Abstract

The purpose of this study was to investigate the effect of eight weeks of high intensity interval training (hiit) on osteopontin and some bone mineral indexes (calcium, phosphorus, magnesium, iron) on active young women. Materials and methods: in this semi-experimental study, 16 active women were selected and divided into two groups: hiit with mean age 22.2±1.2 years, height 162.8±6 cm, weight 57.6± 9 kg, and control with average age of 22.3±4 years, height 156.2±4 cm, and weight 54.6±7 kg. Running- based anaerobic sprint test (rast) with a 3-minute break-in interval, which was added every two weeks. Blood samples of participants were taken before and after the protocol, data were analyzed using independent t-test (p≤0.05) was analyzed. Results: the results showed a statistically significant difference between the two groups in the osteopontin index (p = 0.0001). The results also showed that there was no significant difference between calcium (p =0.259), magnesium (p =0.19), fe (p =0.883) and phosphorus (p =0.153) in the young women of the two research groups. Conclusion: the results of this study showed that the implementation of 8 weeks hiit exercises improved bone mineral density.

Abstract

Introduction
The bone is a transplanted tissue that together with the cartilage forms the body's skeleton. This tissue is
composed of osteoblastic cells and bone matrix. Unlike other transplanted tissues, the bone matrix has the unique
ability to calcify. One of the major components of bone marrow is osteopontin. Osteopontin which was formerly
called bone sialoprotein, secreted phospho protein-1, and activator-1 t cells, is an acidic glycoprotein and an
important factor in bone turnover and osteoclast tightness, cell adhesion, body chemistry control, cell retention,
tissue alteration, and inflammation regulation (kiefer, zeyda et al. 2008; de fusco, messina et al. 2017).
Osteopontin is a protein at very low levels in normal skeletal muscle. However, the induction of muscle damage
has been reported in the increase of 100 equalities of the opn transcription site (hoffman, gordish-dressman et al.
2013; yang, ramachandran et al. 2014).
On the other hand, the bone is interacting with many external and internal factors that affect calcium
homeostasis, half of the bone tissue is formed by hydroxyapatite, carbonate, citrate, magnesium, sodium, and
steronium crystals. Give that there are many factors affecting the production of osteoblast cells and restoration of
osteopontin of a person whose genetics and race are the most important, but nutrition and exercise are among the
most important factors that one can control them (menuki, mori et al. 2008). The results show that performing
appropriate sporting movements before reaching the maximum bone density can produce 2 to 20% difference in
bone density (nordström, karlsson et al. 2005).
Sport activity is a preservative and stimulant of bone formation, which strengthens muscles and
improving individual balance through the accumulation of minerals, and also can reduce the risk of bone
fractures. In fact, exercise activities in two ways lead to the transfer of force to the bone: muscle tension and
gravity. These forces can increase bone density; however, people who have active life, have a significantly
higher bone mass compared to their non-aged people, and their pre-eminence is preserved until the seventh and
eighth decades of their lives. Appropriate intensity, regular exercise and balanced exercise, along with proper
nutrition, have a beneficial effect on bone density (ginty, rennie et al. 2005; zernicke, mackay et al. 2006; barry
and kohrt 2008; moayyeri 2008; langsetmo, hitchcock et al. 2012; alghadir, gabr et al. 2015).
Still, no studies have been conducted on the effects of intense periodic exercises on osteopontin and
bone mineral density, and because of the limited studies in the field of the effect of these exercises on the
performance indicators of athletes, according to if the positive effects of such exercises are confirmed to the
athletes, it can be used as an appropriate training protocol to improve the performance and physiological indices
of athletes. However, the difference in the effect of these exercises on osteopontin and bone mineral density
(calcium, phosphorus, magnesium, iron) is not yet clear in people who are on a regular basis. Therefore, the

