Abstract

and associated weeds in Latakia Province in Syria. Arab Journal of Plant Protection, 33(1): 50-54.
A survey of Potato virusY (PVY, genus Potyvirus, family Potyviridae) on some solanaceous crops and associated weeds was conducted
between March and November 2013. In this survey, 983 field samples (703 solanaceous crops, 280 associated weeds), and 218 greenhouse
samples (190 solanaceous crops, 28 associated weeds) were collected. Tissue blot immune-binding assay (TBIA) was used to detect PVY in
infected plant samples. TBIA results showed that the natural infection of PVY in Lattakia by 40.72%. The infection rate intested symptomatic
samples was 52.63% in solanaceous field crops and 25.09% in associated weeds, and 21.58% in solanaceous greenhouse crops and 27.27% in
associated weeds.

The effects of seedborne potato virus Y (PVY) in Shepody and Russet Norkotah, two new cultivars that express mild disease symptoms, were determined by comparing tuber yield and specific gravity of plants grown from PVY-infected seed with those of adjoining plants grown from virus-free seed. Seedborne PVY-infected Shepody plants produced 28.7% less total yield in 1994, and 41.1 and 47.3% less in two trials in 1995. Marketable yield was reduced 55.8 and 79.4% in 1995 trials. PVY infection did not affect specific gravity in Shepody. Seedborne PVY infections in Russet Norkotah reduced total yield by 45.5 and 48.8% in the 1994 and 1995 trials, respectively. During the 1995 trial, marketable yield from seedborne infected plants was reduced 65.1% compared with adjacent plants grown from noninfected seed. Additionally, specific gravity was lower for tubers from seedborne infected Russet Norkotah plants than from plants grown from noninfected seed in 1994. Seedborne infected plants of both varieties had fewer and smaller tubers than did plants grown from virus-free seed. No compensatory yield increase was measured for plants originating from PVY-free seed and growing adjacent to seedborne infected plants for either variety. Rather, yield reduction was observed in Russet Norkotah plants growing adjacent to seed infected plants in 1995. Enzyme-linked immunosorbent assay (ELISA) on Shepody in 1995 showed most plants grown from PVY-free seed became infected within 6 weeks of emergence.

This thesis constitutes a comprehensive analysis of the molecular and biological characteristics of three potyviruses (genus Potyvirus, family Potyviridae) naturally occurring in cultivated and wild species of family Solanaceae: Peru tomato virus (PTV), Potato virus V (PVV) and Wild potato mosaic virus (WPMV). In addition, the studies presented in this thesis focus on the genetic variability of isolates of PTV and PVV and on the role of the Potato virus A (PVA) 6K2 protein as a host-specific determinant of virus movement and symptom induction. Determination of the complete genomic sequences of PVV, PTV and WPMV demonstrated that these viruses are typical members of the genus Potyvirus. Furthermore, comparison of the polyprotein amino acid sequences and the biological and serological characteristics of these three viruses supported their current taxonomic position as independent species of the genus Potyvirus. The nucleotide sequences of the P1 protein, coat protein and non-translated regions of European and South American PVV isolates were determined and compared. Results showed limited genetic variability among the European isolates, in contrast to the higher variability found among the South American isolates of PVV. Phylogenetic analysis defined two distinct clusters, grouping the European isolates together but placing two South American isolates to a different group; these two isolates of PVV did not induce a hypersensitive response in an Nv gene-carrying potato cultivar in contrast to the European PVV isolates. Thus, European and South American PVV isolates belong to different strain groups. In addition, great genetic variability was detected among PTV isolates. Analysis of phylogenetic relationships among PTV, PVV, WPMV and other members of the genus Potyvirus commonly found infecting solanaceous crop plants showed that PTV, PVV and WPMV are the most closely related viruses which together with Potato virus Y, Pepper mottle virus, Pepper severe mosaic virus and Pepper yellow mosaic virus constitute a group distinguishable from other potyviruses. Thus, members of this group seem to share a common ancestor. The 6K2 protein of PVA was modified by deleting various portions or by introducing six histidine residues (6xHis) into various positions of this protein. These modifications disturbed functions required for viral infection in Nicotiana tabacum. Furthermore, inoculation of the insertion constructs to N. benthamiana plants did not result in systemic infection with the exception of one plant. This plant lacked typical PVA symptoms but had virus titers similar to the plants infected with the wild type virus: a single point mutation (Gly2 ® Cys2) in the 6xHis-containing 6K2 had restored the viral movement functions. However, partial deletion of the 6xHis-tag to gain the original size of the 6K2 protein was required to restore the induction of symptoms in N. benthamiana and to enable systemic infection of N. tabacum. Taken together, these results indicate the 6K2 is a host-specific determinant for long-distance movement and exemplify that mutations that arise during viral propagation represent a mechanisms by which viruses can evolve and adapt to different hosts.

