Curcumin: A Potential Bioactive Agent

Abstract

Curcumin (diferuloylmethane), the main yellow bioactive component of turmeric has shown a wide spectrum of biological actions. Clinically, curcumin has already been used to reduce post-operative inflammation. Safety evaluation studies indicate that curcumin is well tolerated at a very high dose without any toxic effects. Moreover, curcumin have the potential for the development of modern medicine for the treatment of various diseases. This review paper serves to highlight the extensive work done to establish the biological and pharmacological actions, safety and stability aspects along with the structure and possible modifications in curcumin to enhance its bioeffectiveness.

INTRODUCTION
Curcumin is the active ingredient in the herbal remedy and dietary spice turmeric
(Curcuma longa Linn). Curcumin was identified to be responsible for most of the biological
effects of turmeric [1].This vibrant yellow spice, derived from the rhizome of the plant, has a
long history of use in traditional medicines of China and India [2]. Use of curcumin as a folk
remedy continues today. As part of the ancient Indian medical system, ayurveda, a poultice of
turmeric paste is used to treat common eye infections, and to dress wounds, treat bites, burns,
acne and various skin diseases [3]. In Northern India, women are given a tonic of fresh turmeric
paste with powder of dried ginger roots and honey in a glass of hot milk to drink twice daily
after childbirth. A poultice of turmeric is also applied to the perineum to aid in the healing of
any lacerations in the birth canal [4]. Powdered turmeric is taken with boiled milk to cure cough
and related respiratory ailments [3] and roasted turmeric is an ingredient used as an
antidysenteric for children [3]. This ancient remedy is also used to treat dental diseases,
digestive disorders such as dyspepsia and acidity, indigestion, flatulence, ulcers, as well to
alleviate the hallucinatory effects of hashish and other psychotropic drugs [5]. In food and
manufacturing, curcumin is currently used in perfumes and as a natural yellow coloring agent,
as well as an approved food additive to flavor various types of curries and mustards [5-6].
Recent emphasis on the use of natural and complementary medicines in Western medicine has
drawn the attention of the scientific community to this ancient remedy.
CHEMICAL COMPOSITION OF TURMERIC
Turmeric contains a number of phytoconstituents namely protein (6.3%), fat (5.1%),
minerals (3.5%), carbohydrates (69.4%) and moisture (13.1%). The essential oil (5.8%) obtained
by steam distillation of rhizomes has a-phellandrene (1%), sabinene (0.6%), cineol (1%), borneol
(0.5%), zingiberene (25%) and sesquiterpines (53%) [7]. Curcumin (diferuloylmethane) (34%) is
responsible for the yellow colour, and comprises curcumin I (94%), curcumin II (6%) and
curcumin III (0.3%) [8]. Demethoxy and bisdemethoxy derivatives of curcumin have also been
isolated. Curcumin was isolated in 1815 and its chemical structure was determined by Roughly
and Whiting [9] in 1973. It has a melting point at 176177°C; forms a reddish-brown salt with
alkali and is soluble in ethanol, alkali, ketone, acetic acid and chloroform.
PHARMACOLOGICAL ACTION OF CURCUMIN
Effect on cardiovascular system (CVS)
Curcumin protects from damage caused by myocardial infarction [10] by decreasing the
severity of pathological changes. Curcumin improves Ca2+ transport from the cardiac muscle
sarcoplasmic reticulum, thereby raising the possibility of pharmacological interventions to
correct the defective Ca2+ homeostasis in the cardiac muscle [11]. Curcumin has significant
hypocholesteremic effect in hypercholesteremic rats [12]. Oral administration of curcumin