present study intends to investigate the effect of eight weeks of hiit on osteoponetin and some bone mineral
indices in active young women.
Materials and methods
The present study was semi-experimental and conducted as a pre-test and post-test design. The study
was approved by alzahra university faculty exercise physiology research ethics committee and enrolled
participants gave their written informed consent. The statistical population of this study was the young female
activists of alzahra university in the age range of 20 to 24 years old, among them 16 female students volunteered
to participate in this study. Individual data, their medical and sport records were collected. Also, in an individual
session, the anthropometric and physiological characteristics of the subjects were also measured. Subjects were
randomly divided into two groups of hiit and control with 8 people due to the criteria of no drug and sports
supplements usage, no history of physical and metabolic disease and non-smoking.
Exercise protocol
The exercise program included 8 weeks of 3 sessions per week for 20-30 minutes at alzahra university
under the supervision of a trained specialist and supervised by the researcher. The protocol was started with 3
sets consisting of 6 repetitions of 35-meter distance (80% of maximum heart rate), while in the second week, 3
sets were maintained, but the intensity increased to 85%. The rest was 10 seconds between repetitions and 3
minutes between sets. The third and fourth weeks there were 4th sets, and at the fifth and the sixth week, there
were 5 sets with full power.
Blood samples were taken from nurse's sitting position for 48 hours before and after the exercise period
at a rate of 5 milliliters of blood from the right hand vein of the subjects. Blood samples were immediately
placed in a centrifuge machine at a rate of 3000 rpm for 20 minutes and frozen at -70 ° c.
serum osteopontin levels were measured using osteopntin kit (opn, zellbio, ulm, germany), with a coefficient of
variation of 4.9% and a sensitivity of 0.2 ng / ml, serum calcium levels was measured using pars-chlorometric
enzymatic test kit with a coefficient of variation of 1.9% and a sensitivity of 0.04 mg / dl, serum levels of
phosphorus using the pars- calorimetric enzymatic test kit with a coefficient of variation of 0.8% and a
sensitivity of 0.3 mg deciliters, serum levels of magnesium were measured using the pars- calorimetric
enzymatic test kit with a coefficient of variation of 0.9% and a sensitivity of 0.05 mg / dl and serum levels iron
was measured and analyzed using pars- calorimetric enzymatic test kit with a coefficient of variation of 3.2%
and a sensitivity of 5 milligrams per deciliter. All stages related to the measurement of relevant factors were
performed at the laboratory of endocrine sciences and research institute of endocrine sciences in tehran province.
Using the kolmogorov-smirnov test, the natural distribution of data was analyzed. Then independent t-test was
used to check the changes between the two groups. All statistical research was performed using spss software
version 22 and significance level of p <0.05.
Results
The descriptive characteristics of the subjects are presented in table 1. The results showed that there was
a significant difference between the amount of osteopontin in the two research groups (p = 0.0001). But, the
results also showed that there was no significant difference between calcium (p =0.259), magnesium (p =0.19),
fe (p =0.883) and phosphorus (p =0.153) in the young women of the two research groups.

Discussion
The results of this study showed that eight weeks of hiit caused a significant increase in serum
osteopontin in active women. This result of the present study is consistent with the findings of some of the
previous findings (hoffman, gordish-dressman et al. 2013; barfield, uaesoontrachoon et al. 2014; marędziak,
śmieszek et al. 2015). The amount of osteopontin is altered by various conditions such as the type of exercise,

