Transcriptional regulatory sequences of the housekeeping gene for human triosephosphate isomerase

To examine the functional organization of the human triosephosphate isomerase (TPI) promoter, deletion, insertion, and linker scanning mutations were introduced into the TPI promoter of hybrid TPI/beta-globin genes. These genes were transiently expressed in mouse L and human HeLa cells, and the effect of each mutation on the frequency and position of transcription initiation was assayed by S1 nuclease transcript mapping. Multiple positive regulatory elements reside between positions -595 and +1 in L cells and -920 and -7 in HeLa cells and coordinately promote maximum hybrid gene transcription. These elements include an array of GC boxes (positions -126 to -48) that variably conform to the consensus Sp1-binding site, and a canonical TATA box (positions -27 to -21) that is essential for detectable levels of transcription. In an additive yet position-dependent fashion, the GC boxes function in cis to the TATA box to control both the frequency and position of transcription initiation. Additional positive elements reside upstream of position -131 and are required for full promoter function. Also, an inhibitory element(s) residing between position -7200 and either -2800 in L cells or -920 in HeLa cells reduces transcription approximately 7-fold relative to the level of transcription achieved with the maximally active promoter.

    ABSTRACT: The rat bone/liver/kidney/placenta (BLKP) alkaline phosphatase (ALP) gene is expressed at high level in these particular tissues and at low levels in many other tissues. To study the mechanisms underlying the complex regulation of the rat BLKP ALP expression, we isolated a genomic clone, containing a 10.5-kb insert, which includes the promoter of the BLKP ALP gene with 2kb of 5' flanking region, its first exon (84bp), and over 7kb of the first intron. The promoter of the rat BLKP ALP displays features of a “housekeeping” gene promoter: an atypical TATA-box (TTCATAA); 3 potential Spl binding sites; high GC content (82% in positions -134 to -14); and a high CpG to GpC ratio (60:89 in the 0.85-kb promotor region), indicating an abundance of potential methylation sites. Likewise, transient transfection of CAT fusion genes into ROS 17/2.8 osteoblast-like cells reveals weak expression from the promoter and proximal 5' flanking sequences, which can be elevated by an SV40E enhancer.

    ABSTRACT: We have determined that a 69-base-pair (bp) monkey DNA sequence, previously found to enhance simian virus 40 replication, has transcriptional enhancer activity as well. Consensus recognition sites for the transcription factor AP-1, present at each end of this sequence, are partially responsible for its replication- and transcription-enhancing activities. Other motifs within the 69-bp monkey sequence also act to increase the levels of replication and transcription. The activity of the monkey sequence is augmented by the presence of a simian virus 40 21-bp repeat. The 69-bp sequence enhances transcription but not replication from a distance. We conclude that the stimulation of replication and transcription can be uncoupled, suggesting that different mechanisms may be involved.

    ABSTRACT: The gene for acetyl-CoA carboxylase, the rate-limiting enzyme in the biogenesis of long chain fatty acids, contains two promoter regions which control the generation of different forms of carboxylase mRNA. At least five different forms of carboxylase mRNA are generated by differential splicing of the two transcripts formed under the influence of two promoters. One of the two promoters is mainly responsible for the generation of a class of carboxylase mRNA species, pAU type, induced tissue specifically under lipogenic conditions; the other generates ACC mRNAs (FL-type) which are expressed under normal conditions but this expression is also stimulated under lipogenic conditions. In the present studies, we have characterized the promoter that is responsible for the FL-type of ACC mRNA. This promoter contains no TATA or CAAT boxes, but five G/C motifs whose sequences are typical of transcriptional factor Sp1 binding sites. However, the presence of these G/C motifs is not sufficient to drive the transcription of the gene under the control of this promoter. Expression of promoter activity requires three copies of 11 to 13mer enhancer elements which are located in the region upstream to the G/C motifs. The presence of such enhancer elements in a house-keeping gene is unusual, and provides a new example where an enhancer element occurs in the CpG island-type promoter of a house-keeping gene.
