Abstract
Intraditionalpistachiocultivation,priortosowingseedsthelatteraresoakedin
waterfordifferenttimesasfarmersclaimthatthiswillcontributetogermination.To
ourbestknowledge,thisisthefirstattempttoevaluatetheeffectofdifferentwater-
soakingtimeperiodsongerminationinpistachio.Forthispurpose,seedsofpistachio
(P. veraL.‘Badami’)weretreatedwithwaterfor1,6,12and24h.Wemonitored
germinationpercentage,germinationrate,meantimeforgerminationrate,and
seedlingheightatdifferentstages,numberofleaves,shootandrootdryweight,and
trunkdiameter.Ourresultsindicatedthatgerminationpercentageandrate
significantlyincreasedandseedlingheightincreasedinseeds,whichweretreatedfor
24h.Although,plantrootdryweightsignificantlydecreasedinseedsofthis
treatment,meantimesforgermination(MTG)valueswerenotfoundtobestatistically
significantbetweentreatments.Weobservedthatthe6hsoakingtreatmentledtoan
increaseinshootandrootdryweight,andtrunkdiameter.Alsoleafnumbersandroot
lengthvalueswerehighestinthe1hsoakedseeds,althoughdifferenceswerenot
significant.Positiverelationshipwasobservedbetweentheplantshootlengthand
plantleafnumber.

INTRODUCTION
Pistachio species belong to the Anacardiaceaefamily.Pistaciavera L., i.e., cultivated
pistachio,is the most importantspecies in the genusthat are all deciduousand dioecious.
Pistachioisanimportantfruittreegrowninthesub-tropicalareasintheworldwithhighest
production figures for Iran, Turkey, Syria and USA. Seed germination is the common
procedure for large scale propagation of pistachio seedlings. Seedlingsaregraftedto
desirable cultivars in nursery or field conditions to establishthepistachioproductive
orchards(Panahietal.,2002).
Seed quality is affected by several factors and seed germination percentage,
germination rate and seed vigor index have an important role in determination of seed
quality (Black and Bewley, 2000; Rajjou et al., 2012). To begin the germination process in
dried seeds, priming, pre-soaking, scarification, stratification and chemical treatments are
necessaryand the typeof treatment used depends on species, cultivarsandseedfeatures.
Pistachio species have different seed germination patterns; mostofthemhavedormancy
andneedspecialtreatmenttobreakdowntheirdormancy(Panahi et al., 2002). Acid
treatmentandgibberellicacid(GA3)areoftenusedtobreakdownespeciallythickseedcoats
(RahemiandBaninasab,2000)inwildpistachiorootstocks.Khanetal.(1999)reportedwild
pistachioseedstreatedwithKNO3at1%solutionsignificantlycompletedearlygermination
in135days,increasedthegerminationpercentageandproducedseedlings with higher
lengthanddiameterwhensoakedfor24h.Soakingseedsinpolyaminesisausefulmethod
to induce lateral root in pistachio seedlings (Sedaghat and Rahemi, 2011). Studies on
germination of citrus seeds indicated that osmotic stress causes decline of seed water
content,whichresultsininhibitionofseedgerminationandseedlinggrowth(Zekri,1993).
Thereisno dormancy in P. veraseedsand additionallythey haveahighgermination