the intensity of exercise, and how to practice based on the type of genotype of individuals (barfield,
uaesoontrachoon et al. 2014). Increasing the amount of osteopontin in the present study indicates that the
exercise protocols used in this study have increased the density and bone mineralization of the subjects.
However, the results of the present study are contrary to the findings of duggan et al. Probably the reason for
inconsistent of the results of this study are related to the type of subjects and the type of protocols. In a study by
duggan et al., the effect of aerobic exercise on serum osteopontin was evaluated for 12 months as an
inflammatory factor in obese people. Their results showed that serum osteopontin in the experimental group did
not have a significant difference with the control group (duggan, xiao et al. 2014). The type of subjects in this
study was young women. Differences in the age, type, intensity and duration of exercise protocols used in this
study and most importantly, the subjects in this study were not obese and had good fitness can be the reasons for
difference between the results of the research and the results of the present study. Several stimuli coexist during
exercise and provide the background of the skeletal muscle to be vascular while the most important of which are
skeletal muscle hypoxia, increased blood flow, or shear stress, vascular expansion due to increased lacrimal
gland, the mechanical pulling of the tissue, the contraction of the skeletal muscle and the resulting metabolites
(brown and hudlicka 2003). In the present study, given that the subjects performed hiit protocol, this type of
activity caused a general increase in the blood flow of the skeletal muscle, which leads to increased tensile forces
and shear stress. These reasons for the intensity of exercise can be attributed to the increase in the mean of the
current study variable, regardless of hypoxia conditions. Other possible reasons for increase in osteopontin
following severe activity include secretion of important interleukins such as il-1, il-6, il-10, and tnf-α (lund,
giachelli et al. 2009). However, in the present study, the values of immune factors were not measured. During
the intensive exercise, the production of activated oxygen species increases and it is believed that the main
source of these materials is the mitochondria of activated muscle cells. Two integrin receptors are involved in the
angiogenesis of αvβ5 and αvβ3 (the ligand to osteopontin) expressed in endothelial cells. Based on this
hypothesis, shear stress, by causing turbulence in the skeletal columns of the cell, changes the site of the
endothelial cell in a local adhesive bond and activates the integrin, thereby producing an intracellular message
that ultimately transpires the genes involved in the capillary growth process (jensen, schjerling et al. 2004).
The results of this study showed that there was no significant effect on the amount of calcium in active
women after 8 weeks of intensive exercise. The result of this study is consistent with the findings of gibson and
michell. The researchers studied the impact of two-way sports in young girls and concluded that the higher the
intensity of exercise, the greater the bone mass content would be compared to the lighter exercises in the entire
body and pelvic bone (gibson, mitchell et al. 2004). Appropriate intensity, regular and balanced exercise, along
with proper nutrition, has a beneficial effect on bone density. In contrast to the beneficial effects of exercise on
bone density, severe physical activity may increase the risk of osteoporosis, especially in women, and the
athlete's triple syndrome includes amenorrhea, malnutrition and osteoporosis. However, the results of this study
were not consistent with the findings of some studies (florindo, latorre et al. 2002; vanaky, sadeghi et al. 2015).
The contradiction of the present research with the above research can be due to the type of subjects. Subjects in
this study were postmenopausal women with osteoporosis and elderly men whose exercise program was
continuously and entertaining along with supplementation of calcium, vitamin d, and other inorganic minerals.
Exercise in elderly people, along with the use of mineral supplements, leads to the improvement of structural and
physiological variables. The subjects were active women aged 20-24 years and did not use any supplement
before exercise. Also, the type of exercise program in the study was different from the present study. The effect
of the training period, mostly on inactive people and non-athletes, especially postmenopausal women, and
middle-aged men, make changes.
Also, the results of this study showed that eight weeks of hiit had no significant effect on the magnitude
of magnesium in active women for eight weeks of hiit. The results of studies by kilic et al, brilla et al., are
consistent with the results of the present study (brilla and conte 2000; kilic, baltaci et al. 2004). The results of the
studies have shown that minerals, especially magnesium, may be excited during sweat during intensive
exercises, and this decrease leads to occasional fatigue, with reduced endurance, and the performance of anabolic
factors is not easily conducted. However, the results of sadeghi et al. Are not consistent with the results of the
present study that may be due to the type of subjects who were women and men with osteoporosis and their
sports activities may be associated with the administration of certain vitamins and minerals which is prescribed
by a specialist physician are the reasons for the contradiction of the above study with this research. It has been
shown that magnesium intake is lower than daily intake, and this shortage is more pronounced in athletes, and it
may be necessary for athletes to use supplements to take advantage of their proper amount in their body (brilla
and conte 2000). In addition, the results of the present study showed that there was no significant effect on the
amount of active female iron in eight weeks of hiit. The results of this study are consistent with the findings of
baker et al.(baker, de lisio et al. 2011). There is empirical evidence that during the tolerance exhausting exercise
activities, moderate to severe ros production, as well as oxidative stress and tissue damage associated increase
with physical activity. During the exhausting sporting activities, oxygen consumption is increased by 100 to 200
times the resting time. It has been shown that iron and the group of hemoglobin and myoglobin are potential
sources of ros but it has not yet been determined how much they act in oxidative stress during or after the
exhausting exercise(cooper and beasley 1999). The lack of reduction of iron in the subjects of the present study

