Comparative Effect of Wasp Density in Unisexual and Bisexual Strains of Lysiphlebus fabarum, on Superparasitism in the Black Bean Aphid, Aphis fabae

Abstract

Lysiphlebus fabarum Marshall (Hymenoptera: Braconidae: Aphidiinae) is a common aphidiine parasitoid in central Europe where it attacks more than 70 species of aphids. Although both unisexual (thelytokous) and bisexual (arrhenotokous) strains of L. fabarum have been reported in Iran, the former appear to be more widely distributed. Aphid control in Iran relies heavily on pesticides in both field and greenhouse settings, whereas any availability of a new biological control agents such as L. fabarum would likely aid in reducing pesticide use. One of the most important aspects of implementing a parasitoid as a biological control agent is to determine the best density of parasitoid in the host access, in mass rearing. Parasitoid wasps display a wide array of strategies to dispose of host resources. Superparasitism is parasitism of a host by parasitoids of the same species. In solitary parasitoids, superparasitism almost occurs when hosts are limited or parasitoid density in a patch is high. Acceptance or rejection of a host usually depends on a female parasitoid’s ability to distinguish parasitized and unparasitized hosts. Previously, superparasitism was thought to be a maladaptive strategy, wasteful of both eggs and time, while it is now recognized as adaptive in a number of situations, e.g. under conditions of low host availability, as a mechanism for overwhelming the immune response of hosts. Materials and methods: In the present study, stock colonies of the unisexual and bisexual strains of the parasitoid were established from materials collected from Zanjan province and Khuzestan province fields, respectively. Both populations of the parasitoid were collected from parasitized black bean aphids feeding on broad bean, Vicia fabae L. A stock colony of black bean aphids was established with material collected from bean fields in Khuzestan province. The synchronous cohorts of both strains of L. fabarum were produced at 21±1 ºC, 50–60% r.h., and L14:D10 photoperiod, and then introduced (one day old) separately into 40 2nd instar nymphs in different densities (1, 5, 10, 15 and 30). The females were removed after 24 hours and parasitized aphids were dissected three days after parasitism to determine the number of wasp larvae in each host. As well as, in another experiment the superparasitized aphids were dissected in three consecutive days (3.5, 4.5 and 5.5) to determine the process of competitors’ elimination. According to results, in both strains, wasp densities affected on parasitism rates. In bisexual strain, females parasitized more aphids in 20 and 30 densities treatments than one density treatment, while this rate did not differ significantly between 1, 5, 10 and 15 densities treatments. In unisexual strain, single females significantly parasitized less hosts than other treatments, but there was no difference between other densities treatments. When in each density, percentage parasitism was compared between two strains, in density of one female, bisexual females parasitized significantly higher than unisexual ones, in contrast to density of 30, which significantly more larvae were counted in hosts of unisexual strain than bisexual ones. In other densities, there was no significant difference between two strains. In the second experiment, which the rate of superparasitism was compared in different densities, in bisexual strain, rate of superparasitism significantly was lower in density of one female than other treatments. The number of larvae did not differ significantly between other densities treatments. In unisexual strain, rate of superparasitism significantly was lower in density of one female than other treatments. The most rate of superparasitism was observed in 20 and 30 density treatments. When in each density, rate of superparasitism was compared between both strains, in 1, 5 and 10 densities, bisexual females superparasitized significantly higher than unisexual ones, in contrast to density of 30 females which significantly more larvae were counted in hosts of unisexual strain than the hosts of bisexual ones. In association with the process of competitors’ elimination, bisexual strain larvae eliminated rival larvae over time (in three consecutive days, including days 3.5, 4.5 and 5.5), as the numbers of larvae were decreased in densities of 5, 20 and 30 females. While in unisexual strain, elimination of rival larvae was not observed over time. Wasp densities affected on parasitism rates and rate of superparasitism in both strains. In mass rearing, the best density of wasps can be defined as the density that most available hosts are parasitized with a minimum of superparasitism, because superparasitism can depredate the time and eggs of female foragers. The results showed that densities of one and 5 females (for 40 individuals of 2nd instar nymphs) can be suggested as the proper densities of females in bisexual and unisexual strains, respectively. The results of this study can be useful in mass rearing and release of both L. fabarum strains. Keywords: Mutual interference, competition, parasitoid, mass rearing, Braconidae