significantly decreases the lipid peroxides (33%), increased HDL cholesterol (29%), decreased
total serum cholesterol (11-12%) [13].
Effect on gastrointestinal system (GI)
Stomach: Turmeric powder has beneficial effect on the stomach. It increases mucin secretion in
rabbits and may thus act as gastroprotectant against irritants [14]. Controversy exists regarding
antiulcer activity of curcumin. Both antiulcer [15] and ulcerogenic [16-17] effects of curcumin
have been reported but detailed study is still required. Curcumin has been shown to protect the
stomach from ulcerogenic effects of phenylbutazone in guinea pigs at 50 mg/kg dose [18-19]. It
also protects from 5-hydroxytryptamine-induced ulceration at 20 mg/kg dose [18-19]. However,
small concentration of 0.5% curcumin failed to protect against histamine-induced ulcers [20].
Intestine: Curcumin has some pharmacological effects on the intestine also. Antispasmodic
activity of sodium curcuminate was observed in isolated guinea pig ileum [21]. Anti-flatulent
activity was also observed in both in vivo and in vitro experiments in rats [22]. Curcumin also
enhances intestinal lipase, sucrase and maltase activity [23].
Liver: Curcumin and its analogues have protective activity in cultured rat hepatocytes against
carbon tetrachloride (CCl4), D-galactosamine, peroxide and ionophore-induced toxicity [24].
Curcumin also protects against diethyl nitrosamine and 2-acetylaminofluorine-induced altered
hepatic foci development [25]. Increased level of bile was reported in dogs by both curcumin
[26].
Pancreas: Curcumin increases the activity of pancreatic lipase, amylase, trypsin and
chymotrypsin [27]. A synthetic derivative of p-tolylmethylcarbinol (an ingredient of C. longa) 1-
phenyl-1-hydroxy-n-pentane increases bicarbonate levels as well as plasma secretion [28].
Effect on nervous system
Curcumin and its complex with manganese offer protective action against vascular
dementia by exerting antioxidant activity [29-30]. Curcumin also reduces oxidized proteins in
amyloid pathology in Alzheimer transgenic mice [31]. It also decreases lipid peroxidation in rat
liver microsomes, erythrocyte membranes and brain homogenates [32].
BIOLOGICAL ACTION OF CURCUMIN
Turmeric plant extract (curcumin and other derivatives) have anti-inflammatory, anti-
arthritic, antioxidant, anti-microbial, anti-leishmanial, hepato protective, anti-cancer, anti-ulcer
and anti diabetic activity.

Anti-inflammatory agents
The water extracts of C. longa along with volatile oil, petroleum ether and alcohol shows
anti-inflammatory effects [33]. The natural analogues of curcumin, i.e. FHM and BHM, are also
potent anti-inflammatory agents. Curcumin is effective against carrageen induced oedema in
rats [33] and mice [34]. Curcumin offers anti-inflammatory effect through inhibition of NFkB
activation [35]. Curcumin has also been shown to reduce the TNF-a-induced expression of the
tissue factor gene in bovine aortic-endothelial cells by repressing activation of both AP-1 and
NFkB. The anti-inflammatory role of curcumin is also mediated through down regulation of
cyclooxygenase-2 and inducible nitric oxide synthetase through suppression of NFkB activation
[36].
Anti-cancer activity
Anti-cancer activity of curcuma is by inhibiting tumour cell proliferation, induction of
analysis, inhibition of transformation of normal cells to tumour cells and inhibition of invasion
of metastasis. A number of animal studies have shown that curcumin has a dose-dependent
chemopreventive effect in colon, duodenal, stomach, esophageal and oral carcinogenesis [37].
Colon carcinoma is prevented by curcumin through arrest of cell-cycle progression independent
of inhibition of prostaglandin synthesis [38]. Curcumin was an effective cytotoxic agent against
the mouse bladder tumor line MBT-2 and the UMUC human bladder tumor cell line, and
effectively inhibited implantation and growth of bladder tumor cells in C3H mice [39]. Curcumin
inhibits cancer development in rat stomach initiated by Nmethyl- N-nitro-N-nitrosoguanisine
(MNNG) [40] and reduces the incidence and/or multiplicity of esophageal tumors and
preneoplastic lesions in rats with N-nitrosomethylbenzylamine-induced esophageal
carcinogenesis [41].
Anti-coagulant activity
By inhibiting collagen and adrenaline-induced platelet aggregation Curcumin shows
anticoagulant activity both in vitro as well as in vivo in rat thoracic aorta [42].
Anti-fibrotic effect
Curcumin suppresses bleomycin-induced pulmonary fibrosis in rats [43]. It also
suppresses bleomycin-induced alveolar macrophage-production of TNF-a, superoxide and nitric
oxide. Thus curcumin acts as a potent anti-fibrotic agent. Oral administration of curcumin at
300 mg/kg dose inhibits bleomycin-induced pulmonary fibrosis by increase in total cell counts
and biomarkers of inflammatory responses.

