Biochemical Evaluation of the Ulcer Curative Effect of Aloe vera on Experimental Rats

Abstract

Objective: The present work was aimed to evaluate the curative properties of Aloe vera leaf gel on experimentally induced ulcer in rats. Methods: Ethanol was used for the induction of ulcer. An aqueous extract of Aloe vera leaf gel was administered orally to ulcer rats. Total number of lesions in the gastric area, total volume, acidity, levels of protein and glycoprotein components were determined in the gastric juice. Results and conclusions: The observed decrease in the number of gastric lesions in the treated rats suggested the cytoprotective and acid regulating properties of the leaf gel.

The production of gastric ulcers in rats treated with indomethacin may be linked to the indomethacin inhibitory effect on prostaglandins formation, where the inhibition increases gastric acid secretion, reduces mucosal blood flow and reduces bicarbonate secretion (Wallace, 2008). The anti-ulcerogenic activities of Securinega virosa aqueous leaf extract on gastric ulcer induced with indomethacin in albino rats were studied. Thirty rats weighing 120 - 200 g were grouped into six groups of five rats each. All groups except the uninduced-untreated (group A) were starved for 24 hours prior to indomethacin administration. After 4 hours of 30 mg kg-1 b.w. indomethacin administration, the groups (A, B, C, D, E and F) received once daily oral doses of distilled water (5ml kg-1 b.w.), cimetidine 60 mg kg-1 (b.w.) and the S. virosa extract at doses of 35, 70 and 140 mg kg-1 b.w. respectively for 11 days. At the end of the treatment, animals in groups B, C, D, E and F were starved for 18 hours then sacrificed. The extract significantly (p < 0.05) decreased gastric secretion volume, mean ulcer index, total acidity, total protein and pepsin secretion relative to the induced-untreated rats. The extract significantly (p < 0.05) increased the gastric pH and total carbohydrate content relative to the induced-untreated. These results were similar to those achieved by treatment with cimetidine. Catalase and SOD activities in the 35, 70 and 140 mg kg-1 bw S. virosa extract-treated groups were increased significantly (p < 0.05) over the untreated group. Similarly, the extract reversed the indomethacin-induced decrease in reduced glutathione level (GSSH) and the increase in malondialdehyde concentration in the serum. The histological analysis showed positive effect of the extract on the indomethacin-induced ulceration. It was concluded that the extract has anti-ulcerative and antioxidant activity in indomethacin-induced ulcerative rats.

The objective of the present study is to investigate the effect of ethanolic extract of Anethum graveolens L. on Aspirin-induced gastric ulcers in guinea pigs. thirty guinea pigs divided in to five groups (6 each group), first group received oral administration 2ml of normal saline served as negative control. 500mg/kg B.W of Aspirin was used to induced ulcer in guinea pigs for 3 days. after checking for presence of ulcers, the ulcerogenic guinea pigs were divided in to four groups, 2 groups were treated with ethanolic extract of Anethum graveolens L. at doses of (150mg/kg, 300mg/kg) respectively, while a group received cimetidine at dose 20mg/kg for 10days and the last group was the ulcer untreated control (control positive). At the end of the study the animals were sacrificed, the stomach was prepared for estimating, gastric PH , gastric ulcer index, total gastric area, gastric fluid volume, serum MDA and tissue MDA, in addition a gross and histological examination of the stomach. There were significant decrease in PH in control positive , while it showed significant increase(P≤ 0.05) of gastric ulcer index, total gastric area, gastric fluid volume, serum MDA and tissue MDA compared with the negative control group. The study also revealed significant decrease (P≤0.O5) in gastric ulcer index, gastric volume, total gastric area, serum MDA and tissue MDA with a significant increase(P≤0.05) in gastric PH in groups treated with ethanolic extract and in the group treated with cimetidine compared with the positive control group. Histological studies showed many pathological changes in stomach in control positive group but in the treated with ethanolic extract of Anethum graveolens L at dose 300mg/kg group the histopathological changes near to the normal status, whereas, higher dosage of extract (300mg/kg) was more superior to cimetidine drug in its protection of the stomach

In this study, the phytochemical screening and anti-anaemic effect of Aloe barbadensis leaves was investigated in nutritionally stressed rats. Preliminary phytochemical screening test of the plant revealed that it contained tannins, flavonoids, anthraquinones, saponins, alkaloids and cardiac glycosides. Sets of male albino rats of the Wistar strain, weighing between 100 and 105g, were fed on normal diet (20% protein) and low protein diet (2% protein) supplemented with the plant leaves (10% of diet) for ten (10) weeks. The results revealed that rats fed on the protein deficient diet supplemented with the leaves (PDDV) had significantly (p < 0.05) increased packed cell volume (PCV), haemoglobin (HB), red blood cells (RBC), platelets, total protein and albumin levels when compared with the protein deficient diet fed rats (PDD – negative control) which had significantly reduced (p < 0.05) levels of these parameters when compared with the normal diet fed rats (ND – positive control). The histopathological examination of the liver and kidney tissues showed evidence of severe fatty changes especially in the PDD fed rats whereas the PDDV fed rats showed moderate fatty changes relative to the control. These findings confirm local claims on the efficacy of A. barbadensis leaves in the treatment of anaemia as a result of its bioactive principles.

