Monitoring of genealogical structure development of East Bulgarian breed horses originating from stallions of other sporting breeds

Abstract and Figures

The origin of horses from the East Bulgarian breed, which descent from the other sport breeds stallions used for grading has been investigated. Genealogical structure development was traced by pedigree analyses of 1123 horses, born during the period from 2000 to 2018. The East Bulgarian Horse Association set of statistical data, concerning stallions' breeding activity and productive life of their progeny were used. Two new lines with founders Hanoverians Da Kapo (GER) and Eistanzer (GER) were set up during the last four generations. Nowadays the line of stallion Da Kapo (GER) has developed in second and third generation after the founder and the line of stallion Eistanzer (GER)-in fourth generation after the founder. A small bulk of genealogical groups with origin traced to stallions Ladykiller, Cottage Son, Cor de la Bryere and Alme Z have developed.

Introduction
The East Bulgarian horse is a national breed with an endangered
status. The Stud Book is recognized by the World Breeding Federation
for Sport Horses (WBFSH). The main method for improving the traits
that are subject of selection is pure breeding. The genealogical struc-
ture and variation of genealogical units existence is the subject of nu-
merous scientic studies, the results of which are used in the design
of the respective breeding programs of populations (Popov, 1959;
Sabeva, 1991; Kierkegaard, 1993; Silvestrelli et al., 1993; Flanagan,
1993; Zechner et al., 2002; Sabeva, 2007; Sabeva and Kaschiev, 2010;
Barzev, 2011; Vicente et al., 2012; Bokor et al., 2013; Hamann and Distl,
2014; Medeiros et al., 2014; Popova, 2017).
By the early 1990s, horses of the East Bulgarian breed have
been tested in at races and steeplechase. When evaluating the
breeding value, the data from their participation in equestrian sport
disciplines was complementary. Over the last three generations, the
increased demand for horses suitable for jumping has led to a wider
application of grading with stallions from acknowledged sport breeds.
Study of Sabeva (2009) showed that in terms of body measurements
and development of the bone system, the descendants of the used
Hanoverian stallions signicantly exceeded the mean of the breed.
The aim of the current study is to establish the existence of new
genealogical lines from stallions representing acknowledged sport
breeds which were used in the population for grading of the East
Bulgarian horse.
Material and methods
Subject of the study was to trace genealogical belonging of horses
from the East Bulgarian breed, which on male side descend from the
other sport breed representatives. Genealogical structure develop-
ment was traced by pedigree analyses of 1123 horses, born during
the period from 2000 to 2018. The East Bulgarian Horse Association
set of statistical data, concerning stallions’ breeding activity and pro-
ductive life of their progeny were used.
Results and discussion
Two new lines with founders Da Kapo (GER) and Eistanzer (GER)
were set up during the last four generations:
Line of the Hanoverian stallion Da Kapo (GER) – Devils Own
Da Kapo1977 (Direx1966 and Heidschnucke) was imported from
former East Germany. He originated from the line of thoroughbred
Stallion Devils Own1887 through the branch of the Duel II1959, which
produced horses with exceptionally good sport qualities. Da Kapo
himself had a very successful sports career in Bulgaria as a jump-
er. He produced 36 East Bulgarian foals, including 22 females.
Based on official statistics, 6 of his sons were used for breeding.
Nowadays the line is developing in the second and third genera-
tions after the founder (Figure 1).

Line of the Hanoverian stallion Eistanzer (GER) – Adeptus
Eistanzer1985 was imported from Germany and originates from
Eistvogel and mother Pera, the daughter of a renowned in the past stal-
lion Adept. He was one of the most widely used stallions in our country
during the period 1988-2002. Eistanzer gave over 102 foals, 79 (includ-
ing 42 females) of which were representatives of the East Bulgarian
breed. From the male descendants, 7 of his sons were used for grading.
Nowadays, the line develops in second, third and fourth generation after
the founder, through the progeny of Avantaj by Jenniffer (GER), Aden by
Erigona and Argo Men by Membrana (Figure 2).