Anti-mutagenic activity
Curcumin has shown the reduction in the number of aberrant cells in
cyclophosphamide-induced chromosomal aberration with the dose of 100 and 200 mg/kg body
wt in Wistar rats [44]. Curcumin exerts both pro- mutagenic and anti-mutagenic effects.
Curcumin has also been shown to be non-protective against hexavalent chromium-induced
DNA strand break. In fact, the total effect of chromium and curcumin is additive in causing DNA
breaks in human lymphocytes and gastric mucosal cells [45].
Anti-fertility activity
100% anti-fertility effect has been observed with aqueous extracts of turmeric rhizomes
in rats on oral administration [46]. Curcumin inhibits 5a-reductase, which converts testosterone
to 5a-dihydrotestosterone, thereby inhibiting the growth of flank organs in hamster [47]. Ability
of curcumin to inhibit human sperm motility has developed the potential for its application in
the development of novel intra-vaginal contraceptive [48].
Anti-bacterial activity
Growth of several bacteria like Streptococcus, Staphylococcus and Lactobacillus [22]
could be suppressed by curcumin. The aqueous extract of turmeric rhizomes has reported the
antibacterial effects [49]. In vitro growth of Helicobacter pylori can also be prevented by the use
of curcumin [50].
Anti-diabetic effect
Curcumin decreases advanced glycation end products induced complications in diabetes
mellitus [51]. At very low doses, curcumin prevents galactose-induced cataract formation [52].
Curcumin decrease blood sugar level in alloxan-induced diabetes in rat [53]. Curcumin also
reduces oxidative stress in diabetic induced rats having increased NADPH/NADP ratio as well as
increased activity of oxidative enzymes [54].
Anti-protozoan activity
In vitro curcumin has anti-Leishmania activity [55]. Several synthetic derivatives of
curcumin have anti- Leishmania. amazonensis effect [56]. Anti-Plasmodium falciparum and anti-
Leishmania major effects of curcumin have also been reported [57].
Anti-viral effect
Curcumin acts as an efficient inhibitor of Epstein-Barr virus (EBV) key activator Bam H
fragment z left frame 1 (BZLF1) protein transcription in Raji DR-LUC cells [58]. EBV inducers such
as 12-0-tetradecanoylphorbol-13-acetate, sodium butyrate and transforming growth factor-

beta increase the level of BZLF1 m-RNA at 1248 h after treatment in these cells, which is
effectively blocked by curcumin [58]. Therefore, curcumin has been shown to have antiviral
activity. More importantly, curcumin also shows anti-HIV (human immunodeficiency virus)
activity by inhibiting the HIV-1 integrase needed for viral replication [59-60]. It also inhibits UV
light-induced HIV gene expression [61]. Thus curcumin and its analogues may have the
potential for novel drug development against HIV.
BIOAVAILABILITY ASPECTS OF CURCUMIN
Curcumin has got poor bioavailability but is remarkably well tolerated and does not
appear to be toxic to animals [62] or humans [63] even at high doses. Curcumin has also been
measured in human tissue, i.e. in the liver and portal blood of 12 patients undergoing resection
of hepatic metastases of colorectal cancer. They administer dose of 450 3600 mg curcumin
daily for 1 week prior to surgery [64]. Low nano-molar levels of curcumin and its metabolites,
curcumin glucuronide and curcumin sulfate, were detected in portal serum of all 3 patients who
received 3600 mg of curcumin. Metabolic reduction products of curcumin (hexahydrocurcumin
and hexahydrocurcuminol) were found in the liver of 1 patient. Therefore, the researchers
concluded that the bioavailability of curcumin is poor in tissues remote from the
gastrointestinal tract, including the liver.
An independent dose-escalation study on 15 patients with advanced colorectal cancer
was conducted in the United Kingdom [65]. Patients consumed a single daily dose of 440 2200
mg curcuma extract, equivalent to 36 180 mg curcumin, for up to 4 months. The treatment
was well tolerated and there was no dose-limiting toxicity. Lao et al. studies also gave a similar
result in healthy human volunteers consuming a single dose of curcumin ranging from 500 to
12,000 mg [66]. No dose-limiting toxicities were observed, and low levels of curcumin were only
detected in the serum receiving the highest doses of curcumin (10,000 or 12,000 mg/day).
Interestingly, curcumin was only detected in 2 of these 6 patients, perhaps indicating the
existence of genetic modifiers of curcumin metabolism.
STABILITY ASPECTS OF CURCUMIN
The stability of curcumin toward chemical degradation by alkali has been investigated
by several laboratories with varying results, possibly due to differences in the media used [67-
70]. Tonnesen et al. [68] identified degradation products including ferulic acid and
feruloylmethane, and studied kinetics of degradation in a MeOH/aqueous buffer medium (1:9),
with phosphate buffer (pH 6 9) or carbonate buffer (pH 9 10). The rate behavior was
complex showing several peaks and valleys in the 7 10 pH regimes, showing second order rate
kinetics in curcumin. Wang et al. [69] found that curcumin decomposed 90% within 30 min in
0.1 M phosphate buffer at pH 7.2 at 378C, and tentatively identified the decomposition product
trans-6-(4_-hydroxy-3_-methoxyphenyl)-2,4-dioxo-5-hexenal, from which they identified
vanillin as a final product along with ferulic acid and feruloylmethane.

