Chemical constituents from aerial parts of Fagopyrum dibotrys

Abstract

Objective: To study the chemical constituents from the aerial parts of Fagopyrum dibotrys. Methods: The compounds were isolated and purified by means of chromatographic techniques and their structures were identified on the basis of spectral features. Results: Fourteen known compounds were isolated in the methanol extract from the aerial parts of F. dibotrys and their structures were identified as benzoic acid (1), p-hydroxybenzoic acid (2), p-hydroxy benzaldehyde (3), 3,4-dihydroxy benzoic acid (4), succinic acid (5), caffeic acid (6), methyl caffeate acid (7), luteolin (8), tricin (9), quercetin (10), afzelin A (11), 2α,3β,29-trihydroxyolean-12-en-28-oic acid (12), yarumic acid (13), and 3α-hydroxy-urs-12,15-dien (14). Conclusion: Compounds 2-3 and 6-9 are firstly obtained from the aerial parts of F. dibotrys. Compounds 11-14 are isolated from the genus of Fagopyrum Mill. for the first time. © 2016, Editorial Office of Chinese Traditional and Herbal Drugs. All right reserved.

The genus Fagopyrum (Polygonaceae), currently comprising 15 species of plants, includes three important buckwheat species: Fagopyrum esculentum (F. esculentum) Moench. (common buckwheat), Fagopyrum tataricum (F. tataricum) (L.) Gaertn. (tartary buckwheat) and Fagopyrum dibotrys (F. dibotrys) (D. Don) Hara. (perennial buckwheat), which have been well explored due to their long tradition of both edible and medicinal use. This review aimed to present an up-to-date and comprehensive analysis of the phytochemistry and pharmacology of the three Fagopyrum buckwheats. In addition, the scope for future research was also discussed. All available references included in this paper were compiled from major databases, such as MEDLINE, Pubmed, Scholar, Elsevier, Springer, Wiley and CNKI. A total of 106 compounds isolated from three Fagopyrum buckwheats can be mainly divided into six classes: flavonoids, phenolics, fagopyritols, triterpenoids, steroids and fatty acids. Flavonoids and phenolic compounds were considered to be the major active components. Considerable pharmacological experiments both in vitro and in vivo have validated that Fagopyrum buckwheats possess antitumor, anti-oxidant, anti-inflammatory, hepatoprotective, anti-diabetic activities, etc. All reported data lead us to conclude that Fagopyrum buckwheats have convincing medicinal potential. However, further research is needed to explore its bioactive constituents, the relationship to their structural activities and the molecular mechanisms of action.

Fagopyri Dibotryis Rhizoma (FDR) is an effective Chinese herbal medicine with a long history of use in China. FDR is effective in heat clearing and detoxifying, promotion of blood circulation, relieving carbuncles, dispelling wind, and removing dampness. Its seeds also have high nutritional value, are rich in protein, and contain a variety of mineral elements and vitamins. Therefore, FDR is considered a natural product with medical and economic benefits, and its chemical composition and pharmacological activity are of interest to scientists. The current review provides an overview of the available scientific information on FDR, particularly its botany, chemical constituents, and pharmacological activities. Various sources of valid and comprehensive relevant information were consulted, including the China National Knowledge Infrastructure, Web of Science, and PubMed. Among the keywords used were “Fagopyri Dibotryis Rhizoma”, “botanical features”, “chemical composition”, and “pharmacological activity” in combination. Various ailments are treated with FDR, such as diabetes, tumor, sore throat, headache, indigestion, abdominal distension, dysentery, boils, carbuncles, and rheumatism. FDR is rich in organic acids, tannins, flavonoids, steroids, and triterpenoids. Experiments performed in vitro and in vivo showed that FDR extracts or fractions had a wide range of pharmacological activities, including antitumor, anti-inflammatory, immunomodulatory, antioxidant, antimicrobial, and antidiabetic. The current review provides an integrative perspective on the botany, phytochemistry and pharmacological activities of FDR. FDR may be used as a medicine and food. Based on its chemical composition and pharmacological effects, the main active ingredients of FDR are organic acids, tannins, and flavonoids, and it has obvious antitumor pharmacological activity against a variety of malignant tumors. Therefore, FDR is worthy of further study and application as a potential antitumor drug.

