The Gene 4 of Rice Yellow Stunt Rhabdovirus Encodes the Matrix Protein

The complete nucleotide sequence of the gene 4 of rice yellow stunt rhabdovirus (RYSV) was determined from cDNAs corresponding to the viral genomic RNA. Gene 4 is 913 nucleotides (nt) long, comprising a 17-nt untranslated 5' region, a 786-nt open reading frame encoding a polypeptide with a molecular mass of 29,125 Da, and a 110-nt untranslated 3' region. Western blot analysis of the RYSV proteins using the antiserum raised against the protein expressed from the cloned gene in Escherichia coli indicates that gene 4 encodes the M protein of RYSV. Comparisons of the deduced amino acid sequence of the M protein of RYSV with those of other rhabdoviruses revealed no significant homologies. However, it shared a similar basic property and a similar distribution of charges with the other rhabdovirus matrix proteins and showed a relatively closer relationship to the sonchus yellow net virus (SYNV) M1 protein.

    ABSTRACT: The nucleotide sequence of the glycoprotein (G) gene of rice yellow stunt rhabdovirus (RYSV) was determined. The G gene is 2158 nucleotides long and contains an open reading frame of 2007 nucleotides encoding a protein with a calculated molecular mass of 75,358 Da. Furthermore, the 5'- and 3'-terminal sequences of the G protein mRNA were defined by the RACE method. Non-viral nucleotides appear to be present at the 5' end of G mRNA. The G protein contains an N-terminal signal peptide of 32 amino acids, C-terminal transmembrane and cytoplasmic domains, ten potential glycosylation sites and four stretches of a-d hydrophobic heptad-repeats.

    ABSTRACT: The 3′ leader and the 5′ trailer of the rice yellow stunt rhabdovirus (RYSV) genomic RNA have been cloned and sequenced. Sequence data indicate that the RYSV leader region is composed of 203 nucleotides (nt) and the trailer region 191 nt. The terminal 9 nt of the two regions are complementary and capable of forming a putative panhandle structure common to rhabdovirus genomes. In comparison with the leader or trailer sequences of other rhabdoviruses reported so far, both the leader and trailer of RYSV are the longest and there is no obvious sequence homology between the counterparts except for a few terminal nt and the UGUU motif in the leader sequences. Polyadenylated plus-strand leader RNA has been detected in RYSV-infected rice plants by 3′ RACE. This is the second example in rhabdoviruses following the report for sonchus yellow net virus (SYNV) for existence of a polyadenylated leader RNA. No polyadenylated plus-strand transcripts of the RYSV trailer have been found using the similar method.

    ABSTRACT: Taxonomic relationship: A member of nonsegmented, negative-sense, single-stranded RNA viruses of the genus Cytorhabdovirus (type member: Lettuce necrotic yellows virus), family Rhabdoviridae, order Mononegavirales. Members of the family Rhabdoviridae can infect vertebrates, invertebrates and plants. Physical properties: Virions are bacilliform, 74–88 nm in diameter and 163–383 nm in length with surface projections probably composed of trimers of the glycoprotein G, occurring in the cytoplasm in either the coated or the uncoated form (Fig. 1). The nucleocapsid is enclosed in a host-derived envelope. Within the virion, the SCV genome consists of a single negative-sense single-stranded RNA molecule of approximately 13 kb. Figure 1. Download figure to PowerPoint Viral proteins: The SCV genome encodes at least five proteins: the nucleocapsid (N) protein (45 kDa), the matrix (M) protein (77 kDa), the nonstructural protein [Ns, 55 kDa, also known as phosphoprotein (P)], the glycoprotein (G, 23 kDa) and the large (L) protein. Hosts: The natural host range of SCV is limited to species of the genus Fragaria L. Experimental hosts include Physalis pubescens L., P. floridana Rydb., Nicotiana occidentalis, N. glutinosa L. and N. clevelandi Gray. SCV also replicates in its insect vectors Chaetosiphon fragaefolii Cockerell and C. jacobi Hille Ris Lamberts. When injected as purified virus, SCV replicates in aphids Hyperomyzus lactucae (L.), Macrosiphon euphorbiae Thomas, Myzus ornatus Laing, Megoura viciae Buckton, and Acyrthosiphoa pisum (Harris).