Light, as an environmental factor, affects the biology, physiology and behavior of insects. In this study, the feeding behavior of Hippodamia variegata and parasitism rate of Lysiphlebus fabarum were studied under light and dark conditions. Synchronous cohorts of different growth stages of H. variegata and female parasitoid wasps were produced and then they were singly introduced into experimental arena containing third instar nymphs of Aphis gossypii (20 nymphs for second instar larvae and female wasp; 30 nymphs for other growth stages of ladybird beetle) on a cucumber leaf. The results revealed that, although light did not influence foraging behavior of second instar larvae of the ladybird beetle, the number of killed aphids was significantly higher for fourth instar larvae by female and male adults of H. variegata during foraging in the light condition in comparison with the dark condition. The ratio of partial prey consumption (No. of partially consumed aphids to No. of aphids killed) was significantly greater in fourth instar larvae than in second instar larvae in the dark condition. Female adults displayed significantly more partial consumption of prey in the dark relative to the light condition. Moreover, in both light and dark phases, this ratio was higher in female adults than in males. The results revealed that female wasps parasitized fewer A. gossypii nymphs in the dark than in the light. Based on the acceptable performance of H. variegata in the dark condition, it can be said that in addition to the benefits of simultaneous application of the parasitoid wasp and the ladybird beetle during the day, aphid population decline continues at night.

In superparasitism, a host is parasitized by one or more conspecific females, a phenomenon resulting in immature competition within the host’s body. In the present study, the competition between sexual and asexual strains of the parasitoid wasp, Lysiphlebus fabarum (Marshall) (Braconidae: Aphidiinae), was studied in simultaneous and successive introductions. The biological traits by both strains were determined in surviving individuals of superparasitized aphids as well. For this purpose, eight wasp females of each strain were introduced into 40 second instar of black bean aphid, Aphis fabae Scopoli, simultaneously or by a 24-hour priority. Our results revealed that the early introduction of each strain had a significant effect on the number of emerging adults of that strain. However, in simultaneous introduction, a greater number of sexual progeny emerged than asexual ones. The development time of sexual progeny was significantly decreased when their females were introduced earlier into the host aphids. Other morphological and biological traits (hind tibia length, head width, ovary area, egg load and egg size) of surviving individuals of superparasitized aphids were not affected by the oviposition sequence of the strains. The findings showed that sexual L. fabarum females were successful in competition with asexual females, the superiority of which could be more important in a host limitation conditions.

In this study, the reproductive activity and olfactory response of the parasitoid wasp Lysiphlebus fabarum to the presence of an intraguild predator, the ladybird Hippodamia variegata, was investigated on the cotton aphid Aphis gossypii on cucumber plants. For this purpose, in addition to comparing the predator feeding of healthy and parasitized adult aphids, reproductive characteristics of the parasitoid wasp (percent parasitism, percent emergence, and sex ratio) were investigated in three different host patches, including those of only aphids alone, aphids with a female ladybird, and aphids with aphid carcasses. Moreover, the preference of the female wasps to these patches was determined using a Y-tube olfactometer. Ladybirds significantly preferred healthy aphids to previously parasitized ones (F2,27=12.53, P<0.001), and the proportion of parasitism by parasitoid wasps were lower in the patch containing a female ladybird (G2,42=6.99, P=0.03). According to olfactometer test results, the parasitoid wasp significantly avoided the patch containing the intraguild predator ladybird (χ2=6.40, df=19, P=0.01). In conclusion, based upon the findings of the avoidance of ladybirds by female wasps and also preference of healthy aphids by ladybirds, it is expected that female wasps forage more in patches lacking the intraguild predator ladybirds, and that ladybirds prefer feeding on healthy aphids, resulting in a reduction in the intraguild predation and improving the cotton aphid control.