could be due to the lack of continuity and long distance in the exercise protocol. Intensive exercises composed of
a short period of time from several intermittent periods with both low and high intensity (boutcher 2010).
Contrary to tolerance exercises with long distances, further sweating, along with a greater number of foot
strokes, can lead to the destruction of red blood cells.
The results of this study showed that there was no significant effect on the amount of phosphorus in
active women for 8 weeks of hiit. The results of sedegi et al are consistent with the results of the present study
(vanaky, sadeghi et al. 2015). However, the results of duggan et al. And mason et al are not consistent with the
results of this study (mason 2007; duggan, xiao et al. 2014). In the above studies, the elderly and postmenopausal
women were at risk of osteoporosis, which had severe exercise for two years. Usually, people with lower levels
of physiological characteristics have a better response to training, and older people can have such a feature. In a
study by mason and colleagues in swimmers, women's parasols and runners who consumed 2 g sodium
dihydrogen phosphate one hour before training all improved exercise performance and recorded better records.
The contradictory reasons in this study can be difference in the intentional training protocols with these exercises
as well as differences in the ability of individuals to perform these exercises. Another reason for not having a
significant difference after the exercise period in this study may be due to the activity of these subjects, their
compatibility with exercise. There is evidence of phosphate regulation of the gene expression, which may play a
role in the mineralization of the bone. Phosphate expresses glycoprotein phosphorylation of osteopontin.
Osteopontin plays a role in prolonging crystalline hydroxyapatite.
Conclusion
Regarding the results of this study, it can be said that the hiit program with the present training method
makes it compatible with the biological ability and degree of mineralization of bone marrow cells in athletes.
These results can be used to design a workout program by athletic instructors, especially coaches who lack the
time to have athletes (for example, in academic teams).

Modern toplumda bir norm hâline gelmiş olan hareketsizlik, birçok hastalık için risk faktörüdür. Fiziksel aktivite ve egzersiz için birçok engel bulunmakta olup, başlıca sebebi zaman eksikliğidir. Yaygın metabolik hastalıkların tedavisi ve yönetiminde rol oynayan egzersiz, sağlıklı yaşamın temel prensiplerinden biridir. Daha kısa egzersiz ve dinlenme süreleriyle uygulanan antrenman birimleri, aerobik fitness gelişimi için gelecek vaat eden bir egzersiz stratejisidir. Sağladığı kardiyovasküler, metabolik ve fiziksel yararları ile evrensel bir antrenman seçeneği olabilir. Background and Study Aim. This study investigated the effects of HIIT (high-intensity interval training) implementations on some serum elements, resting heart rate, total body weight, and BMI parameters in tennis players. Material and Methods. The study group consisted of 20 athletes (aged 18-22) who were actively playing tennis. The athletes were divided into experimental (n=10) and control (n=10) group. To assign participants to these groups homogeneously, preliminary measurements were made before the study, and the results were used for clustering analysis. While the control group continued their regular tennis training, the experimental group was given additional HIIT three days a week besides regular tennis training. Measurements of BMI, resting heart rate, and body weight were taken, and serum samples were collected before and after training from both groups. The SPSS package program was used in data analysis. The level of significance was set at p<0.05 in the tests. Participants showed significant differences in sodium, calcium, iron, resting heart rate, weight, and BMI parameters between groups (p<0.05). Results. However, no significant differences between the groups were found in potassium, chlorine, and magnesium parameters (p>0.05). The within-group measurements of sodium, calcium, resting heart rate, weight and BMI levels decreased in the experimental group in the post-test, while iron levels were higher in the post-test compared to the pre-test value (p<0.05). On the other hand, the pre-test and post-test values were similar within the control group, and there were no significant differences (p>0.05). Conclusions. Training remodelled with HIIT implementation can affect athletes' element levels, BMI, total body weight, and resting heart rate. In addition, the increase in iron levels due to HIIT in the experimental group was important. We believe that this may affect both muscle contraction functions and the efficiency performance output. Lastly, due to the decrease of calcium and sodium elements with HIIT implementations, we suggest that HIIT applications should not be performed unsupervised. In addition, by tracking the element levels of athletes, they should be supported with supplements or nutritional meals in line with expert advice when necessary.