Adeptus
A characteristic feature of sport horse-breeding at the present stage
of breeds development is that the designation of stallions to a given
breed is made depending on the licensed test they passed through and
sometimes on the willingness of the owner. That is why in the genealog-
ical development schemes there is breed designation of the representa-
tives other than that of the founders, that of the parents or that of the other
ancestors. Genetic plasma from some specialized lines of Hanoverian,
Holsteiner, Trakehner, KWPN, Zangersheide and other breeds has an
international spreading. During the last decades, the large scale imple-
mentation of articial insemination has begun with deep-frozen semen
from world-wide stallions. A few genealogical groups in East Bulgarian
breed with origin from famous founders are set up:
Genealogical group of thoroughbred stallion Ladykiller (GB)
Ladykiller1961 (GB) was born in England out of Lone Beech and fa-
ther Sailing Light from the Fairway line. He was imported to Germany,
where he became one of the leading stallions in the Holsteiner breed.
Ladykiller left behind 35 licensed stallions. His sons, Landgraf I1966 and
Lord1967, had no less contribution to the creation of the modern type of
Holsteiners. Semen from 7 stallions was imported from the branch of
Landgraf I, numerous offspring gave the stallion Limnos (GER). The
East Bulgarians Lafaet2004 (Leonardo(GER) and Tafta from Tigar) and Lord
Primero2008 (Limnos(GER) and Primera from Don Primero) have been
rarely used for breeding during the studied period. The imported stal-
lions Lamar1999 (GER) and Lancelot2001 (GER) were representatives of
the Lord’s branch. From the same branch a deep-frozen plasma was
used from Lord Granus2007 (GER). During the past 11 years 49 horses
from the line took part in breeding tests for young East Bulgarian horses.
Genealogical group thoroughbred stallion Cottage Son (FR)
Cottage Son1944 (FR) originates from mother Wait Not and father
Young Lover from the Dark Ronald’s line. Young Lover is half-sib (by Son-
in-Law) of Tihany, a founder of one of the main lines of the East-Bulgarian
breed. Cottage Son acted in the Holsteiner breed, and his descendants
displayed very good qualities in the military. In Bulgaria the line is repre-
sented through the branch of Capitol I1975 and his sons Calato1987, Cassini
I1988, Cento1988, Cassini II1994 and Concens1994. The imported from Germany
Camargo2001 (GER), Cardinal Lamberto2004 (GER) и Carre2005 (GER) left
behind numerous descendants. From the line 35 horses participated in
the breeding tests for young East Bulgarian horses.
Genealogical group of stallion Cor de la Bryere (FR)
Cor de la Bryere1968 (FR) is the son of the thoroughbred Rantzau
(FR) from the line of Dark Ronald, through the Son-in-Law by branch
of Fox Law. His mother Quenotte, as well as himself, belong to the
Sell Francais breed. Deep-frozen plasma from the branches of the
6 sons of Cor de la Bryere in the Holsteiner breed - Calypso II1974,
Caletto I1975, Caletto II1978, Corrado I1985, Carte Dor1987, Calypso I1989
and stallion Kotrak1997 (GER) was used in the East Bulgarian breed. 32
horses from the line participated in the breeding tests.
Genealogical group of stallion Alme Z (FR)
Alme Z1966 (FR) is a representative of the Sell Francais breed. He
originates from Ibrahim, grandson of the thoroughbred Orange Peel
(the old line St. Saimon) and mother Girondine out of the thoroughbred
granddaughter of Blandford – Ultimate. Alme Z by his descendants has
had signicant inuence on the development of sport horse-breeding in
Europe (420 mares during the period 1971-1990). He is the only stallion
that has produced 2 World and 3 Olympic champions. His licensed sons
founded branches of the line in Zangersheide, Holsteiner, Hanoverian
and others sport breeds. Stallions from the branches of Galobet A1972,
Alexis Z1977, Ahorn Z1979 и Quidam de Ravel1982 were used for the grading
of the East Bulgarian breed (Figure 3). During the period 2006-2018, 23
horses with origin traced to Alme Z participated in the breeding tests for
young East Bulgarian horses.

Conclusion
Two new lines have formed during the last four generation in-
tervals, with the Hanoverian stallions Da Kapo (GER) and Eistanzer
(GER) being the founders. Currently the Da Kapo (GER) line is devel-
oping in second and third generation after the founder. The Eistanzer
(GER) line is developing in fourth generation after the founder. A small
bulk of genealogical groups with origin traced to stallions Ladykiller,
Cottage Son, Cor de la Bryere and Alme Z have developed.

