Anti-acne and Anti-atopic Dermatitis Effect of Plant Extracts Including Eucommia ulmoides Oliv and Phellodendron amurense

Abstract

In this study, plant extracts including Eucommia ulmoides Oliv. and Phellodendron amurense were studied to test possible application for cosmetics and skin related medicine. Anti-oxidation effect of plant extracts was measured by DPPH free radical scavenging activity and it was insignificant at low concentration, however, it was as good as vitamin C, excellent anti-oxidation agent, at 1000 . Anti-bacterial effect was tested by disc diffusion method, and plant extracts showed mild anti-bacterial effect for normal skin flora, Staphylococcus epidermidis while it indicated strong anti-bacterial effect for acne inducing Propionibacterium acne. Therefore it had powerful potential for anti-acne material because of selectivity. Anti-atopic dermatitis effect was tested by hairless mouse and plant extracts recovered damaged skin to near normal condition after 14 days of treatment. IgE concentration in treated mouse was decreased 16% compared with control. From the research, plant extracts indicated strong anti-acne and anti-atopic dermatitis effect, and showed strong potential for cosmetics and skin related medicine.

Genetic diversity and genetic structures were estimated in seven natural populations of Phellodendron amurense Rupr in South Korea using ISSR markers. Objectives : The purpose of this study was to investigate the effects of Polygonum tinctoria Niram(PTN) on atopic dermatitis(AD) in hairless mice induced by 2,4-dinitrochlorobenzene(DNCB). Method : They were divided into four group, Nor, Con, 1X and 5X, for evaluating the change of appearance of skin surface, skin hydration, thickness of epidermis and mast cell numbers during 30 days. Results : PTN suppressed symptoms of AD in appearance of skin and increased skin hydration for 1X and 5X. In histopathological examination, 1X and 5X significantly reduced the thickness of epidermis and number of mast cell in dermis. These results suggested that the PTN improved the AD-like symptoms of hairless Mice.

Atopic dermatitis (AD) is a common chronic inflammatory disease associated with a cutaneous hypersensitivity reaction to an allergen. Although the incidence of AD is increasing these days, therapeutics has yet to be developed for its treatment. The aim of this study was conducted in order to compare and investigate the characteristic between the Castanea crenata inner shell extract (CS) and the Castanea crenata inner shell extract fermented by Lactobacillus bifermentans (FCS) for an anti-atopic medication. The total polyphenol and flavonoid contents were similar to CS and FCS. In the DPPH and superoxide anion radical scavenging, the CS and FCS had the potential for antioxidant activities. Both of them did not exhibit cytotoxicity to HS68 cells. The evaluation of the anti-inflammatory activity in Raw264.7 cells demonstrated that the FCS has inhibited the LPS-induced production of nitric oxide as compared to the CS. The anti-atopic dermatitis test was done through the induction of DNCB in AD hairless mice. The FCS has inhibited the development of the atopic dermatitis-like skin lesion by transdermal water loss, melanin and erythema of the skin as compared to the CS. Moreover, the pro-inflammatory cytokine IL- and TNF- production in hairless mice were inhibited by the FCS treatment. It indicates that the fermentation of the Castanea crenata inner shell has the potential for the treatment of atopic dermatitis.

In this study, ethosomes loaded with Eucommia ulmoides extracts that have anti-inflammatory and anti-aging activities on the skin were prepared as cosmetic materials. The particle size, physical property, and entrapment efficiency of ethosomes to identify their stabilities during 4 weeks were measured. Ethosomes containing 0.01~0.05% E. ulmoides extracts did not show either precipitation or separation. The mean particle size of these ethosomes was 136.4 nm, and monodisperse. The particle size of ethosome containing 0.05% E. ulmoides extracts was 151.15 nm and the entrapment efficiency was 81.79%. Therefore, in vitro skin permeation experiment was performed with ethosome containing 0.05% E. ulmoides extracts that showed the high loading efficiency and stability during 4 weeks. The order of skin permeation of E. ulmoides extracts was ethosome (77.16%) > liposome (62.80%) > ethanolic solutoin (41.59%). These results suggest that ethosomes containing 0.05% E. ulmoides extracts are effective for the skin permeation and may be used as a cosmetic formulation containing plant extracts.

In this study, mixed plant extracts including Phellodendro namurense and Eucommia ulmides Oliv were tested for antimicrobial activity and safety. Antimicrobial activity was measured by disc diffusion method using normal skin flora and opportunistic microbes such as Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli and Candida albicans. As safety test, single dose oral toxicity test, single dose inhalation toxicity test and repeat dose inhalation toxicity test were done. From antimicrobial test, plant extracts showed significant antimicrobial activity Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans, however, did not indicate any antibacterial effect on Escherichia coli. From 3 kinds of safety tests, toxicity of mixed plant extracts was not observed. From experimental results, mixed plant extracts including Phellodendro namurense and Eucommia ulmides Oliv showed good potential for natural antimicrobial agent.

Astradalus membranaceus root extract (ARE) contains saponins, flavonoids, and their glycosides. Two flavonoids, calycosin and formononetin, were predominant constituents with the different amount by diverse extraction conditions. ARE showed the anti-oxidative activities, inhibited tyrosinase activity, and increased type I procollagen expression in proportion to calycosin and formononetin contents. The clinical study also showed that skin wrinkles were significantly reduced in the test group treated with a cream formulation containging 0.1% ARE compared with a group treated with a cream formulation without 0.1% ARE (p < 0.05). These results suggest that ARE may have potential as an anti-aging ingredient in cosmetics.

Patients with atopic dermatitis (AD) often present with a dry skin. To clarify the relationship between dry skin and lipid abnormalities within stratum corneum, stratum corneum lipids were collected from six AD patients aged 15 to 25 years and from sex- and age-matched controls. All major stratum corneum lipid classes were separated and quantitated by high-performance thin-layer chromatography/photodensitometry. Six ceramide fractions were also isolated and quantitated by thin-layer chromatography/photodensitometry. Esterified fatty acids of both ceramide 1 (acylceramides) and wax esters were analysed by capillary gas chromatography. The relative amounts of all the stratum corneum lipid classes including squalene, cholesterol esters, wax esters, triglycerides, free fatty acids, cholesterol, ceramides, cholesterol sulphate and phospholipids did not differ statistically between AD patients and controls. However, a significant decrease in proportion of ceramide 1, which is believed to be a carrier of linoleate responsible for a water-barrier function, and increased levels of esterified C18:1 fatty acids (oleate) of ceramide 1 were observed in AD patients. On the other hand, the fatty acid compositions as well as the proportions of C16:1 straight-chain component in sebum wax esters of AD patients were very similar to those of controls. These results suggest that a significantly reduced amount and/or structural alterations of ceramide 1 deriving from epidermal keratinocytes may be responsible for the impaired water-barrier function of the skin in AD.