The purpose of this research was to improve the handball athletes’ physical fitness through Tabata training during the outbreak of COVID-19. This pre-experimental research used a One-Group Pretest-Posttest Design. The research participants were male handball athletes at amateur level (N = 30) (age = 21.8 ± 1.3 years, height = 169.6 ± 2 cm, weight = 60.7 ± 7 kg). All participants performed the Tabata training for 23 sessions with a frequency of 3 times a week. The athletes’ physical fitness level was measured using push-ups, curl-ups, V-sit and reach, as well as shoulder stretch. All research data were then analyzed using SPSS version 22 with a significance level of α 0.05. The results of this research found that the physical fitness components consisting of strength endurance (P<0.05), muscle strength (P>0.251), flexibility in the area of lower back and hamstrings (P<0.05) as well as upper arm flexibility (P<0.05) experienced differences before and after performing the Tabata training program. It is concluded that in response to the outbreak of COVID-19 throughout the world, Tabata is one alternative training to be performed by each handball athlete at home since Tabata training may provide various benefits in improving the handball atlethes’ physical fitness components. Original Article Abstract Purpose: Osteopontin is a novel adipose tissue derived cytokine correlating with obesity and its related metabolic diseases such as insulin resistance and type II diabetes. The purpose of this study was to investigate effect of high intensity interval training (HIIT) on serum levels of Osteopontin, insulin and fasting glucose and insulin resistance index (HOMA-IR) in young sedentary overweight and obese women. Methods: Twenty overweight and obese women randomly divided into two groups including experimental (mean age 32.5 ± 4.14 years, body mass index 33.18 ± 6.12 Kg/m2) and control (mean age 35.45 ± 2.33 years, body mass index 31.58 ± 1.65 Kg/m2). Subjects in experimental group participated in eight weeks of HIIT (one minute of running interval at intensity of 90 percent of maximal heart rate and one minutes of rest interval of running at intensity of 50-60 percent of maximal heart rate, 10 sets per session, 20 minutes per session, three times per week). Blood factors and body composition indices were assessed before and after training protocol. Statistical analysis was done by Covariance, paired t-test and Pearson correlation at significant level of P < 0.05. Results: According to paired t-test, HIIT resulted in significant decrease in serum levels of osteopontin (P = 0.0001) and weight (P = 0.0001), body mass index (P = 0.0001) and body fat percent (P = 0.0001) in post-test compared to pre-test. However, Covariance showed that only changes in levels of insulin (P = 0.039), weight (P = 0.001), body mass index (P = 0.005) and body fat percent (P = 0.0001) were significantly different between training and control groups. Moreover, there were negative correlation between changes in osteopontin level and changes in body mass index (P = 0.0001) and body fat percent (P = 0.044). Conclusion: High intensity interval training (HIIT) decreased osteopontin level and improved body composition and metabolic status, and therefore, can be suggested as a therapeutic procedure in obese and overweight young women. Osteopontin (OPN) is a multifunctional protein mainly associated with bone metabolism and remodeling. Besides its physiological functions, OPN is implicated in the pathogenesis of a variety of disease states, such as obesity and osteoporosis. Importantly, during the last decades obesity and osteoporosis have become among the main threats to health worldwide. Because OPN is a protein principally expressed in cells with multifaceted effects on bone morphogenesis and remodeling and because it seems to be one of the most overexpressed genes in the adipose tissue of the obese contributing to osteoporosis, this mini review will highlight recent insights about relation between adipose tissue and bone homeostasis.

