A Study on the Free Radical Scavenging Activity of the Rehmannia glutinosa Extracts

Several in vitro methods have been conducted in order to investigate the antioxidant activities of ethanol extract from Rehmannia glutinosa. Both the total phenolic and flavonoid component were expressed as the equivalent of positive control with the values of 1432.3±16.2 mg TAE/100 g and 97.6±14.1 mg QE/100 g, respectively. In addition, the antioxidant activity with an IC50 of 469.87±12.42 μg/g was measured via the DPPH scavenging activity. The study also showed that Rehmannia glutinosa ethanol extract exhibited antioxidant activities on metal-chelating, reducing power, total antioxidant activity assay.

The solvent used for extraction is ethanol with 3 variations in concentration: 50%, 70%, and 96% (absolute ethanol). TPC and TFC were determined by colorimetric method using a UV-Vis spectrophotometer. TPC was stated to be equivalent to gallic acid, while TFC was stated to be equivalent to quercetin. DPPH free radical scavenging activity was measured based on the IC50 value. The results showed that Katuk leaf extract produced from 50% ethanol solvent was able to produce TPC (42.18 ± 0.30 mg GAE / g), TFC (11.18 ± 0.38 mg QE / g) and reduction activity against DPPH radicals (IC50 = 88.33 ± 3.53 ppm). These were higher than ethanol with other concentrations. However, various things need to be considered when using this solvent given the high water content in the solvent.

The results indicated that EEP of Geniotrigona thoracica had the highest TPC and TFC at 259.84 ± 4.97 mg/g GAE and 435.00 ± 6.57 mg/g QE, respectively. Similarly, EEP of G. thoracica exhibited the greatest free radical scavenging activity with the percentage of inhibition at 88.00 ± 13.34% and IC50 of 104.20μg/mL. Antioxidant activity was highly correlated with TPC (R2 = -0.744, p < 0.05) whereas no correlation was observed between TFC and antioxidant activity. This study indicates that the antioxidant property of the propolis extract may be influenced by elevated levels of phenolic content. In conclusion, ethanolic extract of G. thoracica is the greatest source of antioxidants among the six tested Malaysian stingless bee propolis and this suggests that the extract may serve as a promising source of antioxidant agent.

In vitro antimicobial, antioxidant and antidiabetic potential of Cephalaria gigiantea

This study evaluates the antimicrobial, antioxidant and antidiabetic potential of leaf and root extracts of Cephalaria gigantea. The results showed that the extracts inhibited both bacteria and fungi tested at MIC values which ranged from 3.1 to 12.5 mg/mL. The root water and leaf hydro-ethanol extracts had the highest content of phenolics exhibiting IC50 values of 310.9 mg/g and 275.2 mg/g ... [Show full abstract] respectively. The leaf water extract possessed the highest amount of flavonoids with IC50 value of 145.4 mg/g followed by root hydro-ethanol extract with IC50 value of 53.4 mg/g. The leaf and root water extracts had the best DPPH radical scavenging activity with IC50 values of 0.6 μg/mL and 2.8 μg/mL compared to gallic acid at 14.0 μg/mL. The leaf hydro-ethanol and root water extracts scavenged ABTS radical best. Also the leaf and root hydroethanol extract exhibited the best reducing power activity when compared to gallic acid. The root water and leaf ethanol extracts displayed the most potent inhibition of α-amylase and α-glucosidase respectively. It can be concluded that C. gigantea extracts possessed antioxidant and antidiabetic potentials but has poor antimicrobial activities.

Antioxidant evaluation of A.mexicana was carried out using 1, 1-diphenyl-2- picrylhydrazyl radical (DPPH) free radical scavenging assay and exhibited considerable antioxidant potential and showed good correlation with the total phenolic and total flavonoidal content. The highest radical scavenging effect was observed in flowers of A. mexicana with IC50 = 23.75μg/ml. The results of the study showed that A.mexicana possess significant antioxidant activity. Owning to these results, the plant has the potential to be used as a medicine against the diseases caused by free radicals.

The total phenolic compounds responsible to the antioxidant activity of ethanol extract of Wali seeds derived from maceration and soxhletation were 4.946,35 mg EAG/100g and 3.830,72 mg EAG/100g, respectively. The ethanol extract of Wali seeds derived from maceration has higher antioxidant activity and total phenolic content than the extract from soxhletation with a significant different (p<0.05).

The AEAC values of the L. guyonianum ranged from (1.8 to 2.16mM) in LO, and ranged from (0.55 to 2.14mM) in LE. The Phosphomolybdate antioxidant activity of the plant extracts ranged from (1.25 ± 0.07 to 7.94 ± 0.06 mM).

DPPH free radical scavenging activity of ashwagandha extracts varied widely and it increased with increase of concentration levels. Among aqueous and ethanolic extracts, aqueous extracts exhibited higher antioxidant activity. Hence, aqueous extracts of ashwagandha roots are better source of antioxidants in comparison to ethanolic extracts.