percentage after being soaked in water (Panahi et al., 2002), thedurationofsoakingis
howeveraveryimportantparameterinachievingthemaximumgerminationpercentage.As,
too lengthy periods of soaking cause bacterial decay of the embryo, prior to the actual
germination, and inadequate duration causes inactivation of the enzyme facilitating the
germination. Thus, both of these conditions cause a decrease in seed germination
percentageandrate,andanoveralldeclineinthesubsequentseedlinggrowthparameters.
The objective of this study was to evaluate the effects of seed soaking with varying
durationsinwatertofind,optimizeandfinallystandardiseadurationtimeforoptimalseed
germinationfeaturesandsubsequentgrowthparameters of seedlingsofthemostcommon
pistachiocultivar,whichisusedaspistachiorootstockinIran.
MATERIALSANDMETHODS
TwoexperimentswereconductedinagreenhouseattheBioscience Engineering
Faculty, Ghent University (51°3’N, 3°42’E). Certified seeds of pistachio, PistaciaveraL.
‘Badami’ were obtained from the Iranian Pistachio Research Institute (30°39’N, 55°94’E),
Rafsanjan,Iran.Seedsizevariedfor17.9mm(length),10.6mm (width), 11.4 mm (thick)
and0.87g(weight).
Experiment1
Seedsof‘Badami’pistachiocultivarwerepre-treatedwithdistilledwaterfor1,6,12
and24hassoakingtimetreatments.Thenallpre-treatedseedsweredisinfectedwith0.01%
Captan,abroad-spectrumfungicidefor20min(Panahietal.,2002).Disinfectedseedswere
placed in petri dishes on Whatman filter paper and moistened with distilled water. Then
seedswerekeptinlabconditionsfor21daysandtheirrigationofthem wasperformedby
distilled water regularly. The germinated seeds were recorded every day and germination
percentagecalculatedonthe21stday.
Allgerminatedseedsweresownimmediatelyin4-Lpotscontainingsandandorganic
materialandkeptingreenhouseconditions. Plant cultural requirements were done
according to good agricultural practices during the experimental time. After 30, 40 and
50daysofsowingdate, shootheightandrootlengthweremanuallymeasuredwitharuler,
shootdiameterdeterminedwithadigitalcaliperandseedling(shoot and root) dry weight
wasdeterminedafterdryingat85°Cfor48hinanelectricoven. Germination percentage
andrate,mean of germinationtimeandvigorindexwere calculatedaccordingtofollowing
equations(EllisandRoberts,1981):
- Germination percentage (GP): GP (%) = SNG/SNO×100; where SNG is the number of
germinated seeds and SNOisthenumberofexperimentalseedswithviability,
respectively.
- Germinationrate(GR):GR(%)= SNM/SNG×100;whereGRisthegerminationrateand
SNMisthenumberofgerminatedseedsonagivenday.
- Mean of germination time (MGT): MGT is an index for germinationrate.MGT=
∑ (nd)/∑n (day-1);wherenisnumberofgerminatedseedsindayd,∑nistotal
germinatedseeds,Ddayofcounting.
- Vigorindex(VI):VI=GP×seedlingdryweight(g)(Abdul-BakiandAnderson,1973).
Experiment2
The similar seeds of ‘Badami’ pistachiowereputinpetridishes.Threepetridishes
were considered for each treatment and control. Polyethylene glycol (PEG) solutions were
preparedin0.2,0.8,and1.5MPaconcentrationsand5mLofeach solution was added to
petri dishes containing seeds. Also distilled water was applied to control petri dishes. All
petridisheswereplaced in lab conditions and every24hthenumberofgerminatedseeds
wascountedandincaseofmoisturedeficiencydistilledwaterwasused.
Statisticalanalysis
Treatmentswere arranged in a randomized completedesign with fivereplicatesper
treatmentsinexperiment1andthreereplicatespertreatmentsinexperiment2.Atwo-way

analysisofvariancewasusedtotestfortreatmentdifferencesandeffects.Meanswere
comparedbyaDuncantest.AllanalyseswereperformedinSPSS20(IBMCorporation,USA).
RESULTSANDDISCUSSION
Experiment1
1.Germinationpercentage(GP).
Thegerminationpercentage ofseedsincreasedwithincreasing thesoakingtimesfor
pistachioseeds(Figure1).Seedssoakedfor24hwasthemosteffectivetreatmentonGPand
treatmentfor1hshowedthelowestpercentage.Statisticalanalysisshowedthattherewere
significant differences between these two treatments (Figure 1). These results are in
agreementwiththeresultsofSedaghatandRahemi(2011)thatreported application of
polyamines for 12 h compared to 6 h increased percentage of germination capacity in
‘Ghazvini’pistachioseeds.While,Khanetal.(1999)showedthatnormalwatersoakinghad
better germination percentage in 48 h duration compared to 12, 24 and 36 h duration in
wildpistachio.