Purpose] The purpose of this study was to assess the possible role of physical activities, calcium consumption and lifestyle factors in both bone mineral density and bone metabolism indices in 350 young adult volunteers. [Subjects and Methods] All volunteers were recruited for the assessment of lifestyle behaviors and physical activity traits using validated questioners, and bone mineral density (BMD), serum osteocalcin (s-OC), bone-specific alkaline phosphatase (BAP), and calcium were estimated using dual-energy X-ray absorptiometry analysis, and immunoassay techniques. [Results] Male participants showed a significant increase in BMD along with an increase in bone metabolism markers compared with females in all groups. However, younger subjects showed a significant increase in BMD, OC, BAP, and calcium compared with older subjects. Osteoporosis was more common in older subjects linked with abnormal body mass index and waist circumference. Bone metabolism markers correlated positively with BMD, physically activity and negatively with osteoporosis in all stages. Also, moderate to higher calcium and milk intake correlated positively with higher BMD. However, low calcium and milk intake along with higher caffeine, and carbonated beverage consumption, and heavy cigarette smoking showed a negative effect on the status of bone mineral density. Stepwise regression analysis showed that life style factors including physical activity and demographic parameters explained around 58–69.8% of the bone mineral density variation in young adults especially females. [Conclusion] body mass index, physical activity, low calcium consumption, and abnormal lifestyle have role in bone mineral density and prognosis of osteoporosis in young adults.

A promoter polymorphism of the osteopontin (OPN) gene (rs28357094) has been associated with multiple inflammatory states, severity of Duchenne muscular dystrophy (DMD) and muscle size in healthy young adults. We sought to define the mechanism of action of the polymorphism, using allele-specific in vitro reporter assays in muscle cells, and a genotype-stratified intervention in healthy controls. In vitro reporter constructs showed the G allele to respond to estrogen treatment, whereas the T allele showed no transcriptional response. Young adult volunteers (n = 187) were enrolled into a baseline study, and subjects with specific rs28357094 genotypes enrolled into an eccentric muscle challenge intervention [n = 3 TT; n = 3 GG/GT (dominant inheritance model)]. Female volunteers carrying the G allele showed significantly greater inflammation and increased muscle volume change as determined by magnetic resonance imaging T1- and T2-weighted images after eccentric challenge, as well as greater decrement in biceps muscle force. Our data suggest a model where the G allele enables enhanced activities of upstream enhancer elements due to loss of Sp1 binding at the polymorphic site. This results in significantly greater expression of the pro-inflammatory OPN cytokine during tissue remodeling in response to challenge in G allele carriers, promoting muscle hypertrophy in normal females, but increased damage in DMD patients.

The effect of regular aerobic exercise on body fat is negligible; however, other forms of exercise may have a greater impact on body composition. For example, emerging research examining high-intensity intermittent exercise (HIIE) indicates that it may be more effective at reducing subcutaneous and abdominal body fat than other types of exercise. The mechanisms underlying the fat reduction induced by HIIE, however, are undetermined. Regular HIIE has been shown to significantly increase both aerobic and anaerobic fitness. HIIE also significantly lowers insulin resistance and results in a number of skeletal muscle adaptations that result in enhanced skeletal muscle fat oxidation and improved glucose tolerance. This review summarizes the results of HIIE studies on fat loss, fitness, insulin resistance, and skeletal muscle. Possible mechanisms underlying HIIE-induced fat loss and implications for the use of HIIE in the treatment and prevention of obesity are also discussed.

