15 mg NaF/L for 30 days) while sinusoidal dilation with enlarged central vein surrounded by deep-blue erythrocytes were preponderant when treated with the same dose for a period of 90 days. Blood filled spaces, disintegration of tubular epithelium, and atrophy of glomeruli were also recorded in the kidney of the same treatment group. Change in reduced glutathione level (GSH), glutathione-s-transferase (GST) activity, malondialdehyde (MDA) production in both liver and kidney, disturbances in liver function, induction of heat shock protein 70 (Hsp 70) expression in kidney and its down regulation in liver were positively correlated with histopathological lesion.

In traditional African communities, repellent volatiles from certain plants generated by direct burning or by thermal expulsion have played an important role in protecting households against vectors of malaria and other diseases. Previous research on volatile constituents of plants has shown that some are good sources of potent mosquito repellents. In this bioprospecting initiative, the essential oil of leaves of the tree, Suregada zanzibariensis Verdc. (Angiospermae: Euphobiaceae) was tested against the mosquito, Anopheles gambiae s.s. Giles (Diptera: Culicidae) and found to be repellent. Gas chromatography (GC), GC-linked mass spectrometry (GC-MS) and, where possible, GC-co-injections with authentic compounds, led to the identification of about 34 compounds in the essential oil. About 56% of the constituents were terpenoid ketones, mostly methyl ketones. Phenylacetaldehyde (14.4%), artemisia ketone (10.1%), (1S)-(-)-verbenone (12.1%) and geranyl acetone (9.4%) were the main constituents. Apart from phenylacetaldehyde, repellent activities of the other main constituents were higher than that of the essential oil. The blends of the main constituents in proportions found in the essential oil were more repellent to An. gambiae s.s. than was the parent oil (p < 0.05), and the presence of artemisia ketone in the blend caused a significant increase in the repellency of the resulting blend. These results suggested that blends of some terpenoid ketones can serve as effective An. gambiae s.s. mosquito repellents.

The potential haematological effects associated with the administration of ethanolic leaf extract of AGERATUM CONYZOIDES was investigated in rats. 27 rats were randomly divided into four groups. The first group had 6 rats and served as control, the remaining 3 experimental groups and had 7 rats each. These later groups were gavaged with the extract of Ageratum conyzoides in concentrations of 200mg/kg, 400mg/kg and 500mg/kg respectively for 30days at a dose of 0.1ml/body weight. The control group was gavaged with 0.9% sodium chloride at a dose of 0.1m1/body weight as placebo. The extract at the doses administered was found to increase in a dose-related fashion PCV and Hb (p<0.01 for 200mg/kg and p<0.001 for 400mg/kg and 500mg/kg), RBC (P<0.05 for 400mg/Kg and 500mg/kg) and marginal increases that were not significant (P>0.05 for 200mg/kg); MCH and MCV (P<0.05 and P<0.01 for 400mg/kg and 500mg/kg respectively) 200mg/kg was not significant. MCHC recorded no significant change. WBC recorded marginal increases that were not significant (P>0.05), similarly, the differential white blood cell recorded marginal increases that were not significant, except lymphocytes that recorded significant increase in group 4 (P<0.05). Marginal Decreases in body weight were also observed, these decreases were however not significant. The result of this study thus indicate haematopoietic potentials of the extract and could possibly remedy anaemia.

An ethnomedical survey in Coast, Dar es Salaam, Morogoro and Tanga regions of Tanzania has resulted in the identification of 36 plant species belonging to 21 plant families that are used traditionally for the treatment of Candida infections. Twenty-one plants constituting 58.3% of all collected plants are used to treat of oral candidiasis (Utando) one of the important signs of HIV/AIDS. The knowledge of traditional healers for the treatment of Candida infections has been highly supported by the literature in that 13 (36.1%) out of the 36 plants identified have been proven to be active against Candida albicans and/or other species of Candida. Also, some of the plants were reported to be active against other species of fungi including Cryptococcus neoformans, one of the important pathogenic fungi in HIV/AIDS. It can be seen that ethnomedical information from traditional healers provides a solid lead towards development of new drugs than random screening. The task that remains is to screen extracts prepared from these plants and perform a bioassay-guided fractionation of the active extracts so as to isolate the active compounds from these plants.

