Antiinflammatory, antinociceptive and antioxidant activities of Phyllanthus acidus L. extracts

Abstract

Objective To evaluate analgesic, anti-inflammatory and in vitro antioxidant potential and determine total phenolic, total flavonoid content of leaves extracts of Phyllanthus acidus, a folk medicinal plant of India.Methods Anti-inflammatory activity was evaluated using carrageenan induced paw oedema, cotton pellet induced granuloma, membrane stabilizing activity method. Analgesic activity of the extracts was estimated against acetic acid induced writhing, tail immersion method, formalin test. Free radical scavenging and antioxidant potential of the extracts of Phyllanthus acidus leaves was performed using several in vitro and ex vivo assay models. Total phenolic and total flavonoid contents of the extracts were determined using standard chemical methods.ResultsThe extracts exhibited significant anti-inflammatory and analgesic activities at dose dependent manner. Methanol extract at a dose of 500 mg/kg showed superior activity which was comparable with the standard drugs. Ethyl acetate extract showed moderate activity while petroleum ether extract showed least activity. Total phenolic and total flavonoid content in methanol extract were 73.08±0.682 mg GAE/g and 61.28±0.062 mg QE/g respectively. The extracts possess significant antioxidant activity, methanol extract showed highest IC50 value. The contents of flavonoids and phenolic compounds could be correlated with the antioxidant, analgesic and anti-inflammatory activities observed for Phyllanthus acidus leaves.Conclusion Our findings suggest that Phyllanthus acidus contains potential antioxidant, analgesic and anti-inflammatory compounds which could be tested as drug candidates against oxidative stress, pain and inflammation related pathological diseases.

The Phyllanthus acidus (L.) Skeels has been used as traditional medicine. It has been proved to have hepatoprotective, anti-inflammatory, antidiabetic, and antioxidant properties. Toxicology data in pregnant women are still unknown. The purpose of this study is to assess the safety of an ethanol extract of Phyllanthus acidus (L.) Skeels leaves (EEPA) on pregnant women and fetuses. The pregnant Wistar rats were divided into five groups: a negative control (vehicle), a positive control (5-fluorouracil 10 mg/kg), and three experiment groups (22.5, 90, and 1000 mg/kg). The extract was given orally between the 6th and 15th days of pregnancy, during the organogenesis phase of fetal growth. The EEPA found light maternal toxicity. It was demonstrated by the lower weight of fetuses from rats given EEPA and the lower number of fetuses than the vehicle. EEPA did not cause abnormalities in the skeleton or organs of the fetus, but the pale color and smaller size of the fetal's kidneys, heart, and lungs were observed after a dose of 1000 mg/kg. Although the EEPA affected fetal weight during pregnancy and the genetic disorders require further research.

Antioxidants of natural origin have become the target of a great number of researches in finding the sources of potentially safe, effective, and cheap antioxidants. This study investigated the antioxidant potential of the methanol extract Dalbergia saxatilis leaf using the cotton pellet-induced granuloma method in Wistar rats. Thirty rats of both sexes weighing 150-200 g were divided into five groups of six rats each. Group one received distilled water 1ml/kg, group two received ascorbic acid 250 mg/kg, while groups three, four, and five received graded doses of the methanol leaf extract of Dalbergia saxatilis (250 mg/kg, 500 mg/kg and 1000 mg/kg) respectively daily for 9-days. The rats were sacrificed on the 10th day. Blood samples were collected in plain bottles for evaluation of oxidative stress markers which include superoxide dismutase (SOD), catalase (CAT), glutathione (GSH), and malondialdehyde (MDA). Results showed that the extract at the highest dose (1000 mg/kg) significantly (p < 0.001) decreased MDA concentration similar to the vitamin C at 250 mg/kg, while the other doses of the extract decreased MDA concentration but was not significant. Increased CAT concentration was observed in the 500 and 1000 mg/kg though not significant. Increased concentrations of SOD and GSH were also observed across all doses of the extract but only significant at highest and lowest doses respectively. These findings suggest that, the methanol leaf extract of Dalbergia saxatilis possesses antioxidant properties which may be via its action on MDA, SOD, CAT, and GSH.

