Functionalizing and derivatising organic molecules is a centerpiece in organic synthesis. Succinctly manipulating and installing acyl moieties in organic molecules spurred the interest of chemists owing to its occurrence in natural products, bioactive molecules, pharmaceuticals, and advanced materials. Traditionally, access to acylation reaction was achieved by Friedel‐Crafts reaction, Schotten‐Baumann, and Vilsmeier‐Haack acylation, however, these protocols own pitfalls. Further to make the acylation process attractive and environmentally friendly, toluene, aldehydes, alcohols, α‐keto acids, amines, amides, esters, ethers, nitriles, alkynes, alkenes, ketenes, N‐acylbenzotriazoles, ketones, thioacids, oximes, thiazolium carbinols, PIDA, diacyl disulfides and acyl salts were used as an acyl surrogates/reagents. Amusingly, these acylating reagents are considered uncommon and alternative to carboxylic acids, acid chlorides and acetic anhydrides. This short review aims to encompass the usage of acylating agents in transition‐metal, metal‐free, light‐driven and other demanding conditions, and thus reveals their practicality.

BACKGROUND The present work describes the facile, fast and economic base‐catalyzed acylation of benzenediols with vinyl acetate. The Hirshfeld surface analysis of benzene‐1,4‐diol O,O′‐diacetate is also described. NaOH, KOH and K2CO3 were the bases used for the catalysis of the acylation of benzene‐1,x‐diols (x = 2, 3, 4) with vinyl acetate to obtain the corresponding O,O′‐diacetyl derivatives. RESULTS The yields were quite good, up to 99% under solvent‐free conditions. This method represents an alternative ‘green’ process for the preparation of O‐acetyl derivatives of hydroxyaryls. The benzene‐1,4‐diol O,O′‐diacetate crystal structure was obtained and described using Hirshfeld surface analysis. A noticeable contribution of the C⋯H contacts is shown (16.7%), despite the insignificant area of the Hirshfeld surface occupied by the carbon atoms (9.4%). By using two‐dimensional fingerprint plots, it was proved that C⋯H contacts were due to the presence of π‐stacking. The O⋯H contacts (41.4%) were the most numerous ones, relating mainly to typical hydrogen bonds (de + di ≈ 2.6 Å). The high saturation of the C⋯H and O⋯H contacts (enrichment ratios (E) = 1.30 and 1.36, respectively), despite their smaller contribution to the overall surface as compared to the H⋯H contacts (E = 0.84), is shown. CONCLUSION The novelty of this work is the development of a method based on an inexpensive, safe and easy‐to‐use catalyst (K2CO3). This method is fast with high yields. Besides, it may be used for acylation at an industrial scale. © 2024 Society of Chemical Industry (SCI).

Mono α‐acylation of acetone has been achieved for the first time by reacting with bench‐stable acyl azolium salts under violet‐LED light at room temperature. The intermolecular hydrogen atom transfer (HAT) from acetone to triplet state of azolium salts under violet LED irradiation resulted in thermodynamically less favourable (Z)‐α,β‐unsaturated ketones with up to 99 : 1 selectivity via C−C bond formation. This compelling protocol access the desired α‐C(sp³)−H acylation product under metal‐, ligand‐ and oxidant‐free conditions on a wide range of substrates.

A synthesis of pyrrolo[2,1-a]isoquinolines based on intramolecular condensation of an enaminone intermediate obtained by C-acylation of an N-alkylated 6,7-dimethoxy-1-methyl-3,4-dihydroisoquinolinium salt was developed. This methodology was further applied to the total synthesis of lamellarin G trimethyl ether from commercially available starting materials compatible with xylochemistry with an overall yield of 26% in 7 steps based on homoveratrylamine.

Acyl moiety is a common structural unit in organic molecules, thus acylation methods have been widely explored to construct various functional compounds. While the traditional Friedel–Crafts acylation processes work to allow viable construction of arylketones under harsh acid conditions, recent progress on developing acylation methods focused on the new reactivity discovery by exploiting versatile and easily accessible acylating reagents. Of them, alcohols are cheap, have low toxicity, and are naturally abundant feedstocks; thus, they were recently used as ideal acyl precursors in molecule synthesis for ketones, esters, amides, etc. In this review, we display and discuss recent advances in employing alcohols as unusual acyl sources to form C-C and C-heteroatom bonds, with emphasis on the substrate scope, limitations, and mechanism.