STRUCTURAL ASPECTS OF CURCUMIN
Curcumin is a lipophilic molecule and rapidly permeates cell membranes [71]. Curcumin
was found to affect the structure and function of cellular membranes and mimic typical events
occurring during apoptosis; however, the cellular response to curcumin contrasted with typical
apoptotic cell death because loss of membrane integrity was immediate, partly reversible, and
cells could recover in a relatively short time [71]. To overcome its limited water solubility, a
number of new approaches have been explored to deliver curcumin effectively, such as
liposome encapsulation [72-73]. Arezzini et al. [74] has sought to improve solubility by
modifying the structure by covalent linking of a sugar to curcumin and have studied its potential
as an agent for treatment of iron-overload disease. Metal ion complexation of curcumin, those
with the cations [VO] 2+ [75,76], Mn2+ [77,78], Fe2+/3+ [79-82] and Cu2+ [83-85] have findings of
particular biological interest.
CONCLUSION
On the basis of various literatures survey, curcumin was considered to be an effective
bioactive agent. The low water solubility and poor bioavailability of curcumin can be overcome
by various structural modifications. Stability aspect shows better effect and less toxicity offering
better pharmacodynamic characteristics. A lot of work has been done and a lot to do in this
field.

Adult Anopheles tessellatus is a vector of Malaria, which is responsible for increased morbidity and mortality in many tropical and subtropical countries. During epidemics, the emphasis is given to the use of insecticides as a control measure against mosquitoes. As an alternative to synthetic insecticides, the use of biodegradable phytochemicals against mosquito larvae is considered to be one of the safest approaches to control mosquito-borne diseases. This project was carried out to monitor the effect of ethanolic rhizome extract of Curcuma longa on second larval instar of Anopheles tessellatus. Bio-assay was performed to determine the percentage mortality of Anopheline larvae following exposure to different concentrations of plant extract. The study indicated that the mortality rate following exposure increases in a time and dose-dependent manner. The plant demonstrated its strongest larvicidal activity as it caused 100% mortality by its rhizome extract at 500 ppm following 48 hours of exposure. Anopheles tessellatus larvae were subjected to histological analysis after 48 hours of exposure to 300 ppm of the extract. The histology revealed a disruption in the cuticle and mid-gut epithelium of the larvae. Additionally, a DNA fragmentation analysis was also performed to detect possible induction of apoptosis and the results indicated no significant DNA degradation. The results of this study suggest a potential utilization of the rhizome extract of Curcuma longa for the control of Anopheles tessellatus larvae.