Poisonous plants are plants containing a poison that can cause pain or death. The people of the Dawan Fatuleu Tengah tribe have used poisonous plants in their daily lives. However, these types of poisonous plants have not been well documented. The purpose of the present study was to determine the types of poisonous plants and their utilization by the Dawan Fatuleu Tengah tribe. The data collection was conducted by interviewing the respondents for the use of poisonous plants. The obtained results showed that eight types of poisonous plants from six families used by the Dawan Fatuleu Tengah tribe, namely arbila nuts (Phaseolus lonatus L.), tubaroot (Deris elliptica L.), cassava (Manihot esculent), sengon (Paraserianthes faltacataria L.), ginje (Thevetia peruvianan), kecubung (Datura metel L.), ceremai (Phyllanthus acidus L.), and biduri (Calotropis gigantean L.). The Dawan Fatuleu Tengah tribe community uses these poisonous plants to poison fish in the river for consumption, as a natural pesticide, to kill insect disease vectors, and for attacking the enemy. Abstrak Tumbuhan beracun merupakan tumbuhan yang mengandung racun dan dapat menyebabkan rasa sakit atau kematian. Masyarakat suku Dawan Fatuleu Tengah telah lama memanfaatkan tumbuhan beracun dalam kehidupan sehari-hari. Namun, sampai saat ini jenis-jenis tumbuhan beracun tersebut belum terinventarisasi dengan baik. Tujuan dari penelitian ini adalah untuk mengetahui jenis-jenis tumbuhan beracun dan pemanfaatannya oleh suku Dawan Fatuleu Tengah. Teknik pengambilan data berupa pengambilan sampel responden dapat dilakukan dengan menghimpun keterangan melalui wawancara tentang pemanfataan tumbuhan beracun. Hasil penelitian yang diperoleh menunjukkan bahwa terdapat 8 jenis tumbuhan beracun dari 5 famili yang dimanfaatkan suku Dawan Fatuleu Tengah yaitu kacang arbila (Phaseolus lonatus L.), akar tuba (Deris elliptica L.), ubi kayu (Manihot esculenta), sengon (Paraserianthes faltacataria L), ginje (Thevetia peruviana), kecubung (Datura metel L), ceremai (Phyllanthus acidus L), biduri (Calotropis gigantean L). Masyarakat suku Dawan Fatuleu Tengah memanfaatkan tumbuhan beracun tersebut untuk meracuni ikan di sungai untuk dikonsumsi, sebagai pestisida alami, untuk membunuh serangga vektor penyakit, dan juga untuk menyerang musuh.

Two experiments were conducted for standardizing feeding ration and frequency of goldfish Carassius auratus using commercial shrimp feed. In both the experiment duration was 60 days and the stocking density of gold fish is one number per four litres of water. In first experiment of standardization of feeding ration five treatments were given i.e. T1 (2%Body weight), T2 (4%Body weight), T3 (6%Body weight), T4 (8%Body weight) and T5 (10%Body weight) with four replicates each. The initial size of gold fish fry was in the range of (Average Weight-1.25±0.014 g to1.33±0.014 g and Average Length- 2.60±0.012 cm to 2.81±0.017cm). There is significant increase in gain in weight and length in treatment T4 (2.80cm and 1.57 g) as compare to other treatments T1 (2.12cm and 1.22 g), T2 (2.16cm and 1.27 g), T3 (2.41cm and 1.43 g) and T5 (2.52cm and 1.45 g) was observed at the completion of experimental period. In second experiment standardization of feeding frequency was done by giving five treatments i.e. T1 (2times per day), T2 (3times per day), T3 (4times per day), T4 (5 times per day) and T5 (6 times per day) with four replicates each. The initial size of gold fish fry was in the range of (Average Weight-1.24±0.011 g to 1.41±0.011 g and Average Length- 2.51±0.013 g to 2.66±0.021 g). There is significant increase in gain in weight and length in treatment T4 (1.74 cm and 1.70 g) as compare to other treatments T1 (1.66 cm and 1.50 g), T2 (1.67cm and 1.51 g), T4 (1.74cm and 1.70 g) and T5 (1.70 cm and 1.68 g). Survival percentage in both experiment is 100% at feeding ration (8%) and feeding frequency (4 times a day). It is concluded that rearing goldfish using commercial shrimp feed @ 8% feeding rate and 4 times per day feeding frequency is giving optimum gain in length, weight and survival during sixty days rearing period.