2.Meangerminationtime(MGT).
Meangerminationtimesignificantlydecreasedbyincreasingsoakingtimedurationof
seedsto12h.Althoughsoakingseedsfor24hsignificantlyincreasedcompared to 12 and
6htreatments(Figure2).Therewasnosignificantstatisticaldifferencebetween1and24h
treatmentsin ‘Badami’pistachiocultivar(Figure2).SedaghatandRahemi(2011)indicated
thatmeangerminationtimeofpistachiowasenhancedbysoakingtimeofseedsin
polyamines.

AsGPsignificantlyincreasedinseedswhichweresoakedinwaterfor24hcomparedamong
othertreatments,soakingofseedsfor6hhadthelowestpercentage of GP in ‘Badami’
pistachio cultivar (Table 1). There were significant differences between soaking times on
germinationpercentageinthiscultivar.

4.Vigourindex(VI).
The results of analysis showed that prolonging seed soaking times decreased the
vigourindexinpistachioseeds,althoughtherewerenosignificant differences among
treatments (Table 1). These results are in agreement with the finding of Oskouie and
Divsalar(2011)whichshowedthatvigourwasnotaffectedbydifferent level of nitrogen
fertilizeronmotherplantsofrapeseed.
5.Seedlingheight.
Observationof experimentshowedthattherewerenosignificantdifferencesinplant
height (cm) between different treatments in different periods after sowing the seeds
(Figure3),although,thereweremoreheightvaluesinthe24-htreatmentafter40daysin
‘Badami’ pistachio cultivar (Figure 3). These results are in agreement with the results of
SedaghatandRahemi(2011)thatreportedapplicationofpolyaminesfor12hcomparedto6
hincreased the shoot lengthin ‘Ghazvini’ pistachio seeds. While, Khan etal. (1999) found

6.Rootdryweight.
There were significant differences in root dry weight between 6and24soaking
treatments. Seeds which treated for 24 h had the lowest root dryweightamongother
treatments(Figure4).Applicationofpolyaminesanddistilledwater (control) for 12 h
comparedto6hdecreasedthe dry weight of roots in ‘Ghazvini’pistachioseeds(Sedaghat
andRahemi,2011).
There were no significant differences among different seed soaking times for leaf
number,rootlength,shootdryweight,plantdryweight,andtrunk diameter (Khan et al.,
1999)ofpistachioseedlings(Table1).

Evaluationsofthisexperimentshowedthattherewerenogerminatedseedsinanyof
thePEGsolutiontreatmentsexceptthecontrol.Aswecouldnotrecordanydataonthispart
ofourexperiment,wewerenotabletocomparethePEGtreatments with control.Inother
words,osmoprimingcausedbyallPEGtreatmentspreventedthegermination of pistachio
seeds in this experiment. These results are in agreement with thefindingofZekri(1993)
that indicated in germination of citrus seeds osmotic stress causes decline of seed water
content, results in inhibition of seed germination and seedlinggrowth.Althoughsome
references indicated that increasingPEG concent rationsdecreased germination percent in
canolacultivars(Shahverdikandietal.,2011)andinZiziphuslotus(Maraghnietal.,2010).
CONCLUSIONS
Theresultofthisgreenhouseexperiment revealedthat soakingseedsofpistachiofor
24hindistilledwaterimprovedgerminationcapacity,germination rate and plant length
growthcomparedtoothertreatments.Although,effectsofthistreatmentonrootdryweight
androotlengthweredecreasedcomparedto1and6htreatments, a positiverelationship
wasobservedbetweentheplantshootlengthandplantleafnumber. Thus, it is
recommendedtousedistilledwaterfor24hforPistaciavera(differentcultivars)seedsthat
lack dormancy to achievethe bes t germination parameters and plant length growth rates.
Theauthors suggestedevaluatingtheeffectsofPEGasprimingorosmoprimingtreatments
onotherpistachiocultivarsandspeciesseeds.