This study aimed to investigate the dietary mineral and vitamin supplementation on the hematological, the circulating biochemical parameters, and the reproductive performance of cyclic mares aged 4–12 years old. Purebred Arabian (PAB, n = 8) and East-Bulgarian (EBB, n = 12) breeds were equally divided into treated and control groups. The nutritional supplement was given orally (50 g daily/animal/day) for seven consecutive days and repeated for 4 consecutive months. All mares were examined daily with ultrasound from exhibiting oestrus signs till ovulation. Blood samples were collected with and without anticoagulant before supplementation, on Day 7 and Day 120 for the measurement of ALT, lactate dehydrogenase, bilirubin, calcium, phosphorus, and creatinine and to perform full blood picture. The nutritional supplement decreased lactate dehydrogenase (p < .05) and creatinine (p < .05). Compared to the PAB mares, control mares had one day shorter oestrous cycle length (20.2 ± 2.13 vs 21.0 ± 2.46 days), but the treated EBB mares had two days shorter oestrus cycle (20.00 ± 1.202 vs 22.00 ± 0.93 days). The oestrus phase of the treated mares was shorter (p < .001) than that of the control mares (3.63 ± 0.38 vs 5.50 ± 0.31 days). The diameter of the dominant follicle varied (p < .05) between the two breeds that was 38.25 ± 1.90 and 36.94 ± 1.70 mm for treated and control EBB but was 35.38 ± 1.41 and 34.89 ± 1.35 mm for treated and control PAB. In conclusion, the nutritional supplement had a good effect on the reproductive status, and on the blood parameters (hematological and biochemical) of the mares, but more research is necessary to recommend its use. • Highlights • The addition of the nutritional supplementation to the mares from the East-Bulgarian breed lead to shorter oestrus. • There was a tendency to shorten the sexual cycle (up to 2 days) and increase the size of the dominant follicles in mares receiving the supplement.

In the mares of purebred Arabian horse (n-8) and East Bulgarian horse (n-12) breeds, raised in the State stud farm `Kabiuk` near Shumen, Bulgaria, were studied basic hematological and biochemical blood parameters in the beginning of spring (March). It was established that the average levels for the mares from both breeds are as follows: alanine aminotransferase ALT U/L - 73.00±14.47, lactate dehydrogenase LDH U/L - 1523.88±93.11, creatinine Creat. U/L -284.56±53.08, calcium Ca U/L - 2.06±0.4, total bilirubin TBL U/L - 78.40±12.45, phosphorus P U/L - 6.63±0.51, white blood cells WBC - 8.87±0.71, lymphocyte count Lym - 32.94±2.48, monocytes Mon-3.98±0.30, granulocytes Gra -61.45±2.74, red blood cells RBC - 8.01±0.14, erythrocyte volume MCV - 67.07±28.42, hematocrit Hct - 35.12±1.54, hemoglobin MCH - 12.34±0.16, erythrocyte distribution width RDW-16.90±1.42, hemoglobin Hb-11.16±0.67, platelets THR -255.17±40.66, platelet volume MPV -7.81±0.19, platelet distribution width PDW-9.22±0.30, hemoglobin in erythrocytes MCHC - 27.29±0.29. The breed is a reliable (p<0.01) source of variation of LDH, with higher values found in the East Bulgarian breed (EBB). The mares of different ages can differ significantly (p<0.05) in the content of Mon in the blood. The age group in the breed had a significant effect (p<0.05) on Ca and PDW, with the oldest mares from EBB (10-12 years old) had decreasing amount of Ca and increasing amount of POW.

Genetic diversity and population structure were analyzed in the Lusitano horse breed based on pedigree information of animals registered in the Studbook, to identify factors which may have affected the genetic variability of the breed, and provide the bases for the establishment of sustainable utilization programs. Pedigree records collected from 1824 to 2009, including information on 53,411 animals, were used in the analyses. The mean generation interval was 11.33±5.23 and 9.71±4.48 years for sires and dams, respectively, while the mean number of offspring registered was 13.13±22.53 for stallions and 4.00±3.38 for mares. All great-grandparents were known for the animals in the reference population (registered foals born from 2005 to 2009, n=9712), which had a mean number of equivalent generations known of 11.20±0.71 and an average inbreeding of 11.34±7.48%. For this population, the rate of inbreeding per year was 0.173±0.070, and the effective population size computed from this rate was about 28. The mean relationship among animals from the same and from different studs was 0.31±0.16 and 0.15±0.10, respectively. In spite of the high within-stud relationship, inbreeding has been kept lower than expected due to restrictions imposed by breeders on selection and allocation of mates. The effective number of founders, ancestors and studs contributing to the current genetic pool was 27.5, 11.7 and 5.4, respectively. Over the last 30 years, contributions to the genetic pool of Lusitano have been decreasing, with a reduction to about one-half in the effective number of founders and ancestors. Of the 267 founder sires, only 9 Y-chromosome sources are currently represented, with an effective number of 2.23. The diversity of mt-DNA sources is broader, with 129 lineages represented and an effective number of 42.8. The results of our study reflect the intensive emphasis that has been placed on a few sire-families over the years, and raise concerns regarding the conservation of genetic diversity for the future. Methods designed to minimize inbreeding and maximize the maintenance of genetic contributions from different founders and ancestors should be envisaged, to prevent further losses of genetic variability in the Lusitano horse breed.