Hyperuricemia is a risk factor for gout and other cardiovascular diseases. One of the therapies used is Allopurinol. Unfortunately, it has unwanted side effects. These conditions made researchers continue to seek and develop alternative treatments from natural products. One of which is from plants of the Phyllanthus genus. One of their contents was polyphenols, especially flavonoids. It is an alternative treatment for hyperuricemia because of its minimal side effects. The flavonoids in this genus were reported to have xanthine oxidase inhibitory: quercetin, kaempferol, rutin, apigenin, luteolin, myricetin, catechin, epicatechin, and epigallocatechin with IC 50 values from 0.44 M to > 100μM. The presence of π-π interactions between planar rings A and C on flavones with phe 1009 and phe 914 and the addition of hydroxyl groups on flavonoid compounds plays a crucial role in inhibiting xanthine oxidase.

Six diterpenoids including three ent-kauranes (1–2, 4) and three cleistanthanes (3, 5–6) were isolated from the roots and stems of Phyllanthus acidus (L.) Skeels. Of them, (16S)-ent-16,17,18-tri-hydroxy-19-nor-kaur-4-en-3-one (1), phyllanthone A (2), and 6-hydroxycleistanthol (3) are new compounds, while the ent-kaurane diterpenoids were reported from the titled plant for the first time. Their structures were elucidated on the basis of the extensive spectroscopic analyses. Compounds 2 and 4–6 displayed cytotoxic potential with IC50 values ranging from 1.96 to 29.15 μM. They also showed moderate anti-inflammatory activities (IC50 = 6.30–12.05 μM). Particularly, the new ent-kaurane 2 displayed cytotoxic potential against HL-60 (IC50 = 2.00 μM) and MCF-7 (IC50 = 3.55 μM) cells, and anti-inflammatory activity (IC50 = 6.47 μM).

Mishing community is one of the major tribal communities residing in Northeastern India. They have adapted their lifestyle with one of the adverse conditions and their reliance on plants for food and medicine is widely prevalent. This article presents an explorative ethnobotanical survey on documentation of plants used by the Mishing community of Assam in treating the common ailments as well as their mode of preparation and administration. Research was conducted in eight villages from four districts of Assam. Various participatory interaction methods, group discussions and semi-structured questionnaires were conducted with a total of 80 respondents. Plant importance analysis was done using four quantitative indices (Use value, Use report, Informant consensus factor and Fidelty level). In total, 153 plants under 126 genera and 62 families are recorded. Fabaceae (11 spp.) was the most dominant family and Clerodendrum and Solanum (5 spp. each) were the most dominant genera. The highest use values were recorded for Musa balbisiana (0.087) followed by Paederia foetida (0.075). Kidney stone has the highest Informant Consensus Factor value of 0.97 with 74 use reports for 3 species. A total of 7 species were found to have ≥ 90% Fidelty Level and Cissus quadrangularis recorded the highest value, i.e. 93.7% FL for treating bone fracture with 75 use reports. In addition, a review of ethnomedicinal plants published in earlier literature for the community is presented, and 77 plants are reported as new ethnomedicinal records for the community. A list of bioactive compounds found in the presently reported plants is also listed. Though modern health facilities have gained pace in the developing countries like India, traditional medicine still co-exists with tribal communities as these are the nearest and reliable forms of treatment in Assam, exemplified by the high degree of reliance on it.

Medicinal plant markets provide not only a snapshot of a country's medicinal flora, but also of the importance of herbal medicine among its inhabitants and their concerns about health and illness. During a market survey in 2006, we collected data on the diversity, source, and volume of plants being sold and exported, and the preferences of urban consumers in Suriname. More than 245 species of medicinal plants were sold at the markets of Paramaribo. The annual value of the domestic and export market was estimated to be worth over US$ 1.5 million. Prices of medicinal products were determined by resource scarcity, processing costs, distance to harvesting sites, and local demand. The growing numbers of urban Maroons with their cultural beliefs regarding health and illness, and their strong family ties to the interior are the moving force behind the commercialization of herbal medicine in Suriname.

The present study was aimed to evaluate the free radical scavenging activity of two fractions from the leaves of Indigofera Aspalathoides. Four invitro models namely- DPPH radical, ABTS radical, Nitric oxide radical and hydroxyl radical scavenging assays were used to analyze the activity. Amount of polyphenolic compounds, major phytochemical with antioxidant activity was also estimated. Both fractions displayed significant antioxidant activity when compared to standard antioxidants. Even though Chloroform fraction contained less amount of polyphenolic compounds, it showed more radical scavenging activity than the Ethanol fraction which indicates the role of structural features of polyphenolic compounds with respect to their Antioxidant potential.

