Synthesis and antiemetic activity of 1,2,3,9-tetrahydro-9-methyl-3-(4-substituted-piperazin-1-ylmethyl)-4 H -carbazol-4-one derivatives

Ondansetron analogs possessing piperazine ring as side chains were synthesized in our lab. Thus, one of the two carbonyl groups
of starting material 1,3-cyclohexandione (1) was condensed with phenylhydrazine hydrochloride to form monophenylhydrazone (2). 1,2,3,9-Tetrahydro-4H-carbazol-4-one (3) was prepared from 2
via cyclization and rearranged in the presence of ZnCl2. Through a methylation reaction, compound 3 was converted to 1,2,3,9-tetrahydro-9-methyl-4H-carbazol-4-one (4). 3-Dimethylaminomethyl substituted compound (5) was synthesized from 4 by a Mannich reaction in glacial acetic acid. Nine novel 1,2,3,9-tetrahydro-9-methyl-3-(4-substituted-piperazin-1-ylmethyl)-4H-carbazol-4-one derivatives (6a–6i) were synthesized through nucleophilic substitution reaction of 5 with piperazines. The structures of all the target compounds were determined by elemental analysis, IR, MS, 1H NMR and 13C NMR spectra. The results of preliminary pharmacological test show that part of the novel compounds have antiemetic activity
comparable to that of the control Ondansetron.

    ABSTRACT: According to the principle of combination of active groups, a series of piperazine derivatives containing pyridinemethyl/thiazolemethyl were designed and synthesized from 2-chloro-5-(chloromethyl) pyridine or 2-chloro-5- (chloromethyl)thiazole via N-alkylation and addition reactions. Some compounds showed weak herbicidal activity and insecticidal activity against Aphis fabae but good insecticidal activity against Culex pipiens, among which compound 4f exhibited 100% mortality against Culex pipiens at 50 mg/L.

    ABSTRACT: Carbazole and its derivatives are an important type of nitrogen-containing aromatic heterocyclic compounds, possess desirable electronic and charge-transport properties, as well as large φ-conjugated system with rigid fused rings. These special structures of carbazole compounds endow their distinct various functions, properties and biological activities, as well as wide potential application in photoelectrical materials, dyes, medicinal agents, supramolecular recognition and so on. More importantly, the carbazole compounds are easily modified and various functional groups could be conveniently introduced into the carbazole ring. The syntheses of carbazole derivatives and their potential applications were extensively investigated and have been become highly active highlight in recent years, and the progress is quite rapid. Combining with our researches and referring other works from literatures, this paper systematically reviews the recent advance in the research and development of carbazoles in organic electro-luminescence materials, organic photorefractive materials, solar cells, dyes, medicinal chemistry, supramolecular recognition and so on. The perspectives of the foreseeable future are also presented.

    ABSTRACT: In order to search for efficient sulfonylurea herbicides, the title compounds were designed by introducing triazolinones into some known sulfonylurea structures. 23 novel sulfonylurea compounds had been synthesized and characterized by H-1 NMR, HRMS and X-ray diffraction. Herbicidal activities of title compounds were determined by pot-culture method. IC50 values of some effective sulfonylurea compounds were tested by culture dish method. It was found that compound 5h attained the super-active level which was comparable to the controls as triasulfuron and cinosulfuron. It was worthy to note that compound 5h had potent inhibitory activity against Echinochloa crusgalli by pot-culture method at 3.75 g/ha.