Several lines of evidence indicate that hypoxia is a stimulus to vascular smooth muscle cell (VSMC) proliferation that occurs in pulmonary hypertension. The present study tested the hypothesis that low O2 tension directly stimulates human VSMC proliferation by inducing them to produce interleukin (IL)-1, a potent autocrine growth factor for human VSMC. Human VSMC derived from pulmonary artery, aorta, or saphenous vein were incubated in either a normal in vitro O2 environment (20% O2) or in chambers containing low (~1%) or moderate (5%) O2. Levels of IL-1α and IL-1β mRNA increased in human VSMC after 24-48 h of incubation in low O2 compared with levels in normoxic cells and then decreased upon subsequent reoxygenation. Levels of cell-associated IL-1α also increased progressively after 24-48 h in low O2; however, detectable IL-1α was not released from the cells in the media. IL-1β was detectable in cell lysates and supernatants; however, the levels were not affected by exposure to low O2. mRNA encoding for tumor necrosis factor-α (TNF-α), a related cytokine and VSMC mitogen, was not detectable in human VSMC exposed to either low or 20% O2. Proliferation of human VSMC was not stimulated during exposure to low O2, despite the fact that cells remained responsive to the mitogenic effect of exogenous IL-1. Interestingly, however, exposure to 5% O2 enhanced proliferation of human VSMC but did not induce IL-1α production. Inhibition of IL-1 binding to the type I IL-1 receptor by exogenous addition of IL-1-receptor antagonist (10 μg/ml) did not attenuate the proliferation rates of human VSMC incubated in 20%, 5%, or low O2 or in human VSMC that were reoxygenated after exposure to low O2. These results demonstrate two direct and distinct effects of hypoxia on VSMC. Exposure to moderately low O2 tension induces VSMC proliferation, independent of IL-1, whereas exposure to very low O2 tension induces production of IL-1α.

The aim of this study was to determine the relationship between habitual physical activity (HPA) during life and bone mineral density (BMD) in men aged 50 years and older. A total of 326 men aged 50 years and older, volunteers living in São Paulo city, Brazil, were studied. BMD was measured in the whole body, femoral neck, Ward's triangle, trochanter, and lumbar spine (L2-L4) with a dual-energy x-ray absorptiometer. The HPA data were collected with questionnaires inquiring about physical exercise and occupational physical activity in the past and during the past 12 months and leisure and locomotor physical activity in the preceding 12 months. The relationship between BMD and HPA was analyzed using multiple linear regression models adjusted for age and body mass index (BMI). Practice of physical exercise in the past 10-20 years and leisure and locomotor physical activity in the preceding 12 months showed a significant positive correlation with BMD of whole body, femoral neck, trochanter, and lumbar spine, and this association was independent of age and BMI. HPA can contribute to preserving BMD in men aged 50 years and older in Brazil, when it is practiced in the past 10-20 years and even in the present.

Muscle attributes and selected blood hormones of football players were assessed in response to a nightly supplementation regimen during spring football, over an 8-week period, with pre-post measures. A double-blind randomized study was conducted with ZMA (30 mg zinc monomethionine aspartate, 450 mg magnesium aspartate, and 10.5 mg of vitamin B-6) and placebo (P), n=12 and n=15, respectively. Plasma zinc and magnesium levels were ZMA (0.80 to 1.04 mg/ml; 19.43 to 20.63 mcg/ml) and P (0.84 to 0.80 mg/ml; 19.68 to 18.04 mg/ml), respectively (P<0.001). Free testosterone increased with ZMA (132.1 to 176.3 pg/mL), compared to P (141.0 to 126.6 pg/mL) (P<0.001); IGF-I increased in the ZMA group (424.2 to 439.3 ng/mL) and decreased in P (437.3 to 343.3 ng/mL) (P<0.001). Muscle strength via torque measurements and functional power were assessed with a Biodex dynamometer. Differences were noted between the groups (P<0.001): ZMA (189.9 to 211 Nm at 180°/s and 316.5 to 373.7 Nm at 300°/s) and P (204.2 to 209.1 Nm at 180°/s and 369.5 to 404.3 Nm at 300°/s). The results demonstrate the efficacy of a Zn-Mg preparation (ZMA) on muscle attributes and selected hormones in strength-trained, competitive athletes.

