Abstract

Background Loss of p53 is associated with the occurrence of cancer. P53 is a tumor suppressor gene that serves the repair of DNA, inhibits proliferation, and increased apoptosis. P53 is often called Guardian of the genome and Policeman of the oncogenes. Recently, p53 also regulated metabolic energy pathway. Excess energy is the risk of cancer. This paper wants to know dietary interventions against the development of cancer involving p53. Method This paper was written based on literature review with key word “cancer AND fasting OR diet restriction AND p53” from PubMed, Science direct, and Google scholar. Results Intervention fasting in cancer cells showed an increase expression of p53 and apoptosis genes. Fasting or restriction of glucose in cancer cells with mutant p53 increased cancer cell death. Furthermore, in studies of p53+/- or p53-/- mice demonstrated that the caloric restriction significantly delayed the onset of tumors and decreased the size of the tumor volume. Cancer cells in treatment with cisplatin and fasting revealed a decrease in proliferation through increased the activation of ATM/Chk2/p53 signaling pathway. This combination increased the sensitivity of human tumor xenografts in mice better than only cisplatin treatment. Conclusion Fasting can inhibit the growth of cancer cells through regulation of the p53 gene.