У статті представлено наукове обґрунтування балкового комплексу неподалік с. Вакалівщина Сумського району Сумської області як перспективного ландшафтного заказника місцевого значення «Вакалівські схили» та запропоновано природоохоронний режим цієї території. За даними публічної кадастрової карти України території перспективного ланд- шафтного заказника зазначені як землі запасу. Площа заказника «Вакалівські схили» складає 52,37 га. Дослідження біорізноманіття проводили з 2014 по 2020 роки. При цьому використовували маршрутно-польовий ме- тод для вивчення флори і ліхенобіоти та стандартні ентомологічні методи ‒ для вивчення ентомофауни. Інвентаризація флористичного біорізноманіття показала наявність 128 видів вищих судинних рослин, з яких чотири види є регіонально рідкісними (Campanula persisifolia L., Muscari neglectum Guss. ex Ten., Ranunculus illyricus L., Jurinea arachnoidea Bunge.) та один вид (Adonis vernalis L.) представлений у Червоній книзі України і Міжнародному списку охорони природи. На території перспективного заказника знайдено рідкісні лишайники Sarcosagium campestre (Fr.) Poetsch & Schied. та Peltigera didactyla (With.) J. R. Laundon. Останній є кандидатом до запланованого Червоного списку Сумської області, а біотопи, в яких ці лишайники зростають, потребують збереження. В результаті ентомологічних досліджень було зареє- стровано шість видів комах, що занесені до Червоної книги України (Papilio machaon (L.), Iphiclides podalirius (L.), Parnassius mnemosyne (L.), Zerynthia polyxena (Den. & Schiff), Aglia tau (L.), Catocala fraxini (L.)) та двадцять видів комах, що входять до списку регіонально рідкісних видів. Для забезпечення ефективного збереження біорізноманіття та природних комплексів зазначеної території розроблено природоохоронний режим його території відповідно до Закону України «Про природно- заповідний фонд України». Балкова система з лучно-степовим фітоценозом має особливе природоохоронне, наукове, естетичне та освітнє значення. Перспективна природоохоронна територія повинна бути створена, з метою збереження ландшафтного та біологічного різноманіття, генофонду тваринного і рослинного світу, підтримання загального екологічного балансу в ре- гіоні та забезпечення фонового моніторингу навколишнього природного середовища. Quinoa crop is considered as a source of healthy nutrition due to its energy and nutritional value. The nutritional parameters of quinoa seeds exceed the characteristics of traditional cereals. The high protein content distinguishes quinoa seeds from other plant food. Quinoa contains all the important amino acids, it is rich in minerals and vitamins. The special chemical composition of grain can ensure the production of gluten-free food products. Quinoa due to its unique chemical composition and lack of gluten, can be used as a valuable raw material for the creation of specialized (gluten-free) products for people suffering from celiac disease and for products of high nutritional value. This crop may be an alternative one in areas with water deficiency, droughts and salinization, where other crops are unprofitable. Problematic aspects of the crop production are level of seed germination in the field condition, low and unstable yields. The main condition for the quinoa spread, like other plant species, is the success of the initial phases of plant development. The ability to form in new growing conditions a crop with certain parameters of standing density provides the possibility of further steps in the beeding process and technological optimization of crop indicators. The evolutionary way of the trait formation such as the absence of the seed dormancy period are analyzed, the mechanisms and dynamics of the decrease in the viability of seeds in the modern crop of quinoa are revealed. Significant differences in the seed characteristics of 2019 and 2018 harvest years (storage period of 14 and 28 months, respectively) indicate the difficulty of maintaining the economic indicators of seeds for the 2-nd and subsequent storage years. Based on the experiment result to determine the dynamics of water absorption of seeds, their reactions to soaking and heating, the basic characteristics of quinoa breeding samples (perspective for introduction in the zone of the north-eastern forest-steppe of Ukraine) were determined. It was found that the level of passive water absorption was in the range of 79.7–81.5 % of the mass of dry seeds. The maximum intensity of water absorption was fixed within 1-st hour. Seeds with a longer storage period are characterized by more intense initial water absorption. At temperature of 18 °C, the total duration of the seed swelling period is 7‒8 hours. The highest efficiency of seed soaking was noted for seed samples of the 2019 harvest year, compared with which the growth of indicators of germination energy and laboratory germination was + 9.5 and + 5.8 %. The same seed sample had the best parameters according to the results of heating: + 6.9 and + 2.4 % respectively.

