This study succeeded in inducing the genetic transformation of broccoli (Brassica oleracea var. italica) plants sued direct injection by Ri plasmids of A. rhizogenes R1601, its high concentration 1385.5 ng µl-1 was superior in the percentage for hairy roots production at 54.5%, followed by plasmids with a lower concentration 1094.5 ng µl-1 were its percent 21.6%. When the hairy roots isolated at aggregates and placed on a solid MS medium produced model culture, which characterized by their non-contamination by bacteria. Callus initiated at 50% only from hairy roots achieved from the high concentration of plasmids. The results of the gel electrophoresis of the isolated and amplified DNA samples with Polymerase Chain Reaction (PCR) technology resulted in the appearance of one band for each of the hairy roots and callus samples with a molecular weight of 850bp identical to specific primer used for rol B genes. This results improved the occurrence of genetic transformation by transfer T-DNA genes from plasmids and their inclusion in the cells genome and then successful genetic expression.

Electron microscopic examination of thin sections of infected faba bean leaves showed pinwheels , lamellae , crystals and different components in the cytoplasm of infected cells , as well as satellite bodies in the nucleoli .
