Spermatogenic activity of rhizomes of Curculigo orchioides Gaertn in male rats

Abstract and Figures

The rhizomes of Curculigo orchioides Gaertn have been traditionally acclaimed as aphrodisiac. In the present study ethanolic extract of rhizomes was evaluated for its effect on orientation behavior and spermatogenesis inalbino rats. A change in orientation behavior was assessed by orientation towards female, towards environment, towards self and type of mobility. Administration of 100 mg/Kg b. w. of ethanolic extract had pronounced effect onorientation of male towards the female rats. Males treated with the extractdisplayed more frequent and vigorous anogenital sniffing and mounting as compared to untreated animals. The increased spermatogenesis in treated groupwas confirmed by change in histoarchitecture as evidenced by increase in number of spermatocyte and spermatids. These findings support the folk use ofthis plant as aphrodisiac. Industrial relevance: Study proposes to elucidate and demarcate the potential of Rasayan herbs in the treatment as well as uprooting the causes of disease; which is the underlying principle of Rasayana therapy. This leads to discovery of newer phytoconstituents with betteractivities and provide source of new biomolecules for biotechnologists to work on. The study explore the utilization of Rasyana herbs for effective management of sexual dysfunction. It brings out a competent literature on rasayana, validating their utilization. It gives a larger platform for the herbal cultivators by providing scientific support and data to the traditionalunvalidated herbal drugs as rejuvenative tonics.

The aim of this study was to evaluate the effect of ethanol leaf extract of V. amygdalina on the testicle of alloxan induced diabetic wistar rats. Freshly harvested leaves of V. amygdalina was washed with clean tap water and subsequently dried at room temperature prior to being ground to fine powder. Twenty (20) adult male wistar rats were divided into four (4) groups of five (5) rats per group. · Group I was the normal control and was fed rat chow and water · Group II was induced diabetes without treatment, while · Groups III and IV were diabetic rats administered with 100 mg/kg and 200 mg/kg bw of V. amygdalina extract respectively. Sperm morphology, motility and count evaluated indicated that the aforementioned parameters were significantly (P<0.05) low in diabetic rats which were not treated but however improved in a dose dependent manner to values which though were significantly (P<0.05) lower than that of the normal control. In conclusion, this study shows that extract of V. amygdalina can ameliorate damage imposed on the testicle by diabetes mellitus.

The present study investigated the effect of oral administration of aqueous extract of Symphonia globulifera root on fertility in addition to its effect on some biochemical indices in adult male Wistar rats. After the aqueous extraction, phytochemical screening was carried out. Twenty-four (24) mature and healthy adult male wistar rats of weights between 150-220 g were used. The rats were randomly divided into four (4) groups each containing six (6) rats. Group 1 rats were gavaged with distilled water which served as control group. Group 2, 3 and 4 rats were orally treated with 100, 200 and 400 mg/kg body weight of the aqueous extract of S. globulifera respectively for 21 days. After the treatment, the rats were sacrificed and their organs (heart, liver, kidney and testes) were excised. Hormonal analysis which include seminal fluid analysis (SFA), follicle stimulating hormone (FSH), leutenizing hormone (LH), testosterone and prolactin were carried out following standard procedures. The data were analysed using students t-test and ANOVA with the aid of SPSS with P < 0.05 level of statistical significance. The phytochemical screening of the extract revealed presence of major phytochemicals. There was dose-dependent decrease in the body weight of the rats after the treatment. In hormonal analysis, SFA increased significantly when compared to that of the control rats. The analysis also revealed that the blood level of follicle stimulating hormone (FSH) was significantly increased whereas; there was decrease in testosterone level when compared with the control animals. In conclusion, aqueous extract of S. globulifera was found to be safe and showed significant fertility enhancing effects.