Plectranthus esculentus N.E.Br. (family Lamiaceae) also known as Livingstone potato (vat or rizga in Nigeria), is a dicotyledonous perennial shrub growing up to 2 m tall. While it is cultivated mainly for its edible tubers, the plant is potentially valuable as phytomedicine. Three varieties (vat-long’at, vat-riyom and vat-bebot) are well known among the Berom of Plateau State, Nigeria. The vat-bebot variety (which showed good promise in bioactivity studies) was used in this study. The leaves and tubers were extracted successively with hexane, ethyl acetate, methanol and water. Fractionation of the active ethyl acetate extracts was carried out using open column and preparative High Performance Liquid Chromatography (prep HPLC). This led to the isolation of β-sitosterol and oleanolic acid from the leaves; while 19-dehydroursolic acid and yarumic acid, as well as β-sitosterol were isolated from the tubers. Nuclear Magnetic Resonance (NMR) and Electron Impact Mass Spectroscopy (EIMS) were used to characterize isolated compounds. Comparing acquired spectral data of isolated compounds with those from literature helped to confirm the identity of the compounds. The isolation and characterisation of these compounds, from Plecthranthus esculentus, have not been hitherto reported in literature.

Three new sterols, (24R)-5,28-stigmastadiene-3β,24-diol-7-one (1), (24S)-5,28-stigmastadiene-3β,24-diol-7-one (2), and 24R and 24S-vinylcholesta-3β,5α,6β,24-tetraol (3), together with three known sterols (4–6) were isolated from the green alga Ulva australis. The structures of the new compounds (1–3) were elucidated through 1D and 2D nuclear magnetic resonance spectroscopy as well as mass spectrometry. Compounds 4–6 were identified as isofucoterol (4), 24R,28S and 24S,28R-epoxy-24-ethylcholesterol (5), and (24S)-stigmastadiene-3β,24-diol (6) on the basis of spectroscopic data analyses and comparison with those reported in the literature. Compounds 4–6 were isolated from U. australis for the first time. These compounds, together with the previously isolated secondary metabolites of this alga, were investigated for their inhibitory effects on human recombinant aldose reductase in vitro. Of the compounds, 24R,28S and 24S,28R-epoxy-24-ethylcholesterol (5), 1-O-palmitoyl-3-O-(6′-sulfo-α-d-quinovopyranosyl) glycerol, (2S)-1-O-palmitoyl-3-O-[α-d-galactopyranosyl(1→2)β-d-galactopyranosyl] glycerol, 4-hydroxybenzoic acid, 4-hydroxyphenylacetic acid, and 8-hydroxy-(6E)-octenoic acid weakly inhibited the enzyme, while the three new sterols, 1–3, were almost inactive.

Background Pyrethrum tatsienense (Bureau & Franch.) Ling ex C. Shih (PTLCS) belongs to the family Compositae, which is a perennial medicinal plant mainly distributed in the Qinghai-Tibet Plateau of China. This review provides a comprehensive summary of the ethnopharmacology, phytochemistry, and pharmacology of PTLCS. This review offers valuable references and guidance for researching PTLCS in depth. Methods The related references of PTLCS were retrieved from an online database, such as Web of Science, Google Scholar, SciFinder, PubMed, SpringLink, Elsevier, Willy, CNKI, and so on. Results PTLCS is widely reported for treating headaches, head injuries, traumatic injuries, anabrosis, impetigo, hepatitis, and other diseases in the medical field. Phytochemical research revealed that this plant contained flavonoid aglycones, flavonoid glycosides, xanthones, triterpenoids, coumarins, polyacetylenes, volatile oils, and other compounds. Meanwhile, PTLCS exhibited extensive pharmacological activities including anti-cardiac ischemia, anti-hypoxia, hepatoprotective, anti-inflammatory and analgesic, and antioxidant activities. Conclusions PTLCS is widely used as a Tibetan medicine, which has a variety of chemicals with diverse bioactivities. Therefore, further studies are necessary to perform on the PTLCS to assay biological activities, discover their bioactive constituents, and reveal pharmacological mechanisms. This review may supply an important theoretical basis and valuable reference for in-depth research and exploitations of PTLCS.

