Histological aspects of in situ hybridization

This study examined the detection of cellular poly(A) sequences in mouse liver sections by in situ hybridization using a 3H-labelled poly(dT) probe. Parameters examined included possible losses of target poly(A) sequences from sectioned cells, access of probe to target sequences, section thickness, hybridization conditions, autoradioigraphic efficiency, specific activity of probes and specificity ... An improved protocol was devised that resulted in good preservation of histological detail in sectioned tissue blocks, and a calculated hybridization efficiency of 50%–100%. With the use of probes of defined sequence, the protocol should allow detection of unique mRNA sequences within single cells with an estimated sensitivity of 6–12 unique mRNA molecules per sectioned cell.

Vitellogenin diversity in the Perciformes

[Show full abstract] messenger RNA of distant percoid relatives under stringent conditions of hybridization, suggesting that the vitellogenin gene is well conserved at least within the suborder Percoidei. Multiple vitellogenin gene transcripts were detected in the Cichlidae and these are correlated with the finding of multiple vitellogenin protein monomers in this family. In other families of non-cichlid percoids, only one vitellogenin transcript, and, correspondingly, a single vitellogenin monomer was observed. No cross hybridization was, however, observed with a representative of the suborder Gobioidei. The pattern of protein bands seen in SDS-PAGE also differed. The main features of vitellogenin protein diversity in the Perciformes are interfamily variability in size and subunit number. © 1992 Wiley-Liss, Inc.

The morphological variability of 367 skulls of the American mink from Tver, Pskov, and Novgorod oblasts (Caspian-Baltic watershed) are investigated using quantitative and qualitative traits. Out of the six investigated samplings, five represent feral populations and one represents ranch minks. It is shown that significant morphological differences between feral mink populations exist within a ... [Show full abstract] relatively small area (about 250 × 350 km). Differences in the phenotype are determined by different history of the populations and by the degree of affection of escaped ranch animals. The possible consequences of interactions between the feral and ranch mink populations, which may influence the phenotype, are discussed. The morphological differences observed between the feral mink populations are lower than those theoretically expected when taking into account the considerable influence of domesticated animals. Among the possible reasons of morphological stability of skulls of feral populations of the American mink, we specify naturalization of sizes of ranch escapees under pressure of environmental conditions, as well as possible elimination of the first-generation hybrids between feral and domesticated minks owing to outbreeding depression.

The applicability of our new steric parameter for ortho substituents, E(s)(AMD), in evaluation of organic reactivity and physicochemical data was examined. The E(s)(AMD) constant was derived from the rate constants for acidic hydrolysis of ortho-substituted benzamides, and its scale was put as close as possible to that of the E(s), steric parameter of Taft, and Kutter and Hansch (TKH). [Show full abstract] and halogen substituents, the E(s)(AMD)value was very close to the TKH E(s) value, but for methoxyl, isopropoxyl, and such planar pi-bonded substituents as NO2 and Ph, it was not. Reactivity and physicochemical data taken from the literature were analyzed quantitatively by use of steric and electronic parameters, and the results obtained by E(s)(AMD) and E(s) were compared. The E(s)(AMD) value gave better results than TKH E(s) in various reactions including those of benzoic acids and their derivatives, if the change in the hybridization geometry of the reaction center between the initial and intermediate states was similar to that of the reference reaction.
