Abstract

Thermal rearrangements of 4-isoxazolines from C,N-diphenyl-nitrone and substituted alkynes have been directed towards the formation of substituted indoles. Detection and isolation of the intermediates of the process elucidate the reaction pathway. The appropriate choice of substituents and experimental conditions has allowed the control of the single steps involved in the total process.

The palladium-catalyzed deoxygenation of o-vinylnitrobenzenes employing carbon monoxide as a terminal reductant produces indoles in a continuous flow environment. The reaction proceeds with catalyst loadings of 1 to 2 mol% Pd(OAc)2 in the presence of suitable ligands/additives and generates carbon dioxide as the only stoichiometric side-product. The reductive cyclization proceeds in a clean fashion with high initial reaction rates in the pressurized flow reactor, ultimately leading to deposition of catalytically inactive palladium(0) inside the channels of the flow device, allowing for an efficient catalyst recovery. A variety of o-vinylnitrobenzenes (o-nitrostilbenes and -styrenes) were converted to the corresponding indoles within 15 to 30 min at a reaction temperature of 140 °C to furnish products in good to excellent yields (10–20 bar CO pressure). Mechanistic aspects and the scope of the transformation are discussed.

High yield conversion of 3-aryl-5-ethoxy-isoxazolidines into 3-styryl nitrones has been achieved by 1,5 h refluxing in aq. H2SO4 or catalytic p-toluensulfonic acid/ethanol media. The rearrangement pathway is interpretable on the basis of the ring-opening process of intermediate 5-hydroxy-isoxazolidines. Formation of a masked 5-OH function has been also developed by basic or acid treatment of 5-acetoxy-isoxazolidines.

Treatment of 4-isoxazolines, e.g., I [R1 = Me, CHMe2; R2 = Me, R3 = Me, Ph; R2R3 = (CH2)5; R4 = H, Ph, CO2Et; R5 = H, Ph, CO2Et] with MeI affords α,β-enones II in excellent yields. Indole lactones 8 and 14 were synthesized as a means of functionalizing the 1-isopropyl substituent of the indole nucleus. Lactone 8, upon reduction with diisobutylaluminum hydride, produces lactol 9 which behaves like a masked hydroxy aldehyde and undergoes a Horner-Emmons reaction with triethyl phosphonoacteate to give α,β-unsaturated ester 3 which possesses a hydroxyl group on the 1-isopropyl moiety.

The configurations of some isoxazolidinium salts have been determined by nuclear Overhauser effect difference spectroscopy (NOEDS). The stereochemical findings give a good explanation of the different reactivities experienced in the five-membered ring-opening process as a function of the steric requirement of the system.

A novel reaction including the sequence of hydroamination of alkynes and subsequent oxidative C-H bond functionalization has been developed in the presence of diacetoxyiodobenzene (DIB). The method allows us to synthesize a wide range of tetrasubstituted (E)-alkenes and provides a potential method to construct densely functionalized carbonyl compounds.

Ionic diamine rhodium complex (1) catalyzes the reductive N-cyclization of 2-vinylnitroarenes using carbon monoxide as a reducing agent to afford functionalized indoles. The catalytic system allows direct access to indoles with ester and ketone groups at the 2- or 3-position, in good yields.

A novel synthesis of 1,2-disubstituted 1,2-dihydroquinazoline 3-oxides 8 and the first ever examples of 1,3-dipolar trapping of these nitrones to homonuclear dipolarophiles is described. The new dipoles 8 reacted with N-methyl maleimide, generating diastereomeric adducts 14-16. In the reaction between 8 and dimethyl acetylenedicarboxylate, primary cycloadducts 17 and/or stable rearrangement products, azomethine ylides 18, are formed depending on the substitution pattern of the dipole. The structure of 18c is unambiguously assigned by X-ray crystallographic analysis. An X-ray crystal structure determination is also presented for the cyclopropylisoxazoloquinazoline 22 formed by a [3 + 2] addition of 8a to 21, the dimethyl acetylenedicarboxylate tetramer.