Intraguild predation is defined as killing and eating another predator species as potential competitors that depends upon the same, limited resources. In this study, intraguild predation between the ladybird, Hippodamia variegata Goeze and the parasitoid wasp, Lysiphlebus fabarum (Marshall), were investigated on the cotton aphid, Aphis gossypii, on cucumber plants. Host preference for healthy or previously parasitized aphids in third instar larvae and adult females of H. variegata were studied in a choice access situation as well as in a Y-tube olfactometer bioassay. In these tests, an antenna of the aphids was cut to distinguish between aphids of each treatment. All experiments were performed at 22±1°C, 60%–70% RH, and a 16:8 (L:D) photoperiod. Based on the pre-test results, cutting the antenna in healthy and parasitized aphids did not significantly affect the host preference of adult and third instar larvae ladybirds. In a choice access test, female ladybirds significantly preferred healthy to previously parasitized aphids (24 and 72 hours: t =2.15, df =1,18, P =0.04 and t =6.35, df =1,18, P <0.001, respectively), but third instar larvae did not exhibit a preference for healthy or previously parasitized aphids (t =0.58, df =1,18, P =0.56). According to olfactometer test results, female ladybirds had no preference for healthy or parasitized aphids (χ2 =1.6, df =1,19, P =0.21). In conclusion, preference for healthy over parasitized aphids by female adult H. variegata constitute a feature of the simultaneous use of these two biocontrol agents (H. variegata and L. fabarum).

Superparasitism is a widespread phenomenon in insect parasitoids and may be advantageous in certain circumstances. In this paper, we distinguish two kinds of superparasitism with different adaptive consequences for an ovipositing parasitoid: self superparasitism, where a parasitoid oviposits in a host in which she has previously oviposited, and conspecific superparasitism, where she oviposits in a host containing egg(s) from a previous oviposition by a conspecific. Our studies on Trichogramma evanescens Westwood show that females lay smaller clutches on parasitized than on healthy hosts but make the same overall allocation of males and females, although the sequence of sexes during oviposition differs. No difference was found between allocation in self and conspecific superparasitism. These results are discussed in the light of theories for progeny and sex allocation.

1. In solitary insect parasitoids hosts can only support the development of a single larva. We review the experimental evidence for the ability of parasitoid females to distinguish between hosts parasitized by a conspecific, hosts parasitized by a female of a different species, and self-parasitized hosts. 2. We consider the case of a female that is able to distinguish between unparasitized, self- and non-self-parasitized hosts and a dynamic programming model is developed to predict circumstances under which this female will not discriminate between host types. Host patches deplete when the wasp is foraging in them and the decisions of the wasps concerning whether or not to accept a non-self-parasitized host and how long to stay in a patch are based on the numbers of each host type left in the patch. 3. Handling times, the fitness increments gained through oviposition in a particular host, travel time between patches and the initial numbers of each host type in the patch are shown to influence both patch residence time and the number of parasitized hosts accepted for oviposition. Mortality risks within and between patches and the age of the female affects its foraging strategy. 4. The results of this study are compared with the predictions of a rate-maximizing equivalent of the dynamic programming model and it is shown that the predictions derived from dynamic programming not only include those from rate-maximizing models as a special case but also that the predictions differ when mortality risks are different within and between patches.

