The end products of catabolism of tryptophan (Trp), an essential amino acid, are known to affect mechanism(s) of aging, a neurodegenerative condition. This review focuses on the possible role of the initial step of Trp catabolism, kynurenine (Kyn) formation from Trp, in aging mechanism(s). Rate-limiting enzymes of Trp conversion into Kyn are trypto...

Aim. To compare reproductive indices and stress resistance of Drosophila at outbreeding and inbreeding. Methods. Drosophila melanogaster congenic strains with incomplete development of the radial wing vein – radius incompletus – were used: the laboratory one and the strain, in which the mutation was placed into the genetic background of wild type s...

Aim. To analyze viability and life span of drosophila at genetic and pharmacological inhibition of tryptophan-kynurenine metabolism, when cultivating on the standard nutritive medium and at a high sugar diet. Methods. Wild type stock and the stock with vermilion mutation have been used. Viability (number of individuals, mortality at the pupal stage...

Aim. To analyze life span in mutant Drosophila stocks with impaired tryptophan-kynurenine metabolism. Methods. Wild type stocks Canton-S and Oregon, stocks with mutations of the locus white: white, whiteapricot, whitesatsuma, and stocks with the mutation vermilion have been used. The average life span of imago has been determined, survival curves h...

Kynurenic acid (KYNA) is one of the metabolites of evolutionary conserved tryptophan (Trp)/kynurenine (Kyn) metabolic pathway. Elevation of KYNA contributes to development of psychosis in schizophrenia but attenuates neurodegeneration in Drosophila model of Huntington's disease. We have reported that KYNA increased lethality of pupae of wild-type f...

Exposure to high sugar diet (HSD) is an experimental model of insulin resistance (IR) and type 2 diabetes (T2D) in mammals and insects. In Drosophila, HSD-induced IR delays emergence of pupae from larvae and eclosion of imago from pupae. Understanding of mechanisms of IR/T2D is essential for refining T2D prevention and treatment strategies. Dysregu...

Background Up-regulation of tryptophan (Trp) conversion into kynurenine (Kyn) and increased formation of down-stream metabolites of Kyn is one of the mechanisms of aging and neurodegenerative disorders. Kyn is an immediate precursor of kynurenic acid (KYNA), an antagonist to NMDA and α7nAChR receptors and activator of aryl hydrocarbon receptor. Inc...

Exposure to high sugar diet (HSD) serves as an experimental model of insulin resistance (IR) and type 2 diabetes (T2D) in mammals and insects. Peripheral IR induced by HSD delays emergence of pupae from larvae and decreases body weight of Drosophila imago. Understanding of mechanisms of IR/T2D is essential for refining T2D prevention and treatment...

Cerebrolysin® (CBL) is a neuroprotective drug used for the treatment of neurodegenerative diseases. CBL's mechanisms of action remain unclear. Involvement of tryptophan (TRP)–kynurenine (KYN) pathway in neuroprotective effect of CBL might be suggested considering the modulation of KYN pathway of TRP metabolism by CBL, and protection against eclosio...

We have observed that berberine prolonged life span and improved viability of pupae and climbing activity of imagoes of wild-type Drosophila melanogaster maintained at 23°C. As a continuation of our studies of berberine effect on life span, we were interested to evaluate the effect of berberine of life span in flies maintained at a higher temperatu...

Up-regulation of kynurenine (KYN) pathway of tryptophan (TRP) was suggested as one of the mechanisms of aging and aging-associated disorders. Genetic and pharmacological impairment of TRP - KYN metabolism resulted in prolongation of life span in Drosophila models. Minocycline, an antibiotic with anti-inflammatory, antioxidant and neuroprotective pr...

Drosophila melanogaster mutants with deficient kynurenine (KYN) formation from tryptophan (TRP) have longer life span than wild type flies. Administration of alpha-methyl-TRP and 5-methyl-TRY, the inhibitors of TRP-KYN metabo-lism, prolonged life span in wild-type flies. Both inhibitors are not available for human use. Berberine, an isoquinoline al...

Upregulation of kynurenine (KYN) formation from tryptophan (TRY) was associated with aging in animal and human studies. TRY - KYN metabolism is affected by the activities of TRY 2,3-dioxygenase 2 (TDO) and ATP-binding cassette (ABC) transporter regulating TRY access to intracellular TDO. We studied the effects of TDO inhibitor, alpha-methyl tryptop...

The state of chromatin in human buccal epithelium cell nuclei upon the influence of sport trainings was investigated. Chromatin state was evaluated in interphase buccal cell nuclei after orcein staining. The heterochromatin granule quantity (HGQ) was estimated in 30 nuclei per sample, and for every donor the mean HGQ value per 30 cells was determin...
Article
Intracellular microelectrophoresis was applied to investigate the electrokinetic properties of human buccal epithelium cell nuclei after exposure of cells to microwaves of wavelengths of 8 mm (f = 37.5 GHz) and 16 mm (f = 18.75 GHz) at a surface power density of 0.2 mW/cm(2). Irradiated or nonirradiated cells were suspended in a flat microelectroph...
Conference Paper
The authors studied changes in electronegativity (ENN) and cell membrane permeability under the influence of microwave radiation of wavelengths 8 and 16 mm. Experiments were performed on human buccal epithelium cells of four female donors of age 20-21 years. The data presented show the increase of stainability of cells after microwave irradiation a...
Article
The influence of electromagnetic fields of wavelengths of 7.1 mm and 1.6 cm on human buccal epithelium cells was investigated. The low-level irradiation (200 μW/cm2) of cell samples induced an increase in cell membrane permeability and the decrease in electronegativity of cell nuclei.
