Abstract

In present scenario the research on plants for medicinal purposes is taking place at a phenomenal level. The traditional medicine systems like Ayurveda have been using plant extracts for curing various ailments. Nowadays even allopathic system of medicine is relying on plant extracts for various medicinal compounds. Among many beneficial medicinal plants, one of them is Vitex trifolia. This plant is widely found in coastal areas and is well distributed in Indian coasts as well as the south East Asian coasts. Almost all parts of this plant like leaves, flower, fruit and root are all filled with medicinal properties. It is used for curing ailments like fever, stomach related issues, rheumatic pain and problems caused due to inflammation. It is also used topically for treatment of sore injuries and inflammation. Recent research done on this plant has shown that certain compounds of this plant have potential to act as an anti-cancer drug as well but further research on this area is yet to happen. This paper includes complete description of Vitex trifolia in terms of pharmacognosy, phytochemistry and pharmacology.

INTRODUCTION
Plants have consistently assumed a significant place in innovation and development of both
western and traditional medicine. In traditional systems of medicine like Ayurveda and Siddha plants
have played a significant role in the process of medicine making. Not only in India but in various parts of
the world plants have been used for treating various ailments traditionally. Recently it has been studied
that the majority of the medicinal compounds of allopathic medicine have been derived from plants.
Studies havefound that more than 25% of medicines contain plant-derived ingredients yet only a small
percentage of the plants in the world have been evaluated for potential pharmaceutical use. To
accelerate the discovery of new bioactive compounds from the plants, consistent efforts in the field of
pharmaceutical research are made.Medicinal plants are defined as those plants that are used in treating
and preventing various ailments that harm the well being of humans. Medicinal plants are sources of a
large variety of potent drugs to alleviate infections and various diseases. Hence, the use of plant-based
drugs all over world is increasing. Recent studies indicate that through recent advancements, there have
been great developments in the pharmacological evaluation of various plants used in traditional systems
of medicine. One such plant of great medicinal value is Vitex trifolia. This plant is a shrub, found in the
coastal regions of tropical and sub-tropical countries like India, China, and Indonesia etc. This plant is of
great medicinal value and has been used in traditional medicine since time immemorial. Even in today’s
time extensive studies are done on this plant to study possible bioactive ingredient present in it, so that it
could be used in medicines in future.
Plant Description: Vitex trifolia:
Vitex trifolia Linn. is of the family Verbenaceae. It is also called chaste tree in english and
jalanirgundi in Sanskrit. This plant is found in the tropical and subtropical regions around the world
including India, Sri Lanka, china and Indonesia etc. It has also been found in East African countries.The
plant grows upto the height of 1-3.5 meter. Flowering happens during summer season. Leaves are
variable, simple and usually three foliolate, some leaflets are elliptic or oblongobovate but usually
obtuse. The terminal leaflets are sessile, 5-6.3 by 2.5-3.8 cm in size, the lateral smaller, sessile. There is a
common petioles 1.3-1.6 cm long. The leaves are dark green in colour and bitter in taste.It is a fast-
growing shrub. It is popular for its variegated foliage and beautiful blue flowers. The trifoliate evergreen
leaves are gray-green with white marginal variegation. The soft leaves have grayish pubescence on their
underside and smell pungent when crushed. During the summertime attractive blueflowers with white
spots appear in terminal clusters.