Solid lipid microparticles reach the site of its action in a controlled rate and do show controlled release for a better therapeutic result. A good drug carrying and release system involve a controlled drug delivery that improves bioavail-ability, to enrich stability and to minimise the toxic effects followed with a targeted drug at the site of its action. The solid lipid microparticles of cur-cumin were prepared in a view to achieving high permeability of curcumin in the brain through blood-brain-barrier. The lipid microsphere solids were prepared by hot melts microencapsulation technique to formulate solid lipid microspheres. Twelve lipid formulations were prepared with varying concentration of surfactants (span 40, span 70, span 90 and Tween 100). The developed formulation was subjected to various parameters such as the particle size, % entrapment ef iciencies, yield productions, % cumulative release, percentage yield and drug loading, based upon highest entrapment ef iciency, drug release and % cumulative release, the F 3 formulation was considered as the best formulation. The prepared microsphere was subjected to different evaluation parameters such as thin-layer chromatography, melting point, FTIR, solubility, compatibility study and In-vitro drug release. The developed formulation shows spherical and smooth surface. The percentage release of drug F 3 formulation has been found highest of about 86.23% after 12 hr.

Present study was aimed to optimize the conditions for the encapsulation of curcumin using maltodextrin and gelatin and evaluating its storage stability in different packaging materials. Spray drying process was optimized for core to wall material ratio, inlet air temperature and feed flow rate based on highest encapsulation efficiency and encapsulation yield. Maximum encapsulation efficiency (73.9 %) and encapsulation yield (79.1 %) was obtained at inlet air temperature of 190 0C and feed flow rate 60 ml/min for core to wall material ratio 1:40. Core to wall material ratio of 1:30 and 1:20 also exhibited maximum encapsulation efficiency and encapsulation yield of 83.3 % and 88.2 %, and 66.3 % and 72.1 % at inlet air temperature of 190 0C and feed flow rate 60 ml/min. Packaging in glass bottles allowed maximum retention of curcumin followed by PET jars and LDPE exhibited minimum retention. Thus, core to wall material ratio of 1:30 using gelatin and maltodextrin in 1:1 wall material proportion can be used for the encapsulation of curcumin for incorporation in food products to allow stability towards processing.

Leishmania parasites are intracellular haemoflagellates that infect macrophages of the skin and viscera to produce diseases in their vertebrates hosts. Antileishmania therapy is based on pentavalent antimony compounds which toxicity of these agents and the persistence of side effects are severe. Curcumin was identified to be responsible for most of the biological effects of turmeric. Turmeric plant extracts (curcumin and other derivatives) have anti-inflammatory, anti-arthritic, antioxidant, anti-microbial, antileishmanial, hepato protective, anti-cancer, anti-ulcer and anti diabetic activity. Stock solutions of curcumin, indium curcumin, diacetylcurcumin and Gallium curcumin were made up in DMSO. From the each stock solution serial dilutions were made with phosphate buffered saline and 100 µl of each prepared concentration was added to each well of 96-well micro plate. All tests were performed in triplicate. Negative control only received RPMI-1640 medium with a parasite density of 106 parasites /ml and the positive control contained varying concentrations of standard antileishmania compound, Amphotericine B. MTT solution was prepared as 5 mg/ml in RPMI-1640 and 20 µl of this concentration was added to each well. Antileishmania effects of test agents and control were evaluated by using the MTT assay. Mean growth inhibition of triplicate for each concentration of test agents and control were measured. The IC50 values for curcumin, gallium curcumin [ga (CUR) 3], indium curcumin [in (CUR)3], Diacethyle Curcumin (DAC ) and Amphotericine B were 38 µg/ml, 32 µg/ml, 26 µg/ml, 52 µg/ml and 20 µg/ml respectively. Indium curcumin [in (CUR) 3] with IC50 values of 26 µg/ml was more effective than other three test agents against Leishmania. Mean growth inhibition of triplicate for Amphotericine B as control drug, was 20 µg/ml. Indium curcumin and Gallium curcumin complex showed more antileishmanial activity than curcumin and diacetylcurcumin and could be suitable candidates for further investigations.