Chlorination has been used in the fields of organic solar cells (OSCs) for a long time, and impressive progress has been made over the years. Recently developed chlorinated OSCs have achieved an efficiency of over 18%. For better understanding and application of chlorination in the fields of OSCs, we will briefly introduce the general properties of chlorine and recent advances in its introduction and applications in OSCs in this cluster article. Finally, we also provide a short discussion of current questions regarding chlorination in OSCs and future developments in this area.

A diverse range of urea, N-acyl urea, and thiocarbamates has been synthesized in good to excellent yields by reacting N-acylbenzotriazoles individually with amines or amides or thiols or phenols in presence of diphenylphosphorylazide (DPPA) as a suitable azide donor in anhydrous toluene at 110 oC for 3-4 hours. In this route, DPPA is found to be a well alternative of trimethylsilylazide and NaN3 for the azide donor in Curtius degradation. The high reaction yields, one-pot and metal-free condition, straight-forward nature, easy to handling, use of readily available reagents, and in many cases avoid of the column chromatography are the notable features of the devised protocol.

The indole scaffold is a ubiquitous and useful substructure, and extensive investigations have been conducted to construct the indole framework and/or realize indole modification. Nevertheless, the direct selective functionalization on the benzenoid core must overcome the high activity of the C-3 position and still remains highly challenging. Herein, a palladium-catalyzed direct and specific C-7 acylation of indolines in the presence of an easily removed directing group was developed. This strategy usually is considered as a practical strategy for the preparation of acylated indoles because indoline can be easily converted to indole under oxidation conditions. In particular, our strategy greatly improved the alkacylation yield of indolines for which only an unsatisfactory yield could be achieved in the previous studies. Furthermore, the reaction can be scaled up to gram level in the standard reaction conditions with a much lower palladium loading (1 mol %).

A highly chemoselective method is reported for the aroylation of simple diarylmethane derivatives via direct acyl C–C cleavage with 2-acyl-imidazolium salts under transition metal-free conditions. This represents a straightforward way to access a variety of sterically and electronically diverse 1,2,2-triarylethanones, a class of compounds with biological activities and various applications.

From the viewpoint of meeting the current green chemistry challenges in chemical synthesis, there is a need to disseminate how the cocktail of acylation and activation can play a pivotal role in affording bioactive acylated products comprising substituted ketone motifs in fewer reaction steps, with higher atom-economy and improved selectivity. In recent years, a significant number of articles employing the title compounds “aldehydes” as magnificent acylation surrogates which are less toxic and widely applicable have been published. This review sheds light on the compounds use for selective acylation of arene, heteroarene and alkyl (sp³, sp² and sp) C–H bonds by proficient utilization of the C–H activation strategy. Critical insights into selective acylation of diverse moieties for the synthesis of bioactive compounds are presented in this review that will enable academic and industrial researchers to understand the mechanistic aspects involved and fruitfully employ these strategies in designing novel molecules.

Redox-triggered metal-catalyzed cross-coupling reactions from readily available alcohols provide a step economical means to synthesize ketones and related compounds. Prior examples for hydroge-native remote C(sp 3)−H functionalization of olefins via a metal walking process featured external reducing agents. Here, we report a strategy for redox-triggered hydrogenative remote C(sp 3)−H acylation of olefins with primary alcohols both as an acylating agent and a reductant, which is validated by the base-free 1,3-diketone synthesis. Mech-anistic studies have confirmed that the reaction takes place via an aldehyde intermediate and a metal walking process.

Acyl radicals have been generated from α-keto acids using inexpensive and commercially available 2-chloro-thioxanthen-9-one as the photoredox catalyst under visible light illumination. These reactive species added to olefins or coupled with aryl halides via a bipyridyl-stabilized Ni(II) catalyst, enabling easy access to a diverse range of ketones. This reliable, atom-economical, and eco-friendly protocol is compatible with a wide range of functional groups.

Acyl oximes are directly used as the acyl radical precursors in the hydroacylation reactions for the first time. In this work, acyl radicals can be effectively generated via β-scission of a phosphoranyl radical under photocatalytic conditions. As a result, the hydroacylation of alkenes triggered by the resulting acyl radicals leads to facile syntheses of a range of valuable ketones.

A 3,6-di(pyridin-2-yl)-1,2,4,5-tetrazine (pytz) catalysed efficient, mild and metal-free method has been developed for direct amide bond synthesis from simple thioacids and amines as starting materials. This methodology is useful for aromatic, aliphatic, and heteroaromatic thioacids as well as primary, secondary, heterocyclic, and even functionalized amines. A wide substrates scope, operationally mild conditions, and acylation of amines without affecting other functional groups such as alcohols, esters, carbodithioates, among others make this strategy very attractive and practical.