Curculigo orchioides , commonly called “Kali Musli,” is an endangered medicinal plant commonly found in Asian countries such as India, Japan, China, and Nepal. The plant holds a significant position in Ayurvedic and the Chinese traditional medicine system; it is documented as an aphrodisiac herb. The plant is also reported to be used in the treatment for asthma and jaundice. The botany, traditional uses, phytochemistry, and pharmacological activities to evaluate the plant's importance and relevant information are reviewed and summarized. We discern that a total of 61 phytochemicals are identified and reported in C. orchioides . These belong to the various phytochemical group of glycosides, lignans, polysaccharides, alkaloids, saponins, triterpenes, and aliphatic compounds. The most explored bioactive compound is a phenolic glycoside, curculigoside, isolated from the plant's rhizome. In vitro and in vivo research is conducted globally to provide primary and robust evidence to support this herbal medicine's traditional uses. A large lacuna regarding the mechanisms involved in the biological activity of the plant is evident. There is a need to conduct in-depth studies to understand the relationship between traditional and modern pharmacological uses of C. orchioides.

Both modern and traditional medicine options are being practiced worldwide to alleviate and treat diseases. However, some drugs used for certain diseases have deleterious effects on other systems and functions of the body. This study aims to investigate the impact of aqueous extracts of the stem bark of Anthocleista schweinfurthii on some parameters of the reproductive function of adult albino male rats, and to perform the phytochemical screening of the extract. To achieve this objective, 18 male rats aged 60 days and weighing 150 g were divided into three groups of six rats each and were orally treated for 28 days with: Distilled water (10 mL/kg) for group 1 (control group), aqueous extract of A. schweinfurthii at respective doses of 300 and 400 mg/kg for groups 2 and 3 (the two test groups). At the end of the 28-day period, the animals were anesthetized and decapitated. Blood samples were collected for serum analysis. Some androgen-dependent organs were collected and weighed. The epididymis and sperm were collected for evaluation of sperm count, mobility and viability. A sample of each organ (testis, epididymis and seminal vesicle) was homogenised for biochemical analysis and other parts were kept in Bouin's solution for histological examinations. The phytochemical screening revealed the presence of many secondary plant metabolites in the extract, including alkaloids, flavonoids and saponins. The results obtained show that the aqueous extract induced a significant reduction of the relative weight of the testis, the epididymis and the seminal vesicles. A reduction in sperm density, mobility and viability were also observed. The rates of testicular, seminal vesicle and epididymal proteins, serum cholesterol and seminal vesicle fructose decreased significantly. Otherwise, plant extracts increased testicular cholesterol concentration. Anthocleista schweinfurthii caused a significant decrease in serum gonadotrophin levels (LH, FSH) and serum testosterone concentrations and an improvement in parameters of oxidative stress. In addition, an alteration in the structure of the androgen-dependent organs was depicted on histological sections. From these findings, it is concluded that Anthocleista schweinfurthii stem bark alters some parameters of the reproductive function of adult albino male rats and suggest a limitation of their use in male subjects.

The aim of this study was to evaluate the antistress activity of rhizomes ethanolic extract of Curculigo orchioides Gaertn in vivo, using mice with stress-induced injury. The effect of Curculigo orchioides on motor coordination, spontaneous locomotor activity, immobility time, and analgesia was evaluated in mice using standard procedure. The ethanolic extract of Curculigo orchioides at dose levels, i.e., 100 and 200 mg/kg shows pronounced antistress activity evidenced by reduced immobility time in forced swimming and tail test. It also improved motor coordination and enhanced analgesic effect compared to standard diazepam. The Curculigo orchioides was found to be a antistress activity.
Chapter
In this chapter, the main causes of male factor infertility, specifically endocrine abnormalities and effect of endocrine disrupters, will be outlined. For many patients, conventional therapy presents considerable financial strain and moral dilemma. In this context, many patients are preferring complementary medicine (CAM). Hence, the promises offered by herbal medicine including Ayurveda, Arab, and Chinese medicine will be explored in this chapter. Many naturopathic medicaments such as Withania somnifera, Asparagus racemosus, Curculigo orchioides, Zingiber officinale, etc. are being routinely used as part of traditional medicine practice in Ayurveda and Arab medicine and gaining wider acceptance in other countries. Common herbs, targeted infertility condition, and results thereafter shall be discussed. In the concluding part of the chapter, the potential deleterious contraindications of alternate medicine such as lead toxicity from Ayurvedic medications shall be briefly discussed.