The modes of evolution of the proteins of Potato virus Y were investigated with a maximum-likelihood method based on estimation of the ratio between non-synonymous and synonymous substitution rates. Evidence for diversifying selection was obtained for the 6K2 protein (one amino acid position) and coat protein (24 amino acid positions). Amino acid sites in the coat proteins of other potyviruses (Bean yellow mosaic virus, Yam mosaic virus) were also found to be under diversifying selection. Most of the sites belonged to the N-terminal domain, which is exposed to the exterior of the virion particle. Several of these amino acid positions in the coat proteins were shared between some of these three potyviruses. Identification of diversifying selection events in these different proteins will help to unravel their biological functions and is essential to an understanding of the evolutionary constraints exerted on the potyvirus genome. The hypothesis of a link between evolutionary constraints due to host plants and occurrence of diversifying selection is discussed.

The requirement for helper component(s) (HC) facilitating virus transmission by living organisms, termed vectors, is characteristic of plant viruses in several genera, potyviruses (genus Potyvirus, family Potyviridae) and caulimoviruses (genus Caulimovirus, family Caulimoviridae) being the most intensively studied groups of viruses. The potyviral HC-Pro fulfils several functions, including aphid transmission, long-distance movement and suppression of post-transcriptional gene silencing. The HC of Cauliflower mosaic virus plays an important role in aphid transmission, whereas no direct role in viral infectivity has so far been attributed to this protein. This review shows the current knowledge of major functions of HCs encoded by potyviruses and caulimoviruses.

Potato virus Y (PVY, genus Potyvirus, family Potyviridae) is transmitted non-persistently by aphids. It causes major losses in potato production (Solanum tuberosum), especially following seed tuber-borne infection of plants. To limit the risk of PVY infection, seed potato production is located preferably in regions where vector pressure is low. The northern-most high-grade seed potato production area (HG zone) of Europe is in Finland. The aim of this study was to determine the incidence of aphid species with documented ability to transmit PVY and to use a modelling approach to determine their relative importance as vectors of PVY in the HG zone of Finland. Winged aphids were caught from six to seven potato fields in each of three growing seasons (2007–09) using yellow pan traps that were examined twice a week. Identification of more than 30 000 individuals indicated that 37% of the aphids belonged to nine species reported to transmit PVY. Incidence of PVY in seed lots was low (0–5.6%) and the seasonal increase of PVY incidence was also low in the potato crops. No potato-colonising aphids were found on the plants in any of the years. The seasonal increase in PVY incidence was modelled using aphid counts in traps, the relative vector efficiencies of the aphids, virus resistance of cultivars, and the initial infection rate of the seed tubers as explanatory variables in generalised linear mixed modelling. Akaike's information criterion was employed to find the best set of explanatory variables for PVY in harvested tubers. Results of this modelling approach showed that the incidence of seed-borne PVY infection and the early-season vector flights are the most important factors contributing to the incidence of PVY in the yield. Compared to models with data from all potential vector species, models containing data from Aphis fabae only showed a better model fit with regard to the incidence of PVY in the harvested tubers. The explanatory power of the models was lost when A. fabae was omitted from the vector data, suggesting that other species play a negligible role as vectors of PVY in the HG zone of Finland. Results can be used to devise appropriate strategies for enhanced control of PVY.

An antiserum against potato Y potyvirus (PVY) was produced and the double antibody sandwich (DAS)-ELISA technique with a detection sensitivity of 10 ng/ml purified virus, was applied. One-step immunocapture (IC) — reverse transcription (RT) — polymerase chain reaction (PCR) was assessed using published primers; a gain in sensitivity of 1000-fold over ELISA was achieved. Furthermore, the use of PCR-ELISA to detect the produced amplicons after their direct adsorption on a microplate offered a supplementary 100-fold gain in sensitivity. Thus, 100 fg/ml purified virus in healthy tobacco extract became readily detectable, as did an infected potato sample at a dilution end point of 10-7, equivalent to 1 ng tissue. This highly sensitive procedure coupled with the print-capture (PC) modification to avoid specific sample preparation, offered a most handy and useful tool for effective PVY detection in single aphids(Myzus persicae).