An ethnobotanical survey was carried out to collect information on the use of medicinal plants in Southern Western Ghats of India (Madurai district, Tamil Nadu). Information presented in this paper was gathered from the paliyar tribes using an integrated approach of botanical collections, group discussions and interviews with questionnaires in the years 1998 - 1999. The informants interviewed were 12 among whom 4 were tribal practitioners. A total of 60 ethnomedicinal plant species distributed in 32 families are documented in this study. The medicinal plants used by paliyars are listed with Latin name, family, local name, parts used, mode of preparation and medicinal uses. Generally, fresh part of the plant was used for the preparation of medicine. We observed that the documented ethnomedicinal plants were mostly used to cure skin diseases, poison bites, stomachache and nervous disorders. The results of this study showed that these tribal people still depend on medicinal plants in Madurai district forest areas.

An increase in incidences of tinea infections paves the way to discover the novel antifungal drugs from unexplored natural resources. The quality of life in patients with tinea infection may be affected by different factors, including morbidity, length of illness, social and demographic factors. The present investigation explores the functional principle of a bioactive compound isolated from actinomycetes, S. albidoflavus STV1572a by in-silico and in-vitro studies. In continuation of our previous reports on the antidermatophytic potential of S. albidoflavus STV1572a, this study progresses with the in-silico molecular docking study of the seven GC-MS discovered ligands, and six dermatophytic modelled targets. Through virtual screening, it was revealed that a docking score −8.8 between 1-heneicosanol and squalene epoxidase favoured partially in understanding the mode of action. Further validation of in-silico study was performed by a sterol quantification assay which confirmed the antidermatophytic mechanism of 1-heneicosanol. Taken together, the evidence from this study suggests that 1-heneicosanol has a potential antidermatophytic compound and can be considered for dermatophytic treatment.

Abstract Ethnopharmacological relevance The increasing national prevalence of diabetes mellitus (DM) and its complications have overstretched the health care system in Tanzania and influenced patients to use herbal medicines as alternative therapeutic strategies. Therefore, an urgent need exists to validate the safety and efficacy of plants used locally. Aim of the study To identify plants used for the management of DM in Tanzania and analyses their pharmacological, phytochemistry, and safety evidence with a special focus on the mechanism of action. Methods Researchers searched Medline, web of science, and Scopus for published articles. Also, specialized herbarium documents of Muhimbili Institute of traditional medicine were reviewed. Articles were assessed for relevance, quality, and taxonomical accuracy before being critically reviewed. Results We identified 62 plant species used locally for DM management. Moringa oleifera Lam. and Cymbopogon citratus (D.C) stapf were the most mentioned. Fifty-four phytochemicals from 13 species had DM activities. These were mainly; polyphenolics, phytosterols, and triterpenoids. Extracts, fractions, and pure compounds from 18 species had in vitro antidiabetic activities of which 14 had α-glucosidase and α-amylase inhibition effects. The most studied -Momordica charantia L. increased; glucose uptake and adiponectin release in 3T3-L1 adipocytes, insulin secretion, insulin receptor substrate-1 (IRS-1), GLUT-4 translocation, and GLP-1 secretion; and inhibited protein tyrosine phosphatase 1 B (PTP1B). Preclinical studies reported 30 species that lower plasma glucose with molecular targets in the liver, skeletal muscles, adipose tissues, pancreases, and stomach. While three species; Aspilia mossambiscensis (Oliv.) Willd, Caesalpinia bonduc (L.) Roxb, and Phyllanthus amarus Schumach. & Thonn. had mild toxicity in animals, 33 had no report of their efficacy in DM management or toxicity. Conclusion Local communities in Tanzania use herbal medicine for the management of DM. However, only a fraction of such species has scientific evidence. A. mossambiscensis, C. bonduc., and P. amarus had mild toxicity in animals. Together, our findings call for future researches to focus on in vitro, in vivo, and phytochemical investigation of plant species for which their use in DM among the local communities in Tanzania have not been validated.