Annona reticulate (AR) is indigenous to the tropical areas of India and worldwide. The use of plant as remedy for diarrhea and ulcer is well documented in Ayurvedic system of medicine. However, pharmacological evidence does not exist to substantiate its therapeutic efficacy for the same. The aim was to investigate the antiulcer and anti-diarrheal activity of methanolic (ME) and aqueous extracts (AE) of A. reticulate in animal model. The antiulcer activity of extracts was investigated using ethanol and pylorus ligation-induced ulcer. The anti-diarrheal activity of MEAR and AEAR extracts was evaluated by castor oil induced diarrhea and gastro intestinal motility using parameters such as onset of diarrhea, number of wet stools, total number of stool and weight of total number of stools. The antiulcer activity of the extracts was confirmed by a reduction in ulcer index along with the decrease in gastric volume, total acidity, and an increase in pH of gastric content in both the models. A. reticulate extracts were more efficacious in reducing number of total stools in both the models of diarrhea and showed a dose-dependent antidiarrheal effect. The obtained results have established a pharmacological evidence for the folkloric use of the A. reticulate as antiulcer and antidiarrhoeal agent.

Gastric hyperacidity and gastro duodenal ulcer is a very common global problem today. Among various causes of gastric ulceration lesions caused by stress, alcohol consumption, H.pylori infection and use of NSAIDs have been shown to be mediated largely through the generation of reactive oxygen species (ROS), especially hydroxyl radical (*OH). Hyper secretion of gastric acid is a pathological condition, which occurs due to uncontrolled secretion of hydrochloric acid from the parietal cells of gastric mucosa through the proton pumping H+ K+ ATPase. The modern approach to control gastric ulcer is to inhibit gastric acid secretion, to promote gastro protection, to block apoptosis and stimulate epithelial cell-proliferation for effective healing. On contrary most of the green pharmaceuticals reduces the offensive factors and have proved to be safe, clinically effective, better patient tolerance, relatively less expensive and globally competitive.

The purpose of this study was to systematically assess 100% ethanol-induced gross and histologic injury of the antral and the corpus mucosa of the rat stomach and the effect of two protective agents, a prostaglandin analog and meciadanol (a flavenoid) on this injury. The gross antral injury was much more subtle than the corpus injury. Therefore, different scoring criteria were developed for the antrum. Surprisingly, however, the extent and severity of histologic injury was similar in both areas. Pretreatment with either meciadanol or 16, 16-dimethyl prostaglandin E2 significantly protected against this injury in the antrum as well as in the corpus.

Piracetam, used clinically for cognitive disorders, was found to have significant anti-ulcerogenic activity against immobilization stress- and aspirin-induced gastric ulcers in rats. The anti-ulcer effect of piracetam was exerted by augmentation of mucosal resistance. This was indicated by the significant attenuation of the decrease in total carbohydrate: protein ratio induced by aspirin. It also reversed the marked increase in gastric juice protein and DNA induced by aspirin, indicating that piracetam attenuated the augmented mucosal cell exfoliation induced by the ulcerogen. The drug also increased gastric mucosal serotonin concentrations. Piracetam, thus appears to have a profile of activity associated with cytoprotective agents.

We investigated whether the trophic actions of prostaglandins, omeprazole, and indomethacin on gastric mucosa lead to accelerated healing of gastric ulcers in the rat. Cryoulcers were produced in the corpus area and treated with 16,16-dimethyl prostaglandin E2 (5 or 100 micrograms/kg b.i.d., intragastrically), omeprazole (40 mumol/kg once daily, subcutaneously), indomethacin (2 mg/kg b.i.d., subcutaneously), or placebo. At the end of the treatment, plasma gastrin, cell labeling index (autoradiography with [3H]thymidine), and the size and depth of mucosal defects were measured. Compared with placebo, omeprazole accelerated ulcer healing as indicated by a smaller ulcer area [1.1 +/- 0.2 vs. 4.8 +/- 1.2 mm2 (mean +/- SEM)] and smaller ulcer depth (383 +/- 31 vs. 488 +/- 41 microns) after 10 days of treatment. Prostaglandins did not affect ulcer healing despite thickening of gastric corpus mucosa. Indomethacin delayed ulcer healing and reduced the labeling index. Omeprazole induced a marked hypergastrinemia (208 +/- 12 vs. 66 +/- 12 pmol/L on day 5, and 469 +/- 23 vs. 58 +/- 16 pmol/L on day 10). The results indicate that abolishment of acid secretion by omeprazole accelerates healing. Trophic actions and "cytoprotective" effects by prostaglandins are not relevant for ulcer healing in this model.

Copper salicylate has been shown to be an inhibitor of gastric lesions in rats induced by aspirin, indomethacin and cold stress whereas cimetidine was only effective against those produced by cold stress. The results are in line with those reported earlier for the anti-ulcer activity of sodium salicylate and metiamide respectively. On this basis, it appears that the animal model of stress-induced gastric lesions in the rat is the most suitable for testing the activity of new-antiulcer drugs.