A phytochemical study of the whole plants of Polygonum aviculare L. led to the isolation of 21 compounds, including fourteen flavonoids (1–14), six phenols (15–20), and one lignan (21). The structures of these isolated compounds were identified using NMR spectroscopy (¹H and ¹³C) by comparison with previously reported data. To the best of our knowledge, compounds 3, 7, 15–16, and 18 were reported from this plant for the first time, compound 19 has never been isolated from the genus Polygonum, and compounds 8 and 21 were only acquired from Polygonum (Polygonaceae). The chemotaxonomic significance of the isolates was also discussed.

Ethnopharmacological relevance The root of Dendropanax dentiger (Harms) Merr. is a pivotal folk Chinese medicine against rheumatoid arthritis (RA) with no scientific validation. Aim of the study This study was conducted to explore the anti-RA effect of the D. dentiger extract on complete Freund's adjuvant-induced arthritis (AIA) in rats and identified its major bio-constituents. Materials and methods Dendropanax dentiger roots extracts (127.5, 255.0 and 510.0 mg/kg, once daily) were orally at day 7 post-administration adjuvant and lasting for 22 days. The therapeutic effects of D. dentiger roots extract on AIA rats were investigated by body weight growth, arthritis score, thymus and spleen indices, and histopathological analysis. Moreover, the levels of rheumatoid factor (RF), C-reactive protein (CRP), tumor necrosis factor alpha (TNF-α), interleukin-1β (IL-1β), IL-4, IL-6, IL-10, IL-17, cyclooxygenase-2 (COX-2), 5-lipoxygenase (5-LOX) and matrix metalloproteinase-2 (MMP-2) were also evaluated. Finally, the major constituents were isolated and identified from D. dentiger roots extract with COX-2 inhibitory and antioxidant activities. Results Dendropanax dentiger roots extract remarkably alleviated the histological lesions of knee joint, increased body weight growth, decreased arthritis score, and reduced thymus and spleen indices in model rats. In parallel, the levels of RF, CRP, TNF-α, IL-1β, IL-6, IL-17, COX-2, 5-LOX and MMP-2 were observably downregulated, while the levels of IL-4 and IL-10 were prominently upregulated in D. dentiger roots extract-treated rats. Meanwhile, 14 compounds were isolated and identified from D. dentiger roots extract, and four phenol derivatives (1, 4, 6 and 7) exhibited remarkable COX-2 inhibitory and antioxidant activities. Conclusions Dendropanax dentiger roots extract possessed persuasive anti-RA effect may be partly responsible for phenol derivatives via modulation of inflammatory biomarkers, and supports the traditional folk use of D. dentiger in China.

One previously undescribed polyacetylene, erythro-(5E)-1,5-didecadiene-7,911-triyne-3,4-diacetate (1), two previously undescribed flavonoids named tricin-7-O-ethylglucuronide (2) and tricin 4′-O-(erythro-β -guaiacylglyceryl) ether 5-O-β-glucopyranoside (3), together with four known polyacetylenes (4-7) and fourteen known flavonoids (8-21) were isolated from alcoholic extracts of the stems and leaves of Pyrethrum tatsienense (SLPT). Their structures were elucidated by extensive analysis of NMR and mass spectroscopic data as well as chemical reactions. Among all isolates, polyacetylenes were found as a new structural type from P. tatsienense for the first time. Tricin and its derivatives were common compositions between SLPT and the flowers of P. tatsienense (FPT). According to previous reports, tricin and its derivatives are responsible for the anti-inflammatory activity of FPT. These findings suggested that SLPT might be worthy of further development and research as medicinal parts.
