The neuronal apoptosis inhibitory protein (NAIP) is a constituent of the NLRC4 inflammasome, which plays a key role in innate immunity, and an antiapoptotic protein. Recently, we reported the previously undescribed role of NAIP in cell division. The liver is one of the body's most actively regenerative organs. Given the novel mitotic role of NAIP,...

Monocytes and macrophages constitute the first line of defense of the immune system against external pathogens. Macrophages have a highly plastic phenotype depending on environmental conditions; the extremes of this phenotypic spectrum are a pro-inflammatory defensive role (M1 phenotype) and an anti-inflammatory tissue-repair one (M2 phenotype). Th...

Data
Expression of polarization markers in primary human monocytes, M0-, M1- and M2-macrophages. Peripheral blood monocytes, M0 macrophages, and M1 and M2 polarized macrophages were harvested and RNA extracted. mRNA expression of the M2 markers CD206 and CCL18, CD206, considered both an M2 and a macrophage marker, and of the M1 marker CXCL10 were analyz...
Data
Flow cytometry gating strategy for THP-1 cells. Cells were first gated for monocytes or macrophages (SSC-A vs FSC-A) and then for singlets (FSC-A vs FSC-H). The expression of different markers (CD11b, CD14, CD163, CD86 or CD206) were then analyzed in the singlets gate by measuring their median fluorescence intensity. The gating strategy for CD86, r...

We have previously reported that administration of Lactobacillus paracasei CNCM I-4034, Bifidobacterium breve CNCM I-4035 and Lactobacillus rhamnosus CNCM I-4036 to obese Zucker-Leprfa/fa rats attenuates liver steatosis and exerts anti-inflammatory effects. The goal of the present work was to investigate the modulation of gene expression in intesti...

The neuronal apoptosis inhibitory protein (NAIP) is a constituent of the inflammasome and a key component of the innate immune system. Here we use immunofluorescence to position NAIP within the cytokinetic apparatus, contiguous to chromosomal passenger complex (CPC), Centralspindlin, PRC1 and KIF4A. During metaphase, NAIP accumulates in the mitotic...

Question
I have THP-1 on an early pass (they are 5th pass).
So I bought 5mg of PMA (sigma), and diluted it in 1mL of DMSO. I tried 3 different concentrations to differentiate the cells: 10ng, 50ng and 100ng/mL.
After 48 hours, 50 and 100ng were mostly dead, and the10ng weren´t doing so good either.
Does anyone have any advice? or a hint of why it istn´t working at all?
Thank you!
Question
I use PMA to differentiate thp-1 and u937 cells to macrophage. I usually use CD11b as differentiation marker but I need another one.
Question
I've been using beta-actin or GAPDH but they are both in the nucleus.
