I have THP-1 on an early pass (they are 5th pass).
So I bought 5mg of PMA (sigma), and diluted it in 1mL of DMSO. I tried 3 different concentrations to differentiate the cells: 10ng, 50ng and 100ng/mL.
After 48 hours, 50 and 100ng were mostly dead, and the10ng weren´t doing so good either.
Does anyone have any advice? or a hint of why it istn´t working at all?