This study was carried out to evaluate the impact of gamma irradiation and turmeric powder (TP) on microbial quality (total aerobic bacteria and coliforms), physicochemical quality (pH, Hunter’s parameter, oxidative and microbial stabilities, haem pigment), stability, and antioxidant status of chicken meat. Accordingly, two doses (1 kGy and 2 kGy) of gamma irradiation alone and in combination with 3% TP along with the control (0 kGy) were applied. Aerobic and vacuum packaging were used for storage of chicken meat on the 0, 7th, and 14th days of storage at refrigeration temperature (4°C). The microbiological results showed that the contamination level decreased as the dose of gamma irradiation was increased for both total bacteria and coliforms, whereas no contamination was documented in the group treated with 2 kGy+TP for both aerobic and vacuum packaging. The results further showed that pH, haem pigment, and Hunter’s colour were also significantly influenced with respect to different groups. The peroxide value (POV), thio-barbituric acid reactive substances (TBARS), and total volatile basic nitrogen (TVBN) differed significantly in chicken meat with different treatments and storage intervals. Higher POV and TBARS were noticed in chicken meat treated with 2 kGy under aerobic packaging after 14 days of storage, and TVBN was higher in the control on the 14th day under aerobic packaging. Total phenolics and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity were also higher in chicken meat treated with 2 kGy + TP on 0 day of storage. Furthermore, higher sensory attribute scores for attributes like appearance, taste, texture, flavour, and overall acceptability were found in the 2 kGy-treated group. It is concluded that chicken meat treated with 2 kGy+TP was considered better for microbial and physicochemical quality, antioxidant activity as well as sensorial properties of chicken meat.

Human population can be considered as a subject of combined exposure to chemicals. Hexavalent chromium is a well-known mutagen and carcinogen. Curcumin, a popular spice and pigment, is reported to have antineoplastic properties. The single cell gel electrophoresis (Comet assay) is a sensitive technique that allows detecting double- and single-strand DNA breaks caused by a broad spectrum of mutagens. In the present work the ability of curcumin to reduce DNA damage induced by chromium in human lymphocytes and gastric mucosa (GM) cells was investigated by using the comet assay. Chromium at 500 μM evoked DNA damage measured as significant (P < 0.001), about a two-fold increase in comet tail moment of both lymphocytes and GM cells. Curcumin at 10, 25, and 50 μM also damaged DNA of both types of cells in a dose-dependent manner: the increase in the tail moment reached about twenty times of the control value (P < 0.001). The combined action of chromium at 500 μM and curcumin at 50 μM resulted in the significant (P < 0.001) increase in the comet tail moment of both types of cells. In each case, treated cells were able to recover within 60 min. Our study clearly demonstrates that curcumin does not inhibit DNA damaging action of hexavalent chromium in human lymphocytes and GM cells. Moreover, curcumin itself can damage DNA of these cells and the total effect of chromium and curcumin is additive. Alkaline degradation of the pigments curcumin (CC), demethoxycurcumin (DMC) and bis-demethoxycurcumin (BDMC) in aqueous solutions of purified and commercial oleoresin sources was investigated. Alkaline degradation of the compounds corresponded to pseudo-first-order kinetics. Responses were similar for pigments combined or alone in solutions. Profiles of rate constants were closely associated to polynominal curves. Degradation rate constants rapidly increased from pH 7.45 to a maximum at about 10.2 and then tended to decline at higher pH. Rates of alkaline degradation were lower for BDMC than for CC and DMC. Halflives of CC, DMC and BDMC at pH 7.45 were 900, 1700 and 2200 hr, respectively. In contrast, half-lives of CC, DMC and BDMC at pH 10.2 were 0.4, 1.0 and 5.0 hr, respectively. Use of BDMC instead of CC in alkaline foods would be expected to improve color stability.

