I performed indirect in cell-elisa using HRP cojugated secondary antibody, where OPD was used as peroxidase substrate. The substrate produces a soluble end product that is orange - brown in color which can be read spectrophotometrically at 450nm. Now I would like to do in cell- elisa using flurescent tagged secondary antibody. But I am unable to find good refernces to perform the experiment.
Once I tried by collecting the spheroids by centrifugation at low speed Aafter fixing with 4% paraformaldehyde the cells were permeabilized with triton x 100. Incubation of primary antibody was done overnight at 4 degree, followed by tagging it with alexa fluoro secondary antibody. Washed and mounted on slides. Dot like structures were observed @ 40x. No rigid spheroids were seen.
Kindly suggest some proper protocols.