Phytochemistry of vitex trifolia:
This plant is known to possess various active constituents such as essential oils, diterpenes,
vitetrifolins with several pharmacological properties such as antipyretic, antibacterial, works against
asthma and allergic diseases.Five compound are isolated from the fruits of Vitex trifolia and are identified
to be rho-hydroxybenzoic acid, beta-sitosterol, beta-sitosterol-3-O-glucoside, casticin and, 3,6,7-
trimethylquercetagetin. Several oils were extracted from the leaves of the plant that showed
considerable mosquito repellent activity. The active compound identified was rotundinal, a cycloterpene
aldehyde. The plant also showed considerable potential as a botanical pesticide.Vitex trifolia extracts
exhibited anticancer activity on the proliferation of mammalian cancer cells, evaluated by
sulforhodamine B, which is widely used in traditional Chinese medicine. The fruit extracts of this plant
showed analgesic, antipyretic and anti-inflammatory activity. Flavonoids, isolated from V.
trifolia exhibited bacteriostasis activity. Vitetrifoline E, isolated from V. trifolia leaves has been reported
to exhibit tracheospasmolytic activity. V. trifolia exhibited antimalarial activity in the range of 10-100 ×
106 g/mL against Plasmodium falciparum . Aqueous and ethanolic extracts of leaves of V. trifolia were
investigated for hepatoprotective activity against carbon tetrachloride (CCl4) induced liver damage.
Results showed significant reduction in total bilirubin and serum marker enzymes.
Medicinal and Pharmacological importance of Vitex trifolia:
Leaves of vitex are commonly used as poultice for rheumatic pains, inflammations, sprains. It is
used in curing fever, improves memory, favours hair growth, improves vision and treats leucoderma.
Roots of vitex are used in treatment of painful inflammations, cough and fever. Flowers are found useful
in treating fever and fruits in treating amenorrhoea.
Traditionally Vitex trifolia is usd for curing the following ailments:
1. Ring worm Infection: In traditional medicine systemvitex leaves are used to cure fungal infections.
It is done so by using completely crushed leaves mixed with ghee, this mixture is applied on
infected area.
2. Leprosy and Skin Rashes:Vitex leaves and roots are useful in curing leprosy. The leaves of vitex are
ingested along with honey. This according to the traditional medicines is a probable cure for skin
diseases like leprosy. Roots and leaves can also be applied topically.
3. Blood Pitha:Vitex leaves along with several other herbs can be used to treat a problem called blood
pitha according to ayurveda. Vitex helps control blood vomiting ,blood diarrhoea,urinal blood and
other blood related diseases.
4. Joint pains:Extract of Vitex trifolia leavesare given orally to help ease joint pains and sciatica pains.

Other medicinal use which is under research by the western medicine system, are as follows:
Anti- inflammatoryThe Immune-mediated inflammatory diseases generally hold a root cause in
chronic inflammation. Such diseases include rheumatoid arthritis, inflammatory bowel disease, and
psoriasis. V. trifolia is traditionally used for various inflammatory ailments. In traditional Chinese
medicine dried fruits of V. trifolia are used to treat ailments like eyeinflammation, rhinitis and
rheumatoid arthritis. Leaves of V. trifolia are also used to treat inflammatory conditions, caused by
poising from a certain harmful substance. The decoction of the leaves is used to treat oral
inflammation and poultice is used to find relief in rheumatic pain and sprains.
Anti-oxidant Some compounds found in Vitex trifolia exhibit anti-oxidant properties that
safeguard the cells from the oxidation process through which free radicals are produced which lead
to aa chain reaction and results in damage of the cells. One of the main anti-oxidant found in this
plant is ascorbic acid, commonly known as vitamin c which is involved in strengthening immunity
and healing skin related problems and is a significant part of collagen protein. It is involved in
wound healing as well.
Hepatoprotective According to the studies it is found that leaves and flowers of Vitex trifolia
possess potent hepatoprotective activity against carbon tetra chloride induced liver damage. It was
found in the study that Vitex trifolia significantly reduced total bilirubin, total protein and activities
of serum enzymes in the CCl 4 treated groups. Declinein the bilirubin level indicates the
effectiveness of the extract in the normal functioning of the liver.
Anti-fungalAccording to a research several compounds present in Vitex show significant growth
inhibition in various fungal strains of tricoderma and aspergillus genus. But more elaborate and in
depth study still has to be done to prove above theories.
Wound healing- In a recent research it was found that extracts from Vitex trifolia
exhibitedprofound wound healing activity. This was proved by an increase in the wound contraction
rate,decrease inepithelialization period, breaking strength of skin and there was increased
collagenation which showed the signs of speedy wound recovery. This plant is useful not only in
treating cuts but also helps in healing inflammation due to its anti inflammatory properties as well.
Anti-cancer activity-among all the compounds obtained from Vitex, 6 compounds, which are
flavinoids, persicogenin, artemetin, luteolin, penduletin, vitexicarpin and chrysosplenol-D were
isolated and identified for the anti-proliferative effect on mammalian cancer cells. It was found that
the extracts inhibited the growth of cancer cells by inducing apoptosis and inhibiting the cell cycle.
Insecticidal (larvicidal for mosquito)-A crystalline compound named Methyl-p-hydroxybenzoate
extracted from Vitex trifolia showed insecticidal properties and it works exceptionally well in
irradicating the larvae of the mosquito of genus Aedes.
Some other ailments which can be cured by extracts of vitex trifolia are asthama, bronchitis and
tuberculosis. This plant has properties like analgesic, vascular relaxation, trachea-spasmolytic, anti-
helminth etc. In general it is a very useful plant and can be very effective in finding cures of diseases like
cancer if effective research is done on the phytochemistry of this plant.This plant is also used in
treatment of skin diseases, spleen disruption, and rheumatism. This plant is also helpful in regulationof
menstrual cycle and bowel function. It stimulates healing of sore wounds, neutralizes poisons, and
promotes vitality. The juice of the Vitex leaf is used to treat varicose veins and other circulation related
problems. The leaf juice is applied topically to heal chronic sores and is put inside the ear to eradicate
earaches and to cure ear infection. Leaves boiled in water are ingested for weakness and weight loss.
Vitex leaves are also used for curing diseases like malaria due to its larvicidal properties. Vitex is also
useful in curing menstrual problems, and complications related to child birth. Dried leaves are used for
curing insomnia and brain abnormalities.