The stem bark extract of Suregada zanzibariensis afforded a previously undescribed ent-abietane diterpenoid trivially named mangiolide (1) and a known jolkinolide B (2) via anticancer bioassay-guided fractionation. The CH2Cl2:MeOH extract of S. zanzibariensis was initially analysed for its anticancer properties against three cancer cell lines, renal (TK10), melanoma (UACC62), and breast (MCF7) and was found to be potent at low μg/mL ranges. Compound 1, 6α-acetoxy-14-keto-ent-abieta-7(8),13(15)-diene-16,12-olide (mangiolide) inhibited the growth of renal (TK10) with a GI50 of 0.02 μg/mL; a GI50 of 0.03 μg/mL for melanoma (UACC62) and a GI50 of 0.05 μg/mL for breast (MCF7) cancer cell lines. Compound 2, 8,13-diepoxy-13,15-ent-abietene-16,12-olide (jolkinolide B) inhibited the growth (GI50) of the cell lines at 3.31 μg/mL for renal (TK10), 0.94 μg/mL for melanoma (UACC62) and 2.99 μg/mL for the breast (MCF7). The structures were established on the basis of their spectroscopic analysis and the absolute stereostructures assigned using electronic circular dichroism (ECD).

Biosynthesis of long‐chain n‐3 fatty acids from precursors is limited. In vivo effect of squalene (SQ) on the metabolic fate of n‐3 fatty acid precursors in obese/diabetic KK‐A y rodent model was evaluated in our work. Soybean oil, being rich in ALA (18:3 n‐3; a known precursor of EPA/DHA), was chosen as the n‐3 fatty acid precursor rich source. A high‐fat diet (20%) containing 7% soybean oil (SO) was fed to obesity/diabetes‐prone male KK‐A y mice (control). In the case of diets fed to test groups, soybean oil was replaced with 5% SO and 2% SQ. Hepatic DHA levels increased (four fold) in SQ fed group over control (p < 0.05). Gene and protein expressions of Δ ⁵ and Δ ⁶ desaturases, key enzymes involved in the fatty acid metabolism, further supported the results. Also, SQ exhibited a hypotriglyceridemic and hypoglycemic effect. The results clearly indicated the effect of SQ in modulating the n‐3 fatty acid metabolism, including EPA/DHA synthesis in the presence of n‐3 fatty acid precursor. This is the first report of enhancement of in vivo DHA/EPA by SQ and in turn, modulating the physiological fatty acid profile. Practical applications: Squalene (SQ) is an important marine biofunctional material that is found in some terrestrial sources as well. Squalene, being a cholesterol precursor, forms an interesting subject of research for its effect in vivo. SQ significantly enhanced proportions of EPA and/or DHA when their n‐3 fatty acid precursors were available in the diet. The study further establishes the usefulness of SQ in functional food formulations. The work provides an important basis for further evaluation of the role of SQ in normal and disease conditions. KK‐A y mice were fed high fat/sucrose diet to induce obesity/diabetes. Fat source in control diet was lard and soybean oil while experimental group diet contained 2% squalene + 13% lard + 5% soybean oil. Feeding squalene for 4 weeks modulated fatty acid metabolism with increased docosahexaenoic acid (DHA) and decrease in triglycerides (TG), compared to control. The enhanced DHA in the fatty acid profile was supported by upregulated mRNA expression of Δ ⁵ ‐desaturase enzyme and protein expression of Δ ⁵ and Δ ⁶ desaturases (FADS1 and FADS2). Additionally, squalene had a hypoglycemic effect in the mice.