The aim of the study was to analyse the pedigree information of Thoroughbred horses which were participating in gallop races between 1998 and 2010 in Hungary. Among the 3043 individuals of the reference population there were imported animals from foreign countries (e.g. Germany, England or Ireland) and horses that were born in Hungary. The number of complete generations was 15.64 (varied between 0 for the founders and 25.20), the mean number of full generations was 6.69, and the mean maximum generations were 28.96. The number of founders was 1062, and the effective number of founders was 42. Two hundred and thirty-two founders were born before 1793 (when the stud book of the Thoroughbred breed was closed), therefore these founders are considered as true founders of the breed. These 232 founders were responsible for 88.58% of the gene pool in the reference genome. The significant difference between the number of founders and effective number of founders indicate that the genetic diversity decreased greatly from the founders to the reference population. The number of ancestors was 908 and only 6 of them were responsible for 50% of the genetic diversity in the examined population. The effective number of ancestors was 15.32. From the ratio of the effective number of founders and effective number of ancestors we concluded to a bottleneck effect that characterizes the pedigree under study. Generation interval was more than a year longer for stallions (12.17) than it was for mares (10.64). More than 94% of the individuals in the pedigree were inbred, and the average inbreeding of the population was 9.58%. Considering the changes of the inbreeding status of the examined population 4 large time periods were appointed. The first lasted until 1780, the second period was from 1780 until 1952, the third period was between 1946 and 1998 and the last one was from 1998 until 2008. Rate of inbreeding in the last generation was 0.3%, which forecasts further increase in inbreeding. The effective population size was above 100 in the last 30 generations, proving the genetic diversity did not decrease by a level that would make long-term selection impossible.

The pedigrees of 565 Lipizzan horses from eight European studs were traced back to the individuals considered as the founding population of the breed. The length of pedigrees was up to 32 generations, with an average of 15.2 complete generations. Average inbreeding coefficients varied from 8.6 to 14.4% between studs, with an overall mean of 10.8%. Increase in inbreeding was relatively small in the last generation. Other measures of genetic variability based on contributions of founders or important ancestors were also calculated, leading to partly different conclusions about the variability of the population at stud level. Average coancestries between studs showed that the two Romanian studs were most similar but also that the Austrian, Slovenian and Italian studs formed a cluster of genetically similar individuals. The pedigrees were also used to calculate gene proportions of founder breeds and individual founders. About 52% of the current genes are of Spanish or Italian origin, 21% Arabian, 8% Fredriksborg, 4% Kladruby, 3% English and 2% Shagya-Arab. The most important founder individual was Toscanello Hedera, followed by Neapolitano, one of the founders of the classical stallion lines, each contributing more than 6% of the current gene pool.

A data set constituting a total of 310,109 Hanoverian warmblood horses was analyzed to ascertain the genetic variability, coefficients of inbreeding, and gene contributions of foreign populations. The reference population contained all Hanoverian horses born from 1980 to 2000. In addition, Hanoverian stallions born from 1980 to 1995 and Hanoverian breeding mares from the birth years 1980 to 1995 with registered foals were analyzed for the same genetic parameters. The average complete generation equivalent was approximately 8.43 for the reference population. The mean coefficient of inbreeding was 1.33, 1.19, and 1.29% for the reference population, stallions, and breeding mares, respectively. The effective number of founders was largest in stallions (364.3) and smallest in the reference population (244.9). The ratio between the effective number of founders and the effective number of ancestors was 3.15 for the reference population, 3.25 for the stallions, and 3.06 for the breeding mares. The effective population size in the Hanoverian warmblood reference population was 372.34. English Thoroughbreds contributed nearly 35% of the genes to the Hanoverian reference population and even slightly greater contributions (39%) to the stallions. Trakehner and Arab horses contributed approximately 8 and 2.7%, respectively, to the Hanoverian gene pool. The most important male ancestors were Aldermann I from the A/E line, Fling from the F/W line, and Absatz from the Trakehner line, whereas the breeding mare Costane had the greatest contribution to the reference population, stallions, and breeding mares. From 1996 onward, the stallions Weltmeyer and Donnerhall had the largest genetic impact on the Hanoverian horse population.
