Seed germination of the endangered Viola kitaibeliana and other Italian annual pansies (Viola Sectio...

Here, successful germination protocols for the following four annual pansies are presented: Viola arvensis subsp. arvensis, V. hymettia, V. kitaibeliana, and V. tricolor subsp. tricolor. Seeds were collected mainly in Central Italy (Tuscany, Umbria, and Latium) and in Sicily. Typification of the name Iris benacensis A.Kern. ex Stapf (Iridaceae)

[Show full abstract] Iris benacensis A.Kern. ex Stapf (1887: 649), a species of hybrid origin, often regarded as a heterotypic synonym of I. aphylla Linnaeus (1753: 38) or I. variegata Linnaeus (1753: 38), even if morphologically and genetically distinct (Colasante & Mathew 2008). Iris benacensis was first observed by A. Kerner in northern Italy, near Arco (Trento), and formally described by Stapf (1887). To date, the occurrence of I. benacensis is confirmed only for the Southern Alps, in Trentino Alto Adige near the Lake Garda (known to the Romans as Benacus, from which the name of the species), while the report for Austria needs to be confirmed (Colasante 2014). To the best of our knowledge, the name Iris benacensis was not yet typified.

A rapid double-staining technique to improve seed viability testing in terrestrial orchids

The need to optimize seed banking efforts has stimulated research for rapid methods to estimate quality in seed-lots. For terrestrial orchids, viability testing using tetrazolium (TTC) staining requires chemical scarification, as seeds have an impermeable testa. Different seed-coat permeability may affect TTC staining, thus affecting the results. The aim of this study was to perform a ... [Show full abstract] permeability test to assess the effectiveness of the used scarification method and its usefulness to correct TTC viability results. Mature seeds of Anacamptis laxiflora were subjected to eight scarification treatments with sodium hypochlorite solutions with different concentration and duration. Viability tests were performed using the basic tetrazolium (TTC) methodology, followed by a permeability test performed by means of trypan blue dye (TB). The different scarification methods resulted in estimated TTC viability ranging from 0 and 94% for the same seed lot of A. laxiflora seeds. Our results proved that the used scarification protocol significantly affects both seed coat permeability and subsequent TTC staining (2-way ANOVA, p < 0.0001). We describe a new rapid protocol that can be used to test terrestrial orchid seed viability. This double-staining method, providing rapid information on seed coat permeability, can be useful to avoid under-estimation of TTC results.