CONCLUSION
Plants are one of the most extensive sources of medicine for both traditional medicines like
Ayurveda and siddha as well as the western allopathic medicine. On one hand traditional medicine is
purely plant based, on the other hand western system of medicine has also started acknowledging th
importance of plants and now majority of bioactive ingredients present in western medicines are plant
derived ingredients.Vitex trifolia is one such plant which is highly important in traditional medicine. The
bioactive compounds found in leaves, roots, flower and fruits of Vitex are rich in chemicals that help in
treatment and prevention of various ailments like inflammatory diseases, fevers and skin diseases
etc.The plant Vitex trifolia is a flavonoid rich medicinal plant which has been traditionally used to treat
rheumatic pain as well. Also, pharmacological properties have been linked with Vitex which are anti-
microbial, anti-cancerous, insecticidal, anti-inflammatory and hepatoprotective activity.Through the
review it can be concluded that the main phytochemical constituents of Vitex trifolia have anti-oxidant,
anti-inflammatory, anti-microbial, anti- carcinogen, anti-asthmatic and hypatoprotective
properties.Though these findings tell a promising prospect for use of Vitex trifolia in modern medicine
but still a lot of research in this area is required for any substantial incorporation of the compounds of
this plant in medicine. Especially in the field of oncology and hepatology the compounds of this plant can
pose of great benefit in producing anti-cancer and hypatoprotective drugs [1-17].

Vitex trifolia var. purpurea leaves (Lemuni leaves) is considered as a traditional medicine due to their antioxidant property. The antioxidant properties of Lemuni leaves depends on the concentration of ethanol-water solvent. Therefore, this current study intended to identify the antioxidant activity of Lemuni leaves (Vitex trifolia var. purpurea) in different concentrations of ethanol-water solvent extraction. Four ethanol-water concentrations were prepared (water extract (0% ethanol), 25% ethanol extract, 50% ethanol extract, and 75% ethanol extract). The antioxidant properties of Lemuni leaves were evaluated by Folin-Ciocalteu assay, aluminium chloride calorimetric method while 2,2’-diphenyl-1-picrylhydrazyl (DPPH) assay and 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assay were used to measure the antioxidant activities of the extracts. The experimental results showed that 75% ethanol extract had the highest (p<0.05) total phenolic content (94.69 ± 4.75mg GAE/g DW) and total flavonoid content (30.76 ± 2.85mg QE/g DW) significantly. The 75% ethanol extract also had the significantly lowest (p<0.05) IC50 value for DPPH (3.56 ± 0.11 mg/mL) and ABTS (2.01 ± 0.05 mg/mL) assays. Strong correlations were discovered between the total flavonoid content, total phenolic content with antioxidant properties of Lemuni leaves at different ethanol-water ratio by DPPH (r= -0.816; r= -0.824) and ABTS assays (r= -0.929; r= -0.920).The significantly elevated antioxidant properties of the 75% ethanol extract suggest promising applications in health supplements, pharmaceuticals, skincare products, biomedical research, and commercial ventures.