Some plants scavenge nitric oxide (NO) with high affinity. For this purpose, forty extracts from 26 medicinal plants, growing extensively in Elburz mountains, were evaluated for their NO scavenging activity. Total phenolic and flavonoid contents of these extracts were also measured by Folin Ciocalteu and AlCl3 colorimetric assays, respectively. Sambucus ebulus fruit aqueous extract and Pterocarya fraxinifolia leaf methanol extract showed better activity than others with IC50 equal to 29 and 65 μg ml-1, respectively. NO is also implicated in inflammation and other pathological conditions, therefore very powerful scavenging activity of S. ebulus fruit extract may explain its very good anti-inflammatory activity. Phenolic and flavonoids contents of the extracts varied between 10.2 - 200.4 and 2.1 - 90.9 mg g-1 of extract, respectively. Good correlations could be found between total phenolic contents and NO scavenging activity through linear regression analysis (R2 = 0.63). There was also a better correlation between total flavonoids and NO scavenging activity (R2 = 0.73). High NO scavenging activity in plants can candidate them for testing their anti-inflammatory property.

This paper presents a robust direct adaptive control scheme for the active control of nonlinear base isolated buildings subjected to near-fault earthquakes. The control architecture is based on the premise of direct adaptive control, where the system is made to follow a desired trajectory without the need of an identifier. The control force is calculated using a single hidden layer non-linearly parameterized neural network in conjunction with a Proportional-Derivative (PD) type controller. Stable tuning laws for the free parameters of the nonlinearly parameterized network are derived based on Lyapunov theory. To achieve good performance and to ensure that the network parameters remain bounded, initialization of the weights is required. A perturbed model is used for the initialization purposes in order to simulate the uncertainty typical of the mathematical models of civil engineering structures. The initialized parameters provide a starting point for the subsequent online adaptation of the controller under earthquake excitations. Set in the framework of adaptive control, the proposed control architecture addresses important issues related to the stability of the closed loop system and parameter bounds, issues that have previously not received the attention they deserve in a majority of the neural network based structural control approaches available in the literature. The robustness of the controller is investigated under actuator failure conditions. Simulations are performed on a full-scale nonlinear three-dimensional base isolated benchmark structure incorporating lateral-torsion superstructure behavior and bi-axial interaction of the nonlinear bearings in the isolation layer. Results are presented in terms of a comprehensive set of performance indices to reflect the tradeoffs in performance commonly associated with structural control methods.

The in vitro activity of the petroleum ether extract of fruit part of Phyllanthus acidus was tested for cytotoxic antibacterial and antioxidant activities as well as for phytochemical screening. The plant was collected from Savar, Dhaka. Phytochemical screening of petroleum extract of phyllanthus acidus revealed the presence of carbohydrate, glycoside and steroid. The extract exhibited antibacterial activity was determined by the disc diffusion method against thirteen pathogenic bacteria and the cytotoxic activity was performed by brine shrimp lethality bio-assay method. The higher concentrations showed antimicrobial activity against a number of bacteria including Salmonella typhi, Staphylococcus aureus, Bacillus cereus, Escherichia coli, Bacillus megaterium. In brine shrimp lethality bio-assay, the LC 50 value was 3.12. The phenolics contents was 159.601 mg/g GAE and the amount of flavonoid was 24.183 mg/g of quercetin equivalent. The DPPH radical scavenging activity of Phyllanthus acidus was found to slight increase with increasing concentration of the extract and IC 50 value showed 1192.263 µg mL −1 for plant extract compared to 13.37 µg mL −1 which was the IC 50 value for the reference ascorbic acid.

The antibacterial and antifungal activities of Phyllanthus acidus was investigated against S.aureus (gram+ve), E.coli (gram-ve) and C.albicans using the Stokes disc diffusion, the Pour plate, Well diffusion and Streak plate methods. The solvent type extracts were obtained by three extractions with hexane, CH2Cl2, EtOAc and CH3CH2OH respectively. Solvents were removed in vacuo to yield viscous oils and paste which were made up to a concentration of 0.035g in 0.01L(10 mL)of the respective solvents. These were tested in varying volumes of 0.2-0.6 ml/plate. The solvents were used as control whereas ampicillin and nystatin were used as references for bacteria and fungal species respectively. The solvents had no effect on the microorganisms whereas ampicillin and nystatin inhibited microbial growth. Phyllanthus acidus showed antimicrobial inhibitory activity at 0.18mg/10mL plate of medium with activity most prominent with the ethanol extracts and negligible with the hexane. This study suggests that the ethanol extracts of Phyllanthus acidus ,can be used as herbal medicines in the control of E.coli and S.aureus following clinical trials.