The experiment conducted with lettuce, tomato and maize seeds pre-soaked in 5, 10, 20 per cent vermicompost concentrations and NPK fertilizer for 10 hrs. Result obtained showed that vermicompost at 10 per cent concentration produced maximum seed germination in lettuce and tomato, while maximum germination in maize seeds was obtained with 20 per cent vermicompost concentration. There was a significant difference in the number of leaves, length of shoot, length of root, wet and dry weight of plants tested at (P<0.01) across treatments. It was concluded that pre-soaking seeds in vermicompost leachate was beneficial in maximising plant growth.

The rapid rate of deforestation in Côte d’Ivoire has led to loss of plant species diversity and also threatens some commercial tree species with extinction. Some reforestation and afforestation has been completed. However, for some species, the numbers of surviving transplants has declined. Hence, it is necessary to develop properly adapted and resilient genotypes that can effectively support the programs of forest restoration. In this context we evaluated the effects of the provenances of seeds and of various treatments on the germination rates and on plant growth of four commercial trees species (Pycnanthus angolensis, Terminalia superba, Mansonia altissima and Pterygota macrocarpa). We analyzed these parameters for seeds of three provenances (Daloa, Akoupé and Daoukro) of four species and for thirty plants aged 6 months per provenance. Time to begin germination and time to reach maximum germination were similar among species, and were not influenced by either treatment or provenance. Untreated seeds and those soaked for 24 and 48 h in water at room temperature yielded the highest germination rates, irrespective of species or provenance. The one exception was P. angolensis, for which no germination was recorded. Within species, some provenances produced higher germination percentages than did others. No clear relationship was computed between the germination rate and the morphometric characteristics of the seeds which varied significantly between provenances. Plant growth and morphological variability also depended greatly on provenance. Genetic factors might be implicated, therefore provenance trials should be undertaken and evaluated. Furthermore, molecular analysis should be undertaken to confirm the implications of genetic factors and allow for genetic selection.

This experiment was conducted as factorial experiment on completely randomize design with 4 replications. Experimental factors was canola varieties (RGS003, SUNJA, JEWEL, OPERA, ADDER, HYOLA60, ELITE, ELRIS, WESTER and COANTOM) as a first factor and different osmotic levels, made by PEG6000 (0, 0.4 and 0.9 MPa) as second factor. Results showed that main effects of variety and drought interactions of them were significant on germination percent, mean time to germination, means of daily germination, daily germination rate, coefficient of velocity of germination and seedling vigor index. Increasing drought level decrease germination percent in all variety. RGS003 was tolerant to drought stress and ELITE was susceptible one. Variety with high germination percent had lowest germination rate and variety with low germination percent exhibit more daily germination rate.

Ziziphus lotus (L.) Lam. is a deciduous shrub with intricately branched stems in the Rhamnaceae family. It's a dominant and economically important species widely distributed in active sand dunes in the southern desert of Tunisia. To provide basic information for its conservation and reintroduction, we studied the influence of environmental factors on seed germination patterns. The germination responses of seeds were determined over a wide range of constant temperatures (10–50 °C), polyethylene glycol (PEG)-6000 solutions of different osmotic potentials (0 to − 1 MPa) and burial depths (1–10 cm). Temperatures between 15 and 45 °C seem to be favorable for the germination of this species. Germination was inhibited by either an increase or decrease in temperature from the most suitable temperature found (35 °C). The highest germination percentages (100%) were obtained under control conditions without PEG, and increasing moisture stress progressively inhibited seed germination, which was less than 5% at − 1 MPa. When tested for germination in distilled water, after PEG treatments, seeds germinated to the same extent as when fresh. When seeds buried deeply, there was a significant decrease in seedling emergence percentage and rate. Seedlings of Z. lotus emerged well at depths of 1–2 cm and could not emerge when sand burial depth was > 4 cm.

Germination vigor is driven by the ability of the plant embryo, embedded within the seed, to resume its metabolic activity in a coordinated and sequential manner. Studies using "-omics" approaches support the finding that a main contributor of seed germination success is the quality of the messenger RNAs stored during embryo maturation on the mother plant. In addition, proteostasis and DNA integrity play a major role in the germination phenotype. Because of its pivotal role in cell metabolism and its close relationships with hormone signaling pathways regulating seed germination, the sulfur amino acid metabolism pathway represents a key biochemical determinant of the commitment of the seed to initiate its development toward germination. This review highlights that germination vigor depends on multiple biochemical and molecular variables. Their characterization is expected to deliver new markers of seed quality that can be used in breeding programs and/or in biotechnological approaches to improve crop yields.