Traditional healers from four wards of the Kilosa district, Morogoro region (Tanzania) were interviewed to assess their knowledge and competence in the management of noninsulin dependent diabetes mellitus (NIDDM). Among 65 reputed healers who were interviewed, 21 (32.3%) recognised and were treating one or more symptoms related to NIDDM, but none could unambiguously relate the symptoms they were treating with NIDDM. Based on what the traditional healers revealed, 54 plants representing 54 species, 48 genera and 33 families were collected. Two of the Kilosa plants, Cassia auriculata and Ricinus communis are reported to have a hypoglycaemic activity, while Securinega virosa has been shown to improve oral glucose tolerance in rabbits. Albizzia anthelmintica has hypocholesterolemic activity and some of the plants also have antibacterial and antifungal activity which may benefit NIDDM patients. Some of the ethnomedical reports of the Kilosa healers, related to management of diabetes, have been reported from other areas. However, one of the reported plants, Albizzia versicolor, is toxic. We conclude that the traditional healers of Kilosa district can not unambiguously diagnose diabetes mellitus as a disease entity, but some of them recognise symptoms characteristic of the disease and they are well-known with plants that may be useful in alleviating its symptoms. Two of the plants they mentioned, Cassia auriculata and Ricinus communis, have a proven hypoglycaemic effect. Careful sieving of the knowledge in the custody of the Kilosa district healers is needed to sort out possibly toxic plants and to determine the other properties of these plants that may be useful in the management of diabetes mellitus.

Context: Polyalthia longifolia (Sonn.) Thw. var. pendula (Annonaceae), a tall evergreen tree, is cultivated all over India. The plant is used in traditional systems of medicine for the treatment of fever, skin diseases, and hypertension. Objective: The present study evaluated the acute oral toxicity of Polyalthia longifolia var. pendula leaf extract in Wistar albino rats. Material and methods: The parameters evaluated daily after oral drug administration of the extract (540, 1080, 2160 and 3240 mg/kg body weight) were mortality, signs of toxicity, feed and water consumption and body weight changes up to 14 days. The effect of different doses of the extract on organ weight, biochemical and hematological parameters were evaluated on the 15th day. Results and conclusion: Methanol extract of Polyalthia longifolia leaf up to the dose level 3240 mg/kg body weight did not produce any toxic effects or deaths; the extract was well tolerated by the rats. It did not alter body weight, feed and water consumption. The organ weight, biochemical and hematological analysis did not show any dose-dependent changes in any of the parameters examined in animals of both sexes. The acute oral administration of the methanol extract of Polyalthia longifolia leaf was not toxic and safe in a single dose.

Squalene is a natural triterpene and an important intermediate of sterol and hopanoid biosynthesis in various types of cell from bacteria to human. Synthesis and further conversion of squalene are key steps in the metabolism of sterols and related components. Here we summarize the recent knowledge of squalene biochemistry, its molecular properties, and its physiological effects. We compare squalene biosynthetic pathways in different cell types and describe biotechnological strategies to isolate this lipid. Finally, applications of squalene in nutrition, pharmacy, and medicine are discussed.

Aberrant DNA hypermethylation is an important mechanism for the inactivation of tumor-related genes in human tumors. Gastric mucosa-associated lymphoid tissue (MALT) lymphomas arise from Helicobacter pylori-associated chronic gastritis; most patients are H. pylori-positive and eradication therapy is highly effective. In the present study, we used methylation-specific PCR to analyze the DNA methylation status of 11 tumor-related genes (Kip2, p16, hMLH-1, p15, p73, MGMT, DAPK, MINT1, MINT2, MINT31 and HCAD) in 21 specimens of MALT lymphoma, 5 specimens of MALT lymphoma with large cell component (high-grade MALT lymphoma), 15 specimens of diffuse large B-cell lymphoma (DLBCL), 8 specimens of complete remission of MALT lymphoma after eradication therapy, 5 specimens with no evidence of malignancy and PBMCs from 10 healthy donors. The average number of methylated genes was significantly greater in gastric lymphomas as compared to normal controls (P<0.001). The CpG island methylator phenotype (CIMP) was observed in 93.3% (14/15) of DLBCLs, 100% (5/5) of high-grade MALT lymphomas and 61.9% (13/21) of MALT lymphomas; in contrast, CIMP was not found in the control group (0%). The average number of methylated genes and the CIMP incidence significantly increased with H. pylori infection. Furthermore, aberrant CpG methylation of specific genes, such as p16, MGMT and MINT31, was consistently associated with H. pylori infection. These findings strongly suggest that H. pylori infection causes the aberrant DNA hypermethylation of specific genes and induces CIMP, which is an important epigenetic mechanism for the development and progression of gastric MALT lymphoma; additionally, our findings provide new epigenetic markers.