One of the foraging decisions facing parasitoids is whether to accept (superparasitize) or reject hosts that have already been parasitized. An important distinction is whether the host has been parasitized by the female parasitoid herself or by a conspecific. In solitary parasitoids, the pay-off from an egg laid in the latter host type (conspecific superparasitism) is the probability that the second egg wins the competition for the host and results in an offspring. The pay-off from an egg laid in the former type (self-superparasitism) increases with an increasing probability that another female will superparasitize the host in the near future. When this probability equals one, self-superparasitism and conspecific superparasitism have the same pay-off. However, conspecific superparasitism will generally have a higher pay-off than self-superparasitism. It will therefore be beneficial for a female parasitoid to be able to distinguish between a host she parasitized and one parasitized by a conspecific. The degree of benefit depends on the probability of conspecific superparasitism in the near future. Using an optimal diet model, I show that when a parasitoid encounters a patch containing a mixture of unparasitized and already-parasitized hosts, a female that can distinguish between the two types of parasitized hosts gains more offspring than a female without this ability. However, when parasitoids search a patch together with conspecifics, it is adaptive to self-superparasitize, and the pay-off from this ability may be negligible. It is therefore predicted that when a female parasitoid searches a partially depleted patch alone, it will reject the hosts parasitized by itself more frequently than hosts parasitized by conspecifics. In contrast, female parasitoids searching together are predicted to accept hosts that they parasitized themselves much more often. The results show that the solitary parasitoid Leptopilina heterotoma (Hymenoptera: Eucoilidae) is able to distinguish between hosts that it parasitized and hosts parasitized by conspecifics. The predictions of the model are met in a second experiment that shows that L. heterotoma self-superparasitizes when the probability of conspecific superparasitism is high.

(1) Parasitic wasps (Nemeritis canescens) discriminated between hosts (larvae of the flour moth Plodia interpunctella) containing their own progeny and those containing the progeny of other individuals when deciding whether or not to avoid superparasitism. This differential discrimination lasted for about 48 hours. (2) The ability of a parasitoid to detect prior parasitism, and avoid subsequent superparasitism, seemed to depend on the operation of a specific chemical marker, originating in Dufour's gland, and introduced into the host with each egg laid. (3) A specific mathematical model, based on game theory, supports our experimental data in indicating that the patterns of differential discrimination we have observed are evolutionarily stable.

: The black bean aphid (Aphis fabae) was heavily parasitized by the aphid parasitoid, Lysiphlebus fabarum both on agricultural crops (broad bean, fodder beet) and on associated weeds (goosefoot, chamomile, tansy). Parasitization rates increased during the season and were lower on broad bean than on other host plants. Habitat diversity contributed to an increase of parasitism on fodder beet but not on broad bean. Ant-attendance lead to increased parasitism on all host plants. The findings of the field studies were confirmed by laboratory experiments on foraging and oviposition behaviour of L. fabarum females on the various host plants.

Egg distribution by the solitary parasitoid Aphidius nigripes was studied experimentally at 14 densities of the host aphid, Macrosiphum euphorbiae. The number of hosts parasitized increased with host density, the functional response being of Holling's type II. However the average number of eggs/female was not significantly affected by host density, and consequently superparasitism increased with decreasing host density. When egg distributions were tested against random models, higher than predicted numbers of both unparasitized and superparasitized hosts were observed at all densities tested. Further analysis assuming random search suggested that female A. nigripes tend to deposit several eggs per host while experiencing a low rate of encounter with hosts. The ecological significance of this behaviour is examined.

Self-superparasitism – ovipositing in or on a host that was previously parasitized by the same individual – had generally been considered maladaptive for solitary parasitoids. Recently, however, some researchers have suggested that self-superparasitized hosts produce higher fitness returns per host than singly parasitized hosts when these hosts are parasitized by conspecifics. The present study is the first to verify this possibility in the parasitoid Haplogonatopus atratus. Fitness returns from the first and second ovipositions under triple parasitism were compared with those from the first oviposition under double parasitism. Fitness returns were assessed in terms of survival rate, adult size, and developmental time. No differences were found between the two groups in adult size and developmental time. When the interval between the second and third ovipositions was 1 h, the total survival rate of the first and second comers under triple parasitism was higher than the survival rate of the first comer under double parasitism, and as high as the total survival rate of two first comers under double parasitism. This suggests that the value of self-parasitized hosts is as high as that of unparasitized hosts. When the second-to-third interval of triple parasitism was 24 h, self-superparasitism produced no fitness returns. Moreover, fitness returns from ovipositing into self-superparasitized hosts were similar to those from ovipositing into singly parasitized hosts.