Beneh (Pistacia mutica F. & M.) and kolkhong (Pistacia khinjuk Stock) are wild species of pistachio which grow naturally with other trees in some parts of Iran. Because of their adaptibility to severe environmental conditions and their resistance to some pests and diseases, they can be used as rootstocks for pistachio cultivars. Poor germination and very low seedling vigour of these two species have been a major problem in using them as rootstocks for pistachio cultivars. In this study gibberellic acid (GA3), at five concentrations (100, 250, 500, 750 and 1000 mg l21), were used during and after stratification to enhance seedling growth. The results showed that GA3, applied during and after stratification, significantly increased the length, trunk diameter, internode length, leaf area and fresh and dry weight of seedlings of both beneh and kolkhong species. However, application of GA3 after stratification was more effective on seedling growth of beneh. GA3 applied at higher concentrations (500 and 750 mg l21) increased the rate of growth, but growth malformations were clearly evident in seedlings of kolkhong. GA3 at 250 mg l21 enhanced seedlings growth of kolkhong. It was concluded that application of GA3 to the seedlings might be a useful method for promoting rapid shoot growth of beneh and kolkhong and reduce the expense of seedling production in glasshouse conditions.

A multiple criteria approach is presented by which vigor was evaluated in 16 lots of soybean ( Glycine max L., cv. ‘Lee‐68’) seed by determining O 2 uptake, CO 2 production, uptake of labeled glucose or leucine, conversion of isotopes into ¹⁴ CO 2 and polysaccharides or proteins, and leaching of metabolites through membranes of excised embryonic axes after 5 hours of imbibition. The concept upon which this approach is based is that excised embryonic axes from vigorous seed lots take up more sugars and amino acids from imbibing media, incorporate these metabolites faster into polysaccharides and proteins, and permit less leaching of nonused metabolites into surrounding aqueous media than axes from less vigorous seed lots. These measurements focus on the biosynthetic capacity of the axis and the integrity of its cellular membranes.

The effects of isosmotic solutions of sodium chloride (NaCl), polyethylene glycol (PEG), and mannitol with or without supplemental calcium (Ca) were compared to separate osmotic effects from toxic ion effects of salts on seed imbibition, seedling emergence, and the early stage of seedling growth of three citrus rootstocks. The rate of seed imbibition was relatively low for sour orange (SO) in PEG treatments which caused a delay in seedling emergence. These results of PEG supported earlier findings which suggested the existence of a critical level of hydration that seeds must reach before germination can proceed. Final percent emergence (FPE) was reduced the most by NaCl and mannitol compared with PEG. Furthermore, Ca addition to mannitol and NaCl solutions improved FPE in Cleopatra mandarin (CM) and trifoliate orange (TO). These results demonstrated that the effects of mannitol and NaCl on FPE went beyond the osmotic effects. Seedling growth was reduced the most by mannitol not only due to its uptake and transport to the leaves and stems where it caused dehydration and visible leaf injury but also to reduction in shoot Ca and potassium (K) concentrations. This study showed that the effects of salts on seed imbibition and time to emergence of the first seedling was primarily osmotic, while their effects on FPE and seedling growth was mainly toxic.

A research project was envisaged to standardised a stratification technique for better seed germination and to produced a uniform rootstock for cultivation of pistachio.Seed were immersed in different solutions of KNO3 1 per cent, MS salt, GA3 1 ppm, normal water and control (no treatment), for 12, 24, 36 and 48 hours duration before sowing in pots. It was found that KNO3 at 1 per cent solution treated seeds significantly completed early germination in 135 days and increased the germination percentage to 26 per cent when soaked for 24 hours. Lengthy seedling of wild Pistachio (39.50 cm) was noted in the same solution for 24 hours immersion. Diameter of the seedling was pronounced in the above chemical at 1 per cent level of concentration.
