[The CIC chloride channels and their role in plant cell]

The chloride channel superfamily (CIC) is the large group of anion transporters expressed in nearly every cell of eukaryotic as well as prokaryotic organisms. Functionally, CIC proteins can be divided into two classes: anion channels and secondary-active anion/proton transporters. Most CIC's have also large C-termini cytosolic domains containing two cystathionine beta-synthetase domains (CBS) that are crucial for regulation of their activity. In plant cell, CIC proteins are present in membranes of various organelles including vacuole, Golgi and chloroplast. Although most of them is involve in Cl(-) transport, recent studies on Arabidopsis thaliana have revealed that at least tonoplast AtClCa can act as a NO3(-)/H+ exchanger, suggesting the role of proteins from CIC family also in nitrate transport. Here we summarize recent advances in the molecular characterization of this family and its role in plants, especially in NO3(-) distribution within the cell.

CYTOPLASMIC ORGANIZATION AND RHYTHMIC STREAMING IN GROWING BLADES OF CAULERPA PROLIFERA

Light- and electron-microscopic studies of the growing blades and their meristematic tips in Caulerpa prolifera have been correlated with time-lapse photographic studies. The cytoplasm of these growing regions has an abundant ... [Show full abstract] endoplasmic reticulum, Golgi bodies, amyloplasts, and nuclei, but lacks chloroplasts and a central vacuole. An intermediate zone lies between the white meristemplasm and the green, mature basal zone. The basal zone contains a parietal cytoplasm with organelles typical of the Chlorophyta and a dominant central vacuole. Two distinct systems of cytoplasmic streams occur in the basal zone and both contain packets of microtubules orientated parallel to the axis of the streams. As the blade matures and growth ceases, the dominant central vacuole forms up to the tip. In developing blades the tips become white in the absence of light as a result of basipetal movement of the chloroplasts. In plants kept on a 12-hr L: 12-hr D cycle, blade growth and chloroplast migration at the blade tip are rhythmic, and the peak occurs about 2-4 hr after initiation of the dark phase. Experiments are reported using continuous light or darkness after three 24-hr periods of 12-hr L: 12-hr D phasing.

[Show full abstract] described an autophagy process for chloroplast degradation, during which a portion of chloroplasts are mobilized into the vacuole via autophagic vesicles termed Rubisco-containing bodies. Our recent study demonstrated that the activation of autophagy in photoassimilate-limited leaves is required for the production of free amino acids (AAs) as an alternative energy source. The catabolism of free branched-chain amino acids (BCAAs) is particularly important for survival under starvation conditions. These recent findings suggest an additional role for chloroplasts as a reservoir of AA when photosynthetic energy production is limited.

Chlorophyll catabolism precedes changes in chloroplast structure and proteome during leaf senescence

We observed that initiation of chlorophyll catabolism precedes other breakdown processes. Chloroplast size, stacking of thylakoids, and efficiency of PSII remain stable until late stages of senescence, whereas the number and size of plastoglobules increase. Unlike catabolic enzymes, whose level increase, the level of most proteins decreases during senescence, and chloroplast proteins are overrepresented among these. However, the rate of their disappearance is variable, mostly uncoordinated and independent of their inherent stability during earlier developmental stages. Unexpectedly, degradation of chlorophyll‐binding proteins lags behind chlorophyll catabolism. Autophagy and vacuole proteins are retained at relatively high levels, highlighting the role of extra‐plastidic degradation processes especially in late stages of senescence. The observation that chlorophyll catabolism precedes all other catabolic events may suggest that this process enables or signals further catabolic processes in chloroplasts.