The genus Vitex is also known as a chaste tree, in which it is a large shrub native to the tropical and subtropical regions of the world. A diverse range of species is distributed throughout Southern Europe, the Mediterranean, and Central Asia. The Vitex tree, including its leaves and fruits, has been used for herbal remedies in the form of pastes, decoctions, and dried fruits since ancient times. This article aimed to prepare a comprehensive review of traditional uses and secondary metabolites derived from Vitex sp., including the chemical compounds, biological activities, application of Vitex in human clinical trials, toxicology and safety, marketed products, and patents. The scientific findings were obtained using a number of search engines and databases, including Google Scholar, PMC, and ScienceDirect. Vitex species are well known in pharmacology to have medicinal values, such as anti-inflammatory, antibacterial, antifungal, antimicrobial, antioxidant, and anticancer properties. Previous studies reported that some species are proven to be effective in treating diseases, such as diabetes, and improving female health. A total of 161 compounds from different Vitex species are reported, covering the literature from 1982 to 2022. A chemical analysis report of various studies identified that Vitex exhibited a wide range of phytoconstituents, such as iridoid, diterpenoid, ecdysteroid, and flavonoid and phenolic compounds. Apart from that, the review will also discuss the application of Vitex in human clinical trials, toxicology and safety, marketed products, and patents of the genus. While the extracts of the genus have been made into many commercial products, including supplements and essential oils, most of them are made to be used by women to improve menstrual conditions and relieve premenstrual syndrome. Among the species, Vitex agnus-castus L. is the only one that has been reported to undergo clinical trials, mainly related to the use of the genus for the treatment of mastalgia, menstrual bleeding problems, amenorrhea, menorrhagia, luteal insufficiency, and premenstrual syndrome. Overall, the review addresses recent therapeutic breakthroughs and identifies research gaps that should be explored for prospective research work.

Plants, renowned for their rich reservoir of metabolites, play a pivotal role in addressing health-related issues. The Verbenaceae family stands out, showcasing immense potential in preventing and treating chronic diseases. Vitex trifolia L. ( V. trifolia ), a shrub with a rich history in traditional medicine, particularly in Eastern Asia, has garnered attention for its diverse therapeutic applications. This comprehensive review aims to bridge traditional knowledge and contemporary insights by investigating ethnopharmacology, phytochemistry, and pharmacological effects of V. trifolia . The keyword “ V. trifolia ” and its synonyms were searched within the main scientific databases including PubMed, Web of Science, ScienceDirect, Google Scholar, and Baidu Scholar (from 1974 to 2022, last search: 21.10.2023). Phytochemical analyses reveal a spectrum of secondary metabolites in V. trifolia , including terpenoids, flavonoids, lignans, phytosterols, anthraquinones, and fatty acids. Notably, terpenoids and flavonoids emerge as the main bioactive metabolites. Pharmacological studies validate its therapeutic potential, demonstrating significant antioxidant, anti-inflammatory, hepatoprotective, anticancer, anti-amnesic, antimicrobial, antiviral, anti-malaria, antispasmodic activities, and reported insecticidal effects. Despite existing literature exploring pharmacological attributes and secondary metabolites of related species, a conspicuous gap exists, specifically focusing on the pharmacological activities and novel methods of purification of pure metabolites from V. trifolia . This review aimed to fill this gap by delving into traditional medicinal applications, exploring secondary metabolites comprehensively, and providing an in-depth analysis of pharmacological effects of pure metabolites. Combining traditional uses with contemporary pharmacological insights, this article sought to serve as a crucial reference for future research and practical application of V. trifolia . This approach contributes substantially to understanding the plant, fostering scientific inquiry, and facilitating its broader application in healthcare.

Vitex peduncularis, vernacularly known as ‘Charaigorwa’ is a large evergreen tree having numerous ethnopharmacological uses and belongs to the family Lamiaceae. The species is globally known for its usage to cure multifarious human health issues in traditional as well as contemporary medicines. The species exerts high bioactivity due to the presence of several secondary metabolites, including flavonoids, Vitexin, iridoid glycosides, pachypodol, ursolic acid, 2-hydroxy-ursolic acid terpenoids. Due to the high demands of V. peduncularis in pharmaceutical industries, the species is being exploited indiscriminately, pushing it towards the abyss of extinction. Immediate conservation measures are required to sustain the available natural population through multidisciplinary scientific interventions. This article cumulates the importance of the species directing towards its specialized pharmacological characteristics and its therapeutic uses. It also highlights the research gaps needed to be filled up for judicial use of the species conserving its depleting genetic resources.

These medicinal herbs have immense benefits in strengthening the immune system and also are effective in curing symptoms like sore throat, nasal congestion etc. This medicine can be used as a supplement for immunity enhancement but its effectiveness in curing certain symptoms is still under question as extensive research on this medicine is yet to be done.

Aqueous and ethanol extracts of leaf of Vitex trifolia was investigated for hepatoprotective activity against carbon tetrachloride induced liver damage. To assess the hepatoprotective activity of the extracts, various biochemical parameters viz., total bilirubin, total protein, alanine transaminase, aspartate transaminase and alkaline phosphatase activities were determined. Results of the serum biochemical estimations revealed significant reduction in total bilirubin and serum marker enzymes and increase in total protein in the animals treated with ethanol and aqueous extracts. However significant rise in these serum enzymes and decrease in total protein level was noticed in CCl4 treated group indicating the hepatic damage. The hepatoprotective activity is also supported by histological studies of liver tissue. Histology of the liver tissue treated with ethanol and aqueous extracts showed normal hepatic architecture with few fatty lobules. Hence the present study revealed that Vitex trifolia could afford significant protection against CCl(4) induced hepatocellular injury.