A large variety of natural products have been described as anti-HIV agents, and for a portion thereof the target of interaction has been identified. Cyanovirin-N, a 11-kDa protein from Cyanobacterium (blue-green alga) irreversibly inactivates HIV and also aborts cell-to-cell fusion and transmission of HIV, due to its high-affinity interaction with gp120. Various sulfated polysaccharides extracted from seaweeds (i.e., Nothogenia fastigiata, Aghardhiella tenera) inhibit the virus adsorption process. Ingenol derivatives may inhibit virus adsorption at least in part through down-regulation of CD4 molecules on the host cells. Inhibition of virus adsorption by flavanoids such as (−)epicatechin and its 3-O-gallate has been attributed to an irreversible interaction with gp120 (although these compounds are also known as reverse transcriptase inhibitors). For the triterpene glycyrrhizin (extracted from the licorice root Glycyrrhiza radix) the mode of anti-HIV action may at least in part be attributed to interference with virus-cell binding. The mannose-specific plant lectins from Galanthus, Hippeastrum, Narcissus, Epipac tis helleborine, and Listera ovata, and the N-acetylgl ucosamine-specific lectin from Urtica dioica would primarily be targeted at the virus-cell fusion process. Various other natural products seem to qualify as HIV-cell fusion inhibitors: the siamycins [siamycin I (BMY-29304), siamycin II (RP 71955, BMY 29303), and NP-06 (FR901724)] which are tricyclic 21-amino-acid peptides isolated from Streptomyces spp that differ from one another only at position 4 or 17 (valine or isoleucine in each case); the betulinic acid derivative RPR 103611, and the peptides tachyplesin and polyphemusin which are highly abundant in hemocyte debris of the horseshoe crabs Tachypleus tridentatus and Limulus polyphemus, i.e., the 18-amino-acid peptide T22 from which T134 has been derived. Both T22 and T134 have been shown to block T-tropic X4 HIV-1 strains through a specific antagonism with the HIV corecept or CXCR4. A number of natural products have been reported to interact with the reverse transcriptase, i.e., baicalin, avarol, avarone, psychotrine, phloroglucinol derivatives, and, in particular, calanolides (from the tropical rainforest tree, Calophyllum lanigerum) and inophyllums (from the Malaysian tree, Calophyllum inophyllum). The natural marine substance illimaquinone would be targeted at the RNase H function of the reverse transcriptase. Curcumin (diferuloylmethane, from turmeric, the roots/rhizomes of Curcuma spp), dicaffeoylquinic and dicaffeoylt artaric acids, L-chicoric acid, and a number of fungal metabolites (equisetin, phomasetin, oteromycin, and integric acid) have all been proposed as HIV-1 integrase inhibitors. Yet, we have recently shown that L-c hicoric acid owes its anti-HIV activity to a specific interaction with the viral envelope gp120 rather than integrase. A number of compounds would be able to inhibit HIV-1 gene expression at the transcription level: the flavonoid chrysin (through inhibition of casein kinase II, the antibacter ial peptides melittin (from bee venom) and cecropin, and EM2487, a novel substance produced by Streptomyces. α-Trichosanthin (from the root tubers of Trichosanthes kirilowii), MAP30 (from Momordi ca charantia), GAP31 (from Gelonium multiflorum), DAP30 and DAP32 (from Dianthus caryophyllus) would act as RIPs (ribosome-i nactivating proteins), suppressing the translation process. As glycosylation inhibitors, castanospermine and 1-deoxynojirimycin would interfere with the infectivity and secondary spread of HIV-1. And so would bellenamine [(R)-3,6-diamino-N-(aminomethyl)hexanamide] (from Streptomyces nashvillensis), albeit by an unknown mechanism. Hypericin and pseudohype ricin, two aromatic polycyclic diones from Hypericum triquetrifolium (St. Johnswort), could block HIV-1 infection through a variety of mechanisms (direct virucidal effect, inhibition of secondary virus spread, inhibition of virus budding, and inactivation of the preintegration complex). A number of miscellaneous compounds, including propolis, pokeweed antiviral protein, and michellamines inhibit HIV replication by mechanisms that still have to be resolved. Modifying effects of curcumin (derived from the rhizome of Curcuma longa L.) during the initiation or post-initiation phase of N-nitrosomethylbenzylamine (NMBA)-induced esophageal carcinogenesis were investigated in male F344 rats. Five-week-old rats were divided into 5 groups, and groups 1, 2 and 3 were given intraperitoneal injections of NMBA (0.5 mg/kg body weight/injection 15 times) for 5 weeks from 7 weeks old to induce esophageal neoplasms. Groups 2 and 3 were fed the diet containing 500 ppm curcumin during the initiation and post-initiation phases, respectively. Group 4 was given the diet containing curcumin throughout the experiment, and group 5 was kept on the basal diet alone and served as an untreated control. Incidence and multiplicity of esophageal neoplasms of group 1 (NMBA alone) were 66.7% and 0.83 ±0.70, respectively. Those of groups 2 and 3 were significantly less than those of group 1 (39.3%, 0.46±0.64, P < 0.05; 33.3%, 0.36±0.56, P < 0.05, respectively). Furthermore, the incidence and multiplicity of esophageal preneoplastic lesions (moderate or severe epithelial dysplasia) of group 2 (57.1%, 0.61±0.57; 40%, 0.29±0.46) or 3 (56.7%, 0.67±0.66; 23.3%, 0.23±0.43) were less than those of group 1 (100%, 1.67±0.70; 70.8%, 0.92±0.72) (P < 0.05). In this experiment, feeding of curcumin significantly decreased the expression of cell proliferation biomarkers (5-bromo-2′-deoxyuridine labeling index) in the non-lesional esophageal epithelium (P < 0.01). These findings indicate that curcumin inhibits NMBA-induced esophageal carcinogenesis when given during the post initiation as well as initiation phase. This inhibition may be related to suppression of the increased cell proliferation induced by NMBA in the esophageal epithelium.