Background: Increased physical activity is associated with decreased risk of several types of cancer, but underlying mechanisms are poorly understood. Angiogenesis, in which new blood vessels are formed, is common to adipose tissue formation/remodeling and tumor vascularization. Methods: We examined effects of a 12-month 45 minutes/day, 5 days/week moderate-intensity aerobic exercise intervention on four serum markers of angiogenesis in 173 sedentary, overweight, postmenopausal women, 50 to 75 years, randomized to intervention versus stretching control. Circulating levels of positive regulators of angiogenesis [VEGF, osteopontin (OPN), plasminogen activator inhibitor-1 (PAI-1)], and the negative regulator pigment epithelium-derived factor (PEDF), were measured by immunoassay at baseline and 12 months. Changes were compared using generalized estimating equations, adjusting for baseline levels of analytes and body mass index (BMI). Results: VEGF, OPN, or PAI-1 levels did not differ by intervention arm. Participants randomized to exercise significantly reduced PEDF (-3.7%) versus controls (+3.0%; P = 0.009). Reductions in fat mass were significantly associated with reductions in PAI-1 (Ptrend = 0.03; Ptrend = 0.02) and PEDF (Ptrend = 0.002; Ptrend = 0.01) compared with controls, or to those who gained any fat mass respectively. There was a significant association between decreases in VO2max, and increased reductions in PEDF (Ptrend = 0.03), compared with participants who increased their level of fitness. Conclusions: Fat loss reduces circulating PAI-1 and PEDF. Changes in VO2max are associated with alterations in PEDF, but these associations are complex. Impact: Unexpected reductions in PEDF with decreasing fat mass, and with decreasing VO2max, warrant further study, including examining the effects of different types and intensities of exercise; and role of dietary weight-loss with and without exercise.

Purpose: An osteopontin (OPN; SPP1) gene promoter polymorphism modifies disease severity in Duchenne muscular dystrophy, and we hypothesized that it might also modify muscle phenotypes in healthy volunteers. Methods: Gene association studies were carried out for OPN (rs28357094) in the FAMuSS cohort (n = 752; mean ± SD age = 23.7 ± 5.7 yr). The phenotypes studied included muscle size (MRI), strength, and response to supervised resistance training. We also studied 147 young adults that had carried out a bout of eccentric elbow exercise (age = 24.0 ± 5.2 yr). Phenotypes analyzed included strength, soreness, and serum muscle enzymes. Results: In the FAMuSS cohort, the G allele was associated with 17% increase in baseline upper arm muscle volume only in women (F = 26.32; P = 5.32 × 10), explaining 5% of population variance. In the eccentric damage cohort, weak associations of the G allele were seen in women with both baseline myoglobin and elevated creatine kinase. The sexually dimorphic effects of OPN on muscle were also seen in OPN-null mice. Five of seven muscle groups examined showed smaller size in OPN-null female mice, whereas two were smaller in male mice. The query of OPN gene transcription after experimental muscle damage in mice showed rapid induction within 12 h (100-fold increase from baseline), followed by sustained high-level expression through 16 d of regeneration before falling to back to baseline. Conclusion: OPN is a sexually dimorphic modifier of muscle size in normal humans and mice and responds to muscle damage. The OPN gene is known to be estrogen responsive, and this may explain the female-specific genotype effects in adult volunteers.

Endurance exercise is a poorly defined yet powerful mediator of hematopoiesis. The purpose of this study was to directly investigate the effects of endurance exercise training on hematopoiesis and to identify potential mechanisms responsible for any observed changes. Four-week-old male C57Bl/6 mice were trained on a treadmill at progressive speeds over a 10-wk period. Tissues were harvested 2 d following the final training session. Flow cytometry, the cobblestone area-forming cell assay, and the methycellulose colony-forming unit assay were used to assess medullary and mobilized hematopoietic stem and progenitor cells. Quantitative real-time PCR and Western blots were used to measure hematopoietic cytokine production. Histochemistry was also used to assess adaptations to exercise in the bone marrow niche. Depending on the cell type, endurance training increased medullary and mobilized hematopoietic stem and progenitor cell content from 50 to 800%. Training also reduced marrow cavity fat by 78%. Skeletal muscle hematopoietic cytokine expression was also increased at least 60% by training. Sedentary mice served as controls for the above experiments. In conclusion, endurance exercise training greatly promotes hematopoiesis and does so through improvements in medullary niche architecture as well as increased skeletal muscle hematopoietic cytokine production.