Objective: In this present study the methanolic leaf extract of Vitex trifolia was assessed for its anti quorum sensing activity against acyl-homoserine lactone (AHL)-dependent virulence factors in Pseudomonas aeruginosa. Methods: In this study the methanolic leaf extract of Vitex trifolia was screened to control the virulence factors of Pseudomonas aeruginosa by antibacterial analysis, biofilm prevention assay, protease and elastase assay, pyocyanin production, EPS (Exopolysaccaride) assay, Swimming assay and Light microscopic analysis. Results: The methanolic leaf extract obtained from Vitex trifolia exhibited a concentration dependent (50-100 μg/ml) reduction in anti quorum sensing activity in Pseudomonas aeruginosa. The extract inhibited 69% of protease activity, 48% of elastase activity and 96% of exopolysaccharide production in Pseudomonas aeruginosa. The pigment pyocyanin production and swimming motility of the bacteria were also controlled. Conclusion: Thus in this study the methanolic leaf extract of Vitex trifolia was found to be potent against anti quorum sensing activity in Pseudomonas aeruginosa. © 2015, International Journal of Pharmacy and Pharmaceutical Sciences. Plants and their active constituents play an important role in the prevention of a variety of ailments. Most of the species of the Genus Vitex are used therapeutically in ancient Indian systems of medicine especially, Ayurveda and Siddha. The genus Vitex contains about 270 species distributed around the world. These species contain a variety of potentially bioactive molecules, such as iridoids, flavonoids, diterpenoids, derivatives, and phytosteroids. Most of these species possess analgesic, anti-inflammatory, antimicrobial, antioxidant, hepatoprotective, antihistamine, and antiasthmatic properties. This work reviews the pharmacological evidence for the effects of extracts of plants from the genus Vitex, giving an overview of the most widely studied biological effects and the known phytochemical constituents.

Vitex trifolia var.simplicifolia plant was found growing wildly at the beach, especially at Pantai Sri Tujuh, Tumpat, Kelantan, Malaysia. The extracted leaves were commonly used to prepare traditional dessert among Siamese communities in Kelantan called “Khanom Bai Kunthi” for a long time. Water was used as an extraction agent by the peoples in this area. This extracted leaves were used by the Siamese communities in Tumpat area because it has natural color, flavor and fragrance properties. The Siamese communities who consumed “Khnom Bai Kunthi” was found to be healthy life and live longer. This is the starting point of the study. For the purpose of this study, Vitex trifolia var.simplicifolia leaves were extracted by using 80% methanol in the laboratory. The extracted V. trifolia var.simplicifolia was analyzed and found to be high in antioxidant activity of phenolic compound. The result shows that the extracted V. trifolia var. simplicifolia had high amount of total phenolic compound (44.66μg of GAE/mg of fresh weight). The plants secondary metabolites normally play role in cancer treatment. Cytotoxicity activity of the extracted V. trifolia var.simplicifolia was evaluated on MCF-7 (Breast cancer cell line), HT-29 (colorectal cancer cell line), and WRL-68 (normal liver cell line), and the results show that, cytotoxicity activity (IC50 values 78.87μg/ml, 77.50μg/ml, and 78.29μg/ml, respectively).

A protocol was established for mass propagation of thevaluable medicinal plant Vitex trifolia Linn.(Verbenaceae) through in vitro culture using leaf and internodal explants. The highest percentage (88%) of callus induction from internodalexplants was observed on MS medium supplemented with 1.5mg/l2, 4-D and 0.3mg/l KIN and highest percentage (95%) of shoot induction from callus was observed on MS medium supplementedwith 2.0mg/l BAP and 0.3mg/l NAA. The in vitro raised shootswere successfully rooted in MS medium augmented with 0.5 mg/lIBA. The plantlets were subsequently hardened to green house conditions.

A review of the phytoconstituents of the genus Vitex so far reported, has been presented considering that it is one of the largest genus which comprises of 250 species, and distributed all around the world. Only 28 species have been studied phytochemically. Keeping in view of the potential of the genus, an attempt is made to present a review of the phytoconstituents and biological activities of the genus Vitex, which still remain as a source for lead molecules.