The cytotoxic and antitumour activities of four curcuminoid analogues, namely 1,7-diphenyl-1,6-heptadiene-3,5-dione (HL1), 1,7-bis(4-methoxyphenyl)-1,6-heptadiene-3,5-dione (HL2), 1,7-bis(3,4-dimethoxyphenyl)-1,6-heptadiene-3,5-dione (HL3), 1,7-bis(3,4-dihydroxyphenyl)-1,6-heptadiene-3,5-dione (HL4) and their CuII complexes were investigated.The Cu complexes were found to be more active as antitumour agents compared to the free curcuminoids, in both in vitro studies and in increasing the life span of tumour bearing mice. The synthesis and characterization of the curcuminoid analogues and their CuII chelates employing u.v., i.r., 1 H-n.m.r., e.s.r. and mass spectral studies are also included in this report.

An extract of the crude drug "ukon", the rhizomes of CURCUMA LONGA, exhibited intense preventive activity against carbon tetrachloride-induced liver injury IN VIVO and IN VITRO. The extract was subjected to fractionation by monitoring the activity by the IN VITRO assay methods employing carbon tetrachloride- and galactosamine-produced cytotoxicity in primary cultured rat hepatocytes. Curcuminoids were shown to possess significant antihepatotoxic action. The liver protective effects of some analogues of ferulic acid and p-coumaric acid, probable metabolites of the curcuminoids, were also evaluated.

There are a variety of crude drugs which are reputed to be effective for liver disorders. To evaluate liver-protective activity and to elucidate active constituents of those drugs, it is essential that appropriate assay methods be provided. Antihepatotoxic activity has hitherto been assessed by in vivo assay methods employing whole animals which, however, are costly in animals, require fairly large amounts of samples and give marked variation and poor reproducibility of the results and, therefore, are not suitable for activity-guided fractionation of plant extracts. For primary screening of antihepatotoxic activity of extracts, fractions and constituents of liver-protective crude drugs, carbon tetrachloride, D-galactosamine-, peroxide- and ionophore-produced cytotoxicity models utilizing primary cultured rat hepatocytes have been established. The antihepatotoxic effects of the known natural products have evaluated using these methods thus devised. A number of crude drugs have been screened by means of the in vitro assay methods using carbon tetrachloride- and D-galactosamine-induced cytotoxicity models to reveal that some exhibited significant antihepatotoxic effects. A survey for the antihepatotoxic constituents in the active drugs has been conducted and the antihepatotoxic actions of the active constituents of Artemisia capillaris buds, Salvia plebeia herbs, Curcuma longa rhizomes, Zingibir officinale rhizomes, Schizandra chinensis fruits, Atractylodes rhizomes, Swertia japonica herbs, Tetrapanax papyriferum leaves, Dianthus superbus var. longicalycinus herbs, Panax ginseng roots, Aeginetia indica herbs and Ephedra roots have been assessed. The mechanisms of antihepatotoxic action of glycyrrhizin, atractylon and wuweizisu C have been examined.