A previous report showed that infection with the nematode Nippostrongylus brasiliensis stimulates a basophilia as well as an eosinophilia in the blood of August rats. The present study shows that blood levels of basophils and eosinophils were increased in two other rat strains, one inbred and one outbred, after infection with N. brasiliensis, and infection of two inbred rat strains with Trichinella spiralis also stimulated a basophilia as well as an eosinophilia. No increase occurred in basophils or eosinophils in athymic (nude) rats infected with N. brasiliensis, although both these cell types were found in the blood of control, specific pathogen free, nude rats in numbers comparable to those in specific pathogen free, heterozygote controls of the same strain. Rat basophils usually have few granules and in blood smears often appear as if they were partly degranulated. Basophils from uninfected nude rats contained more negative than positive staining granules compared with basophils from parasitized heterozygotes. The possession of small numbers of granules which vary in their reaction to stains of the Romanowski type is a normal feature of rat basophils in blood smears. Consequently rat basophils differ in these respects from those of other species.

Fifty-three Angiosperm species (Sapotaceae to Zingiberaceae), which are used by traditional healers in five regions of Eastern Tanzania (Coast, Dar es Salaam, Kilimanjaro, Morogoro and Tanga) are listed. For each species, the botanical name, vernacular name, collection number, locality, habit, geographic distribution and medicinal uses are given. Additionally, information from the literature on medicinal uses, chemical constituents and pharmacological effects are also provided. Data analysis and conclusions to cover all the six parts of the papers previously published are presented.

The rapidly increasing diabetes mellitus is becoming a serious threat to mankind health in all parts of the world. The control and treatment of diabetes and its complications mainly depend on the chemical or biochemical agents, but the fact is that it has never been reported that someone had recovered totally from diabetes. With the distinctive traditional medical opinions and natural medicines mainly originated in herbs, the traditional Chinese medicine performed a good clinical practice and is showing a bright future in the therapy of diabetes mellitus and its complications. Based on a large number of chemical and pharmacological research work, numerous bioactive compounds have been found in Chinese medicinal plants for diabetes. The present paper reviews 86 natural medicines with regards to their origin, anti-diabetic active principles and/or pharmacological test results, which are commonly used in the traditional Chinese medical system and have demonstrated experimental or/and clinical anti-diabetic effectiveness. Among these natural medicines, 82 originate from plants and 4 from animals or insects, which covers 45 families. It is strongly significant to pay close attention to traditional Chinese medical therapeutics and natural medicines for treatment of diabetes mellitus and its complications.

Diabetes mellitus is caused due to deficiency in production of insulin by the pancreas, or by the ineffectiveness of the insulin produced. It is a global problem and number of those affected is increasing day by day. The plants provide a potential source of hypoglycemic drugs because many plants and plant derived compounds have been used in the treatment of diabetes. Several medicinal plants have found potential use as hypoglycemic in the Indian system of medicines, including ayurveda. Many Indian plants have been investigated for their beneficial use in different types of diabetes and reports occur in numerous scientific journals. This article aims to provide a comprehensive review on various plant species from Indian biosphere and their constituents, which have been shown to display potent hypoglycemic activity. The use of herbs as hypoglycemic is a major avenue in Indian perspectives particularly for treating diabetes, which require to be explored more effectively as there are so many literatures available on these aspects. This paper describes the chemistry, activity and usage of the constituents isolated from these plants from India for the treatment of diabetes.

The antiinflammatory effect of the aqueous leaf extract of Byrsocarpus coccineus was evaluated using the carrageenan and egg albumin induced rat paw edema, xylene induced mouse ear edema and formaldehyde induced arthritis inflammation tests. The extract administered orally at doses of 50, 100, 200 and 400 mg/kg b.w produced a significant (P<0.05) dose dependent inhibition of edema formation in all four methods used. The results obtained suggest that the aqueous leaf extract of B. coccineus is endowed with effective antiinflammatory activity mediated via either inhibition of phospholipase A(2) (PLA(2)) activity or cyclooxygenase cascade and by blocking the release of vasoactive substances (histamine, serotonin and kinins). These findings seem to justify the use of the plant in traditional African medicine in the treatment of inflammation, including arthritic conditions.