Evaluation of hepatoprotective activity, against drug (paracetamol and rifampicin) induced hepatotoxicities in albino rats, of powdered drug as well as various extracts of the rhizomes of Curculigo orchioides Gaertn. (kali musli) was carried out with a view to substantiate the incorporation of this drug in various polyherbal formulations available for the purpose. The methanolic extract against paracetamol and powdered rhizomes against rifampicin induced hepatotoxicities exhibited significant hepatoprotective activity (P < 0.01) like that of silymarin, a known hepatoprotective agent.

Curculigo orchioides Gaertn. (common name: Kali musli; Fam: Hypoxidaceae), a known raditional medicament in the treatment of joint pains and jaundice, was screened for its anti-inflammatory and hepatoprotective activity. The powdered rhizomes and aqueous extracts showed marked hepatoprotective activity when compared to silymarin, a known hepatoprotective agent. The methanolic and aqueous extracts showed significant anti-inflammatory activity when compared to ibuprofen and indomethacin.

Methanol extract of Curculigo orchioides was screened for in- vitro antioxidant activity using DPPH assay, superoxide scavenging, nitric oxide scavenging, determination of reducing power and in-vitro lipid peroxidation. Methanol extract was found to be extremely effective in scavenging superoxide radical (IC50 29.28 μg/ml) whereas activity was moderate in scavenging DPPH radical (IC50 105.94 μg/ml), nitric oxide radical (IC50 90.96 μg/ml) and in inhibition of lipid peroxidation (IC50 94.78 ìg/ml). Methanol extract showed different levels of antioxidant activities in tested models. It also supported anti oxidant activity by showing significant reducing power. Further HPTLC finger print profile of the methanol extract was established to facilitate its identification and characterization. Present study demonstrated antioxidant activity of methanol extract of Curculigo orchioides in-vitro.

The roots of Chlorophytum borivilianum (Liliaceae) Sant. & F. roots are used traditionally in Indian subcontinent as aphrodisiac and sexual stimulant. Ethanolic extract of roots as well as sapogenins isolated from the roots were studied for effect on sexual behavior and spermatogenesis in albino rats. Administration of 100 mg/Kg and 200 mg/Kg b.w. of the sapogenin and ethanolic extract respectively had pronounced anabolic and spermatogenic effect in treated animals as evidenced by weight gains in the body and reproductive organs and histological studies. The treatment also markedly affected sexual behavior of animals as reflected in reduction of mount latency, ejaculation latency, post ejaculatory latency, intromission latency and an increase of mount frequency and attractability towards female.

In the present study, we describe two procedures for isolation and culture of Sertoli cells from the testes of adult Siberian hamsters. In procedure I, collagenase and pancreatin were used for differential enzymatic digestion of the tissue at 32 degrees C, and effects of several attachment factors were examined. Sertoli cells isolated from adult hamsters were not affected by addition of Na selenite, epidermal growth factor, insulin, and transferrin, with or without 5% fetal bovine serum, to Dulbecco's modified Eagle's medium + Ham's F-12. Significant increase in the yield and plating efficiency of Sertoli cells was achieved by developing a novel procedure (procedure II) for Sertoli cell isolation. In this procedure, testicular tissue was exposed to only one enzyme (collagenase I) for two consecutive digestions at 37 degrees C, and the total period of enzymatic exposure was reduced relative to that of procedure I. Another objective of this study was to compare the functions of Sertoli cells isolated from spermatogenetically active and inactive adult testes. Isolated Sertoli cells from 60 +/- 5-day-old hamsters raised in long day conditions (LD, 16L:8D) or in short day conditions (SD, 6L:18D) were stimulated with FSH and testosterone in the presence of 35S-methionine for 24 h on Day 4 of culture. The medium was concentrated, and equal amounts of radioactive proteins from LD and SD hamster Sertoli cell cultures were analyzed by two-dimensional PAGE. Compared to Sertoli cells derived from SD hamsters, Sertoli cells from LD animals produced greater amounts of two secretory proteins and a smaller amount of one.(ABSTRACT TRUNCATED AT 250 WORDS)
